1.In vitro evaluation of apical sealing ability of nanomaterial

Objective:To observe apical sealing ability of two kinds of nanomaterial. Methods:40 straight human maxillary anterior teeth were randomly divided into four experimental groups of 8 samples each and two control groups of 4 samples each. The root canals were prepared using step-back technique and obturated with gutta-percha and one of the sealers by the lateral condensation technique. Group 1: Filled with zinc oxide-eugenol sealer and gutta-percha (ZOE); group 2: Filled with hydroxyapatite sealer and gutta-percha (HA); group 3: Filled with nano-zinc-oxide sealer and gutta-percha (NZO); group 4: Filled with nanohydroxyapatite sealer and gutta-percha (NHA). The teeth were placed into microleakage model. Glucose leakage from the coronal to apical was measured by the concentration of glucose in apical reservoir at 1, 2, 4, 7, 10, 15, 20, 25 and 30 days with GOD (glucose oxidase ) method. 2.Treatment of Senile Viral Myocarditis with Yixin Decoction: A Fifty-four Cases Study

Objective To observe the clinic effects of treating senile viral myocarditis with Yixin decoction plus western medicine. Methods A retrospective analysis was performed on 98 cases with senile viral myocarditis from March 2005 to September 2007 in our hospital. The 98 cases were randomly recruited into a control group with 44 cases and a treatment group with 54 cases. The treatment group was treated with Yinxin decoction plus conventional western medicine and the control group was treated with conventional western medicine exclusively. Results The total effective rate in the treatment group and the control group was 98.15% and 75% respectively. The difference between the two groups were significant (P<0.05). Conclusion Yixin decoction plus western medicine has good effects in treating senile viral myocarditis.
3.The effects of losartan potassium and simvastatin combination therapy on oxidative stress indicators in diabetic patients on peritoneal dialysis

Objective To explore the effects of losartan potassium and simvastatin combination therapy on oxidative stress indicators in diabetic patients on peritoneal dialysis through 12 weeks observation. Methods Diabetic patients with end-stage nephropathy (n=80) who were treated with continuous ambulatory peritoneal dialysis were randomly divided into two groups:control group who received routine treatment (n=40), treatment group who were given losartan potassium 50 mg, once per day and simvastatin 20 mg, once every night (n=40). HbA1C, Insulin dosage, Oxidative stress indicators(SOD, GSH-PX, MDA and Hcy)were compared between two groups before and after peritoneal dialysis. Results There was no significant difference of HbA1C between the 2 groups before and after treatment(P>0.05). The insulin doses increased be?fore dialysis in both groups after CAPD treatment. It is lower in the treatment group than that in the control group ( P<0.05). The expression levels of GSH-PX and SOD in treatment group were higher while the expressions of Hcy and MDA were lower after treatment. The expressions of GSH-PX and SOD were higher while the expressions of Hcy and MDA were lower in treat?ment group than those in control group when comparing the same time point(P<0.05). GSH-PX expression level was lower while the expressions of MDA and Hcy were higher after dialysis than those before dialysis in control group ( P<0.05). Con?clusion Losartan potassium combined with simvastatin treatment can improve curative effect and oxidative stress indicators in diabetic patients on peritoneal dialysis.
4.Clinical study on rt-PA intraarterial thrombolysis for acute ischemic stroke

Objective To investigate the curative effect and complications of intra-arterial thrombolysis (IAT) with recombinant tissue plasminogen activator(rt-PA) for patients with acute ischemic stroke, and analyse the factors related the outcomes.Methods 12 patients were treated by IAT with rt-PA in our hospital from Oct.2002 to Oct.2003. Primary neuroradiological assessment was performed with CT in all patients. Mechanical disruption of clot remnants were attempted after rt-PA infusion. Angiographic recanalization was classified according to thrombolysis in myocardial infarction (TIMI) grades. Clinical evaluation was undertaken 20 d after thrombolysis with classification of modified rankin scale (MRS) scores, good for 0 to 3 and poor for 4 to 6. Results Before thrombolysis the scores for TIMI 1 in 1 case was 8.33% and TIMI 0 in 11 cases was 91.67%. The rates of complete or partial recanalization just after IAT were 75%(9/12), less or no recanalization were 25%(3/12). Good outcome in 66.7%(8/12), poor outcome in 33.33%(4/12). Cerebral hemorrhage occurred in 1 case.Conclusions Intra-arterial thrombolysis ( IAT) with rt-PA is feasible and safe in treatment of acute ischemic stroke.
5.Study on screening protein secretion after TGF-?1 stimulation of gastric cancer cells by SELDI-TOF-MS technique

