1.Relationship between insulin sensitivity index and extent of coronary atherosclerosis in Uygur and Han nationality patients

Objective: To explore the relationship between insulin sensitivity index (ISI)and extent of coronary atherosclerosis in Uygur and Han nationality patients. Methods: A total of 125 patients with coronary heart disease were enrolled, including 59 Uygur nationality patients and 66 Han nationality patients. There were 42 cases with stable angina pectoris, 52 cases with unstable angina pectoris, 31 cases with old myocardial infarction, 53 cases with type 2 diabetes and 35 cases with hypertension. Relevant medical history of patients [age, body mass index (BMI) etc.] was collected and laboratory examination (blood lipid, fasting blood glucose, fasting insulin etc.)were performed, and extent of coronary atherosclerosis was evaluated by coronary angiography in all patients. ISI was calculated and its relationship with extent of coronary atherosclerosis was analyzed. Results: In both nationality patients, ISI was negatively correlated with extent of coronary atherosclerosis (Han nationality: r=-0.71, P=0.03, Uygur nationality: r=-0.52, P=0.04). ISI was negatively correlated with BMI, glycosylated hemoglobin (HbA1c) and low density lipoprotein –cholesterol (LDL-C) (r=-0.45, -0.68, -0.55, P<0.05 all) in Han nationality patients; ISI was positively correlated with high density lipoprotein–cholesterol (HDL-C, r=0.67, P<0.01), and negatively correlated with HbA1c, LDL and lipoprotein a (r=-0.47, -0.35, -0.42, P<0.05) in Uygur nationality patients. Conclusion: Insulin resistance participates and promotes formation and development of coronary atherosclerotic plaques. Controlling weight, blood glucose and LDL-C levels etc. can decrease insulin resistance and prevent occurrence and development of coronary heart disease.
2.Therapeutic effects of endoscopic mucosal resection on gastric polyps and colorectal polyps

Objective To investigate therapeutic effects of endoscopic mucosal resection( EMR) on gastric polyps and colorectal polyps.Methods 172 cases of elderly patients with early gastric and colorectal polyps were col-lected.They were divided into the observation group and control group by random number table,each group 86 cases. The observation group were treated with EMR,the control group were received conventional treatment for surgery.The clinical efficacy and adverse events were compared.Results The total effective rate of the observation group was 96.51%,significantly higher than 70.93% in the control group (χ2 =15.73,P<0.05);the postoperative adverse reactions in the observation group was 22.09%,significantly lower than 47.67% in the control group (χ2 =31.65, P<0.05).Conclusion For patients with pre-senile and senile stomach and colon polyps,the application of endo-scopic mucosal resection in the treatment not only can achieve better clinical efficacy, but also lower incidence of adverse reactions.
3.Analysis of Urinary Metabonomics Pathway in Rats with Chronic Glomerulonephritis Intervened by Tannins from Pericarpium Granati

Objective: To analyze the metabolicomics pathway in the rats with chronic glomerulonephritis intervened by tannins from Pericarpium Granati.Methods: Signal pathway analysis was carried out using the KEGG database and molecular metabolite annotation was performed using HMDB database.The enzyme or transporter and its related properties were analyzed.The metabolite path visualization was carried out by using MetPA network software.Results: The analysis of biological metabolism pathway showed that 12 metabolites involved in 16 metabolic pathways.The pathway of tryptophan metabolism, citrate cycle, phenylalanine metabolism, cysteine and methionine metabolism showed notable changes (P<0.05).Conclusion: The changes of phenylalanine metabolism, citrate cycle, tryptophan metabolism, cysteine and methionine metabolism in the rats with chronic glomerulonephritis intervened by tannins from Pericarpium Granati participate the pathological process.
4.Effect of sufentanil on anoxia/reoxygenation-induced injury to H9c2 cells with high-glucose incubation

