1.Changes of potassium channels in subarachnoid hemorrhage-induced cerebral vasospasm

OBJECTIVE: Cerebral vasospasm (CVS) induced by subarachnoid hemorrhage (SAH) is a difficulty in neurosciences. Because the pathogenesis of CVS remains a matter of discussion, clinical prevention and treatment are currently not sufficient. With the application of advanced experimental technology in recent years, great breakthroughs have been achieved in domestic and foreign studies on potassium channels. Definite mechanism in the region plays a significant role in instruction for clinical prevention and treatment. In this paper, researches related to potassium channels in cerebral vascular vessels after SAH are reviewed.DATA SOURCES: Related articles published between January 1991 and December 2005 were searched by the computer in Medline database with the key words of "potassium channels, subarachnoid hemorrhage, cerebral vasospasm" and the language was limited to English. Meanwhile, relevant articles were also searched in CNKI and Wanfang database with the same key words in Ghinese.STUDY SELECTION: All articles were selected firstly, and literatures related to potassium channel on physiology and pathogenesis of SAH-induced CVS were selected. Researches on biochemistry and gene etc were excluded.DATA EXTRACTION: A total of 45 English articles were collected, among which, 17 met the inclusion criteria. Of 2 Chinese articles collected,one study was in accordance with the inclusion criteria. Eighteen literatures on the pathological and physiological mechanism of CVS after SAH were looked for the full text.DATA SYNTHESIS: Researches showed that remarkable changes occurred in potassium channels in CVS after SAH, while potassium channel modulators had significant interventional effect on CVS in animal experiment by promoting or blocking the ion movements.CONCLUSION: Potassium channels plays an important role in the pathogenesis of SAH-indueed CVS. Some therapeutic intervention may be provided based on the studies of potassium channels in CVS.
2.Professor SHI Yanping's Experience of the Treatment of Adenomyosis

[Objective] To introduce the Professor SHI Yanping's experience in treating adenomyosis with traditional Chinese medicine. [Methods]Through clinical learning with the teacher and systematic reading literatures about adenomyosis, the author summarized the teacher 's academic thoughts and clinical treating experience of the disease from the etiology and pathogenesis,fundamental law of treatment ,and medication characteristics,and one proven case was presented respectively.[Result] Professor SHI thinks the basic pathogenesis of adenomyosis is deficiency of kidney and blood stasis, accompanied by phlegm dampness, the proof is vacuity, treatment should be staged in terms of different periods:non menses-paying attention to eliminating mass and strengthening the body resistance, creatingNei Yi Ting Fang;menses-paying attention to regulating qi and blood by warming drugs, makingXin Gui Zhi Tong Fang;and at the same time,we have to pay attention to the adjustment of life style.All those lead to a good clinical effect. [Conclusion] The rich experience of Professor SHI in treating this disease might be worthy of promotion and learning,which has a good effect on relieving the discomforts and improving the quality of life in patients.
3.Correlation of serum homocysteine,vitamin B12 and folate levels and subclinical hypothyroidism in pregnancy

Objective To analysis the correlation of Serum homocysteine ,vitamin B12 and folate levels and subclinical hypot‐hyroidism in pregnancy .Methods 150 cases of subclinical hypothyroidism during pregnancy were selected as the experimental group ,and 150 cases of pregnant women with normal thyroid function were collected as the control group .Compared the serum thy‐roid‐stimulating hormone ,free thyroid hormones ,three free triiodothyronine ,homocysteine ,serum vitamin B12 ,folic acid levels of pregnant women in the two groups ,and analyzed the related serum markers of thyroid‐stimulating hormone and serum homocys‐teine .Results The homocysteine levels in experimental group was higher than that of control group(P<0 .05) ,the vitamin B12 and folate levels in experimental group were lower than that of control group (P<0 .05) ,the thyroid‐stimulating hormone levels in ex‐perimental group was significantly higher than that of control group (P<0 .05) .Serum thyroid‐stimulating hormone and homocys‐teine levels were positively correlated (P<0 .05) ,serum thyroid‐stimulating hormone and vitamin B12 and folic acid had no signifi‐cant correlation (P>0 .05) .Serum homocysteine and thyroid stimulating hormone levels were positively correlated (P<0 .05) ,ser‐um homocysteine and folate levels were negatively correlated (P<0 .05) ,serum homocysteine levels and vitamin B12 were not corre‐lated (P> 0 .05) .Conclusion Subclinical hypothyroidism during pregnancy have some relevance with homocysteine ,vitamin B12 and folate levels .
4.Nutlin-3 promotes pyroptosis in SMMC-7721 cells

