Objective To survey the quality of life and mental health status of pre-post household service workers who held a high school graduation certificate or above and had been engaged in different work.Methods A total of 254 household service workers completed Symptom Checklist 90 (SCL-90) and World Health Organization Quality of Life-Brief (WHOQL-BREF).Statistical analyses of variance and correlation were carried out to evaluate mental health status and quality of life of the participants.Results Abnormal score of SCL-90 was found in 8% of household service workers who had graduated from high school.The psychological score of household service workers who had worked as workers(60.59± 2.18)was lower than those who had worked as service workers (66.23 ± 12.13) or peasants (65.40 ± 12.24) (P <0.05).Almost all factors of SCL-90 were negatively correlated with WHOQL-BREF (correlation coefficient -0.140 to-0.376).Conclusions Our data suggest that mental health status can be well in pre-post household service workers who held a high school graduation certificate or above.Special training should be given to the household service workers in accordance with their working experience.
3.Protective mechanism of NALP3-siRNA on rat renal tubular epithelial cells from hypoxia/reoxygenation injury

Objective To explore the mechanism of protecting cells from hypoxia/ reoxygenation (H/R) injury by constructing specific small interference RNA (siRNA) to inhibit NALP3 expression in rat renal tubular epithelial cells (NRK-52E).Methods (1) To establish the H/R injury model of NRK-52E by regulating the pressure of N2 in incubator to hypoxia condition,the cells were cultured with hypoxia for 1 h and then with reoxygenation for 1 h,2 h,4 h,8 h,16 h and 24 h.The activity of lactae dehydrogenase (LDH) in the culture medium,cell count and cell viability,the expression of NALP3 were determined by biochemical method,trypan blue exclusion and Western blotting.(2) The siRNA was transfected into NRK-52E.The irrespective siRNA transfected group wasused as control.NALP3 expression was examined by Western blotting.(3) The cells were divided into 4 groups:control group,H/R group,irrespective siRNA transfected group and NALP3-siRNA transfected group.To establish the H/R injury model of NRK-52E by regulating the pressure of N2 in incubator to hypoxia condition,the cells were cultured with hypoxia for 1 h and then with reoxygenation for 4 h.And the expression of NALP3 was determined by Western blotting.(4)Cellular apoptosis was examined by Annexin V/PI staining and flow cytometry.NF-κB DNA binding activity,IκB-α,Bcl-2 and Bax expression were examined by EMSA and Western blotting.Results (1)Compared with the control group,the activity of LDH significantly increased,cell count and cell viability significantly decreased (all P<0.05).The expression of NALP3 significantly increased and peaked at 4 h after H/R.(2)The specific siRNA could efficiently inhibit NALP3 expression in NRK-52E.Compared with the irrespective siRNA transfected group,the protein expression of NALP3 was significantly down-regulated in NALP3 siRNA transfected group (P<0.05).(3)After hypoxia 1 h and reoxygenation 4 h,the activity of LDH and the expression of NALP3 increased.Compared with the irrespective siRNA transfected group,LDH concentration in media and the expression of NALP3 significantly decreased in NALP3-siRNA transfected group.(4)After hypoxia 1 h and reoxygenation 4 h,NF-κB DNA binding activity was increased,IκB-α phosphorylation and degradation,Bcl-2 and Bax were significantly up-regulated.However,compared with the irrespective siRNA transfected group,NF -κB DNA binding activity,IκB-α degradation and Bax/Bcl-2 were significantly decreased (P<0.05) in NALP3-siRNA transfected group.At the same time,the ratio of apoptosis was significantly increased in three groups than that in control.Compared with the irrespective siRNA transfected group,the ratio of apoptosis in NALP3-siRNA transfected group was significantly decreased (P<0.05).Conclusions H/R induces the expression of NALP3 in NRK-52E.The synthesized siRNA can inhibit the expression of NALP3 and protect NRK-52E from hypoxia/reoxygenation injury.The mechanism may be via inhibiting the activation of NF-κB,modulating expression of Bcl-2 and Bax,as well as decreasing cell apoptosis.

Acute kidney injury (AKI), associated with significant morbidity and mortality, is widely known to involve epithelial apoptosis, excessive inflammation, and fibrosis in response to ischemia or reperfusion injury, which results in either chronic pathological changes or death. Therefore, it is imperative that investigations are conducted in order to find effective, early diagnoses, and therapeutic targets needed to help prevent and treat AKI. However, the mechanisms modulating the pathogenesis of AKI still remain largely undetermined. MicroRNAs (miRNAs), small non-coding RNA molecules, play an important role in several fundamental biological and pathological processes by a post transcriptional regulatory function of gene expression. MicroRNA-21 (miR-21) is a recently identified, typical miRNA that is functional as a regulator known to be involved in apoptosis as well as inflammatory and fibrotic signaling pathways in AKI. As a result, miR-21 is now considered a novel biomarker when diagnosing and treating AKI. This article reviews the correlative literature and research progress regarding the roles of miR-21 in AKI.

