Objective To explore the capacity of the Sweden CellaVision DM96 automatic digital cell morphology analysis sys‐tem (DM96) in nucleated cell classification of serous cavity effusion .Methods 36 specimens of serous cavity effusion were selected from the inpatients of Second Affiliated Hospital of Xi′an Jiaotong University in March 2015 and performed the Wright staining by the two kinds of method ,the Japanese Sysmex SP‐1000Ⅰ automatic smearing machine and manual smearing ,after staining ,the smear was performed the nucleated cells classification by DM96 .The consistency and relevance of the classification results by DM 96 with those by the Sysmex XT‐4000i were calculated .Results The classification results by DM96 had better consistency with the results by XT‐4000i ,moreover the cell images taking by DM96 were clear with high automatic degree .Conclusion The DM96 auto‐mated digital nucleated cell morphology analysis system is reliable and effective ,and has a significance for improving the cellular morphological analysis of serous cavity effusion specimen .
3.The Contribution of 3D-TOF MRA in Evaluation of NeurovascularCompression in Patients with Trigeminal Neuralgia

Objective To study the contribution of enhanced 3D time-of-flight MRA in the diagnosis of neurovascular compression in the patients with trigeminal neuralgia.Methods Enhanced 3D time-of-flight MRA and MRI in 37 patients clinical suspected trigeminal neuralgia were analyzed retrospectively.Results We found 27 sides trigeminal nerve in 24 patients were compressed by vascular in the enhanced 3D time-of flight MRA.In 24 patients neurovascular compression was ipsilateral with trigeminal neuralgia,and the side without trigeminal neuralgia, vascular compression in 3 cases were found.There was statistics correlation between neurovascular compression and trigeminal neuralgia.The other 13 patients with trigeminal neuralgia were non-neurovascular compression.The trigeminal nerve was compressed by superior cerebellar artery in 14 patients,anterior inferior cerebellar artery in 5 patients,tortuous vertebral artery in 1 patient,malformation of vascular in 1 patient,other microvascular in 5 patients.Conclusion Enhanced 3D time-of-flight MRA,which is super than MRI,can clearly display trigeminal nerve and neighboring vascular.Int is the optimum technology of radiology to diagnose the neuroveascular trigeminal neuralgia and helpful for diagnosis and treatment.
4.3D-TOF MRA in the Etiologic Diagnosis of Hemifacial Spasm and Trigeminal Neuralgia

Objective To study the clinical significance of 3 dimensional time of flight magnetic resonance angiography(3D-TOF-MRA) for the pathogenesis of hemifacial spasm (HFS) and trigeminal neuralgia(TN).Methods 48 patients with HFS and 46 patients without HFS and 42 patients with TN and 40 patients without TN were examined by MRI and 3D-TOF-MRA by the enhancement of DTPA. Diagnosis of the presence of compressions in the root exit zone(REZ) of facial nerves and trigeminal nerves were done by two radiologists on an independent console. Results (1)In the patients, compression of the REZ of the facial nerves and trigeminal nerves were detected on 45 spastic sides (93.8%,neurovascular on 44 sides and tumor on 1 side) and 36 spastic sides ( 85.7%,neurovascular on 32 sides and tumor on 4 sides ), 8 and 4 on the asymptomatic sides (16.7% and 9.5%, all neurovascular ). In the controls, 4 and 5 sides ( 4.4% and 6.3% ) were found in the compression of the REZ of the facial nerves and trigeminal nerves. ( 2 ) The offending vessels of compression of the REZ of the facial nerves were the anterior inferior cerebellar artery (AICA) in 17 cases ( 38.6% ), the posterior inferior cerebellar artery (PICA) in 12 cases (27.3%), the vertebral artery (VA) in 6 cases (13.6%). The offending vessels of compression of the REZ of the trigeminal nerves were the superior cerebellar artery ( SCA ) in 18 cases ( 56.3% ), the anterior inferior cerebellar artery in 5 cases (15.6%), the difficult identified vessels (DIV) in 4 cases (12.5%). (3)The relative risks of microvascular compressions which cause HFS and TN were 26.6 and 9.84. (4) The compressions of the REZ of the facial nerves and trigeminal nerves were proved in 4 cases (neurovascular 3 cases and tumor 1 case) and 10 cases (neurovascular 6 cases and tumor 4 casee) in the operation.Conclusion MRI and enhanced 3D-TOF-MRA appeare to be the best imaging technology for the pathogenesis of HFS and TN now. The major causes of HFS and TN may be different neurovascular compressions in the REZ of the facial nerves and trigeminal nerves, some cases are caused by tumor compression.

