1.Simultaneous determination of seven active ingredients in Xiongju Shangqing pills by HPLC

Objective To establish a method for simultaneous determination of seven active ingredients(Jasminoidin,Forsythoside A,Caffeotannic acid,3,5-Di-O-caffeoylquinic acid,Jatrorrhizine Hydrochloride,Baicalin,Berberine Hydrochloride).Methods The determination was carried out with Agilent Poroshell 120 SB-C18(100 mm ×3.0 mm,2.7 μm);The mobile phase consisted of acetonitrile(A)-0.1% phosphoric acid solution(B)with gradient elution at a flow rate of 1.0 mL/min;The detection wavelengths were 230 nm in 0~4.5 min(determination of Jasminoidin),348 nm in 4.5 ~9.0 min(determination of Forsythoside A,Caffeotannic acid,3,5-Di-O-caffeoylquinic acid),and 278 nm in 9.0 ~15.0 min (determination of Jatrorrhizine Hydrochloride;Baicalin;Berberine Hydrochloride).Results The calibration curves were linear in the ranges of 4.024~80.48μg/mL for Jasminoidin(r=0.9991),2.042~40.84μg/mL for Forsythoside A(r=0.9993),3.956 ~79.12 μg/mL for Caffeotannic acid(r=0.9996),6.090 ~121.80μg/mL for 3,5-Di-O-caffeoylquinic acid(r=0.9990),4.026 ~80.52 μg/mL for Jatrorrhizine Hydrochloride(r=0.9995),3.924~78.48 μg/mL for Baicalin(r=0.9998),6.030~120.6μg/mL for Berberine Hydrochloride(r=0.9998).The average recoveries(n=6)were 96.5%,97.5%, 95.9%,98.6%,99.8%,98.4% and 97.3% ;RSDs were 1.5%,1.3%,1.2%,1.4%,1.2%,1.1% and 0.9% respectively.Conclusion The method is simple and repeatable,which can be applied to the quality control of Xiongju Shangqing pills.
2.Research progress in heterotopic ossification pathogenesis

Heterotopic ossification is a pathologic bone formation,and can lead to joint dysfunction.There are several related factors for heterotopic ossification,such as surgery and wound,cytokine,genetic factor and perioperative drugs,while the definite pathogenesis is still unclear.Mature heterotopic ossified bone shows similar images as normal bones,and dynamic histomorphologic measurements suggest contrast to normal bone,heterotopic ossified bone exhibits higher metabolic activity.Risk factors for heterotopic ossification include severe central nerve injury,long-term coma,limb spasticity,limb braking and increase of serum alkaline phosphatase.Related factors for heterotopic ossification at molecular level are important in future studies to provide support for the prevention and therapy in heterotopic ossification following trauma or surgery.
3.Differences of clinical results of anterior cruciate ligament reconstruction between two different transplants

4.DNA damage of bone marrow cells in mice induced by radon exposure

Objective To study the DNA damage of bone marrow cells of mice after exposure to Radon.Methods Twenty-four mice were randomly divided into four groups, one control group and three experimental groups with the cumulative doses of radon at 27 WLM (low dose group), 52 WLM (middle dose group) and 105 WLM ( high dose group). DNA damage induced by radon in bone marrow of mice was detected by methods of single cell gel electrophoresis (SCGE), micronucleus(MN) and laser scanning confocal microscope (LSCM) observation. Results The DNA strand breakage, rate of MN and apoptosis increased significantly in the high dose group, but not in the middle and low dose groups. Conclusions Exposure to radon could induce DNA damage in bone marrow cells of mice at high levels.
5.Factors related to heterotopic ossifications after treatment of bi-columnar acetabular fractures

