Objective To establish a recombinant retroviral vector containing insulin-like growth factor-1(IGF-1) gene and to provide the basis for the application of IGF-1 in treating nervous system diseases such as stroke.Methods The plasmid pcDNA3.1-IGF-1 was cut by EcoR I/Xho I,and subcloned to retroviral vector pLXSN,resulting in the recombinant plasmid(pLXSN-IGF-1.)The recombinant IGF-1 expression vector was evaluated by using enzyme cutting and sequencing.By the Lipofectamine 2000,pLXSN-IGF-1 was transferred to packaging cell line-pA317.Culture supernatant of these cells was detected for titration of the recombinant virus.Results The two fragments from recombined IGF-1 eukaryotic expression vector by EcoR I and Xho I represented 400?bp and 6.0?kb by agarose electrophoresis,and PCR showed positive fragment which was about 400?bp long,and sequence analysis showed the same sequence as expected.The cell line pA317-IGF-1 was established,and average virus titer of the recombinant virus in the culture supernatant was about 6.5?10~5 CFU/ml.Conclusion A recombinant retroviral containing IGF-1 gene was successfully constructed.

Background and purpose: Peritoneal metastasis of gastric cancer is mainly discovered in the ad-vanced cancer. Nonetheless, the clinical applicability of each tumor biomarker in peritoneal metastasis of gastric cancer is still ambiguous. Therefore, this study investigated the diagnostic value and clinical significance of CEA, CA125 and CA72-4 in gastric carcinoma patients with peritoneal metastases. Methods: A total of 108 gastric carcinoma patients with peritoneal metastases from Jan. 2008 to Dec. 2013 were studied. All patients were diagnosed by imaging, operations and pathological examination, and also received intravenous or intraperitoneal chemotherapy. Serum tumor markers such as CEA, CA125 and CA72-4 were determined during diagnosis and before each chemotherapy. The diagnostic sensitivity of single marker and combined detection with 2 or 3 markers were analyzed. The correlations among the serum tumor markers and clinical pathological factors, chemotherapeutic effects and survival time were analyzed. Results: Positive rates of CEA, CA125 and CA72-4 were 20.4%, 46.3% and 45.4% in gastric cancer patients with peritoneal metastases, respectively. For these patients, the positive rates of CEA/CA125, CEA/CA72-4, CA125/CA72-4 and CEA/CA125/CA72-4 were 54.7%, 52.8%, 69.5% and 79.6%, respectively. The combined detection of 3 tumor markers was much better than single marker detection (P<0.05). Positive rates of CEA, CA125 and CA72-4 were correlated with the ECOG scale (P<0.05). Positive rate of CA125 was associated with ascites (P<0.001), while positive rate of CA72-4 was associated with ovarian metastasis (P<0.05). Median survival time of patients with positive rates of CEA, CA125 and CA72-4 was significantly lower than that of the patients with normal levels of these markers (P<0.05). Compared with pre-treatment, the levels of all three tumor markers significantly declined after three cycles of chemo-therapy (P<0.05). The decline in CA125 level after chemotherapy was significantly correlated with decreased amount of ascites (P<0.05). The tumor markers turned negative after 3 cycles chemotherapy in patients with positive markers upon initial diagnosis, their survival was significantly prolonged (P<0.001). Conclusion: Combined detection of serum CEA, CA125 and CA72-4 can significantly promote diagnostic rate of gastric cancer with peritoneal metastasis, and may be helpful in evaluating chemotherapeutic effects and predicting prognosis.

OBJECTIVE: To screen the fungi which can induce Aquilaria sinensis to produce active substance. METHODS: Fungus was inoculated in the stem of A. sinensis under natural conditions. The penetration and planting of fungus in stem of A. sinensis as well as the interaction of the two organisms were studied by microscope and scanning electron microscopy, and the main ingredients of the volatile oil in induced agilawood were analysed by GC-MS. RESULTS: Three active strains inducing the formation of agilawood were obtained, which was reported for the first time in the world. The main ingredients of the volatile oil were similar between the induced agilawood and the natural materials. CONCLUSION: It is very important for us to use the fungus in the induction of agilawood and in the sustainable utilization of this special medicinal material. Objective: To observe the effect of ATENS on distance and near vision and stereopsis in amblyopic children patients.Methods: 80 eyes from 70 patients with amblyopia were randomly divided into two groups: the group of ATENS and the group of western medicine.The difference of distance,near vision and stereopsis before and after treatment and between two groups after treatment were observed.Results: Distance and near vision improved and stereopsis diminished significantly after treatment in the TENS group and the western medicine group(P0.05).The curative effect and improving stereopsis in the western medicine group were better than that in the TENS group(P

Objective To evaluate the expressive levels of calcitonin gene related peptide (CGRP) and neuropeptide Y (NPY) in tissue-engineered bone with fascia flap in vivo. Method A segmental bone defect 1.5cm in length was made at the both radius of 12 healthy New Zealand rabbits. Objective To study the characteristics of peptic ulcer (PU) of korean nationality(KN).Method The 4842 cases of PU were detected by gastroscopic technique from Jan 1985 to Apr 1999.The patients were divided into Korean nationality(KN,n=3124) and Han nationality(NH,n=1718) groups,the difference of between two groups was compared and analyzed.Results The morbidity of gastric ulcer C(GU) in KN group was obviously higher than that of HN group,however,the morbidity of duodenal ulcer(DU) was lower in KN group.Morbidity of GU was signifcantly higher than those of DU in KN group.In contrary to KN group,it was lower in HN group.The moraibity of composite ulcer(CU) in KN group was significantly higher than those of HN group in the spring,but in the autumn,the CU morbidity of HN group was higher.PU morbidity of KN group in the spring and winter were markedly higher than those of HN group.The helicobacter pylori(HP) infective rate of the patients with PU in KN group was significantly higher than those of HN group.HP positive rate of the patients with DU was obviously higher than that of the patients with GU.The morbidity of ulcer in KN group was significantly higher than that in HN group at gastric antrum,gastric body,anterior wall of duodenal bulb respectively,but the ulcer morbidity of gastric angle,pyloric canal,greater curvative of duodenal bulb were higher in HN group than that in KN group.The morbidity of all types peptic ulcer in KN group was remarkable higher than that of HN group in age over forty.In constrast,under forty-age-old(or equal to forty),the PU incidence of those anatomical situs of KN group was lower than that of HN group.The average phathogenic attack age of all type ulcer in KN group were significantly higher than that in HN group.The morbidity of ulcer of gastric antrum and anterior wall of duodenal bulb were most in two nationality groups,the autumn was the most season in two nationality groups.Conclusions PU of KN has own characteristics in detectable rate,attack season,pathogenetic situs,age of PU attack as well as HP infection etc.
