1.Prokaryotic expression and preliminary identification of scavenger receptor protein

Objective To express and preliminary identify scavenger receptor B2 (SCARB2)protein.Methods SCARB2 cDNA was amplified by reverse transcription-polymerase chain reaction (RT-PCR)from mRNA extracted from RD cells,and cloned into pMD19-T vector.Expression vector pET28a(+)/SCARB2 was constructed,and SCARB2 protein was expressed in prokaryotic ex-pression system.Obtained recombinant proteins were identified by detection of Western blot using His labeled monoclonal antibody.Results Recombinant SCARB2 proteins,expressed by induction of isopropy-β-D-thiogalactoside (IPTG),were success-fully purified and specifically recognized by His labeled monoclonal antibody.Conclusion SCARB2 proteins could be expressed and identified successfully,which could provide reference for researching mechanism of enterovirus type 71 (EV71)and scavenger re-ceptor protein,and for preparation of SCARB2 monoclonal antibodies.
2.Advances of fulvestrant in treatment of advanced breast cancer in hormone receptor-positive postmenopausal women

Endocrine therapy targeting estrogen pathway is one of the ifrst-line treatment choices of advanced breast cancer. Fulvestrant is a pure estrogen antagonist that blocks and downgrades estrogen receptor, which makes it effective in the treatment of progression after prior endocrine therapy. Fulvestrant 250 mg per month regime was approved for postmenopausal women with hormone-positive advanced breast cancer after progression or recurrence on antiestrogen therapy. Fulvestrant 500 mg per month regime was approved by the EMA and the US FDA in the same population based on the CONFIRM trial which proved improved efifcacy and similar tolerance compared with 250 mg regime. Recent trials were focused in the ifrst-line treatment and combination use with other therapeutics. This review discusses the advances of fulvestrant in postmenopausal women with hormone-positive advanced breast cancer.
3.Analysis of Electrochemical Performances of Different Mesoporous Manganese Dioxide Materials by Quartz Crystal Microbalance

The mesoporous manganese dioxide (MnO2) materials were synthesized by the calcinations of manganese carbonate (MnCO3) precursors in air atmosphere at 300, 350 and 400℃. The slurries of MnO2 and binder were sprayed on the quartz electrodes. Quartz crystal microbalance (QCM) was conducted with cyclic voltammetry to analyze the electrochemical behaviors of the three MnO2 samples in 0.1 mol/L sodium sulfate (Na2SO4). The mass of the three samples were generally increased during potential cycling, especially in the first stage, which suggested that an irreversible reaction process occurred. The as-synthesized MnO2 at 300℃ had the better electrochemical stability and capacity retentivity. The three materials were assembled to (-) active carbon/MnO2 (+) supercapacitors and the charge-discharge tests were conducted. The results showed that 35%-40% capacity loss was occurred in the initial cycling, and the total discharge capacities of MnO2 formed at 300℃, 350℃, 400℃ were 15.9, 12.9, 11.7 mA h/g, respectively. The detection results of QCM method were consistent with that of the charge-discharge tests, suggesting that this QCM method could be used for distinguishing the electrochemical performance of mesoporous MnO2 materials.
4.Renal allograft long-term function protection with enalapril

Objective To observe the protective effects of enalapril on the long-term functions of the allograft in renal transplant recipients. Methods Twenty renal transplant recipients with ~survival time over one year, normal renal functions of the allograft and urine TGF-?_1 levels being more than ~250.0 pg/mg.Cr took enalapril every day for at least one year. Twenty-three recipients under the same conditions who did not receive enalapril served as control group. Three years later renal ~dysfunction cases, loss of creatinine clearance rate (Ccr) and TGF-?_1 levels in blood and urine were compared between the two groups. The changes in the expression of TGF-?_1mRNA in renal biopsy specimens were compared before and one year after enalapril therapy. Side-effects of enalapril were ~investigated in all patients in enalapril-treated group. Results Three years later, the number of renal dysfunction cases was less, the loss of Ccr was less and the level of urine TGF-?_1 was lower in ~enalapril -reated group than those in control group with the differences being significant (P~0.05 ). The effects of tetramethylpy-razine (TMP) on pancreatic blood flow and survival rate were studied in sodium tarocholate-in-duced acute pancreatitis (AP) in rats. The results showed that pancreatic relative blood flow and pancreatic tissue perfusion were significantly increased and the pathologic changes and survival rate were improved in TMP treated group-s. Plasma value of TXB2?6-Keto-PGF1? and platelet aggregation rate (PAR) were also mea-sured. We found that TMP could maintain the balance between TXA2 and PGI2 and lower the elevated PAR. It was suggested that TMP has therapeutic effect on AP in rats through improving pancreatic microcirculation; which was related to the maintanance of the balance between PGI2 and TXA2.
6.Analysis of diagnostic and handling procedures of the first imported case of A/H1N1 influenza in mainland China

