1.Relationship between the angiotensin Ⅱ type 1 receptor gene polymorphisms and cerebrovascular disease

Objective To explore the relationship between angiotensin Ⅱ type 1 receptor (AT1R) gene polymorphisms and cerebrovascular disease (CVD).Methods The 104 patients with CVD and 98 healthy individuals were detected the AT1R genotypes polymorphisms by restriction fragment length polymorphism in order to analysis.Results CC genotype was not found both in CVD and control group. In CVD group, genotypic frequency of AA was 40.4% and AC was 59.6%. The allele frequency of A was 70.1% and C was 29.9%. In control group, genotypic frequency of AA was 91.8% and AC was 8.1%. The allele frequency of A was 95.9% and C was 4.1%. Objective To investigate the relationship between renin (REN) gene G10631A, angiotensinogen (AGT) gene T704C mononucleotide polymorphisms and cerebral infarction and to investigate the mechanisms and characteristics of cerebral infarction from molecular level. Methods REN gene G1063A and AGT gene T704C polymorphisms in 82 patients with cerebral infarction and 89 controls were detected with polymerase chain reactionrestriction fragment length polymorphism. The differences of the genotypes and allele frequencies were compared between the patient group and the control group. Results The frequency of REN 10631AA genotype (31. 7% vs. 10. 1%,χ2 =12. 816, P = 0. 002) and the frequency of A genotype (49. 4% vs. 30. 3% χ2 = 12. 969, P =0. 000), as well as the frequency of AGT 704 CC genotype (63. 4% vs. 34. 8% χ2 = 15. 029, P = 0. 001) and the frequency of A genotype (79. 9% vs. 61. 2% χ2 = 14. 173, P = 0. 000) in the cerebral infarction group were all significantly higher than those in the control group; the frequency of haplotype 704C 10631A was also significantly higher than that in the control group (P=0. 000). Conclusions REN 10631AA genetype and A allele as well as AGT 704 CC genetype and C allele may be the susceptible factors of cerebral infarction. Haplotype 704C-10631 A may be a genetic risk factor for the occurrence of cerebral infarction.

Objective To investigate the ischemic injury of hepatic cell caused by hepatic artery occlusion.Methods The hepatic artery was occluded in 20 dogs via operation,while the portal vein remained patent.Specimens were gained from the right liver at four time points:before occlusion of the hepatic artery,20(minutes),40 minutes and 60 minutes after artery occlusion.Each specimen was examined by HE and BCL-2 by immunohistochemistry.The gray scale of BCL-2 in HE sections was detected.Results Hepatic cellular injury was obvious 20 minutes after occlusion of the hepatic artery.Irreversible hepatic cellular injury was(observed) 60 minutes after hepatic artery occlusion.The results showed that the gray scale of BCL-2 at every time point after hepatic artery occlusion were significantly different from that before hepatic artery occlusion(P
6.Content Determination of Astragaloside in Danggui Buxue Formula Granule by HPLC

OBJECTIVE:To establish a method for content determination of astragaloside in Danggui buxue formula granule, and investigate the influence of different conditions of dissolution on the content. METHODS:HPLC was performed on the column of Aglient C18 with mobile phase of acetonitrile-water(28:72,V/V)at flow rate of 1.0 ml/min,column temperature was 25 ℃,and the injection volume was 10 μl. RESUITS:The linear range of astragaloside was 2.91-17.46 μg;RSDs of precision,stability were lower than 2%,and reproducibility tests was 3.63%,recovery was 95.76%-98.20%(RSD=1.03%,n=6). The optimized use method was mixing the two granules with 80 ℃ hot water and keeping heat preservation for 5 min successively before taken. CON-CLUSIONS:The method is simple and reproducible,and can be used for the content determination of astragaloside in Danggui bux-ue formula granule.
7.Confirm fusion cell line by STR technology of parentage identification

