1.Effect of glucagon-like peptide-1(7-36)NH_2 on insulin secretion and membrane potentials of pancreatic islet ?-cells

AIM: To explore the cellular mechanisms by which GLP-1(7-36)NH 2 stimulates insulin secretion in ?-cells. METHODS: Using adherent cell analysis and sorting cytometer (ACAS) by DiBAC 4(3) as a probe,effects of glucagon-like peptide-1(7-36)NH 2 on membrane potential was investigated in cultured neonatal rat pancreatic islet ?-cells,and radio immunological methods were used to determine the amount of insulin secretion in different glucose concentrations. RESULTS: In the presence of 2.8 mmol/L glucose,GLP-1(7-36)NH 2 could not increase insulin secretion and could not change membrane potentials of ?-cells;In the presence of 5.6,16.7 mmol/L glucose,GLP-1(7-36)NH 2 produced a marked increase in insulin secretion and enhanced the depolarization of ?-cells.In 16.7 mmol/L glucose, EGTA or nifedipine completely blocked GLP-1(7-36)NH 2-induced insulin secretion and changes in membrane potentials of ?-cells. CONCLUSION: GLP-1(7-36)NH 2 stimulates insulin secretion in a glucose concentration-dependent manner,it can enhance the depolarization of ?-cells caused by glucose.
2.The effect of siRNA targeting MIF on the growth of colorectal cancer xenografts and the life quality of tumor-bearing mice

Objective To analyze the effect of siRNA targeting MIF( MIFsiRNA) on the growth of colorectal cancer xenografts and the life quality of tumor-bearing mice.Methods BALB/C mouse model carring colorectal cancer was established.Thirty mice were divided into three groups randomly and managed respectively with intratumor injection of DEPC water, MIFsiRNA(0.15 nmol/g) and non-specific siRNA (0.15 nmol/g), respectively twice a week for consecutively 4 weeks.Drinking water, fodder consumed and body weight was recorded daily, and tumor volume was measured once a week.Mice were sacrificed after four weeks.ELISA and immunohistochemistry were used to detect the expression of MIF in serum and in tumor tissues.Spectrophotometric detection was used to detect caspase-3 protein.TUNEL was used to detect apoptotic cells.Results MIF expression in serum in MIFsiRNA group was lower than the other two groups [(22 ± 6) ng/ml vs (32 ± 8) ng/ml and (33 ± 8) ng/ml, P < 0.01]; MIF expression in tissues was less than the other two groups [(85 ± 20) /500 vs.(423 ± 23) /500 and (442 ± 31) /500, P < 0.01]; Tumor was smaller than the other two groups at third and fourth week (P < 0.01) ; Tumor weight was significantly less than the other two groups [(1.93 ±0.21) g vs (4.40 ±0.30) g and (5.25 ±0.44) g, P<0.01]; Mice in MIFsiRNA group were healthier than the other two groups as judged by water and fodder consumption (P < 0.01 ) , while weight change was not significantly different among the three groups ( P > 0.05 ).Caspase-3 protein in tissues was higher than the other two groups [(0.74 ±0.06) μg vs (0.57 ±0.08) μg and (0.56 ±0.02) μg, P <0.01]; Apoptosis cells in tissues were higher than the other two groups [(12 ± 2)/ 100 个vs 0 and 0, P < 0.01].Conclusions Knockdowning MIF gene expression inhibits the growth of colorectal cancer xenografts and improves life quality of tumor-bearing mice, possibly by a mechanism in which MIFsiRNA activates caspase-3 promoting cell apoptosis.

OBJECTIVE:
To summarize pseudocyst of auricle governor square included surgical curative effect, the advantages of the analysis of the operation.
METHOD:
The governor of the central square in auricle pseudocyst incision, 56 cases were retrospectively analyzed clinical data, surgical curative effect and advantages were summarized.
RESULT:
Adopt the auricle pseudocyst governor of the central square of incision, postoperative follow-up of 6 months did not relapse.
CONCLUSION
Rectangle window method effective method of treating pseudocyst of auricle, able to cure pseudocyst of auricle, is a kind of simple, practical, and value in the basic-level hospital surgical method.