Objective:To evaluate the effect of Nutlin-3,a MDM2 antagonist,on the pyroptosis in SMMC-7721 cells.Methods: The expression of actived caspase-1(p20) and IL-1β was detected using Western blot analysis.Pyroptosis was investigated by a standard lactate dehydrogenase release assay(LDH).IL-1β content in the cell culture supernatant was quantified by ELISA.Results: Nutlin-3 up-regulated the expression level of actived caspase-1 and IL-1β in SMMC-7721 cells.Meanwhile,Nutlin-3 increased significantly the content of LDH and IL-1β in the cell culture supernatant(P<0.05).Conclusion: Nutlin-3 activated pro-caspase-1,promoted pyroptosis and IL-1β release in SMMC-7721 cells.
5.Study on the assessment of carotid plaque stability by Ktrans of dynamic contrast material-enhanced magnetic resonance imaging in elderly patients treated with statin

Objective To investigate the efficacy of statin on carotid plaque stability in elderly patients assessed by K trans (the combination of E and F) of dynamic contrast material enhanced (DCE) magnetic resonance (DCE MR) imaging.Methods 37 elderly patients with carotid artery of low echo or mixed echo plaque in carotid artery patches were screened by B type ultrasound.According to the standard of 2011 ESC/EAS guidelines,all patients were divided into two groups:normal level of LDL-C group achieving LDL-C target and high level of LDL C group not achieving target of LDLC.Plaque area and size of lipid-rich necrotic core were measured by 3.0T enhancement magnetic resonance,and K trans was measured by DCE-MR dynamic contrast material enhanced (DCE) magnetic resonance (MR) imaging.and serum hs-C-reactive protein level was detected within 1 week.Results Plaque area,size of lipid-rich necrotic core,ratio of lipoid core over plaque area,K trans and serum level of hs-CRP were less in normal LDL-C level group than in high LDL-C level group [(2.06±0.45) mm2 vs.(3.63±0.62) mm2,(0.52±0.05) mm2 vs.(1.49±0.01) mm2,2 cases (11.8%) vs.14 cases (70.0%),(0.041±0.009) min1 vs.(0.079±0.011) min-1,(1.60±0.27) mmol/L vs.(2.80 ± 0.34),all P<0.05].Linear regression analysis revealed that there was no significant correlations of K trans with the size of lipid necrotic core (r=0.19,P>0.05) and hs-CRP (r=0.23,P>0.05).Conclusions Dynamic contrast material enhanced (DCE) magnetic resonance (MR) imaging is a quantitative method assaying atheromatous plaque components; K trans may be a new indicator to measure the stability of plaques; Statin can stabilize plaques through inhibiting the proliferation of Vasa vasorum in plaque angiogenesis,anti-inflammatory and reducing the size of lipidrich necrotic core.
6.Silenced NgR gene expression by RNA interference to promote rats facial nerve regeneration in vitro.

OBJECTIVE:
To suppress NgR gene expression in neural stem cells and observe differentiation of neural stem cells in vitro after interfered which provide nutritional support for the facial nerve repair in vivo.
METHOD:
PCR amplification, restriction endonuclease digestion, T4DNA ligase connections were used to connected NgR with rector pGCsi, and constructed recombinant vector (NgR shRNA). Lipofectamine 2000 were used to transfect the NSC. The expression of NgR was examined by Western Blot. The proportion of neural stem cells transformed into neurons after transfection was tested by Immunocytochemistry. Neural stem cells were planted in PLGA tubes after transfected, and were scanned by electron microscopy.
RESULT:
NgR shRNA plasmid was constructed and infected neural stem cells successfully. Western Blot showed that the expression of NgR decreased in neural stem cells after interference. Immunocytochemistry showed that the rate of the neural stem cells transformed into neurons after interfered was significantly higher (P < 0.01).
CONCLUSION
Neural stem cells were transformed into neurons after NgR shRNA plasmid infected neural stem cells, which promoted axonal regeneration more effectively and provided a efficient and stable gene platform for facial nerve repair.

