1.Optimizing the Extracting Process in Diedahuoxue Film With Uniform Design

AIM:To seek a simple and reliable process for extracting effective components in Diedahuoxue film.METHODS: According to uniform design, five factors in extraction were optimized, i.e. ethanol concentration, reflux time, reflux times,ethanol volume and drug particle. The absorption value was taken as evaluating parameter in UV- spectrophotometry. RESULTS: The results showed that the best extracting conditions were 95% ethanol, ten times amount of ethanol, 0.2mm size of particle and reflux time of 1h with 4 repetitions.CONCLUSION:This process is simple and reliable.
2.Analysis of 65 Cases of Ciprofloxacin Lactate-induced Adverse Reactions

OBJECTIVE:To investigate the characteristics of ARDs induced by ciprofloxacin lactate.METHODS:Using philometric method,65 cases developing drug adverse reactions to ciprofloxacin lactate,reported in domestic journals during the past 2 years,were analysed.RESULTS:Of the ARDs in 65 cases,40 were allergic reactions(61.5%),13 reactions of nervous system(12.3%),3 kidney function damage(4.6%),1 liver function damage,1 cardiovascular reaction(1.5%) and 6 reactions of urinary system.CONCLUSION:The ARDs should be under consideration in choice of ciprofloxalin lactate.
3.Clinical study of immediate intravesical instillation combined with short-term maintained intravesical instillation chemotherapy of pirarubicin after operation for preventing postoperative recurrence of superficial bladder tumor

Objective To investigate the efficacy and safety of immediate intravesical instillation combined with short-term maintained intravesical instillat.ion chemotherapy of pirarubicin (THP) after operation for preventing postoperative recurrence of superficial bladder tumor.Methods One hundred and seven patients with superficial bladder tumor were divided by random digits table method into two groups:group A (50 cases ) was treated with immediate intravesical instillation combined with short-term maintained intravesical instillation chemotherapy after operation,and group B (57 cases) was treated with conventional intravesical instillation chemotherapy.Patients in group A underwent intravesical instillation of THP within 24 hours,maintained intravesical instillation once a week for 8 weeks.Patients in group B underwent intravesical instillation of THP in one or two weeks after operation,once a week for 8 weeks.And then once a month for 10 months.All patients were followed up with regular cystoscopy examination.Results Patients were followed up for 13-84 months,the incidence of partial vesical toxic reaction in group A was 26.00 % (13/50),general symptom rate was 4.00% ( 2/50 ),the rate of recurrency was 16.00% ( 8/50 ).The incidence of partial vesical toxic reaction in group B was 22.81%( 13/57 ),general symptom rate was 1.75%( 1/57 ),the rate of recurrency was 19.30% (11/57).There was no significant differences between two groups (P > 0.05 ).Conclusions Immediate intravesical instillation of THP after operation is safe and effective,but it is not suitable for patients with obvious bladder perforation and large wound surface of bladder mucosa after transurethral rescection.Immediate intravesical instillation combined with short-term maintained intravesical instillation chemotherapy of THP after operation for preventing postoperative recurrence of superficial bladder tumor is effective and well tolerated.
5.Frequency of tibial plateau fracture combined with dislocation and imaging features of medial or posteromedial fragments

Objective To evaluate the incidence and imaging features of tibial plateau fracture combined with dislocation.Methods A total of 298 patients with intact imaging data treated for tibial plateau fracture from December 2012 to June 2014 were analyzed retrospcctively.Tilt angle of medial fragment fracture line,ratio of width of medial fragment to total plateau,surface area percentage,fragment height,and major displacement were measured on CT images using picture archiving and communication system (PACS).Results Eighteen patients (6.0%) had tibial plateau fractures combined with dislocation,with tilt angle of medial or posteromedial fragment fracture line of (62.1 ± 14.1)°.Eleven patients (61%) had split of the medial tibial plateau with surface area percentage of (46.3 ± 2.5) %,fragment height of (43.9 ± 6.2) mm,and major displacement of (5.3 ± 2.1) mm.Seven patients (39%) had posteromedial fragment fracture with surface area percentage of (25.1 ± 5.7) %,fragment height of (40.0 ± 10.1) mm,and major displacement of (4.8 ± 6.45) mm.Conclusions Fracture line of fracture-dislocation tibial plateau fracture is much often through the eminence or located at the lateral eminence with large medial or posteromedial fragments.Surgical approaches and directions of nailing should be considered properly according to the morphological characteristics.
6.Research on Effect Factors to Function of Human-machine System as Displaying with LCD.

