1.Anticipated value of ICGR15 and OGTT for hepatic reserving function in patients with primary hepatic cancer

Objective To study anticipated value of the retention rate of indocyanine green at 15 minutes after injection (ICGR15) and oral glucose tolerance test (OGTT) on hepatic function reserve in patients with primary hepatic cancer (PHC). Methods 58 patients with PHC were divided into 3 groups according to the recovery after operation for PHC. Of them, 29 cases showed good recovery (group A), 18 mild liver dysfunction (group B) and 11 severe liver dysfunction (group C) respectively. ICGR 15, OGTT, routine liver function tests were performed in all patients preoperatively. Results ICGR15 in group C(24.17%±6.59%) obviously increased than that in group A(11.42%±3.91%,P<0.01) and in group B(16.28±6.07%, P<0.01). Blood glucose lever of 120 min after glucose loading test in group C (9.91 mmol/L±3.09 mmol/L)was significantly higher than that in group A(5.42 mmol/L±0.83 mmol/L, (P<0.01)and group B(6.68±1.61, P<0.01). The accuracy (81.0%,86.2%), positive (90.6%, 93.5%) and negative prediction value (72.5%, 74.4%) of OGTT and ICGR 15 were respectively significent greater than those of Pugh's scoring (P<0.01).Conclusions ICGR15 and OGTT are worthwhile indicators to assess hepatic function reserve for patients with PHC. Combination of both ICGR15 and OGTT has higher predictive value on hepatic function reserve.
2.Clinical analysis of 55 infants with group B streptococcus blood stream infection

Objective To summarize the clinical characteristics and outcome of infants with group B streptococcus (GBS) blood stream infection.Methods The medical records of 55 cases with GBS blood stream infection who were hospitalized in Shenzhen Children′s Hospital from 1st January 2010 to 31st December 2015 were retrospectively analyzed.Results There were 30 boys and 25 girls in this study.The age ranged from 1 hour to 78 days.Six cases (10.9%) were early-onset and 49 cases (89.1%) were late-onset.Forty cases (72.7%) were neonates and 15 cases were infants.The meningitis was diagnosed in 20 patients (36.4%).Peripheral white blood cell (WBC) counts declined in 10 cases (18.2%), and elevated in 32 (58.2%) cases.Increased levels C-reactive protein were found in 41 cases (74.5%).All of isolates were susceptible to penicillin, ampicillin, linezolid and vancomycin, while the resistance rates to erythromycin, clindamycin, tetracycline and ciprofloxacin were 56.6% (30/53), 77.4% (41/53), 98.1% (52/53), and 1.9% (1/53), respectively.Meropenem was used in 18 cases, and penicillins or cephalosporins were used in 37 cases.Combined therapy with linezolid was used in 13 cases, combined therapy with vancomycin was used in 3 cases, and combined therapy with two kinds of antibiotics was used in 37 cases (67.3%).In clinical outcome analysis, 54 children (98.2%) were improved and 1 child (1.8%) died of complicated multiple organ dysfunction syndrome and intracranial hemorrhage.Conclusions GBS blood stream infection occurs commonly in the infants aged younger than 3 months, more than one third cases complicated with purulent meningitis.All of isolates are susceptible to penicillin, while the resistant rates are high to erythromycin and clindamycin.The percentage of combination therapy is high.The outcomes are not good.
3.Effect of 18F-2-deoxy-2-fluoro-d-glucose on proliferation of Lewis lung cancer cell