Objective SELDI-TOF-MS technology was used to contrastly analyse the changes of protein expression profiles of gastric cancer cells after TGF-?1 stimulation,and provide theoretical and experimental foundation for screening different significant proteins. Methods Serial subcultivation gastric cancer cells BGC823,MKN45,and SGC7901 cultured in vitro were grouped into test and control groups.TGF-?1 was added to the test group,but not to control group.Cell culture fluid was collected and centrifuged after cultured 24h,and crossing with WCX2 protein chip to detect protein differences. Results When the test group was compared with control group,we found:(1) thirteen different proteins in BGC823 cells after TGF-?1 stimulaton,and their M/Z were M4294,M4932,M4945,M4972,M4991,M5015,5036,M5060,M5153,M5180,M5197,M8577,and M8784,respectively;(2) eighteen different proteins in MKN45 cells after TGF-?1 stimulaton,and their M/Z were M4292,M4931,M4945,M4972,M4990,M5014,M5152,M5178,M7055,M8190,M8570,M8652,M8670,M8780,M9963,M10098,M10523,and M11653,respectively;(3) eight difference proteins in SGC7901 cells after TGF-?1 stimulation,and their M/Z were M4945,M4972,M4992,M5015,M5180,M7056,M8573,and M8604,respectively. By comparing three protein expression profiles of gastric cancer cells after TGF-?1 treatment,we found two significcant proteins with common differences,that had M/Z of M4945 and M4972,respectively. Conclusions The biological markers whose M/Z is M4945,M4972 with gastric cancer characteristic and associated with TGF-?1 have been screened,which can be as the basis for early prediction and clinical diagnosis research on metastasis and invasiveness of gastric cancer.
6.Investigation on low doses of metformin induce hepatoma cells senescence and its underlying mechanism

Objective To explore the effect of metformin on hepatoma cells senescence and the underlying mechanism.Methods Cell proliferation,cycle and apoptosis were examined by MTS and flow cytometry assay in response to different concentrations of metformin(0,0.01,0.1,1,10 and 50mmol/L).Senescence-associated β-galactosidase (SA-β-ga1) staining and senescence marker Dec1 protein levels were used to evaluate the effect of metformin on hepatoma cells senescence.In addition,protein expression of p-AMPK,p-ACC and AMPK was detected by Western blot analysis.Results Metformin suppressed proliferation of HepG2 cells in a dose-dependent manner.High concentrations of metformin (10 and 50mmol/L) promoted cell apoptosis,while lower doses of metformin (0.01,0.1 and 1mmol/L) led to enlarged and flatten senescent morphology and increased SA-β-ga1 positive cells.Moreover,cell cycle was blocked in G0/G1 phase and protein levels of senescent marker Dec1,p-AMPK and p-ACC were significantly enhanced,whereas AMPK protein expression was almost unchanged.Conclusion We showed here that high dose of metformin promotes HepG2 cells apoptosis,but low doses of metformin induce cellular senescence,which may be related to the activation of AMPK signaling.These data will provide vital evidence for improving the outcome of comprehensive treatment in HCC patients by driving hepatoma cells to undergo senescence.
7.Analysis of transfection characteristic of two different fluorescently labeled siRNA targeting PIK3CA gene in vitro

Objective To compare transfection properties of two different fluorescently labeled PIK3CA siR-NA and to screen out PIK3CA siRNA with high transfection efficiency and strong anti-quenching ability.Methods Two different fluorescently labeled PIK3CA siRNA was transfected into gastric cancer cell BGC-823 by Lipofectamine 2000.The distribution and quenching of fluorescence were observed by inverted fluorescence microscope.Image J software was used to analyze their transfection efficiency.Real-time quantitative PCR was performed to detect the effect of different fluorescently labeled PIK3CA siRNA on PIK3CA mRNA expression.Results The transfection efficiency and anti-quenching ability of two different fluorescently labeled PIK3CA siRNA were different under the same transfection conditions.The transfection efficiency showed no significant differences between Cy3 or FAM labeled PIK3CA siRNA and negative control siRNA(P > 0.05),but the transfection efficiency of Cy3 labeled PIK3CA siRNA and negative control siRNA was significantly higher than the FAM labeled (P < 0.05).Inhibitory efficacy of target mRNA expression induced by Cy3 labeled PIK3CA siRNA was significantly higher than that of FAM labeled PIK3CA siRNA.Conclusion Cy3 labeled PIK3CA siRNA could act as a good tracer and provide an important evidence for further construction of Cy3 labeled PIK3CA siRNA nanoparticle.

To explore novel ADP receptor inhibitors with anti-thrombotic activity, eighteen compounds were synthesized and their structures were confirmed by 1H NMR and MS. The results showed that the activity of compound C1 was superior to ticlopidine in platelet aggregation inhibition tests in vivo and worthy for further investigation. Compounds A4, B2, C4 and C7 possessed moderate platelet aggregation inhibitory activities.
9.Effect of carbonated hydroxyapatite cement for filling vertebral body on the vertebral heights and pain in patients with osteoporotic vertebral compression fractures