6.Comparison of test bolus and bolus tracking techniques for dual-energy CT lung perfusion scan

Objective To compare the test bolus technique with the bolus tracking technique for dual-energy CT pulmonary angiography in patients suspected of pulmonary embolism (PE).Methods A total of 60 patients were randomized into 2 groups:bolus tracking technique group (group B,n =30) and test bolus technique group (group T,n =30).A standard dual-energy CT pulmonary angiography (CTPA)was performed after injection of 60 ml contrast medium followed by 35ml saline chaser at a speed of 4 mL/s.Attenuation profiles of different vascular segments (the subclavian vein,superior vena cava,left atrium,pulmonary trunk,S1 and S10 pulmonary artery) and enhancement value of lung parenchyma were measured to evaluate the timing techniques.The overall image quality of CTPA and pulmonary perfusion were analyzed by two radiologists.SPSS 19.0,Student t test,ANOVA test,Chi-Square test,Mann-Whitney U test and Kappa test were used for the statistic analysis.Results The attenuation of the subclavian vein [(1042 ± 639),(2200 ± 724) HU,F =43.196,P =0.001],superior vena cava [(529 ± 237),(904 ± 329)HU,F=25.654,P=0.001],pulmonary trunk [(325 ±112),(383±69)HU,F=5.94,P=0.018],S1 pulmonary artery [(320 ± 118),(385 ±73) HU,F =6.549,P =0.013],S10 pulmonary artery [(307 ± 78),(360 ± 82) HU,F =6.658,P =0.012] in group B were lower than those in group T,while the attenuation of the left atrium in group B was higher than that in group T [(270 ± 53),(219 ± 57)HU,F=12.823,P =0.001].The percentage of pulmonary arteries exceeding optimal attenuation (> 250 HU) in groups B was lower than that in group T [n =21 (70.0%),n =29 (96.7%),x2 =5.88,P < 0.05].The enhancement value of pulmonary parenchyma in group B was lower than that in group T (the right lung (29±8)vs(34±8) HU,t=-2.401,P=0.020; the left lung (30±7)vs(36±9)HU,t=-2.493,P=0.016; total (30 ±7)vs(35 ±8) HU,t =-2.495,P =0.016).The image quality of CTPA in group B was poorer than that in group T (U =619.5,P < 0.05,Kappa =0.708),while the artifacts of lung perfusion image in group B was less than that in group T (U =162.5,P < 0.05,Kappa =0.796).Conclusion The test bolus technique has a better image quality of CT pulmonary angiography and a higher lung perfusion blood volume compared with the bolus tracking technique by using 60 ml contrast medium and 35ml saline.
7. Advances in treatment of triple negative breast cancer

Breast cancer is the most common malignant tumor in females in the world. The characteristics of triple-negative breast cancer (TNBC) are defined as estrogen receptor (ER)-negative, progesterone receptor (PR)-negative and human epidermal growth factor receptor 2 (HER2)-negative. TNBC accounts for about 15%-20% in all the pathological types of breast cancer. With poor prognosis, high recurrence rate and high mortality rate, TNBC has become the focus of the research recently. At present, the treatment methods of breast cancer include surgery, neoadjuvant chemotherapy, adjuvant chemotherapy, endocrine therapy and targeted therapy, etc. Due to the lack of ER, PR and HER2 expressions, patients can not benefit from the endocrine therapy and anti-HER2 targeted therapy. Chemotherapy is currently recommended for the treatment of TNBC. Many chemotherapeutic regimens and new drugs are being explored. This paper reviews the progress in chemotherapy, endocrine therapy, targeted therapy and immunotherapy of TNBC.
8.Expression of Bone Morphogenetic Protein-2 in Gunshot Fracture Healing Detected with Real Time Fluorescence Quantitative Reverse Transcript Polymerase Chain Reaction

