1.Vocal Acoustics on Patients with Abnormal Lung Function

Objective To investigate the acoustic f eatures of voice in patients with abnormal lung function, evaluate the effect o f lung dysfunction on acoustics of voice,and observe the relationship between re spiration and phonation. Methods The voice signals of sustained vowel /a/ were mea sured by a micro-computer with Dr.speech science for windows software in 50 pati ents with abnormal lung function and 50 healthy adults. The acoustic parameters( F0,jitter,shimmer,intensity,NNE) were collected and analysed in lowest pitch,no rmal pitch and highest pitch.All patients were divided into two groups according to sex. Results In patients with abnormal lung function ,the pa rameters (shimmer, jitter, NNE,)were significantly increased or others(F0,inten sity) were significantly decreased in lowest pitch. Intensity decreased si gnificantly in highest pitch . There was a positive relationship between the de grees of lung dysfunction and the parameters of acoustics. Conclusion Abnormal lung function affected significantly on acoustics of voice in three different pitches; The changes of the parameters were related to the degrees of lung function.The results showed that respiratory activities play an important role in phonation.
2.Application of Acoustic Analysis by Computer in Differential Diagnosis of Organic and Functional Voice Disorder

Objective To investigate the characteristics of acoustic parameters and spectrograph of organic and functional voice disorder and evaluate the value of acoustic analysis technique by computer in the differential diagnosis of voice disorder. MethodsThe voice signals of sustained vowel "a" were measured with a microcomputer (Dr.Speech Software) in 70 healthy adults, 60 patients with varying organic diseases of vocal cords and 50 cases with functional voice disorder. The acoustic parame ters (jitter, shimmer, NNE) and spectrographic characteristics were analyzed and compared. ResultsIn 60 patients with organic diseases of vocal cords, all the acoustic parameters were significantly increased and the spectrograph showed irregularity,breakage or lack of harmonic waves and formants, and increase of noise. In 50 cases with functional voice disorder, the shimmer and NNE were significantly increased but the jitter was usually normal. The spectrograph showed pathologic changes of harmonic waves and formants in middle and high frequencies. The normal acoustic characteristics could be found in 94% (47/50) of the patients with functional voice disorder. ConcluisionThe acoustic analysis by computer technique is of value in the differential diagnosis of or ganic and functional voice disorder.
3.Research progress of phosphatidylinositol 3-kinase/AKt signaling pathway and mechanisms of resistance to sorafenib in hepatocellular carcinoma

Sorafenib is a molecular targeted drug for the treatment of advanced primary liver cancer.However,along with the occurrence of drug resistance,the therapeutic effect was effected.At present,there is clear evidence that the emergence of drug resistance of live cancer is closely related to the epithelial-mesenchymal transition,liver cancer stem cells and the heterogeneity of liver cancer,and the PI3K/AKt signaling pathway as the vital common signaling channel was involved in the above mentioned process.From this we can conclude that complementary inhibtion of PI3K/AKt signaling pathway at the same time is the method that can strengthen the effect of sorafenib on the treatment of liver cancer so far.
4.Influence on the proliferation of breast cancer with silencing YKL-40 by siRNA interference

Objective:To explore the relationship between YKL-40 and the proliferation of breast cancer and its mechanism.Methods:The expression of YKL-40 in breast cancer MCF-7 cells was detected by immunofluorescence assay.Fluorescence microscope was used to observe the conversion efficiency,and Real-time PCR was used to screen the most effective YKL-40 siRNA.Expression levels of PI3K,P-PI3K,AKT,P-AKT of the PI3K/AKT pathway associated proteins was test by Western blot.At the same time,MTT and flow cytometry were validated by YKL-40 siRNA treatment of human breast cancer MCF-7 ceils,the differences of 24h,48h and 72h groups of cell proliferation ability and cell cycle.Result:MCF-7 cell express YKL-40 protein,mainly located in the cytoplasm.Real-time PCR show that siRNA01,siRNA02,siRNA03 compared with NC group YKL-40 gene silencing effect is remarkable.Among of them the strongest silencing effect is siRNA02 (P<0.01),Western blot show the experimental group than the control group,Total PI3K and AKT remain unchanged while P-PI3K,P-AKT expression decreased (P<0.05).In the experimental group,the number of G1 cells in the control group was increased (P<0.01),while the S phase cells decreased (P<0.01).MTT results showed that the experimental group compared with the control group,the proliferation ability is decreased(P<0.01).Conclusions This study suggests that YKL-40 can be used as the upstream regulatory factor of PI3K/AKT signaling pathway and affect the process of cell cycle in breast cancer,and then regulate the proliferation of breast cancer,YKL-40 may be a crucial target for the treatment of breast cancer.
5.Imaging features of hepatic angiomyolipoma on CT and MRI