7.Comparison of Q-value guide LASIK and standardized LASIK for the treatment of myopia:a meta-analysis

Background Nowadays,customized ablation is widely used in the excimer laser corneal refractive Burgery.And the Q-value guide LASIK is one of the research hotspots. Objective Present study was to evaluate and compare the effectiveness of Q-value guide LASIK and standardized LASIK for myopia.Methods A systematic literature retrieval from 2003 through 2010 was conducted in the MEDLINE,CNKI,Cochrane Library,EMBASE.The literature examine possible difierences in uncorrected visual acuity(UCVA),Q-value,higher order aberrations(HOAs),and spherical equivalent(SE) between Q-value guide LASIK and standardized LASIK for correcting myopia.Statistical analysis was performed using a Review Manager 5.0 software.The data was extracted,and the methodological quality was evaluated by two reviewers independently.The quality of included literature was scored according to the Jadad Scale. Results A total of 14 studies involving 1617 patients(2956 eyes)was included in the meta-analysis.Of these fourteen studies,six studies were randomized trials and the other eight studies were retrospective cohort studies.According to the Jadad Scale,2 studies scored 3 points,and the other 12 studies scored 1~2 points.The results showed that the Q-value guide LASIK group had a better postoperative effectiveness in uncorrective visual acuity(UCVA)(WMD=0.04.95% CI 0.00 to 0.08,P<0.05).Q-value(SMD=-1.52,95% CI -2.23 to-0.81,P=0.00),total HOAs (SMD=-1.63,95% CI-2.57 to-0.69,P<0.05) and spherical-like aberrations ( SMD =-1.49,95% CI-2.22 to-0. 76,P<0. 00) after surgery. However, the number of eyes achieved UCVA≥20/20 ( OR= 1.16,95% CI 0. 61 to 2. 19,P=0. 65) ,coma-like aberrations (SMD=-1. 02,95% CI-0. 36to 0.11,P=0.29) and SE (WMD=0.10,95% CI-0.11 to 0.31,P=0.34) after surgery were comparable.Conclusion The effectiveness of Q-value guide LASIK is superior to standardized LASIK for treatment of myopia.High-quality clinical randomized-controlled study should be performed to further evaluate the comparable outcome of Q-value guide LASIK with standard LASIK.
8.Sress cardiomyopathy:clinical features and imaging findings

Objective One typical case with stress cardiomyopathy was reported and the current knowledge of the syndrome was reviewed to improve relevant knowledge.Methods A 71-year-old female patient presented dyspnea and chest pain due to emotional stress.ECG,echocardiography,selective coronary artery angiography,left ventriculography,~(99)Tc~m-MIBI single photon emission computed tomography (SPECT),~(18)F-FDG SPECT and MRI were performed.Results Electrocardiogram at admission showed ST segment elevation and T wave inversion in leads V1—V4.Pathological Q wave occurred 1 week later,it disappeared 1 month later however and severe T wave inversion occurred.Normal or slightly elevated cardiac enzymes in the blood were found during the course.Left ventriculogram at admission showed left ventricular apical ballooning with LVEF of 30%.The ballooning volume was about 3/4 of left ventricular volume, without any corresponding coronary artery diseases found in coronary angiogram.The abnormal apical ballooning decreased significantly in the follow-up left ventriculogram performed one month later.The LVEF rose up to 63.6%.~(99)Tc~m-MIBI and ~(18)F-FDG SPECT showed mismatch of perfusion and metabolism in the corresponding region,indicating presence of viable myocardium.MRI showed left ventricular apical ballooning without perfusion defect and late enhancement,indicating viability of corresponding myocardium. Conclusions Emotional stress can cause transient left ventricular apical ballooning called"stress cardiomyopathy".Either ~(99)Tc~m-MIBI SPECT associated with ~(18)F-FDG SPECT or delayed enhancement MRI plays an important role in identification of myocardial viability,which can efficiently guide clinical treatment.
10.Preliminary study of odor change mechanism in Crataegi fructus stir-fried process based on correlation analysis.

In order to investigate the mechanism, the correlation between the odor change in Crataegi Fructus stir-fried process and 5-HMF were studied. Required samples were retrieved from Crataegi Fructus stir-fried process. Statistical quality control (SQC) was used to analyze the response values acquired by the electronic nose. At the same time, the content of 5-HMF was detected by high performance liquid chromatography (HPLC). Correlation analysis was used to analyze the relationship between the above two. Experimental results showed that SQC model established by response values of all samples could show the change law of odor in Crataegi Fructus stir-fried process and changes of 5-HMF content was dropped after the first increase. Correlation analysis showed that the odor change in Crataegi Fructus stir-fried process and 5-HMF were significantly correlated (P < 0.05). Sugar degradation reaction and the Maillard reaction may be one of the mechanisms of the odor change in Crataegi Fructus stir-fried process.
