Chemotherapy -induced peripheral neuropathy ( CIPN ) is one of the most common adverse effects in chemotherapy .The mechanism of CIPN has always been attracting researchers′attention.Recently,the intimate relationship between nerve growth factor and CIPN is one of the most hot topics .NGF protects neurons from damage of chemotherapy drugs through inhibiting apoptotsis and other pathway to relieve the neurotoxicity . However ,there are still many problems in the clinical application of exogenous NGF .To improve the mechanism of NGF in the development and application approach of CIPN ,treatment of chemotherapy -induced peripheral neu-rotoxicity is of great significance .

[Objective]To introduce the clinical experience of Pro.Huo Jiege treating lung cancer in the elderly. [Methods]Through copying prescription with the teacher and carding records, and studing on Chinese literature, sum up Pro.Huo Jiege ’s unique insights for the etiology, pathogenesis, principles and methods, and explains it by one case. As the third-generation bisphosphonate,zoledronic acid (ZOL) has been widely used for the treatment of skeletal-related events in breast cancer.Previous studies have demonstrated that ZOL has antitumor activitie in breast cancer.ZOL can induce direct and indirect antitumor activities through inducing apoptosis,inhibiting the proliferation,adhesion and invasion formation of bone microenvironment which is conducive to tumor growth,besides inhibiting angiogensis and modulating immune response.Also evidences of synergistic antitumor effects are found when ZOL combines with radiotherapy and chemotherapy,endocrine,and targeted drugs.The observed improvement in antitumor effects by using combination therapy with ZOL in breast cancer is currently being verified through additional clinical trials.

Discuss the unique views and characteristics of TCM in reducing adverse reactions caused by western medicine treatment, and expound the prevention and treatment of recurrence and metastasis through typical cases.[Results]Pro.HUO Jiege thinks that the etiology and pathogenesis relate to insufficient natural endowment, seven emotions and cancerous toxin. Pro.HUO is expert in treating breast cancer through regulation of Qi, giving consideration to both heart and kidney.In terms of using medication,he is experienced in treating lymph edema after breast cancer operation, toxic reaction induced by chemoradiotherapy,endocrine treatment-related menopausal syndrome and distant metastasis of breast cancer. He usually invigorates Qi and strengthens the spleen, nourishes yin and blood and uses a small amount of attacking poison to help ward off evil spirit. And the case in this article has obtained good treatment results.[Conclusion] Pro.HUO Jiege is good at using traditional Chinese medicine in the treatment of breast cancer,which makes fairly good clinical effect,and his experience is worth learning and promotion.

A cDNA library was constructed from glands of wild Buthus martensii Karsch scorpion.A new sodium channel long-chain toxin BmkNaTx12 was identified by sequence alignment,and its full-length cDNA sequence was cloned.Sequence alignment showed that the sequence of BmkNaTx12 had high structural similarity with the β-toxin Cn12 from Mexican Centruroides noxius scorpion.The predicted structure of BmkNaTx12 was obtained by homologous model construction and the highest scoring model was selected.The protein structure was found to be stable at 300 ns by molecular dynamics optimization,and fluctuations at amino acids 20,35,and 52 are most like-ly due to the location of these amino acids in the loop region.Then,an expression system of recombinant BmkNaTx12-Fc fusion protein was constructed.The optimal expression time of recombinant protein was determined by Western blot and the fusion protein was successfully expressed in HEK293 cells.The purity of recombinant fusion protein which was obtained by protein A affinity chromatography was up to 95％ by SDS-PAGE gel electrophoresis and HPLC.In addition,the whole-cell patch-clamp assay results suggest that 1 μmoL/L BmkNaTx12-Fc can increase 20％ Nav1.7 peak current.These results laid a foundation for the further study of biological function.