Objective To discuss factors influencing the heterotopic ossifications (HO) after treatment of bi-column acetabular fractures. Methods One hundred and ninety-three cases of bi-column acetabular fractures were followed up, including 147 males and 46 females with an average age of 34. 4 years. Thirty-one cases were associated with craniocerebral trauma. The interval between injury and surgery was less than 1 week in 15 cases, 1 to 2 weeks in 121 cases, 2 to 3 weeks in 34 cases, and over 3 weeks in 23 cases. Factors that might have influenced HO, including surgical method, associated injury, and interval between injury and surgery, were analyzed retrospectively. Results All the cases were followed up for an average of 44. 2 (14 to 84) months. The mean operation time was 238 (150 to 330) minutes, and the average blood loss was 1453 (450 to 4400) mL. The incidence rate of HO was 39. 9% (77 in 193 cases), including 39 cases of degree one, 23 cases of degree two, and 15 cases of degree three. Of the 77 cases associated with HO, 14 had craniocerebral trauma and 63 did not (χ~2 = 0. 019, P = 0. 891) . HO was found in 2 cases that had been operated on in less than 1 week, 38 cases in 1 to 2 weeks, 21 cases in 2 to 3 weeks, and 16 cases in over 3 weeks. The Spearman correlation analysis between HO degree and clinical result showed no correlation ( R = 0. 041, P =0. 722). Only 15 cases (7. 8% ) were associated with HO after the debridement of the necrotic gluteus minimus. Conclusions HO after surgery for bi-column acetabular fractures may not be correlated with craniocerebral trauma, but highly correlated with the interval between injury and surgery. Debridement of necrotic gluteus minimus and other muscles can reduce the incidence of HO.
6.New fixation set-up designed for micro-tensile test

Objective:To introduce a new fixation set-up for micro-tensile test.Methods:Dentin-composite were bonded with AdperTM Single Bond 2 (SB2)and sectioned into stick-shaped specimens.Specimens from each tooth(n =6)were equally divided into Ciucchi's jig and the designed set-up(Control and experimental)groups for micro-tensile bond test according to the utilized fixa-tion set-up.The bonding interface failure mode was examined with field-emission scanning electron microscope (FESEM).Three-dimensional models of the two set-ups and the specimen were developed,stress distribution was analyzed by finite element analysis (FEA).Results:The bond strength(MPa)of experimental and control group was 32.76 ±7.43 and 43.58 ±4.72(P <0.05),the ratio of mixed failure was 28 /36 and 20 /36(P <0.05)respectively.FEA showed that the designed set-up for fixing the sticks pro-vided a uniform stress distribution along the long axis of the specimen.FEA and failure mode analysis confirmed such uniform distri-bution was also concentrated at the bonding interface.Conclusion:The new set-up is feasible for micro-tensile test.
7.Relationship between MEK/ERK signal transduction pathway and CX3CR1 in spinal dorsal horns in a rat model of bone cancer pain

Objective To evaluate the relationship between MEK/extracellular signal-regulated kinase (ERK) signal transduction pathway and CX3CR1 in the spinal dorsal horns in a rat model of bone cancer pain (BCP).Methods Fifty female Sprague-Dawley rats,weighing 150-180 g,were randomly divided into 5 groups (n =10 each):sham operation group (group Ⅰ),BCP group (group Ⅱ),sham operation + U0126 group (group Ⅲ),BCP+ 5% dimethyl sulfoxide (DMSO) group (group Ⅳ),BCP + U0126 (MEK inhibitor) group (group Ⅴ).BCP was induced by inoculating Walker 256 mammary gland carcinoma cells into the medullary cavity of the left tibia.In Ⅲ and Ⅴ groups,U0126 10 μg/10 μl was injected intrathecally once a day starting from day 10 after inoculation to day 12 after inoculation,while the equal volume of 5% DMSO was injected in group Ⅳ.Mechanical paw withdrawal threshold (MWT) was measured at 1 day before inoculation (baseline),3,6,9,and 10 days after inoculation,and after administration on 11 and 12 days after inoculation.The rats were sacrificed after administration on day 12 after inoculation and the L4.6 segments of the spinal cord were removed for determination of CX3CR1 expression in spinal dorsal horns (by Western blot and immunohistochemistry).Results Compared with Ⅰ and Ⅲ groups,MWT was significantly decreased at 6-12 days after inoculation,and the expression of CX3CR1 in spinal dorsal horns was up-regulated in Ⅱ,Ⅳ and Ⅴ groups (P <0.01).Compared with Ⅱ and Ⅳ groups,MWT was significantly increased at 10-12 days after inoculation,and the expression of CX3CR1 in spinal dorsal horns was down-regulated (P < 0.01).Conclusion MEK/ERK signal transduction pathway in the spinal dorsal horns is involved in the development and maintenance of BCP in rats possibly through regulating the expression of CX3CR1.
8.Evaluation of the effect of evidence-based nursing on improvement of nursing requirement of patients with ureter external drainage and inner stent drainage after urological operation