ObjectiveTo detect the levels of bone morphogenetic protein-2(BMP-2) mRNA in gunshot fracture healing of the rabbits.MethodsA model of primary treating gunshot fracture of the rabbit with external fixator and the real time fluorescence quantitative reverse transcription polymerase chain reaction(FQ-RT-PCR) technology with SYBR Green I were established.The levels of BMP-2 mRNA 1,2,3 and 4 weeks after operation were detected with FQ-RT-PCR respectively.ResultsIn the control group,BMP-2 mRNA of the bone tissue began to rise at the first week after fracture and the peak of mRNA expression appeared at the second week.The expression returned to normal at the forth week.In the experimental group,the BMP-2 mRNA rose more slowly that it arrived peak stage at the third week and was significantly lower in experimental group than that in control group at the same phases.ConclusionReal time FQ-RT-PCR is a good method for measuring the levels of BMP-2 mRNA during gunshot fracture healing.The mRNA expression rose more slowly and was significantly lower in gunshot fracture than in common fracture at the same phases.
9.Correlation between acupoints and zang-fu organs based on clinical trials.

A systematic analysis was made on the correlation between acupoints and pathological changes of the zang-fu organs through reviewing of literatures in the database of CNKI from 1959--2011 and the database of Pubmed of the past 10 years. The result showed that specificity was found on the pathological changes of zang-fu organs when acupoints were stimulated. And the pathological changes of the internal organs can be perceived from acupoints on the correspondent meridians, mainly regarding the heart, the stomach, the intestines, the lung and the liver, etc. And most of the researches focused on the correlation between acupoints of the heat meridian and the heart. It was also discovered that a lot of acupoints on various meridians could manifest the pathological changes of the same organ. Different effects of the same acupoints on different times could be found on the same internal organ. Therefore, it is concluded that there is a relative specificity between acupoints and pathological changes of the zang-fu organs. However, it is worth to study the regularity of specificity further.

10.Construction and immunogenicity of recombinant porcine parvovirus-like particles with somatostatin.

In order to obtain a virus-like particle vaccine both for porcine parvovirus (PPV) prevention and growth-promotion, VP2 gene of PPV NJ-a strain was amplified with PCR, and four copies of synthetic somatostatin gene were fused to the N-terminal of VP2 gene. The fused gene was cloned into pFast-HT A to construct the recombinant plasmid pFast-SS4-VP2, then the pFast-SS4-VP2 was transformed into DH10Bac competent cells and recombined with shuttle vector Bacmid, followed by identification with blue-white screening and PCR analysis for three cycles, and the positive recombinant was named as rBacmid-SS4-VP2. The positive Sf-9 cells were transfected with rBacmid-SS4-VP2 by Lipofectamine to produce recombinant baculovirus. When the cytopathic effect (CPE) was obvious, the transfected Sf-9 cell was harvested, and the positive recombinant virus was named as rBac-SS4-VP2. The insertion for the target gene into baculovirus genome was confirmed with PCR. SDS-PAGE and Western blotting revealed that the calculated protein of approximately 68 kDa was in the expressed in the insect cells. The Sf-9 cells infected with rBac-SS4-VP2 were stained positive against PPV antibody using the indirect immunofluorescence assay (IFA). Moreover, the virus particle self-assembly was observed under electron microscopy. 90 four-week-old mice were immunized by the recombinant protein coupled with different adjuvants alhydrogel, IMS and oil. VP2-specific ELISA antibodies, PPV-specific neutralizing antibody, somatostatin antibody and growth hormone levels were examined to evaluate the immunogenicity of this virus like particle. Results indicated that mice groups immunized rSS4-VP2 protein with alhydrogel and IMS developed similar humoral immune response comparing with inactived PPV vaccine. Mice group immunized with rSS4-VP2 generated higher level of SS antibody and growth hormone comparing with negative control, mice receiving rSS4-VP2 with alhydrogel developed the highest antibody titre than all other groups, while the oil group developed the lowest antibody level. This study provides not only a new rout for production of safe and effective virus like particle subunit vaccine, but also the foundations for peptide presentation and multivalent subunit vaccine design.