4.Laboratory diagnosis and genetic characterization analysis of the subtype A3 influenza viruses in Quanzhou

Objective To obtain the information of the 2009 influenza outbreak and the variations of influenza virus strains in quanzhou, and explore the relationship between the genetic variation of influenza virus and influenza epidemic. Methods During the influenza outbreak in quanzhou,one hundred and ninetyeight throat swabs specimens from the patients with influenza were collected. Viruses were isolated with MDCK cells and identified with serological test, followed by real-time RT-PCR. RNA of four influenza virus strains were extracted, then HA1 gene was amplified by RT-PCR. The purified PCR products were sequenced. The data were analyzed with the software DNAstar megalign. Results Total 98 pieces of H3N2 subtype influenza virus nucleic acid were detected in 198 throat swabs specimens,among which 62 influenza virus strains were identified as subtype influenza A( H3N2 ). The sequencing results of HA1 gene in these positive strains showed that their genetic characterization were more closed to strains A/Ningbo/333/2008 with a nucleotide homology of 98.7%, which was 96.8% as compared with A/Xiamen/70/2004. The amino acids sequences deduced from the nucleotide sequences in HA1 region of the isolated strain had 7 mutant sites compared with A/Brisbane/10/2007 vaccine strain. One variant amino acids were found located in the antigenic determinant sites A( 144 ), two were in the sites B( 158,189 ). Phylogenetic analysis also confirmed the difference in HAl domain. Conclusion The influenza virus strains causing the flu outbreak among some communities of quanzhou in 2009 are subtype influenza A ( H3N2 ), whose genetic characterization and antigenicity were different from the vaccine strain.
5.Influenza H1N1 surveillance and analysis of genetic characteristics of predominant strains in Quanzhou during 2009

Objective To investigate the influenza H1N1 virus surveillance of 2009 in Quanzhou,and analyze the HA and NA gene of influenza H1N1 virus, explore its genetic variation and molecular characteristics. Methods During the influenza H1N1 virus surveillance in Quanzhou,specimens of throat swabs from the patients with influenza were collected, and detected by real-time RT-PCR. Viruses were isolated with MDCK cells and identified with serological test. Two influenza virus isolates were extracted, and their HA and NA genes were amplified by RT-PCR. The purified PCR products were sequenced. The data obtained were analyzed with the software DNAMAN. Results Of 1020, influenza H1N1 virus RNA was detected in 200 specimens, seasonal influenza virus RNA was detected in 70 specimens. A total of 29 influenza A H1N1 virus strains were isolated. The nucleotide homology in the HA gene was highly homologous with that of pandemic influenza virus in North America. The amino acids sequences deduced from the nucleotide sequences in HA region of the isolated strain had 22 variations compared with A/Brisbane/59/2007 vaccine strain recommend by WHO,the characteristics of α2,6 sialic acid receptor binding remained. The analysis of amino acids sequences of NA indicated that this virus possessed Oseltamivir sensitivity. Conclusion The causative influenza H1N1 strains in Quanzhou is highly homologous with that of pandemic influenza in North America, and it is antigenically and genetically different from the vaccine strain.

Objective Observe the effect of Total paeony glycoside(TPG)has on the T-cell immune action and apoptosis process of Oral lichen planus patients and analyze the mechanism.Methods 20 OLP patients are recruited into this experiment, each is treated with TPG,and their clinical effect is taken notes of.Observe the apoptosis of OLP lesions from each of the patients before and after the treatment,compare the apoptosis condition and the expression of CD4 + ,CD8 + ,and the CD4 +/CD8 + ratio,to analyze the role TPG plays on the immune expression of T-cell and the effect on apoptosis process.Results TPG improve the clini-cal manifestation of OLP patients significantly,resulting in the decrease of reticulum and erosive areas,and the pain index is de-creased obviously.The expression of CD4 + and CD8 + are decreased in OLP lesions after the treatment of TPG.Conclusion TPG may induce the apoptosis of the T-cell in lamina propria of the OLP lesions to regulate the T-cell immune action of OLP patients, and slow the development of inflammatory process of OLP,and in that way,TPG shows its potential in treating OLP,and this ex-periment can provide the primary evidence.
7.CEUS features of hepatitis B virus-related combined hepatocellular-cholangiocarcinoma and hepatocellular carcinoma: Comparative study