Objective To analyze the imaging features of hepatic angiomyolipoma (HAML) on computed tomography (CT) and magnetic resonance imaging (MRI).Methods The imaging fingdings of 18 tumors which were pathologically diagnosed as HAML after surgery were analyzed retrospectively.Before operation,twelve and ten patients underwent CT and MRI non-contrast and dynamic enhanced scans,respectively,and 4 patients received both examinations.The imaging characteristics including the number,diameter,location,appearance of the lesions,plain and dynamic enhancement mode were analyzed.Results Eighteen HAML lesions were found in 18 patients with a diameter ranging from 1.5 cm to 17.2 cm (mean 5.3 cm).Five lesions manifested fatty content,and one showed hemorrhage and necrosis.Five HAMLs enhanced in a fast-in and fast-out mode,eleven in a fast-in and slow-out mode and two other lesions in an irregularly discrete mode.The arterial supply was found in 11 HAMLs in the hepatic arterial phase,all coming from intrahepatic arteries.Intratumoral vessels were observed in 12 HAMLs.Early draining veins to the hepatic vein (n =3) and portal vein (n =1) were detected in 4 HAMLs.One lesion demonstrated delayed enhancement in the pseudocapsule.Conclusion The detection of arterial supply and intratumoral vessels in a hypervascular hepatic tumor on contrast CT and MRI in a patient with a non-cirrhotic liver and normal AFP helps to make a diagnosis of HAML.
6.Detection of EBV-LMP1 antibodies in sera and clinical significance in the patients with nasopharyngeal carcinoma.

OBJECTIVE:
To detect the levels of Epstein-Barr virus (EBV) latent membrane protein 1 Antibodies (LMP1-Ab) in nasopharyngeal carcinoma (NPC)sera and discuss the clinical significance of this test in diagnosis, prognosis, and immune-targeted therapy of NPC.
METHOD:
Enzyme-linked immunosorbent assay (ELISA) and Western blot method were used to detect the LMP1-Ab levels in 61 NPC sera, 30 nasopharyngitis sera, and 55 normal sera. The relationship between the LMP1-Ab level and clinical and pathological features of NPC was analyzed.
RESULT:
ELISA test showed that LMP1 antibodies level was significantly higher in nasopharyngeal carcinoma group than those in nasopharyngitis group and in healthy group and there were statistical significances (all P<0.05). In SNPC group, the LMP1-Ab level was not related to the pathological grade and cervical lymph node metastases (P>0.05). Western blot test revealed that the expression of LMP1 antibodies was higher in NPC sera than in nasopharyngitis sera and in normal sera.
CONCLUSION
LMP1-Ab level was higher in nasopharyngeal carcinoma group than in nasopharyngitis group and in normal group. Therefore, LMP1 may be considered as a tumor correlated antigen to help the diagnosis and immune-targeted therapy of NPC.

7.Characteristic comparison of mouse primary macrophages cultured in L929 cell conditioned medium.

The purpose of this study is to provide a culture for mouse bone marrow-derived macrophages (BMDM) and peritoneal macrophages (PM) and to characterize their molecular and cellular biology. The cell number and purity from the primary culture were assessed by cell counter and flow cytometry, respectively. Morphological features were evaluated by inverted microscope. Phagocytosis by macrophages was detected by the neutral red dye uptake assay. Phenotypic markers were analyzed by real-time fluorescent quantitative PCR. Our results show that the cell number was much higher from culture of BMDM than PM, while there was no significant difference regarding the percentage of F4/80+CD11b+ cells (98.30%±0.53% vs. 94.83%±1.42%; P>0.05). The proliferation rate of BMDM was significantly higher than PM in the presence of L929 cell conditioned medium, by using CCK-8 assay. However, PM appeared to adhere to the flask wall and extend earlier than BMDM. The phagocytosis capability of un-stimulated BMDM was significantly higher than PM, as well as lipopolysaccharide (LPS)-stimulated BMDM, except the BMDM stimulated by low dose LPS (0.1 μg/mL). Furthermore, Tnfα expression was significantly higher in un-stimulated BMDM than PM, while Arg1 and Ym1 mRNA expression were significantly lower than PM. The expression difference was persistent if stimulated by LPS+IFN-γ or IL-4. Our data indicate that bone marrow can get larger amounts of macrophages than peritoneal cavity. However, it should be aware that the molecular and cellular characteristics were different between these two culture systems.
