1.Application of bilateral limb function training in the rehabilitation of stroke patients with hemiplegia

Objective To investigate the effect of bilateral limb function training on the rehabilitation of stroke patients with hemiplegia.Methods According to the random digit table,64 patients were divided into experiment group and control group,32 in each group.The control group was given lateral limb function training and the experiment group bilateral function training for one month. The two groups were compared in terms of the motor function of upper and lower extremities and the activity of daily living. Result In terms of the motor function of the lower extremities and the activity of daily living,the experiment group was superior to the control group(P<0.001).Conclusion The function training of bilateral limbs can promote the function rehabilitation of extremities of stroke patients with hemiplegia and improve the activities of daily living.
2.The value of CD8 +T cell expression in rats with chronic bronchitis

Objective CD8 +T cells increased in the airway of patients with chronic obstructive pulmonary disease and exis -ted constantly .The aim was to investigate the role of CD 8 +T-cells in rats with chronic bronchitis ( CB) which was induced by cigarette smoking and intratracheal injection with lipopolysaccharide ( LPS) . Methods 18 health Wistar rats were radomly divided into sham smoking group(group A), CB group(group B) and N-acetylcysteine prevention group (group C).The rats in group B and group C re-ceived intratracheal injection with LPS twice and exposed to cigarette smoking for 4 weeks to induce CB model .The rats in Group C re-ceiving intragastric administration with N-acetylcysteine (NAC)(200mg/kg) before received LPS and smoking.Group A was the sham smoking group.The lung tissue of all rats were stained by HE then evaluated about pathological scores .The expression of nuclear fac-tor-κB (NF-κB), major histocompatibility complex class I (MHCI), CD8 +T cell and Vascular endothelial growth factor (VEGF) in airway were detected by immunohistochemisty which was stained by labeled streptavidin biotin method . Results The pathological scores of airway ( 10 .83 ±3 .31 ) in group B were higher than (1.17 ±2.40) in group A(P <0.05).The pathological scores of airway(4.66 ±2.25) in group C were less than (10.83 ±3.31) in group B(P <0.05).The expression of NF-κB(4.84), MHC I (2.48),CD8 +T cell(5.35)and VEGF(5.02) in airway increased in group B when compared with (1.18, 1.25, 1.33) and (1.18) in group A respectively(P <0.05).The expression of NF-κB (2.18), MHC I(1.46),CD8 +(2.35)and VEGF(2.02) in airway decreased in group C when compared with (4.84), MHC I(2.48),CD8 +T cell(5.35)and VEGF(5.02) in group B respectively (P<0.05 ). There were positive correlations between the expression of NF-κB, MHC I and CD8 +T cells in airways(r=0.670, r=0.701, respec-tively, all P<0.01).There were positive correlations between the expression of CD 8 +T cells and VEGF the pathological scores of air-ways(r=0.689, r=0.782, respectively, all P<0.01). Conclusion NAC can inhibit airway inflammation which is regulated by CD8 +T-cells and VEGF through suppressing the expression of NF -κB and MHC I.
3.Evaluation of the Immunity Effectiveness on the Engineered Subunit Vaccine of Staphylococcus aureus ?-Hemolysin

Objective:? -Hemolysin of Staphylococcus aureus,which was expressed in E.coli BL21(DE3) with recombinant pET32a+-?-HL plasmid,was purified with gel filtration chromatography(GFC),and then the engineered subunit vaccine was developed. The immunity effectiveness of this vaccine was evaluated on mouse models.Method:The purified fusion protein was analyzed in SDS-PAGE,and subjected to the evaluation of its median hemolytic dose potency(HD50) was finally analyzed with rabbit erythrocyte. Protein concentration was determined by the method of Bradford. Antibody titers were evaluated on ELISA,and then challenged to gain the immunity protect index.Results:There is an expected protein band with molecular mass of 53kDa in SDS-PAGE,and the concentration is 0.1278mg/ml. The hemolysis activity is 8012.5 HU/mg. There are specific antibodies acquired in blood-serum from mice after vaccined and the antibody titers rising until it has arrive the max,then following down.Conclusion:The purified fusion protein has good fineness and hemolysis activity,the antibody titers initiated by the protein vaccine go with regulation and the immunoprotection is satisfied.
4.Expression of calcium handling protein in diastolic heart failure

