2.Application of multislice CT scan combined with 99mTC -MIBI -SPECT in the diagnosis of isolation pulmonary nodule lesions

Objective To discuss the applied value of multislice CT scan in combination with 99 MTC -MI-BI -SPECT for the diagnosis of isolated pulmonary nodule lesions.Methods 60 patients with solitary pulmonary nodules shadow in the X -ray film from July 2014 to August 2015 in our hospital were selected and detected by spiral CT scanning and 99mTC -MIBI -SPECT imaging.Scan results and pathological examination or surgery were com-pared,specificity,sensitivity and accuracy of pulmonary nodules were concluded about the method in the differential diagnosis isolation.Results The specificity,sensitivity and accuracy of multislice spiral CT scanning alone were 73.6%,78.2%,79.3% respectively;The specificity,sensitivity and accuracy of 99mTC -MIBI -SPECT imaging alone,were 82.6%,78.2%,79.3% respectively;The specificity,sensitivity and accuracy of multislice CT scan com-bined with 99mTC -MIBI -SPECT imaging were 93.3%,93.2%,93.8% respectively.The accuracy of combined detection was significantly higher than that of 99mTC -MIBI -SPECT examination(χ2 =6.783,P =0.010)or CT scan (χ2 =8.012,P =0.008).Conclusion Multislice CT scanning combined with 99mTC -MIBI -SPECT imaging has better effect in diagnosing pulmonary nodule lesions,which is superior to the method alone.The combination meth-od can obviously improve the detection rate of lung nodule lesions,which is,worthy of further promotion.
3.The application effect of CT angiography features on qualitative and differential diagnosis of small nodules in the lung lesions

Objective To investigate the effect of CT angiography on the qualitative and differential diagnosis in small pulmonary nodules.Methods 120 patients with pulmonary small nodular lesions from September 2014 to October 2015 in our hospital were selected,and they were divided into the observation group and control group according to pathological type.the patients with malignant pathology were in the observation group,with totaling 64 cases;the patients with benign pathology were selected as a control group,with a total of 56 cases.Lesion type,sensitivity,vascular sign type,CT enhancement value and the ratio of enhancement value to aorta value in the two groups were compared and analyzed after the CT scan.Results In the observation group,positive CT angiography sign was 50 cases while negative ones was 14 cases,the sensitivity was 78.1%;In the control group,CT angiography sign was positive in 16 cases,negative in 40 cases,with the sensitivity of 28.6%.Comparing of two vascular symptoms sensitive case, there was a significant difference(χ2 =6.781,P =0.012).Vascular symptoms genotyping were compared in vascular symptoms positive patients,the results showed significant differences(χ2 =7.694,6.964,5.993,6.012,all P <0.05).Enhanced CT scan value of patients showed positive signs in the observation group was (40.07 ±3.72)Hu, with the control group of (10.54 ±4.83)Hu,the difference was statistically significant(t =11.783,P =0.019).The ratio of enhancement CT value to aorta value in patients with positive reaction showed 24.96% in the observation group,and 4.01% in the control group,the difference was statistically significant(χ2 =7.460,P =0.012).Conclusion
CT angiography features has played a very important role in qualitative and differential diagnosis of small nodular lesions in the lungs.It is of great significance in the clinical diagnosis and treatment,and is worth further promotion.
4.The effect of adenovirus-mediated ING4 expression on breast cancer cells

Objective To investigate the inhibitory effect of inhibitor 4 of growth(ING4)delivered by adenovirus on human breast carcinoma cell MDA-MB-231.Methods MDA-MB-231 human breast carcinoma cells were irfected with Ad-ING4.The expression level of ING4 gene was detected by RT-PCR and Western blot;The growth inhibition,cell-cycle alteration,and apoptosis were detected by MTT,Flowcytometry and Hochest33258 staining.respectively.RT-PCR was used to detect the transcription of Bax,Bc1-2,Survivin genes;The expression level of Ang-1 gene was detected by ELISA;Ad-ING4 was given intratumorally in athymic nude mice beating MDA-MB.231 tumors.and then tumor growth was monitored;The expression of Bc1-2,Bax and Caspase-3 was analyzed by immumohistochemistry.Results ING4 was successfully transcribed and translated iU MDA-MB-231 cells:Ad-ING4 significantly inhibited the proliferation and induced G_2/M phase arrest to(24.86±1.24)% and cell apoptosis of MDAMB-231.Intratumoral injection of Ad-ING4 suppressed the tumor growth obviously with a inhibitory rate of 49%;Immumohistochemistry showed that the expression of Bax,Caspase-3 were up-regulated and the expression of Bc1-2.Survivin,CD34 were down-regulated by Ad-ING4.Conclusions Ad-ING4 can inhibit the growth of MDA-MB-231 cells and induce apoptosis in vitro and in vivo.
5.Recombinant expression of human IL-17F fusion protein in E.coli and the study on its biological activity in vitro