Objective To study the effect of evidence-based nursing on improving the nursing requirement of patients with ureter external drainage and inner stent drainage after urological operation. Methods 62 patients in department of urinary surgery from June 2009 to October 2011 were selected as research object,and they were randomly divided into the control group and the observation group with 31 cases in each group,the two groups adopted routine nursing mode and evidence-based nursing mode respectively.Then the nursing requirement and nursing effect before and three and five days after nursing were compared between two groups. Results All the patients existed needs of related knowledge guidance,unhealthy emotion adjustment and social support,and the needs of patients between ureter external drainage and inner stent drainage showed no significant differences,improvement level of each items of the observation group after nursing were all better than those of the control group. Conclusions The patients with ureter external drainage or inner stent drainage have needs of related knowledge guidance,unhealthy emotion adjustment and social support,and the evidence-based nursing mode can effectively improve the patients'demand situations of these items.
9.Two air disinfection methods for the air cleanliness in interventional operation room: a dynamic monitoring observation

Objective To compare the interventional operation room air disinfection effect of circulatingair disinfection method by an ultraviolet air disinfector with that of air-purifying screen disinfection method in order to provide scientific basis for formulating optimal measures to control hospital infection.Methods By using plate natural sedimentation method,air sampling was conducted at different period of time after the start of air disinfection in operation room.The air sterilization results of the two disinfection methods for the operation room where several consecutive interventional procedures had been performed were compared.Results Both disinfection methods had better preoperative air disinfection effect (P>0.05).When ultraviolet disinfection method was used,the differences in colony detection results among the air samples that were collected at different time periods were statistically significant (P>0.01).With the increasing of time after the start of air disinfection,the number of bacterial colonies was increased.If air-purifying screen disinfection method was employed,the air in the operation could be continuously and dynamically purified,the number of bacterial colonies determined in the whole course of operation met the national hygienic standard for air disinfection.Conclusion Continuous air purification by using air-purifying screen disinfection method can shorten the interval between consecutive surgeries,meanwhile,the operation room can be kept in a condition for any emergency surgery at anytime,besides,nosocomialtion can be effectively prevented.
10.Health care workers' knowledge and attitude towards perioperative warming as well as patients warming status

Objective To investigate the health care workers'(HCWs) knowledge and attitude towards perioperative warming as well as patients' warming status, provide reference for warming in perioperative period and reduce surgical site infection(SSI).Methods Convenience sampling was used to select 198 HCWs in surgical department and operating room as well as 160 patients undergoing operation.HCWs were investigated by referring foreign literatures and designing questionnaires, on-site observation method was used to collect information about warming measures implemented for surgical patients.Results The median (quartile range) of scores of HCWs' knowledge and attitude towards patients' perioperative warming were 9 (2.25) and 25 (1.25) respectively.There was no statistical significance in scores of knowledge and attitude among HCWs with different age, education degree and occupation(all P>0.05).The awareness rates of hypothermia, active warming, and passive warming modes were 21.21%, 43.94%, and 30.81% respectively.80.30%, 89.90%, and 86.36% of HCWs thought that it was essential to keep warming before, during and after surgery.68.13% of surgical patients were warmed by covering cotton quilt and shoulder pad during operation.Conclusion HCWs hold positive attitude towards perioperative warming, but they lack sufficient knowledge about perioperative warming, it is necessary to improve facilities of perioperative warming.
