1.Clinical observation of warm needling moxibustion plus lumbar traction for lumbar disc herniation

Objective: To observe the clinical efficacy of warm needling moxibustion plus lumbar traction for lumbar disc herniation (LDH) and its effect on Oswestry disability index (ODI) score, present pain intensity (PPI) score, and the degree of straight leg raising.Methods: A total of 146 patients with LDH were randomized into a combination group and a control group by the random number table method, with 73 cases in each group. The control group was treated with lumbar traction, and the combination group was treated with lumbar traction plus warm needling moxibustion. The curative efficacy of the two groups was compared. Also, the lumbar function, pain, straight leg raising degree, and inflammatory factor levels before and after treatment were compared. Results: The total effective rate of the combination group was 93.2%, which was higher than 78.1% of the control group (P<0.05). After treatment, the lumbar ODI score, the PPI scores of lumbar pain and lower-limb pain, tumor necrosis factor (TNF)-α and interleukin (IL)-6 levels, and straight leg raising test score in both groups were significantly decreased (P<0.05). The Japanese Orthopaedic Association (JOA) score significantly increased (P<0.05). The improvements of the above items in the combination group were all superior to those in the control group (P<0.05). Conclusion: Warm needling moxibustion plus lumbar traction in the treatment of LDH can improve lumbar function, relieve pain, and reduce inflammatory response of the body.
2.Bone marrow mesenchymal stem cells differentiate into neuron-like cells induced by combination of two cytokines

BACKGROUND:Bone marrow mesenchymal stem cells (BMSCs) can be induced to differentiate into neuron-like cells directional y. Accordingly, BMSCs can be used as seed cells theoretical y in constructing tissue-engineered peripheral nerves.
OBJECTIVE:Using combination of two cytokines to induce BMSCs differentiating into neuron-like cells directional y, and further to discusse its application in peripheral nerve injury.
METHODS:BMSCs were isolated and purificated from the bone marrow of Wistar rats by using the differential adherence method. Basic fibroblast growth factor and epidermal growth factor were used to induce the BMSCs differentiating into neuron-like cells. The morphological change was observed and the neuronal specific markers were detected by immunohistochemistry technique. The morphological and immunohistological changes were also studied after the induce agent were removed.
RESULTS AND CONCLUSION:With presence of morphological and immunohistochemical features of nerve cells induced by neurotrophic factors, BMSCs exhibited two or more processes that were interconnected as a meshwork;cellnucleus and nucleus could be observed with strong light refraction of cytoplasm. After immunohistochemical staining, neuroln specific enolase, neurofilament protein and synaptophysin protein positive cells were detected. A great amount of cells reversed to their original fibroblast-like morphology, and the expression of the three above-mentioned proteins decreased as the induce agent withdrawn. Our study showed that BMSCs can be induced to differentiate into neuron-like cells, but the transdifferentiation is a short-time reversible phenomenon.
3.Investigation on Water Drinking Habit of Residents in Communities Provided with Pipe Direct Drinking Water System in Pudong New District

Objective To know the health habit about water drinking of the residents in the communities provided with pipe direct drinking water system. Methods From June, 2006 till July, 2006, 340 family were interviewed using the same questionnaire. At the same time, the water samples were collected from the pipe direct drinking water system and tap water. Results Of the interviewed family, 80.37% drank the water of pipe direct drinking water system. 47.66% also used the water for washing, gargling, cooking and so on. 61.69% drank the water without boiling. 69.15% did not abandon the old water before using. 67.29% never sterilized the tap and the detection rate of bacteria in the samples was 83.18%. Conclusion The contamination situation of the pipe direct drinking water system in Podong New District, Shanghai is not to be ignored. The health administrative organization must strengthen the inspection of the pipe direct drinking water system and the health education in this field must be promoted.
4.Expression of recombinant human MUC1-MBP fusion protein in E coli DH5?,and its purification and detection of immune responses in mice

Objective:To study MUC1 based cancer vaccine.Methods:MUC1 gene was inserted into pMAL-p2 vector and constructed recombinant pMAL-MUC1. MUC1-MBP fusion protein expression was induced by IPTG in E coli DH5? transformed by the recombinant pMAL-MUC1 .The fusion protein was analyzed by Western blot and purified by amylose affinity chromatography. The antiserum,T cell proliferation and CTL activity of spleen from C57 mice immunized by MUC1-MBP were determined respectively by ELISA,adding 3H-TdR and MTT.Results:Had successfully constructed pMAL-MUC1 expression vector,and purified MUC1-MBP and MBP. C57 mice immunized by MUC-MBP generated MUC1 specific antibody and CTL.The titer of polyclonal antibody to MUC1 was about 1∶5 760?3 221. CTL cytotoxicity to the MCF7 and lewis lung cancer cells respectively were at 47.7%?4.3% and 67.5%?6.5%.Conclusion:Human recombinant MUC1-MBP fusion protein activated T and B cell response in mice.The results suggested that the recombinant Muc1 may be used to develop protein vaccine against carcinoma.
5.Study on inhibitive effect of polysaccharide of cipangopaludina chinensis gray on hela cells in vitro