Objective:To construct a recombinant prokaryotic expression vector pGEX-5X-3/hIL-17F and express it in E.coli and to explore the biological activities of human IL-17F fusion protein.Methods:The coding sequence of the mature human IL-17F(minus the signal peptide) was amplified from pUCm-T/hIL-17F by PCR and subcloned into the prokaryotic expression vector pGEX-5X-3 to express glutathione S-transferase(GST) fusion protein. The fusion protein was induced in E.coli BL21 by IPTG and purified by standard methods reported in prokaryotic system. The purified GST-hIL-17F fusion protein was identified by Western blot. The proliferation of ECV304 cells was observed by incubating them with soluble GST-hIL-17F fusion protein by MTT assay. The concentrations of IL-6, IFN-? and TNF-? in the supernatants of ECV304 cells were determined by ELISA. The effect of GST-hIL-17F on the angiogenesis of the chick chorioallantoic membrane was assessed by CAM assay.Results:A 41 kD fusion protein was effciently induced in E.coli BL21 by IPTG, accounting for about 55% of the total bacterial protein. The purified GST-hIL-17F fusion protein was identified by Western blot. GST-hIL-17F fusion protein had obvious biological activity to inhibit the proliferation of ECV304 cells and enhance IL-6 secretion. GST-hIL-17F had a marked antiangiogenic activity.Conclusion:The preliminary study of hIL-17F recombinant prokaryotic expression and its antiangiogenic effect has been successful, which lays a foundation for future research on the mechanism of antiangiogenesis and clinical application of recombinant hIL-17F protein.
6.Expression of human IL-24 gene in CHO cells and its anti-tumor effect in vitro

Aim To construct the eukaryotic expression vector of hIL-24 cDNA,and express it in CHO cells and detect its anti-tumor effect of recombinant hIL-24 protein.Methods Constructed pcDNA3-hIL-24 was identified by endonucleases digestion & PCR.The recombinant expression plasmids were transfected into CHO cells,human hIL-24 expressed in CHO cells was detected with RT-PCR.The apoptosis-inducing activities of recombinant protein hIL-24 was tested by MTT assay,Hoechst& FCM assay,and the expression of IL-6 and IFN-? from PBMC induced by rhIL-24 was tested by ELISA.Results The eukaryotic expression vector pcDNA3-hIL-24 was constructed correctly.Stable expression of human IL-24 in CHO cells was identified with RT-PCR.The apoptosis of A549 cells induced by hIL-24 was proved by Hoechst & FCM assay,and the expression of IL-6 and IFN-? from PBMC induced by rhIL-24 was identified with ELISA.Conclusion The successful stable expression & experimental study of apoptosis effect of human IL-24 gene lay the foundation for the further study of molecular mechanism of hIL-24 on anti-tumors and potential application.
7.MRI analysis after augmentation mammoplasty

Objective To analyze MRI appearance after augmentation mammoplasty,and to assess the clinical value.Methods Sixty-three cases with augmentation mammoplasty were imaged by MR scanner.MR imaging features of silicone implant and injected implant were analyzed respectively.Results(80 breasts) in 40 patients were with polyacrylamide hydrogel injection.In 50 breasts,the implants were shown as irregular gel in mammary gland,pectoralis muscle,and subcutaneous tissue.Auto-fat injection in both side of breast was performed in 7 cases.Among them,fibrofatty mass was detected in 12 breasts,fat-fluid interface was seen in 6 breasts,and fat in pectoralis major was revealed in 6 breasts.Silicone implant in both side of breast was performed in 16 cases.2 breasts in 2 case were detected as saline-filled implants with intracapsular rupture.14 cases were with silicone gel-filled implants,among them,4 breasts were found to have extracapsular rupture and 10 breasts intracapsular rupture.Conclusion MRI is a perfect method in accessing the patients with augmentation mammoplasty.
8.Long-term efficacy of modified Bristow-Latarjet procedure in treatment of recurrent inferoanterior shoulder dislocation