Objective To provide the experimental data for standard establishment and ergonomics research on manned spaceflight displays basing on relation of six effect factors to function of human-machine system as displaying with LCD,and the best factor-level-composition is chosen.Methods An experimental system was developed.The orhoganal experiment means was adopted.Twenty subjects were asked to finish three tasks: to view parameters of the whole screen interface,to seek local parameters,to identify the fault events quickly.Three indices: reaction time,error times and times of unable to recognize were analyzed.Results The indices were much different for different factors at different levels.According to the indices of mean reaction time,extreme difference of view-angle was the lowest and view-distance the highest.Conclusion The order of the display effects of different factors on human-machine system is successively: viewdistance,illuminant level,character height,color of character/background and viewangle.
7.Changes in corneal nerve fiber length and density before and after eye opening in mouse and its significance

Background Recently,there were many studies on corneal innervations during mammalian development.However,there were fewer studies on discussing corneal innervations before and after mouse eye openings.Objective The present study was to investigate the change in the regulation of corneal nerve fiber length and density before and after mouse eye openings to offer a basis for clinical research in human.Methods Thirty SPF C57BL/6 mice were divided into postnatal 1 day(P1 d),P7 d,P13 d(1 day before eye opening),P14 d(eye halfopened),P17 d(1 day after eye opening)and P23 d(7 day after eye opening)groups,with 5 mice and 10 eyes for each group.Entire corneal stretches were prepared and immunostaining with an anti-neuron-specific β-Ⅲ tubulin antibody was performed to label the corneal nerve fibers.Confocal microscopic pictures from the corneal dorsal-nasal region (DN),dorsal-temporal(DT),ventral-nasal region(VN)and ventral-temporal(VT)were taken using Delta Vision Core.From these pictures,the mouse corneal area,total length and density of nerve fibers in the 4 regions were calculated.The use of the animals complied with Statement of ARVO.Results Corneal areas of P1 d,P7d,P13 d,P14 d,P17 d and P23 d mice were(0.404±0.007),(1.362±0.154),(1.573±0.080),(1.603±0.046),(1.847±0.052),(2.445±0.798)mm2,respectively ; the total lengths of nerve fibers were(3.718±1.044),(19.065±3.350),(23.687±0.907),(27.309±2.477),(31.989±3.976),(41.214±1.573)mm,respectively ; the densities of nerve fibers were(9.592±1.138),(14.506±1.908),(15.088±1.241),(16.772±1.897),(16.821±2.102),(17.660±1.216)mm/mm2,respectively,all showing significant increases with age(F =22.906,P =0.000 ; F =0.424,P =0.000 ; F =2.375,P=0.000).A positive correlation of the increasing corneal areas and increasing lengths of nerve fibers was found(r=0.983,P<0.01).Nerve fiber densities in the four corneal regions significantly increased with age(DN region:F =0.159,P =0.000 ; DT region:F =2.1 72,P =0.001 ; VN region:F =1.998,P =0.000 ; VT region:F=2.352,P=0.000).From P13 d to P14 d,the corneal nerve fiber densities in the DN region decreased by 6.0% without significant difference(t =0.589,P =0.572); and the corneal nerve fiber densities in the DT region,VN region and VT region decreased by 4.6%,5.5% and 0.1%,respectively,without significant difference from P14 d to P17 d(t=0.549,P=0.596;t=0.701,P=0.501 ;t=-0.100,P=0.919).Conclusions The development of nerve fibers in the whole cornea or the four corneal regions is influenced by eye opening in mouse to various extents.From P13 d to P14 d,the corneal nerve fiber densities in the DN region decreased by 6.0% without significant difference.From P14 d to P17 d,the corneal nerve fiber densities in the DT region,VN region and VT region decrease by 4.6%,5.5% and 0.1%,respectively,without significant difference.Afterwards,the growth of nerve fibers increased in pace and the growth rate is recovered.
8.Evaluation on effect of rosuvastatin in treatment of lower extremity atherosclerotic disease in elderly