Objective To comparatively analyze CEUS features of hepatitis B virus (HBV)-related combined hepatocellular-cholangiocarcinoma (CHC) and hepatocellular carcinoma (HCC).Methods Thirty-one patients with HBV-related CHC and 31 patients with HBV-related HCC confirmed by pathology were enrolled and CEUS features were compared.Results On CEUS,HBV-related CHC and HBV-related HCC mainly manifested as hyper-enhanced in arterial phase and hypo-enhanced in portal phase and delayed phase.No significant differences of enhancement level on CEUS were found between HBV-related CHC and HBV-related HCC.When the maximum diameter of tumor ≤3.0 cm,both HBV-related CHC and HBV-related HCC were mainly homogeneous enhancement (P=1.000).When the maximum diameter of tumor more than 3.0 cm,diffuse heterogeneous enhancement and peripheral irregular rim-like enhancement were more commonly observed in HBV-related CHC,while diffuse heterogeneous enhancement was more commonly noted in HBV-related HCC (P=0.001).Conclusion The enhancement pattern of HBV-related CHC >3.0 cm has relative specific performance.
8.A clinical study of virtual endoscopy ultrasound Fly-Thru in the diagnosis of obstructive degree and nature for obstructive bile duct diseases

Objective To investigate the utility of virtual endoscopy ultrasound Fly-Thru in the diagnosis of obstructive bile duct diseases.Methods One-hundred patients with obstructed bile duct diseases underwent Fly-Thru examination.All Fly-Thru images were reviewed by two radiologists with different experience.The capabilities of Fly-Thru for bile duct obstructive degree evaluation and distinguishing malignant lesion from benign one were assessed respectively.Results The accuracy and sensitivity of Fly-Thru image in obstruction degree evaluation were 70.59% and 89.2% (95%CI 74.6%-96.9%).The diagnosis accuracy of 2DUS for lesion characteristics increased from 80% to 84%,accompany with Fly-Thru images,especially for lesions in common bile duct from 77.2% to 86.0% (x2 =14.399,P =0.001).Conclusions The virtual endoscopy ultrasound Fly-Thru is only partly capable to display the obstructed degree of some bile ducts,but it can improve the diagnostic accuracy of common bile duct diseases.
9.Imaging features on contrast-enhanced ultrasound and pathological analysis of combined hepatocellular cholangiocarcinoma

Objective To evaluate the correlation between the imaging features on contrast-enhanced ultrasound (CEUS) and pathological characteristics of combined hepatocellular-cholangiocarcinoma (CHC).Methods Forty patients with pathologically proven hepatic CHC were evaluated,the CEUS imaging findings and pathological characteristics of CHC were retrospectively analyzed.Results On CEUS,peripheral irregular rim-like enhancement,diffuse heterogeneous enhancement and diffuse homogeneous enhancement were illustrated in 13(32.5 %),21 (52.5 %) and 6(15.0%) lesions,respectively.Pathological findings showed that HCC-predominance,CC-predominant,and similar proportions of the two components were illustrated in 16 (40.0 %),19 (47.5 %) and 5 (12.5 %) cases,respectively.The presence of necrosis were illustrated in 28 (70.0%) cases.On CEUS,when the enhancement pattern was peripheral irregular rim-like enhancement,CC-predominance and necrosis were presented in 11(84.6%) and 10(76.9%) cases,respectively.When the enhancement pattern was diffuse heterogeneous enhancement,CC-predominance and necrosis were presented in 11(52.4%) and 18(85.7%) cases,respectively.When the enhancement pattern was diffuse homogeneous enhancement,HCC-predominance and necrosis were presented in 4(66.6%) and 0 (0%) cases,respectively.There were significant differences in relative proportion of HCC,CC components and tumor necrosis among the three types of enhancement pattern on CEUS (P =0.009 and P < 0.001).When CHCs were ≤ 5 cm,peripheral irregular rim-like enhancement,diffuse heterogeneous enhancement and diffuse homogeneous enhancement were illustrated in 5,3 and 5 cases,respectively.When CHCs were >5 cm,peripheral irregular rim-like enhancement,diffuse heterogeneous enhancement and diffuse homogeneous enhancement were illustrated in 8,18 and 1 cases,respectively.There were significant differences in the three types of enhancement pattern between lesion size of ≤5 cm and >5 cm on CEUS (P =0.006).Conclusions The imaging findings of CHC on CEUS depends on the relative proportions of HCC and CC component and on size-dependent patterns.

Neurofibromatosis type Ⅰ(NF1)is an autosomal dominant genetic disease triggered by mutations of nf1gene, nf1 gene and its encoded protein product neurofibromatoprotein play important roles in tumor supressive activity. Plexiform neurofibroma was the main manifestation among some patients For plexiform neurofibroma, surgical treatment did not have satisfactory effect. meanwhile, traditional radiotherapy and chemotherapy are ineffective. All of those above serve as challenges for clinical treatment and have been received much more attention from study of multimoics and targeting therapy In this Review, the clinical features of NF1-associated plexiform neurofibromasand, the progress regarding investigation of drug targets and clinical trials for the drug of plexiform neurofibroma will be presented.
