Objectives:To study the alterations of NF-?B activation in PBMC, in order to clarify the signal transcription of NF-?B concerned in the mechanism of inflammatory reaction in severe burns. To evaluate the alterations of expression of NF-?B dependent proinflammatory cytokines mRNA. Thereby, to observe the regulating effects of NAC on NF-?B, and verify the medial effects of upstream signal molecules on cytokines expression. Methods:Sprague Dawley rats were randomly assigned to each group. A 30% TBSA full-thickness scald was performed by immersing in 100℃ water for 12 seconds. PBMC were isolated at different time point after scalding. From isolated PBMC, the total RNA was isolated and the nuclear protein purified by Trizol reagents. The NF-?B proteins were measured through EMSA. Message RNA expression of proinflammatory cytokines and Th2 cytokines were assayed by RT-PCR. Results:Lots of NF-?B proteins were sequester in nucleus in all burn groups. mRNA expression of proinflammatory cytokines were also enhanced and related to the activity of NF-?B protein in nucleus. NAC could decrease the activity of NF-?B protein in nucleus. NAC had also significantly inhibitory effects on the production of proinflammatory cytokines. However, all of Th2 cytokines measured in this study were enhanced too. Conclusions:NF-?B might be the second signal molecule from cytosol to nucleus and mediate the proinflammatory cytokines transcription in PBMC after burns. In signal transduction levels, enhancing scavengers of oxygen free radicals in PBMC may modulate the activity of NF-?B which mediate the expression of proinflammtory cytokines.

Objective: To study the correlation between intestinal mucosa injuries and inflammatory reaction in cancer cachexia mice. Methods: 14 male BALB/c mice were randomized into two groups: control group and tumor-bearing group.For tumor-bearing group,a suspension of 10 6 murine colon 26 adenocarcinoma cells was inoculated s.c into each mouse.Non-tumor body weight,intestinal villus height,width,basal layer thickness and muscular layer thickness,TNF-?,IL-1,IL-6,IL-10,IFN-?,sTNF-RⅠand sTNF-RⅡ in the serum and intestinal tissue were determined at the 16th day following the inoculation. Results: The non-tumor body weight and intestinal villus height and basal layer width in tumor-bearing group were lost significantly as compared with control group.The concentration of serum TNF-?,IL-6,sTNF-RⅠwere higher and the level of serum IL-10 was less in tumor-bearing group.The concentration of intestinal tissue TNF-? was higher but the IL-1,IFN-?,sTNF-RⅠ and sTNF-RⅡ were less in the tumor-bearing group. Conclusion: The intestinal mucosa injuries have close relation with the inflammatory reactions in cancer cachexia.

Objective: The bacterial superantigen staphylococcal enterotoxin A (SEA) has the potent ability of inducing T cell activation as well as directing activated T cells to kill tumour cells. However, its application in the tumour treatment is limited due to its systemic immune activation.In order to minimize its side effects SEA liposomes is prepared and their toxcicity in vivo is investigated . Methods: SEA liposomes were prepared by the method of reverse-phase evaporation . SEA liposomes were administered intravenously . In vivo the toxcicity of SEA liposomes was investigated by the measurement of blood pressure, colonic temperature and breath rate of New Zealand rabbits. Mice plasma TNF-? and IFN-? level were determined by ELISA . Results:After iv administration of SEA liposomes, significant reduction of mice plasma TNF-? and IFN-? level was observerd. As compared to free SEA, liposomal SEA had less effect on blood pressure, colonic temperature and breath rate of the rabbits. Conclusion:SEA Liposomes had relatively low toxicity. These advantage was probably due to its lower systemic immune activation effect and inducing consequent lower systemic TNF-? and IFN-? level.

Objective: This study was designed to investigate the variations of endotoxin,glutamine and TNF-? concentration in septic rats. Methods: A total of 90 adult male SD rats were divided as follows: control group(normal),experimental group(CLP).They were killed on hour 6,12,24,48,72 after surgery.Plasma,liver,small intestine and skeletal muscle were collected to measure the concentrations of glutamine in plasma and tissues,the levels of endotoxin and TNF-? in plasma. Results: In septic rats,plasma concentrations of endotoxin and TNF-? increased on hour 6,significantly increased on hour 12,attained the peak on hour 24,and decreased evidently on hour 48 and 72.Glutamine concentration in plasma elevated on hour 6 and 12,elevated significantly on hour 24 and 48,and decreased on hour 72.Glutamine concentration in liver increased on hour 6～12,increased significantly on hour 24,and decreased significantly on hour 48 and 72.Glutamine concentration in small intestine and skeletal muscle decreased on hour 6 and 12,and decreased significantly after 24 hour. Conclusion: During the early stage of sepsis,the plasma levels of endotoxin and TNF-? are increased significantly,the glutamine concentration in plasma and liver is also increased significantly,however,it was decreased evidently in skeletal muscle and small intestine.