Objective To investigate the influence of 18F-FDG on the proliferation of Lewis lung cancer cell line,and to elucidate its possible mechanism.Methods Morphological changes of cells after culture for 24 h at different concentrations of 0,0.37,1.85,3.70 and 7.4 (×106) Bq/ml of 18F-FDG were observed by using inverted microscopy and electron microscopy.The apoptosis and phase distribution of cell cycle of irradiated cells were analyzed with flow cytometry.DNA synthesis of irradiated cells was assayed by 3H-TdR incorporation.Lipid peroxidation was measured by chromometry and expression of Bcl-2 and Bax protein was measured by immunohistochemical technique.Results Exposed to (0-7.40) × 106Bq/ml of 18F-FDG for 24 h,the cumulative absorbed doses delivered to cells in five groups were 0,0.11,0.55,1.10 and 2.20 Gy,respectively.Irradiated cells showed morphological changes of apoptosis.The apoptosis rate of irradiated cells was increased from (4.05 ± 0.01)% to (25.6 ± 0.28) % (t = 188,P<0.01).3H-TdR incorporation rate was decreased from 100% to(22.0 ± 0.51)% (t =27.6,P <0.05).The levels of M DA in cells were augmented from (0.08 ± 0.03) to (0.67 ± 0.12) μmol/L (t =11.7,P < 0.01).Cell cycle arrest was found in G2/M phase with the increasing doses from 0 to 2.20 Gy.The expression of Bcl-2 protein was decreased while that of Bax protein increased.Conclusions 18F-FDG could induce the apoptosis of cells and inhibit the proliferation of cells.
4.Analysis on bacterial culture of bronchoalveolar lavage fluid from 1 693 children with refractory pneumonia

Objective To evaluate pathogens and antimicrobial resistance of pathogens causing refractory pneumonia in children.Methods Children with refractory pneumonia who admitted to a hospital between May 2008 and December 2014 were performed bronchoscopy,and bronchoalveolar lavage fluid (BALF)were performed bacterial culture and antimicrobial resistance testing.Results 1 693 patients were recruited in the study,273 bacterial isolates were isolated from BALF speci-mens of 226 children,gram-positive bacteria accounted for 38.10% (104/273 ),the main gram-positive bacteria were Streptococcus pneumoniae (n=71)and Staphylococcus aureus (n=23);gram-negative bacteria accounted for 58.24%(159/273),including 44 isolates of Haemophilus parainfluenzae ,28 Klebsiella pneumoniae ,19 Escherichia coli ,and 17 Pseud-omonas aeruginosa ;10 isolates of fungi were also detected,8 of which were Candida albicans .The sensitivity of Streptococ-cus pneumoniae to quinolones,ceftriaxone and cefotaxime were high.Methicillin-resistant Staphylococcus aureus (MRSA) positive rate was 26.32%.ESBLs-producing rate of Haemophilus parainfluenzae and Klebsiella pneumoniae was 32.72% and 62.96% respectively.Conclusion The major pathogens causing refractory pneumonia were Streptococcus pneumoniae and Haemophilus parainfluenzae ,empirical treatment should be conducted accordingly,antimicrobial resist-ance should be considered if therapeutic effect is poor,and targeted therapy should be performed according to cultured re-sults and antimicrobial susceptibility testing result.
5.Inhibitory effect of antisense miR-30a-5p on glioma cell growth in vivo

Objective To study the inhibitory effect of knocking down miR-30a-5p on the U87 human glioma xenograft growth and its possible mechanism.Methods Nude mice bearing subcutaneous U87 human glioblastoma were established and separated into three groups (eight for each group) by randomized digital table method,including control group,scr-ODN treated group and AS-miR-30a-5p treated group.After relevant subcutaneous injection treatment,tumor size was measured every other day until the observation period ended.Researchers executed the animals after the treatment,stripped tumor tissues and extracted RNA and protein.Real-time PCR was conducted to detect the expression of miR-30a-5p.The histopathological characteristics and proliferation and apoptosis biological characters (including SEPT7,PCNA,cyclin D1,MMP-2,apoptosis related factor P53,bcl-2 and caspase3) were evaluated by HE and immunohistochemical staining,Westem blot analysis respectively,and the cell apoptosis was detected by TUNEL method.Results In AS-miR-30a-5p treated group,the tumor growth was delayed and the final tumor volume was smaller than that in the control and scr-ODN treated group (F =7.167,P <0.05),and the expression of miR-30a-5p was knocked down.The expression of PCNA,cyclin D1 were significantly downregulated while P53,SEPT7 and caspase3 up-regulated.Apoptotic index was increased significantly.Conclusion As-miR-30a-5p suppresses the growth of U87 human gliomas xenografts significantly.Malignant phenotype of tumors are reversed to a considerable degree.Therefore,miR-30a-5p can be a candidate for targeted therapy of human glioma.