6.Video electroencephalogram analysis of 100 cases of intellectual disability children with unexplained pathogenic factor

Objective To observe 100 video-electroencephalogram (EEG)-cases of intellectual disability (ID) children,to analyze their EEG features of different degrees of ID,to explore the relationship between EEG changes and ID.Methods We performed a retrospective analysis of long-range video-EEGs of 100 children with an unexplained reason of ID from July 2016 to May 2018 in the department of pediatrics of Xiangya Hospital of Central South University,sumnarized their EEG manifestations,and compared their differences among different degrees of ID.We carried out x2 test according to the degree of ID,and discussed the relationship between the degree of ID and EEG.Results Among 100 cases of ID children with unexplained pathogenic factor,67 cases (67%) of them were males,33 cases (33 %) of females,38 cases (38%)of mild ID,27 cases (27%) of moderate ID and 35 cases (35%) of severe ID.The EEG background of ID patients showed fast waves (11%),slow waves (15%),generalized discharges (14%) and focal discharges (36%),etc.Focal sharp and spike-wave were the most common in children with moderate ID,while background changes and multifocal sharp spike wave were more common in severe ID children.Conclusions The EEG background changes in children with severe ID have significant differences than mild and moderate degree ones.The specific features of EEG in mild and moderate degree ones is focal discharges,while multiple discharges in severe ID patients.Changes in the EEG may be related to the extent of intellectual disability.
7.Study on the Effect Mechanism of Iridoid Glycosides of Scrophularia ningpoensis Inhibiting Cardiomyocytes Apoptosis in Myo- cardial Infarction Model Rats Based on Different Caspase Apoptosis Pathways

OBJECTIVE: To study the effect mechanism of iridoid glycosides extracted from Scrophularia ningpoensis inhibiting cardiomyocytes apoptosis in myocardial infarction model rats. METHODS: The male Wistar rats were randomly divided into sham operation group, model group and S. ningpoensis iridoid glycosides low-dose, medium-dose and high-dose groups, with 10 rats in each group. Myocardial infarction models were established by ligating the left anterior descending coronary artery of the rats, and sham operation group was only threaded without ligation. After the model was established, each administration group was given S. ningpoensis iridoid glycosides suspension intragastrically at three different doses of 50,100,200 mg/kg (by the amount of total glycosides extract) with 10 mL/time, twice a day, for consecutive 7 days. Sham operation group and model group were given constant volume of normal saline intragastrically with same method. The changes of S-T segment of lead ECG Ⅱ were recorded before, after and during 7 days of administration. Cardiac function of rats was examined. The serum levels of LDH, CK-MB, cTnⅠ, NT-pro BNP and TNF-α were determined by colorimetry, immunosuppression or ELISA. The apoptosis of myocardial cells was observed by TUNEL method. SOD activity and MDA content in cardiac myocytes were detected by colorimetry. The expressions of Bcl-2, Bax, Cyt C, Caspase-8, Caspase-9, Caspase-12, Caspase-3 and Calpain in cardiac myocytes were detected by ELISA, enzymolysis colorimetry or enzymatic fluorescence assay. RESULTS: Compared with sham operation, electrocardiogram S-T segment was significantly elevated and the left ventricular end-diastolic diameter and left ventricular end-systolic diameter were significantly increased in the model group; left ventricular ejection fraction and short axis shortening rate decreased significantly; serum levels of LDH, CK-MB, cTnⅠ, NT-pro BNP and TNF-α were increased significantly; there were a large number of yellow-brown apoptotic cells in myocardial tissue; the activity of SOD in myocardial tissue was significantly decreased while the content of MDA was significantly increased; the protein expression level of Bcl-2 and Bcl-2/Bax were significantly decreased, while the levels of Bax, Cyt C, Caspase-3, Caspase-8, Caspase-9, Caspase-12 and Calpain were significantly increased (P<0.05 or P<0.01). Compared with model group, above indexes and pathological changes of myocardial tissue were improved significantly in administration group; the level of Bcl-2 and Bcl-2/Bax in cardiomyocytes increased significantly, while the levels of Bax, Cyt C, Caspase-3, Caspase-8, Caspase-9, Caspase-12 and Calpain decreased significantly (P<0.05 or P<0.01). CONCLUSIONS: S. ningpoensis iridoid glycosides can inhibit the activation of Caspase-3 by inhibiting three apoptotic pathways related to Caspase-8, Caspase-9 and Caspase-12, and then inhibit the apoptosis of cardiomyocytes.