Objective To investigate the long-term effects of modified Bristow-Latarjet operation for treatment of recurrent inferoanterior shoulder dislocation.Methods A retrospective case series study was done on 90 patients with recurrent inferoanterior shoulder dislocation treated by modified Bristow-Latarjct procedure from January 2001 to January 2016.There were 72 males and 18 females,with an average age of 36.8 years.There were 59 fight shouders and 31 left shoulders,with dislocation for 4-32 times (mean,13 times).Shoulder instability severity index score (ISIS) was 3-10 points (mean,6.4 points).The duration of disease was 6-26 months (mean,13 months).The operation methods were coracoid lateral incision with modification,rotator cuff interval approach,double hollow compression screw fixation,and bone congruent-arc technique in standing position.Clinical evaluation was done on postoperative recurrence of shoulder joint dislocation/subluxation,bone healing evaluated by X-ray and CT,Rowe score,simple shoulder function Test (SST) score,shoulder osteoarthritis score (Samilson-Prieto),bodv side shoulder external rotation angle and the subscapularis muscle strength.Results All patients were followed up for 1-15 years (mean,5.6 years).The rate of redislocation or subluxation during follow-up was zero.All transpositions of the coracoid got bone healing at postoperative 3-6 months.The Rowe score was increased significantly from (45.6 ± 9.4) points preoperatively to (92.6 ± 3.5) points postoperatively (P <0.01);SST score was increased significantly from (5.6 ± 1.2)points preoperatively to (9.6 ± 2.8) points postoperatively at final follow-up) (P < 0.01).Samilson-Prieto score was mild in 2 patients (2%).The body side shoulder external rotation angles were (56.7 ± 13.9) ° preoperatively and (54.6 ±14.1) ° postoperatively (P > 0.05).With regards to subscapularis strength,the lift-off and belly-press in injured side were (4.9 ± 0.9) kg and (4.4 ± 1.0) kg,respectively (P > 0.05).Lift-off and belly-off in normal side were (5.1 ± 0.5) kg and (4.7 ± 1.1) kg,respectively (P > 0.05).Conclusion The modified Bristow-Latarjet operation is a reproducible and effective technique that can restore shoulder stability,with advantages of sound bone healing,free shoulder external rotation,low incidence of shoulder joint osteoarthritis,remaining of myodynamia under scapula and hence has long-term effects and is worth of clinical applicaiton.
9.Expression of Human Interferon-λ1 and Interferon-ε Gene in WI-38 Cells and Comparison of Their Biological Activity

The biological activities i. e. antineoplastic activities and antiviral activity of the two novel kinds of interferons: hIFN-λ1 and hIFN-ε were studied and compared. First the fusion expression vector: pcDNA3.1A-hIFN-λ1-His and pcDNA3.1A-hIFN-ε-His by PCR was constructed, then the two kinds of plasmids were transfected into WI-38 (human embryonic lung cells ) with liposome. And cytopathic effect (CPE) suppression test was used to study and compare the antiviral activities of rhIFNλ1-His and rhIFN-ε-His, meanwhile MTT assay was used to detect their antineoplastic activities. It was found that, antiproliferative activity and MxA protein induction shown by rhIFN-λ1-His is more powerful than of rhIFN-ε -His. The antiviral molecular mechanisms of both hIFN-λ1 and hIFN-ε are related to MxA. The foundation for further study on the bioactivities and mechanism of action of hIFN-λ1 and hIFN-ε was established.
10.Construction of recombinant adenovirus vector expressing hIL-17F and functional study of expressed IL-17 F

Objective To construct a recombinant adenovirus vector ( Ad-hIL-17F) expressing human interleukin 17F (hIL-17F) and to investigate the effects of expressed hIL-17F on angiogenesis. Methods The hIL-17F fragments was amplified by PCR using pUCm-T/hIL-17F plasmids as templates and then cloned into pAdTrack-CMV transfer vector to form pAdTrack-CMV-hIL-17F.The pAdTrack-CMV-hIL-17F transfer vector was linearized with PmeI digestion and then transformed into competent BJ 5183 with pAdEasy-1 backbone vector for homologous recombination .Then it was linearized with PacI digestion and transfected into human embryonic kidney 293 (QBI-293A) cells to construct Ad-hIL-17F.RT-PCR analysis and indirect immunofluorescent assay (IFA) were performed to determine the expressions of hIL-17F.MTT assay was used to detect the inhibitory effects on cell growth of ECV 304 .The expressions of VEGF and Ang-1 in 293 A and ECV304 cells were measured by ELISA .The effects of Ad-hIL-17 F on the expressions of VEGF in 293A cells were analyzed by Real-Time PCR.Results The sequencing result verified that hIL-17F gene fragment was correctly inserted in the vector .The expressions of hIL-17F gene at mRNA and pro-tein levels were confirmed by RT-PCR and IFA.Ad-hIL-17F could significantly inhibit the growth of ECV304 cells and down-regulate the expressions of VEGF and Ang-1 in 293A and ECV304 cells.Conclu-sion Ad-hIL17F expressing hIL-17F was successfully constructed .The expressed hIL-17F could inhibit the angiogenesis through down-regulating the expressions of VEGF and Ang-1.