Objective To investigate the effect of sufentanil on anoxia/rexogenation (A/R)-induced injury to H9c2 cells incubated in high-glucose culture medium. Methods The H9c2 cells were cultured in low-glucose DMEM/F12 culture medium supplemented with 10% fetal bovine serum. The cells were seeded in 96-well or 6-well plates and randomly divided into 5 groups ( n = 12 wells each): normal glucose control group (group NC),high-glucose control group (group HC), high-glucose + A/R group (group HA/R), high-glucose + sufentanil +A/R group (group HSA/R), high glucose + naloxone + A/R group (group HNA/R). The cells were exposed to 95 % N2-5 % CO2 in an incubator at 37 ℃ for 3 h followed by 3 h reoxygenation. In group NC, the cells were incubated in low-glucose culture medium for 48 h. In group HC, the cells were incubated in high-glucose culture medium for 48 h. In group HA/R, the cells were incubated in high-glucose culture medium for 48 h before anoxia.In group HSA/R, after the cells were incubated in high-glucose culture medium for48 h, sufentanil was added to the culture medium with the final concentration of 10-9 mol/L at 15 min before anoxia. In group HNA/R, after the cells were incubated inhigh-glucose culture medium for 48 h, naloxone was added to the culture medium with the final concentration of 10-6 mol/L, 10 min later sufentanil was added with the final concentration of 10-9 mol/L at 15 min before anoxia. The cell viability was measured by MTT assay. The amount of lactic dehydrogenase (LDH) released in the supernatant and superoxide dismutase (SOD) activity were measured. Results The cell viability and SOD activity were significantly lower, while the amount of LDH released was significantly higher in the other groups than in group NC, in groups HA/R and HNA/R than in group HC, and in group HNA/R than in group HSA/R (P < 0.01 ). The cell viability and SOD activity were significantly higher, while the amount of LDH released was significantly lower in group HSA/R than in group HA/R ( P < 0.01 ). There was no significant difference in the parameters mentioned above between group HNA/R and group HA/R ( P > 0.05). Conclusion Sufentanil can attenuate A/R-induced injury to H9c2 cells with high-glucose incubation, and the mechanism may be related to the activation of opioid receptors.
5.Expressions of vascular endothelial growth factor-C,-D and vascular endothelial growth factor receptor-3 correlates with lymph node micrometastasis of patient with pN0 early gastric cancer

Objective To investigate the expressions of vascular endothelial growth factor (VEGF)-C,-D and VEGF receptor-3 (VEGFR-3/Flt-4) in pN0 early gastric cancer (ECG) and their relationship with lymph node micrometastasis.Methods From January 2005 to January 2010,the paraffin-embedded specimens of 61 pN0 ECG were collected.The expressions of VEGF-C,VEGF-D and VEGFR-3 in 61 pN0 early gastric cancer tissue,25 adjacent tissue and CAM5.2 expression in 868 hematoxylin-eosin staining negative lymph nodes were detected by immunohistochemical assay.The rate of lymph node micrometastasis of 61 pN0 ECG was evaluated.According to the data type,t test,x2 test or Fisher exact probability were performed for the relationship analysis between the expressions of VEGF-C,VEGF-D and VEGFR-3 in pN0 stage ECG and lymph node micrometastasis.Results The high expressions of VEGF-C,VEGF-D and VEGFR-3 in the 61 pN0 ECG were 34.4% (21/61),34.4% (21/61) and 44.3% (27/61) respectively,which were higher than those of adjacent tissues [12.0% (3/25),8.0% (2/25) and 16.0% (4/25) respectively] (x2=4.433,6.321 and 6.144 respectively,all P<0.05).There were 10 cases (16.4%) of pN0 ECG with lymph node micrometastasis.In pN0 ECG,the low expressions of VEGF-C and VEGFR-3 in negative Flt-4 vascular invasion (FVI) were common than positive FVI (x2 =15.828,6.879 and 9.244,all P<0.05).The high expressions of VEGF-C and VEGFR-3 were related to the depth of tumor invasion (x2 =5.561 and 5.678,both P<0.05),the density of VEGFR 3 positive vascular (FVD) (t=2.987and 5.652,both P<0.01) and lymph node micrometastasis (x2 =6.705 and 6.192,both P<0.05),but not related to the degree of differentiation (P>0.05).However,the high expression of VEGF-D was not related to depth of tumor invasion,FVD and lymph node micrometastasis (all P>0.05),but related to the degree of differentiation (x2 =8.472,P =0.004).The high expressions of VEGF-C,VEGF-D and VEGFR-3 were not related to gender,age,tumor location,macroscopic type and tumor size (all P>0.05).Conclusions The high expressions of VEGF-C,VEGFR-3 were related to lymph node micrometastasis in pN0 ECG.Although the high expression of VEGF-D was not related to lymph node micrometastasis,it could indirectly affect lymph node micrometastasis through VEGF-C-VEGFR-3 axis in pN0 ECG.
6.Argon-helium cryoablation combined with transcatheter arterial chemoembolization for the treatment of advanced hepatocellular carcinoma:analysis of therapeutic effectiveness