Objective To investigate the distribution of pathogens isolated from clinical samples and the resistance to the com‐mon antimicrobial agents .Methods Of the 3 745 children ,Hand‐foot‐mouth disease was the most prevalent disease with 1 397 (37 .30% ) cases ,followed by the bronchopneumonia ,rotavirus enteritis and bacterial intestinal infection ;784 strains were isolated from the samples mainly including Haemophilus parainfluenzae (16 .20% ) ,Streptococcus pneumoniae (14 .92% ) ,Moraxella ca‐tarrhalis (12 .88% ) ,Staphylococcus aureus (10 .59% ) and Salmonella enterica(10 .8% ) ;The positive rate of Methicillin‐resistance Staphylococcus aureus(MRSA) was 27 .50% and the ESBLs producing Escherichia coli and Klebsiella pneumoniae were 46 .43%and 81 .40% ,and two or more pathogens could be isolated from sputum .Conclusion Haemophilu ,Streptococcus pneumonia and Moraxella catarrhalis are the main bacterial pathogens in the department of infectious .There is a certain resistance to the common antimicrobial agents .It is important for us to focus on the pathogens and we should pay more attention to the control the resistance of the bacteria .
7.CpG-oligodeoxynucleotides induce differentiation and apoptosis in HL60 cells

AIM: To investigate the differentiative and apoptotic effect of CpG-oligodeoxynucleotides on HL60 cells and its mechanism. METHODS: After HL60 cells were exposed to synthetic CpG-oligodeoxynucleotides, non-CpG-oligodeoxynucleotides or ZpG-oligodeoxynucleotides for 72 hours, respectively, the inhibition of HL60 cells were detected using MTT method, NBT test was used and CD14 expression were determined. Apoptosis of HL60 cells were mensurated with flow cytometry and transmission electron microscope, and caspase 3, Bcl-2 and Bax expression of HL60 cells treated with oligodeoxynucletides were detected using immunohistochemistry. RESULTS: Treatment with CpG-oligodeoxynucleotides induced the differentiation and apoptosis in HL60 cells, but non-CpG-oligodeoxynu- cleotide and ZpG-oligodeoxynucleotide had no effect on HL60 cells. CONCLUSION: CpG-oligodeoxynucleotides can induce the differentiation and apoptosis in HL60 cells. It may provide a new approach for the immunological treatment of leukemia.
8.Analysis of the differences of gender and age in six suicide methods in 548 cases(

Objective This study aims to explore the characteristics of gender and age in six different suicide methods in urban area of China. Methods We collected 548 suicides from Shanghai, Jinhua, Wuhan, Loudi and Duyun, and then analyzed the age and gender characteristics in each suicide method. Results In the age characteristics, the mean age for jumping and hanging was oldest (about 55 years old), followed by drowning and hanging (about 49 years old), oral intake poisoning was much younger (about 49 years old), while carbon monoxide poisoning was youngest (about 37 years old). In the gender differences, male were more than females in suicides by sharp object (52:24), hanging (67:33) and jumping (55:45), while females were more than males in suicides by drowning (58:42). Conclusion The age and gender difference was eminent in different suicide methods. Carbon monoxide poisoning was the youngest age group in suicides and males involved more than females in suicides by mechanical methods.
9.Levels and influencing factors of serum pertussis toxin-IgG antibody in children with pertussis

