Objective:To investigate the gene polymorphism distribution of tumor necrosis factor-beta(TNF?) with SLE patient and different race in Chinese Han population of Jiangsu province.Methods:DNA samples were extraced from 168 EDTA-blood of unrelated healthy individuals and 66 SLE patients.TNF? alleles were typed using polymerase chain reaction-restrication fragment length polymorphism(PCR-RFLP). 2.Determination of Hesperidin in Xiangsha Yangwei Pill by HPLC

Objective To establish a method for the determi nation of hesperidin in Xiangsha Yangwei Pill(XYP).Methods Reversed -phrase HPLC was adopted.T he sample was injected into Sep -Pak C 18 cartridge for sample purifica-tion.The chromatographic conditio ns were:Prodigy ODS(150mm?4.6mm,5?m)as analytic column,methanol -wa-ter -acetic acid(35:61:4)as mobile phase,detect wavelength a t 283nmand the flowrate being 1.0mL /min.Results The linear range of hesperidin was from0.148~1.481?g,r =0.9998.The mean recovery was 100.3%,RSD =1.5%(n=6).Conclusion This method was simple,practical,accurate and suitable for the qualit y control of XYP.
3.Research of EDI Technology and Its Application in Field Pharmaceutical Water Preparation

Objective To apply the EDI technology to the field pharmaceutical water preparation, thus improve the ability to prepare pharmaceutical water. Methods The EDI pharmaceutical water production technics was studied, and different kinds of technics were analyzed. The resistance value and TOC content of product water by EDI system were checked. The compact field purified water apparatus was developed, for which self-developed EDI was core technology. Results EDI system had advanced performance, for which the resistance value was high level, and the content of TOC was low. The produced water accorded with the criterion of purified water of Chinese Pharmacopoeia 2005. Conclusion EDI technology adapts to field pharmaceutical water production, and improves the quality of produced water and the adaptability of the system.
4.The mechanism of intercellular adhesion molecule-1 expression in endothelial cells stimulated by advanced glycosylation end products

AIM: To explore the relationship between intercellular adhesion molecule-1(ICAM-1)expression in endothelial cells(EC) and advanced glycosylation end products(AGEs) stimulation. METHODS: Murine bone marrow derived ECs was stimulated by AGEs after pretreated with anti-AGEs, anti-IL-1? and N-acetylcysteine(NAC),then SOD activity and ICAM-1 concentration and adhesion rate(AR) were evaluated. RESULTS: ECs which expressed ICAM-1[(0.65?0.14) vs (0.11?0.02)] induced by AGEs showed lower SOD activity [(0.69?0.19)?10 3 U/L vs (1.71?0.42)?10 3 U/L]. The ICAM-1 expression as well as the increase of AR caused by AGEs stimulation could be suppressed by anti-AGEs(0.12?0.01) and NAC(0.11?0.05). Anti-IL-1? had no influence on these changes. CONCLUSION: AGEs could induce endothelial cells to express ICAM-1 in vitro, most probably due to the formation of free radicals. Besides, AGEs may stimulate other cells to secrete cytokines resulting in ICAM-1 expression in endothelial cells.