Objective To evaluate argon-helium cryoablation combined with transcatheter arterial chemoembolization (TACE) in treating advanced hepatocellular carcinoma (HCC). Methods The clinical data of 66 patients with pathologically-proved HCC were retrospectively analyzed. Based on the therapeutic scheme the patients were divided into TACE group (n=31) and combination group (TACE+argon-helium cryoablation, n=35). All the patients were followed up for 5-35 months. The complete remission rate, total effective rate and survival time were evaluated. The short-term and the long-term effectiveness were compared between the two groups. Results Both the complete remission rate and total effective rate of the combination group were significantly higher than those of TACE group (P<0.05). The median survival time of the combination group was significantly longer than that of TACE group (P=0.038). The half-year, one-year and 2-year overall survival rates of the combination group were higher than those of TACE group, although the differences were not statistically significant (P>0.05). Conclusion For the treatment of advanced hepatocellular carcinoma, argon-helium cryoablation combined with TACE can improve the short-term effect and prolong the progression-free survival time, although its exact effectiveness still needs to be confirmed by large sample, multi-central and randomized controlled studies.
7.Comparison of dosimetry and toxicities between postoperative fixed-field intensity-modulated radiotherapy and image-guided radiation therapy/volumetric modulated arc therapy for cervical cancer

Objective To compare the dosimetry and toxicities between postoperative fixed-field intensity-modulated radiotherapy (FF-IMRT) and image-guided radiation therapy/volumetric modulated arc therapy (IGRT-VMAT) for cervical cancer.Methods A total of seventy patients with stage I b-Ⅱa postoperative cervical cancer who had high risk factors,were divided into FF-IMRT (FF-IMRT group,n =35)and IGRT-VMAT (IGRT-VMAT group,n =35),to compare the difference of target dose and adverse reaction between the two groups.Results In the IGRT-VMAT group,the interfractional setup errors in the x,y,and z axes were (0.25±0.14) cm,(0.26±0.16) cm,and (0.24±0.18) cm,respectively;the intrafractional setup errors in the x,y,and z axes were (0.1±0.09) cm,(0.12±0.09) cm,and (0.11±0.09) cm,respectively;the margins in the x,y,and z axes were 0.75 cm,0.84 cm,and 0.78 cm,respectively.Under the same dosimetric conditions,the IGRT-VMAT group was superior to the FF-IMRT group in terms of conformity index,treatment time,and number of monitor units (P=0.000).The Dmean and volume receiving high-dose irradiation for the bladder,rectum,and small intestine were significantly lower in the IGRT-VMAT group than in the FF-IMRT group (P=0.000).Compared with the FF-IMRT group,the IGRT-VMAT group had a significantly reduced incidence of acute and chronic gastrointestinal,urinary,and hematologic toxicities (P< 0.05).Conclusions IGRT-VMAT can correct setup error online,shorten the treatment time,reduce the dose to organs at risk,and alleviate acute and chronic toxicities,and is especially suitable for patients with postoperative small bowel position changes.
8.MicroRNA-10a promotes granulosa cells tumor development