BACKGROUND: Carbonated hydroxyapatite cement (CHC) s a new kind of biomaterial for bone defect, which is made of powder and fluid, and can be mixed to be pasty to repair various bone defects.OBJECTIVE: To observe the improvement of vertebrae height and pain in patients with osteoporosis vertebral compression fracture (VCF) after vertebroplasty by using a new kind of bone graft biomaterial, taking CHC as the filling material to reinforce the vertebral body.DESIGN: A contrast observation trial taking patients as subjects.SETTING: Department of Orthopaedics, General Hospital of Chinese PLA.PARTICIPANTS: Totally 34 patients with thoracic or lumbar osteoporosis VCF who received the treatment in the Department of Orthopaedics, General Hospital of Chinese PLA between October 2000 and August 2003. Inclusive criteria: ①Definite diagnosis by CT; ② Informed consents were obtained from the patients. Exclusive criteria: The patients with osteoporosis vertebral compression fractures who suffered vertebral posterior wall fracture. There were 6 males and 28 females, and they were aged (72±13)years; Among the patients, 27 were diagnosed as postmenopausal osteoporosis, 1 as cortical hormone-induced osteoporosis and 6 male patients weresenile osteoporosis.METHODS: ①All the patients were randomly divided into two groups: Experimental group (n =23) and control group (n=11). All the patients were performed percutaneous operation with local anesthenia. All cases were performed percutaneous operation under local anesthesia. Under the C-arm monitored, one side pedicle puncture was performed to enter the anterior column of the involved VCF. Patients of the experimental group were filled with CHC. Patients of control group were filled with polymethyl Methacrylate (PMMA) with the same way. ② Referred to McGill-Melzack scoring. Among the scale 0-100 mm (0 was no pain, 100 was acute pain), the value indicated the painful intensity and mental assault degree. < 30 scores indicated good, 30-40 basically satisfied and ≥ 50 poor .③ Referred to the method from Lee et al, the preoperative height (A1) and postoperative height (A2) of compression fracture position of VCF were measured according to the lateral X-ray film. At the same time, the upper vertebral height (A3) and the inferior vertebral height (A4) were measured at the same position. The original height (A) of the involved vertebra was calculated as (A)= (A3+A4)/2,and the preoperative vertebral compression rate =(A-A1 )/A, the postoperative vertebral compression rate =(A-A2)/A, the restoring rate = (the preoperative vertebral compression rate-the postoperative vertebral compression rate)/the preoperative vertebral compression rate. ④ The wounds of the patients were observed after operation. The levels of blood routine, serum calcium and serum phosphorus were detected before, one day and one week after operation. MAIN OUTCOME MEASURES: ① Preoperative and postoperative VAS scoring. ② The vertebral compression rate and restoring rate. ③ Wounds were observed after operation. The blood routine, the serum calcium and serum phosphorus were detected before, one day and one week after operation.RESULTS: Totally 34 patients were involved in the result analysis. ①The preoperative visual analogue scale (VAS) score of experimental group were (91.5±21.7) points, and the postoperative ones were (44.5±27.2) points. The difference of VAS score reduced gradually along with the postoperative time. There was no difference of VAS score between experimental group and the control group 4 weeks after operation. ② The biocompatibility of CHC in the vertebral body was fine. The vertebral compression rate of experimental group was recovered from (43.1±21.4)% preoperatively to (27.3± 18.5)% postoperatively. The rate of restored heights was (27.3±18.5)%. ③ All patients obtained Ⅰ stage wound healing, and none of them had infection, inflammatory secretion and nervous symptom. There were no differences in blood routine test, serum calcium, serum phosphorus between patients in two groups. One case filled by PMMA and two cases filled by CHC presented leakage, and none had nervous symptom.CONCLUSION: As the filling materials for vertebropalsty, CHC can restore the vertebral heights and relieve pain safely and effectively, however, its efficacy to relieve pain is not significant as PMMA in the short term.
10.Analysis of complications of high intensity focused ultrasound in treatment of uterine leiomyoma

Objective To analysis complications and its associated risk factors of high intensity focused ultrasound (HIFU) in treatment of uterine leiomyoma for selecting rationale indicated patients and reducing complications. Methods Medical documents of 171 patients with 231 leiomyomas in total treated by HIFU were studied retrospectively. Common complications were categorized and analyzed, the relationship between risk factors and complications were studied. Results Common complications in treatment of uterine leiomyomas by HIFU were 71.9% (123/171) of abdominal pain, 17.5% (30/171) of vaginal bloody discharge, 8.2% (14/171) of sacroiliac or buttock pain, 7. 6% (13/171) of skin blister,4. 7% (8/171) of leg pain, 2. 9% (5/171) of hematuria and 1.8% (3/171) of febrile. By logistic regression analysis, the factor correlated with abdominal pain included diameter of uterine leiomyomas,sonication time and average power (P < 0. 05). The factor correlated with sacroiliac or buttock pain was uterine leiomyomas located in posteriors of uterine wall(P <0. 05); the factors correlated with vaginal bloody discharge were sonication time and type of uterine leiomyomas (submucous > intramural > subserous, P <0. 05); the factors correlated with skin blister was sonication time (P < 0. 05). There were no statistical relationship between multiple factors and leg pain, hematuria, febrile (P > 0. 05). Conclusion The modality of high-power and short-term treatment might reduce complications of HIFU ablation.