AIM: To elucidate molecular mechanisms underlying calcium handling protein in diastolic heart failure (DHF) from mRNA level and protein expression, including calcium adenosine triphosphatase (Ca 2+ -ATPase), phospholamban, ryanodine receptor, calsequestrin and L-type calcium channel. METHODS: The mRNA of these calcium handling genes were detected by RT-PCR, and the protein levels were analyzed by Western blot analysis. RESULTS: Compared with sham-operated rabbits, the steady-state levels of mRNA encoding the SR Ca 2+ -ATPase and cardiac L-type calcium channel were decreased significantly in rabbits with DHF, and protein level of SR Ca 2+ - ATPase was greatly reduced, whereas the mRNA and protein levels of other calcium handling protein were unchanged. CONCLUSION: L-type calcium channel and the sarcoplasmic reticular Ca 2+ -ATPase were down regulated in DHF. These changes may be a contributory factor for DHF.
5.Expression of β2 microglobin and vascular endothelial growth factors in diffuse large B-cell lymphoma and its clinical significance

Objective To investigate the expression and clinical significance of β2 microglobin (β2-MG)protien and vascular endothelial growth factor (VEGF) protien in diffuse large B-cell lymphoma (DLBCL).Methods The expressions of VEGF protien and β2-MG protien were evaluated in 49 DLBCL patients which started the initial treatment by luminex suspension array.Results Among 49 DLBCL patients,expression of β2-MG protein was high in 37 cases and the expression of VEGF protein was high in 23 cases.The expression of VEGF protien and β2-MG protien were not related with gender,age,B symptoms,clinical stage and lactic acid (LDH).There was positive correlation between the high expression of β2-MG protien and chemotherapy (P =0.037).There was relevant trend between the higher expression of VEGF protien (P =0.067).Conclusion The expressions of VEGF protien and β2-MG protien are detected in DLBCL,both proteins may be the potencial markers of DLBCL and therapeutic targets for DLBCL.

The resource of rare and endangered medicinal plant Swertia mussotii Franch. in Tibet, Qinghai and Sichuan province were surveyed by ways of documents, interview, quadrat and market investigation. The results indi-cated that Swertia mussotii Franch. mainly distributed in Zuogong and Mangkang of Tibet, Yushu of Qinghai province, Shiqu, Daofu, Kangding, Maerkang, Jinchuan and Xiaojin of Sichuan province. According to the height above sea level, the distribution altitude was from 2 300 m (Kangding of Sichuan province) to 3 900 m (Mangkang of Tibet). There are distributions of Swertia mussotii Franch. within the scope of 2 600 m. The illumination, water, soil, temperature and altitude had significant influence on the distribution, growth and reserve of Swertia mussotii Franch. from different angles. In recent years, there was huge increase of market requirement in Swertia mussotii Franch. which were used in Tibetan medicine Zangyinc he n. Excess collection was the primary cause of rapid decreasing in resource of Swertia mussotii Franch.. It was suggested to strengthen the management of rational collection, as well as to accelerate the development of cultivation and production.
7.Characterization and Investigation of Staphylococcus aureus zfb Isolates from Bovine Mastitis

A Staphylococcus aureus strain, designated zfb, was isolated from a clinical bovine mastitis case of a dairy cow. Staphylococcus aureus zfb can have resistance to methicillin and no lipase contrast by ATCC 25923. The production of the capsule was assessed by the diffuse colonial morphology in serumsoft agar. A mouse infection model was used to determine the LD50 and the invasiveness of SA zfb. The LD50 of SA 25923 to experimental mice was 10-2.5/mL, and the LD50 of SA zfb to experimental mice was 10-4.33/mL. The purpose to detect characteristics of SA zfb makes it an interesting candidate for the preparation and assay of an avirulent mutants against staphylococcal infections and further investigate on pathogenic mechanism.