Objective:To investigate the levels and influencing factors of serum pertussis toxin (PT)-IgG antibody in children with pertussis.Methods:The clinical data including age, course of disease and vaccination status of children with laboratory-confirmed pertussis and tested for PT-IgG antibody in Shenzhen Children′s Hospital from July 2015 to December 2018 were collected. Venous blood samples were obtained to detect PT-IgG antibody levels. Nasopharyngeal swabs were taken for polymerase chain reaction (PCR) test to detect Bordetella pertussis nucleic acid and culture of Bordetella pertussis. Mann-Whitney U test was used for comparison between two groups.Kruskal-Wallis test was used for comparison among multiple groups. Multiple linear regression was used to analyze the influencing factors of PT-IgG antibody levels. Results:A total of 871 children aged 4(2, 7) months were included, among whom, 592(68.0%) cases were under six months and 754 (86.6%) cases were under one year old. The course of disease was 15 (11, 20) days. Among 871 cases, 864 (99.2%) cases were PCR test and (or) culture positive, including 696 cases positive only for PCR test, 35 cases positive only for culture and 133 cases positive for both PCR test and culture. There were 452 (51.9%) children who were not vaccinated and 346 (39.7%) children vaccinated with at least one dose. In terms of age, the PT-IgG amtibody levels of children aged 0 to two months, three to five months, six months to two years and ≥three years were 0.7 (0, 8.2) IU/mL, 2.3 (0, 23.0) IU/mL, 24.6 (0, 112.3) IU/mL and 24.9 (0, 114.7) IU/mL, respectively. The PT-IgG antibody levels of children after onset of symptoms at 0 to two weeks, more than two to four weeks, more than four to eight weeks and more than eight weeks were 0(0, 7.9) IU/mL, 8.7(0, 56.0) IU/mL, 26.6(5.1, 82.9) IU/mL and 68.0(15.3, 118.8) IU/mL, respectively. The differences were both statistically significant ( H=88.346 and 94.076, respectively, both P<0.01). The PT-IgG antibody levels in children who were unvaccinated and vaccinated with at least one dose were 0.9 (0, 12.7) IU/mL and 14.6(0, 86.3) IU/mL, respectively. The difference was statistically significant ( Z=-8.520, P<0.01) PT-IgG≥80 IU/mL accounted for 16%(139/871) in the whole range of age, 34.3%(12/35) in children ≥three years old. There were 13 patients aged ≥three years old with a disease course >two weeks, among whom, six patients had PT-IgG≥80 IU/mL. Age, course of disease and vaccination status were independent influencing factors of PT-IgG levels ( β=0.108, 0.189 and 0.250, respectively, all P<0.01). Conclusions:The levels of PT-IgG antibody in children with pertussis are influenced by age, course of disease and vaccination status. The single serum PT-IgG of 80 IU/mL as cut-off value in the diagnosis of pertussis may lead to a increase of missed diagnosis. Therefore, it is necessary to further explore the standards suitable for children in China.
10.Meta-analysis of laparoscopic for elderly patients with colorectal cancer

Objective To evaluate and compare the efficacy and safety of laparoscopic surgery in the treatment of elder colorectal cancer patients.Methods Selected the retrospective or prospective studies which were retrieved in Pubmed,web of Science,Embase as laparoscopic,laparoscopic colectomy,colorectal cancer,rectal cancer,octogenarian,elderly published between Jan.2000 and May 2017.Data regarding operative indexes,postoperative complications and long term outcomes were analyzed by Stata12.0 software.Results A total of 18 articles,included 3 162 patients,were enrolled in this study.The Meta-analysis indicated that there were no significant differences of less blood loss,postoperative wound infection,bowel obstruction and cardiovascular complications in laparoscopic surgery group except the operative time (P < 0.05).No significant difference was found in the number of dissected lymph node,pneumonia,overall survival and disease free survival (P > 0.05).Conclusion With advantages of rapid postoperative recovery and fewer complications,laparoscopic radical resection for colorectal cancer in elderly patients is safe and effective.