Objective To study the effect of polysaccharide of Cipangopaludina chinensis on gray human cervix cancer cells(Hela cells) in vitro. Methods Polysaccharide of Cipangopaludina chinensis gray was extracted by water extraction. MTT assay and microscope were used to observe its effect on the growth inhibition of Hela cells at different concentrations of the polysaccharide. Results Polysaccharide of Cipangopaludina chinensis could more significantly inhibit the growth of Hela cells. Conclusion Polysaccharide of Cipangopaludina chinensis could inhibit human cervix cancer cells in vitro
6.The Anti-Tumor Effect of Recombinant Human MUC1-MBP Fusion Protein

Objective: To study the anti-tumor effect of recombinant human MUC1-MBP. Methods: The C57BL/6 mice were in oculated with MUC1-MBP by subcutaneous. MUC1 specific CTL activity of spleen were determined by MTT; The effects on prevention and treatment of tumor were observed by establishing lewis lung cancer-carrying mice. Results: The cytotoxicity of CTL from immunized mice to the MCF7 and Lewis lung cancer cells respectively was (47.7?4.3) % and (67.5 ?6.5) %; 5?10 5 lewis lung cancer cells following immunization were injected iv into C57BL/6 mice, after three weeks, the number of lung and tail tumor colonies was 51 and 5 for PBS and MUC1-MBP groups respectively and the suvival time was significantly delayed in immunized mice. The average volume of tumors in mice with MUC1-MBP was 386 mm 3 wherea control group was 4 000 mm 3 at tumor treating experiment. Conclusions: Recombinant human MUC1-MBP have significantly effects on prevention, treatment and inhibiting metastases of tumor. Our results suggested that the recombinant MUC1-MBP might be used to develop protein vaccine against human carcinoma.
7.Clinical features of autosomal recessive juvenile Parkinson disease

Objective To explore the clinical features of autosomal recessive juvenile Parkinson disease(AR-JP).Methods The clinical materials of 28 patients from 15 families with AR-JP were analyzed retrospectively.Results The onset of all the patients was insidious and the mean age was 26.1 years old.In 23 patients(82.1%),the symptoms began at one limb or one side and progressed bilaterally in a mean time of((4.7?)3.6) years.Bradykinesia(100%),rigidity(100%),resting tremor (85.7%),postural instability(60.7%),hyperreflexia(53.6%),dystonia(32.1%) and diurnal fluctuations with sleep benefit(89.2%) were the cardinal symptoms.The mean improved Webster score was(11.2?)6.1.The mean maintenance dose of DOPA-preparation was((0.40?)0.28) g/d.The mean UPDRS motor score was(27.9?)10.3 before treatment and it decreased to(6.7?)5.4 after therapy(P
8.Evaluation the diagnostic value of HRCT in a rat model of pulmonary fibrosis induced with bleomycin A5

objective To evaluate the value of HRCT on differential diagnosis of a rat model of pulmonary fibrosis induced with bleomycin A5 and to replicate a rat model of pulmonary fibrosis for radio imaging study to investigate pathogenetic mechanism of pulmonary fibrosis. Methods In 50 of 60 male Sprague Dawley rats bleomycin A5 was given through the trachea, and the other 10 received same volume of normal saline. After 28 days, HRCT and pathological examination of the lungs were performed, and the changes were correlatively studied. Results HRCT showed pulmonary fibrosis in all the 50 rats treated with bleomyein, including ground glass opacity, nodular shadow, abnormality in bronchio vasarlar linings, pleural thickening, and in some with beehive like changes. Lung fibrosis was corroborated by pathological sections. Conclusion HRCT can be used to diagnose pulmonary fibrosis of living rats. It is a simple, accurate, reliable, and it can give good representation.
9.Application values of multislice spiral CT in children malformation of large blood vessels outside the heart

Objective To analyze the application value of multislice spiral CT in diagnosis of children malformation of large blood vessels outside the heart.Methods The diagnosis results were compared between echocardiography and multislice spiral CT in 110 children with congenital heart disease,and the children had been confirmed by operation.Congenital heart disease was divided three kinds according to the region:heart,connections between heart and large blood vessels and large blood vessels outside the heart.Results One hundred and ten children with congenital heart disease were selected,and they had 366 malformations.The accuracy rates of echocardiography and multislice spiral CT were 90.7% (332/366) and 95.6% (350/366) respectively,and there was no statistical difference (x2 =3.284,P =0.070).In 366 malformations,160 malformations were in heart region.The accuracy rates of echocardiography and multislice spiral CT were 98.1% (157/160) and 97.5% (156/160) respectively,and there was no statistical difference (x2 =0.776,P =0.378).There were 55 malformations in connections between heart and large blood vessels.The accuracy rates of echocardiography and multislice spiral CT were 94.5% (52/55) and 100.0% (55/55) respectively,and there was no statistical difference (x2 =3.083,P =0.243).There were 151 malformations in large blood vessels outside the heart.The accuracy rates of echocardiography and multislice spiral CT were 81.5%(123/151) and 92.1% (139/151) respectively,and there was statistical difference (x2 =7.377,P =0.01).Conclusion Multisltce spiral CT has great application value in diagnosis of children congenital heart disease,especially in malformation of large blood vessels outside the heart.