The resource of Sinopodophyllum emodi in Tibet, Qinghai, Gansu, Sichuan and Yunnan province were surveyed by the ways of documents, interview, quadrat and market investigation. The results indicated that S. emodi were mainly distributed in central part from Taibai Shan of Shanxi province along the middle and high mountain on either side of Hengduan mountain which from north to south and in Tibetan plateau of China, the suitable eco-environment of S. emodi was dominated by valleys, wet forests and low bush-woods vegetation which comparatively dry on plateau with the altitude focused on 1 5004 500 meters, and the distribution density of simple alpine meadow was relatively lower. Illumination, water, soil, temperature and altitude had significant influences on the growth, distribution and reserves of S. emodi from different angles. In recent years, there was sharp increase of market requirement in Xiaoyelian (fruit) which were used in Tibetan medicine and the underground part which was used for the extraction of podophyllotoxin, excess collection of the underground part was the primary cause of the rapid decreasing resource of S. emodi. It is suggested that the management of rational collection should be strengthen and the development of culturing and production should be meanwhile accelerated.

9.Determination of xanthones in Tibetan herb Jiadiranguo (Herba Haleniae).

A HPLC method was developed for simultaneous determination of 1-hydroxy-2, 3, 4, 7-trimethoxyxanthone (1), 1-hydroxy-2,3, 7- trimethoxyxanthone (2), 1-hydroxy-2, 3, 4, 5-trimethoxyxanthone (3), and 1-hydroxy-2, 3, 5- trimethoxyxanthone (4) in Halenia elliptica. The analytical column was Welchrom C18 column (4.6 mm x 250 mm, 5 microm). The mobile phase was acetonitrile- water (43:57). The detection wavelength was 265 nm. The flow rate was 1 mL x min(-1) and the column temperature was set at 40 degrees C. There was good linearity between the peak areas and concentration at the ranges of 0.414-16.6, 1.73-69.6, 5.89-117, 3.01-120.5 mg x L(-1) for 1, 2, 3 and 4 respectively. The average recoveries (n = 6) of 1, 2, 3 and 4 were 102.5%, 100.5%, 97.9% and 101.2%. Those four xanthones in thirty samples of H. elliptica. were determined by this method. The method is simple, accurate, repeatable, which could be used for the quality evaluation of H. elliptica. The total content of those four xanthones in H. elliptica should not less than 1.80% by comprehensive analysis.

10.Determination of podophyllotoxin and total lignans in Sinopodophyllum emodi.

OBJECTIVETo determine the content of podophyllotoxin and total lignans in Sinopodophyllum emodi Ying from different areas and evaluate the resource utilization of this endangered medicinal plant.
METHODHPLC and UV spectrophotometry were used to determine the content of podophyllotoxin and total lignans in 126 samples from different habitats and the total lignans content was determined by the reference wavelength method.
RESULTAccording to the results, the highest content was determined from the samples from Yongdeng Nature Reserve in Gansu province, and the lowest ones was found in the samples from Tibet. The former's podophyllotoxin and total lignans contents were 7.40% and 20.66%, respectively, which were 19 times and 4 times more than those of the latter. The content of podophyllotoxin and total lignans in S. emodi were significantly positively related, meanwhile, samples from both low altitude and high latitude showed the higher content.
CONCLUSIONThe two determination methods are simple, rapid, accurate and repeatable. It is more scientific and rational to evaluate the resource utilization of S. emodi with two indicators, those are the content of podophyllotoxin and the content of total lignans. This paper is instructive to the collection of wild resources and the establishment of production bases.