Objective To screen and confirm cell fusion by DNA technology of parentage identification based on detecting of short tandem repeats.Methods With 20% polyethylene glycol (PEG)-6000,human myeloma cell lines and health individual peripheral blood mononuclear cell were fused.Then selected by hypoxantin,aminopterin,thymidin (HAT) medium,and fusion cell were sub-cloned.Morphology of fusion cells was checked by regular microscope.Concentration of DNA was compared to parental cells.Allele genes,identified by short tandem repeats,of fusion cell line were sequenced and compared with each other.Results The fused cells from myeloma cell line and peripheral blood mononuclear cell (PBMC) were slightly larger than primary cells,and the proliferation cycle was not changed significantly.DNA concentration of the fused cell DNA was increased by two times.Sequences of short tandem repeats (STR) showed that the fused cell included all original genetic materials of parent cells.Conclusions DNA technology of parentage identification is a convenient and reliable method to screen and confirm fused cell.
8.Analyzing the influencing factors of occupational stress of firefighters in Shenzhen City

Objective To analyze the current status and influencing factors of occupational stress among firefighters in Shenzhen City. Methods A total of 5 434 first-line firefighters in Shenzhen City was selected as the study subjects using the judgment sampling method. The Job Content Questionnaire was used evaluate the occupational stress level. Results The detection rate of occupational stress in the study subjects was 52.5% (2 854/ 5 434). The result of dichotomous logistic regression analysis results showed that the risk of occupational stress was higher in firefighters who had a bachelor's degree or above than in those who had junior high school or below (P<0.01), the risk of occupational stress in those with irregular meals was higher than those with regular meals (P<0.01), and the risk of occupational stress was lower in those who exercised 1-3 times a month or ≥3 times a week than in those who did no daily exercise (all P<0.05). The risk of occupational stress was higher with older the weekly working hours, the more frequent the injury and the more firefighting workload (all P<0.01). The risk of occupational stress in firefighters who stand for a long time was higher than that in workers who not-stand for a long time (P<0.05). Conclusion The detection rate of occupational stress among the firefighters in Shenzhen City is relatively high. Educational level, dietary habits, daily exercise, weekly working hours, injury frequency, long-standing work and firefighting workload are all influencing factors for occupational stress in firefighters.
9.Mechanism of leukemia cell apoptosis induced by sodium butyrate activating TRAIL pathway.

This study was aimed to investigate the mechanism of leukemia cell apoptosis induced by histone deacetylase inhibitor (HDACI). Flow cytometry was used to detect the apoptosis of leukemia cell lines NB4, U937 and Jurkat, and the changes of mRNA and protein expressions of TRAIL, DR4 and DR5 were detected by Western blot and RT-PCR respectively. The results showed that both TRAIL and DR5 protein and mRNA expressions in NB4, U937 and Jurkat cells increased after treated with sodium butyrate (SB) and in time-dependent manner. However, DR4 mRNA in leukemia cells was not significantly changed after treated with SB. It is concluded that the apoptosis mechanism of leukemic cell lines NB4, U937 and Jurkat induced by SB is closely related to the protein and mRNA expressions up-regulating TRAIL and DR5, but the DR4 may not participate in the apoptosis induced by SB.

10.The effects of Bushen-Gufeigao and breathing movements on the patients with COPD of ;lung and kidney yang deficiency TCM pattern

Objective Evaluating the effects of Bushen-Gufeigao and breathing movements on the patients with chronic obstructive pulmonary disease of Lung and kidney deficiency TCM pattern. Methods The patients with chronic obstructive pulmonary disease of lung and kidney deficiency TCM pattern were recruited and randomly divided into two groups the treatment group (32 cases) and the control group (34 cases). The control group was treated with the conventional medicine, and the treatment group were added Bufei-Gushengao and breathing movements based on the treatment of control group. Both groups were treated for 1 month. The changes of the clinical symptoms, lung function and frequency of attack and duration of each attack in 10 months after treatment were observed. Results After the treatment, compared with the control group, the TCM patterns scores (5.32 ± 2.67 vs. 9.12 ± 4.11, t=4.424) of the treatment group significantly decreased, while the FVC (3.00 ± 0.49 L vs. 2.71 ± 0.47 L, t=2.408), FEV1 (2.27 ± 0.44 L vs. 1.85 ± 0.54 L, t=3.496), and the percentage of FEV1%(75.62 ± 6.84 vs. 66.86 ± 9.32, t=4.333) increased significantly. In the 10 mouths after the treatment, the treatment group showed significantly fewer patients who had attacked more than 6 times than that in the control group, and the duration of attacks showed significantly fewer minutes in the treatment group than the control group (5.56 ± 1.45 d vs. 8.06 ± 1.80 d, t=6.190). Conclusion Bushen-Gufeigao and breathing movements can effectively improve patient's lung function and symptoms.