Objective To observe the clinical effect of rosuvastatin in the treatment of lower extremity atherosclerotic disease in the elderly,and to provide clinical basis of the application of statins in lower extremity atherosclerotic disease in the elderly.Methods 90 patients diagnosed as lower extremity atherosclerotic disease (aged 60 years or over)were randomly divided into simvastatin group and rosuvastatin group (n=45),and they were treated with simvastatin and rosuvastatin,respectively,based on the regular treatment.The total cholesterol (TC),low density lipoprotein-cholesterol (LDL-C), and high sensitivity C-reactive protein (hs-CRP)of the patients in two groups were detected at the begining of treatment and the ending of 32 weeks;at the same time,the lower limb artery intima thickness, strength of hardened, plaque, and artery stenosis degree scores were compared.Results 32 weeks after treatment,the levels of TC,LDL-C and hs-CRP of the patients in two groups were decreased statistically compared with before treatment (P <0.05);the levels of TC,LDC-C,and hs-CRP in rosuvastatin group were lower than those in simvastatin group (P < 0.05 ). 32 weeks after treatment in two groups,compared with before treatment,the lining thickness and plaque,stenosis scores were decreased (P <0.05);the lining thickness,plaque and stenosis scores in rosuvastatin group were lower than those in simvastatin group (P <0.05);the gardening strength grades had no statistical difference between before and after treatment. Conclusion Statins is lipid-lowering and resistant to lower limb arteriosclerosis,and rosuvastatin is better than simvastatin.
9.Surgical treatment of lumbosacral tuberculosis by anterior debridement,bone graft,and posterior pedicle screw-rod internal fixation.

OBJECTIVETo investigate the effectiveness of anterior debridement,bone graft, and posterior pedicle screw-rod internal fixation in the treatment of lumbosacral tuberculosis.
METHODSFrom January 2005 to June 2012,18 patients with lumbosacral tuberculosis undergoing the surgical treatment by anterior debridement, bone graft, and posterior pedicle screw-rod internal fixation were reviewed retrospectively. There were 12 males and 6 females with an average age of 44 years old ranging from 35 to 67. Among them, 2 cases were simple low back pain,3 cases were low back pain combined with radiating pain of lower extremity and 13 cases were muscle strength hypesthesia. According to Fankle grading,the nerve function was grade C in 3 cases,grade D in 10 cases,grade E in 5 cases. Of these cases,L4.5 was involved in 8 cases,L5 in 4 cases and L5S1 in 6 cases. The lumbosacral angle was 150 to 270 and the erythrocyte sedimentation rate (ESR) was 45 to 93 mm/h before treatment. The clinical indexes including the lumbosacral incidence, Frankel grade and ESR were reviewed at follow-up.
RESULTSAll 18 cases were followed up for 14 to 22 months. The mean operation time was 180 min. The amount of bleeding was 400 to 800 ml. Except 1 case with iliac vein injuried and 4 cases with abdominal distension,no spinal injuries and severe relative complication occurred, and neurologic function improved in various degrees. Pain in lumbosacral area and radicular pain in lower extremities disappeared. The X-ray and CT films demonstrated that bony fusion was obtained in all patients during 9 to 13 months postoperatively. The lumbosacral angle and ESR were improved significantly.
CONCLUSIONAnterior debridement, bone graft, and posterior pedicle screw-rod internal fixation is an effective method in dealing with lumbosacral spine tuberculosis.

10.Curcumin induces apoptosis by PTEN/PI3K/AKT pathway in EC109 cells.

OBJECTIVETo study the molecular mechanism of curcumin in human esophageal carcinoma cell line (EC109).
METHODSEC109 cells were cultivated in vitro. When 80%-90% confluence was reached, they were treated with curcumin in different concentrations (15-120 µmol/L). The effects on cell proliferation were examined by CCK-8 colorimetry. The ultrastructure of EC109 cells were detected with transmission electron microscope(TEM). The cells apoptosis was observed with laser confocal microscope(LCM) by AnnexinV-FITC/PI double staining. The proteins level of PTEN, AKT, GSK3β and Caspase 3 were tested by flow cytometry(FCM) .
RESULTSCCK-8 test showed that curcumin could inhibit the proliferation of EC109 cells in a time- and concentration-dependent manner. TEM and LCM examinations indicated that curcumin could make EC109 cells apoptosis. The data of FCM showed that curcumin could increase the expression of PTEN, GSK3β and Caspase 3, decreased the expression of AKT.
CONCLUSIONThe effects of curcumin on inhibiting proliferation and promoting apoptosis of EC109 cells were related with increased expression of PTEN and inhibition of PI3K/AKT signaling pathway.