OBJECTIVE To investigate the effect of microRNA-10a on the development of granulosa cells tumor (GCT). METHODS FISH was used to detect the miR-10a expression in tissues from GCT patients. Several functional assays were performed to investigate the effect of miR-10a on proliferation,migration, invasion, spheroid formation and repressed anticancer drug-induced apoptosis of GCT in vitro.CRISPR- Cas9 system mediated miR- 10a knockout in cancer GC and two mice GCT models wereconstructed to show the knockdown effect of miR-10a on cancer GC both in vitro and in vivo. RNA-seq,Western blot, luciferase reporter assay and FISH were used to identify potential direct functional targets and related pathways of miR-10a in cancer GC. RESULTS Strong miR-10a signal was detected in tissues from malignant GCT patients. And amplification of miR- 10a negatively correlated with overall survival rate of ovarian cancer patients. In addition, ectopic expression of miR- 10a significantly promoted cell proliferation, migration, invasion, spheroid formation and repressed anticancer drug-induced apoptosis in vitro. CRISPR-Cas9 system mediated miR-10a knockout in cancer GC showed opposite phenotype compared to miR-10a overexpressed cancer GC. By using xenograft and orthotropic models, the onco?genic role of miR-10a was further confirmed in vivo. RNA-seq, Western blot, luciferase reporter assay and FISH were used to identified PTEN/TET2 as direct functional targets of miR-10a in cancer GC; Akt and Wnt were found as two associated signaling pathways of miR- 10a in cancer GC. CONCLUSION Taken together, our results demonstrate that the miR-10a is positively involved in development of GCT.
9.Influence of Sanguisorba Officinalis Tannins Extract on Proliferation of Human Renal Tubular Epithelia Cells Induced by TGF-β1

Objective:To study the preventive effect of Sanguisorba officinalis on renal fibrosis by observing the influence of San-guisorba officinalis tannins extract (STE) on the proliferation of human renal epithelia (HK-2) induced by transforming growth factor-β1 ( TGF-β1 ) . Methods:HK-2 cells were cultured in DMEM medium with high glucose containing 10% fetal bovine serum. The cul-tured cells were divided into 5 groups, including the blank control group, TGF-β1 group (5 ng· ml-1 TGF-β1), intervention group 1 (5 ng·ml-1 TGF-β1 +12. 5μg·ml-1 STE), intervention group 2 (5 ng·ml-1 TGF-β1 +25μg·ml-1 STE) and intervention group 3(5 ng·ml-1 TGF-β1 +50 μg·ml-1 STE). The changes of cell morphology were observed under an inverted microscope and the in-fluence of SET on the cell proliferation was detected by CCK8 assay. Results: TGF-β1 could significantly induce the proliferation of HK-2 and promote the cell fibrosis with significant difference when compared with the control group (P<0. 05). However, after trea-ted with STE, the cell proliferation was inhibited obviously (P<0. 05) and the cells morphology tended to be normal in a dose-depend-ent manner. Conclusion:STE can inhibit the proliferation of HK-2 and prevent renal fibrosis to some extent.
10.The diagnosis value of using NBI-ME in serum CEA positive microvascular parting observation of gastric cancer and colorectal cancer

Objective To study clinical diagnosis value of using magnifying endoscopy with narrow-band imaging(NBI-ME) in the CEA positive cases of microvascular parting to gastric cancer and colorectal cancer .Methods 873 cases in our hospital from 2012~2013 ,who had serum CEA positive were choosen ,the 114 patients who first diagnosis for suspected gastric cancer ,35 pa-tients who first diagnosis forsuspected colorectal cancer were included in this study .Using NBI-ME to check and the pathology bi-opsy .NBI-M E check key observation microvascular properties and classification :rules ,slightly irregular ,irregular ,disappeared ,the microvascular parting situation compared with pathological findings .Results 13 patients diagnosed with cancer by pathology ,the positive rate was 8 .72% ;intraepithelial neoplasia in 10 cases ,the positive rate was 6 .71% .No cases of cancer were regular and slightly irregular type ,all cancer cases in the disappearance .The sensitivity of type of disappeared was 100 .00% ,and the sensitivity of irregular type of canceration possibility intestinal large atrophy and high grade intraepithelial neoplasia was 87 .50% ,the differ-ence was statistically significant(P<0 .05) .Conclusion Using NBI-ME microvascular classification diagnosis could help observa-tion CEA positive cases in the screening of gastric cancer and colorectal cancer .
