The aim was to determine the antimicrobial activities of peptide antibiotics hPAB ? and construct the recombinant baculovirus of its mutants. Four mutants of hPAB ? were designed based on molecular autosyndetic modeling and its genes were cloned by PCR. The transfer plasmid and recombinant baculovirus were constructed. The results showed that the hPAB ? cloned previously, is a good peptide antibiotics. Four mutant genes of hPAB ? were inserted into pFAST HTa plasmid and the recombinant pFAST hPAB ? were screened by restriction enzymes analysis and DNA sequencing. The recombinant baculovirus was obtained after transforming pFAST hPAB ? into Escherichia coli DH10Bac. Our work lays a good foundation for further research.

Chinese Journal of Microbiology and Immunology 2016;36(4):277-280
Objective To investigate whether the expression of sterile alpha motif and histidine/aspartic acid domain containing protein 1 ( SAMHD1 ) could be induced by IFN-α and mediated by microRNA-181a (miR-181a). Methods THP-1 and Jurkat cells were treated with different doses of IFN-α(200 IU/ml and 1 000 IU/ml) for 24 h. The expression of miR-181a and SAMHD1 at mRNA level were de-tected by quantitative PCR. Western blot assay was performed to measure the expression of SAMHD1 at pro-tein level. THP-1 and Jurkat cells were transfected with p-181a, an over-expression vector of miR-181a, and then treated with 200 IU/ml of IFN-α. Changes in the expression of SAMHD1 in those cells were analyzed. Results IFN-α significantly enhanced the expression of SAMHD1 at mRNA and protein levels, but inhibi-ted the expression of miR-181a, especially in Jurkat cells. The expression of SAMHD induced by IFN-αwas inhibited in cells over-expressing miR-181a. Conclusion This study suggests that IFN-α could induce the expression of SAMHD1 by down-regulating the level of miR-181a.

Objective To explore ultrasonographic characteristics and prognosis of cesarean scar pregnancy in the ultrasound. Methods To conducted a retrospective study of 8 cases of diagnoses of cesarean scar pregnancy in ultrasound in our hospital from January 2013 to August 2015.All cases had ultrasonographic examinations regularly during the pregnancy and were confirmed placenta increta by cesarean section or prenatal MRI (6 cases). The imaging characteristics of 8 cases of pregnant women in early pregnancy by transvaginal ultrasound, and the positions of gestational sac and chorion frondosum were observed.Ultrasonic follow-up results, numbers of cesarean delivery, final diagnosis, and pregnancy outcome were analyzed. Results Eight cases of patients were diagnosed cesarean scar pregnancy, and all of ceses were diagnosed placenta increta by ultrasound from 11 weeks to 24 weeks of gestation. It was found that the positions of chorion frondosum were located in the lower edge of gestational sac in Two-dimensional ultrasound and the chorion frondosum was covered in cesarean section scar. The echoes of proliferous chorion frondosum were stronger than the rest of chorion leve and decidua reflexa, and the thickness of proliferous chorion frondosum were thicker than the rest of chorion leve and decidua reflexa. In the follow-up ultrasound of 8 cases in the second trimester, the original position of chorion frondosum which covered and the position of placenta were roughly similar. The position of placenta did not move up with the gestational weeks,in addition, appeared as placenta previa and covered in cesarean section scar.Five patients had strong childbearing willing of continue to conceive, one case of which needed hysterectomy, four of which underwent cesarean section delivery with alive births after using bilateral iliac arteries balloon occlusion. The rest 3 cases underwent induction of labor after using bilateral iliac arteries balloon cclusion. Conclusions Cesarean scar pregnancy in first pregnancy may be develop as placenta accreta in second and third trimester pregnancy. If chorion frondosum was observed to cover cesarean section scar by ultrasound in early pregnancy, scar pregnancy diagnosis was established.Pregnant women who have strong willing to continue their pregnancy should be pay more attentin to thiers placenta previa and placenta percreta in second and third trimester pregnancy.Close follow-up are needed.

Aim To investigate the changes of plasticity and distribution in the lungs of asthmatic guinea pigs. Methods Guinea pigs were divided into asthma group and control group.Immunohistochemistry was used in this study to observe the changes of ERK1/2 and Elk phosphoryiation in the lungs of guinea pigs. Results There were significant changes in distribution of EPK1/2 and Elk in the lungs of asthma group. The positive cells were detected on the walls of respiratory bronchioles and alveolar ducts, especially on the smooth muscle layer and basement membrane. In addition, the numbers of positive cell were clearly increased in asthma group(P< 0.01). Conclusion There is close relationship between the EPK1/2 and Elk phosphoryiation and attack of asthma. It may be a reason for persistence and progress of asthma.

Objective To comprehend the status and risk factors of overweight and obesity among urban and rural inhabitants of Shenchi county. Methods With stratified cluster sampling, two residents" committees were selected as investigation spots from each town of Longquan,Yijing,Bajiao and Hezhi of Shenchi county.Permanent inhabitants aged 20 to 74 yrs were investigated on their status of overweight and obesity and its realted risk factors. Results In this study,2231 inhabitants were investigated, among which male 871 and female 1 360.The prevalence rate of overweight and obesity were 28.6%(standardized rate was 26.7%) and 5.1% (standardized rate was 4,5%).There was a statistic significance in prevalence rate of overweight between male and female(X~2=4.595, P0.05).It was discovered that the age,the degree of education, the annual income of family, drinking, exercise status, alimentary control status, family history and so on had the relation with BMI, the exercise and diet control status assumed negative association by multiple linear regression. Conclusion There is a high prevalence rate of overweight and obesity among residents in Shenchi county in Shanxi province. We should make the comprehensive intervention measures according to its status and risk factors, and make the prevalence rate decreased.

This study is to investigate the effects of paeoniflorin on cerebral blood flow and the balance of PGI2/TXA2 of rats with focal cerebral ischemia-reperfusion injury. A total of 72 SD rats (3) were randomly divided into 6 groups: sham operation group, cerebral ischemia-reperfusion model group (I/R gourp), low (10 mg.kg-1), middle (20 mg.kg-1) and high (40 mg.kg-1) doses of paeoniflorin groups and nimrnodipine group. Focal cerebral ischemia in rats was made by inserting a monofilament suture into internal carotid artery for 90 min and then reperfused for 24 h. The effects of paeoniflorin on neurological deficit scores and the infarction volume of brain were detected. Relative regional cerebral blood flow (rCBF) was continuously monitored over ischemic hemispheres by laser-Doppler flowmetry (LDF). The expression of COX-2 in hippocampal CAl region was estimated by immunohistochemistry and the contents of prostacyclin I2 (PGI2), thromboxane A2 (TXA2), and ratio of PGIJ2/TXA2 in serum were measured by ELISA kits. Paeoniflorin significantly ameliorated neurological scores, reduced the infarction volume, and increased regional cerebral blood flow relative to the I/R group. In addition, paeoniflorin could inhibit COX-2 expression and the release of TXA2 and prevent the downregulation of PGI2 induced by I/R injury. The neuroprotective effects of paeoniflorin against focal cerebral ischemia-reperfusion rats might be attributed to improve the supply of injured hemisphere blood flow and adjust the balance between PGI2/TXA2.