Objective The patient started from St. Louis (U. S.), and reached Chengdu (China) after 3 days of flight(St. Louis- Tokyo- Beijing - Chengdu). The patient felt unwell, so he and the two relatives took a taxi to the Emergency Center of Sichuan Provincial People' s Hospital for treatment. The epidemiological survey showed that, the patient had been living in the University of Missouri in the United States, no pigs died locally, there were no markets for the living pigs, farmers markets, farms, and slaughterhouses, and he hadn't contact with pigs. But 4 days before the onset, he closely contacted with a schoolmate, who had cold symptoms. The pa-tient had never been to virus laboratory 7 days before the onset. Clinical examinations showed that, the patient had a fever (37.8 ~ 38.8℃), WBC 7.9×10-9 L-1, N 5.475 × 10-9 L-1, L 19.5% ;the chest X-ray shewed that texture increased and the heart shadow augmented on both lungs; the result of throat swab culture was negative. The result of virus nucleic aeid detected by Sichnan Provincial Center for Disease Control showed H1N1 influenza virus, suspected. Sichuan Province Health organization organized experts on the prevention and control of H1N1 influenza for consultation, and the patient was diagnosed as the first suspected H1N1 influenza case in mainland China based on the epidemiological investigation, symptoms and signs of the patient, the results of laboratory ex-amination, and the result of virus test. Confirmed by the experts from Chinese center for disease control and pre-vention and The Ministry of Health of the People's Republic of China, the patient was diagnosed as the first H1N1 influenza case in mainland China.
7.Influence of exogenous somatostatin on pancreatic blood flow in normal rats or rats with acute necrotizing pancreatitis

AIM: To investigate the influence of exogenous somatostatin (stilamin) on pancreatic blood flow in normal rats or rats with acute necrotizing pancreatitis (ANP).METHODS: Pancreatic blood flow (PBF) was detected with computerized tissue blood flowmeter and rats with ANP were triggered with sodium taurocholate. Metabolites of eicosanoids in plasma were determined with radioimmunoassay. Other laboratory tests including histopathologic observation under optical or electron microscope were used. RESULTS: There was a significant decrease in PBF in normal rats after stilamin administration in comparison with that before use of the drug. There was significant decrease in PBF after onset of ANP, but, compared with that in ANP group, significant increase was shown in SS(stilamin)+ANP group. Plasma thromboxin-B 2(TXB 2) in ANP group at 6 hours after ANP was significantly higher, with increase of 4.5 times, than that in Sham(sham operated) group while TXB 2, detected each time during the course of ANP, significantly decreased in SS+ANP group. 6-Keto-prostagland in F 1? (6-Keto-PGF 1? ) at 6 h after ANP was significantly higher, and the ratio of TXB 2/6-Keto-PGF 1? , significantly lower in SS+ANP group than that in ANP group. Lessened necrosis of acinar cells, along with much fewer microthrombi in microvessels in SS+ANP group, was shown by pathologic scoring or electron microscope than that in ANP group.CONCLUSION: Administration of exogenous somatostatin leads to the decrease in PBF in physiological setting but it attenuates pancreatic ischemia in SS+ANP group, which may be attributed to correction of abnormal metabolism of eicosanoids, improvement of pancreatic microcirculation and cytoprotection of acinar cells as well.
8.White matter microstructure alterations in the patients with generalized anxiety disorder: a diffusion weighted imaging study

Objective To examine the white matter microstructure alternations in the patients with generalized anxiety disorder (GAD) with diffusion weighted imaging.Methods 19 patients with GAD and 20 matched healthy controls were assessed using apparent diffusion coefficient (ADC) and exponential apparent diffusion coefficient (eADC) in the regions of interests (ROIs) approach.The ROIs were the white matter of bilateral cingulate and bilateral hippocampus.Results Significantly increased ADC values were found in GAD patients (0.78±0.02,0.79±0.03) with respect to normal controls in the right and left anterior cingulate white matter.Significantly decreased eADC values were found in GAD patients (0.46±0.01,0.45±0.01) in the right and left anterior cingulate white matter.Conclusion Diffuse cingulate white matter alterations on DWI in GAD denote the disruption of white matter integrity.

10.Effect of Chang'ankang Capsule on rat's colitis and its mechanism

AIM: To observe the effect of Chang'ankang Capsule (Radix Astragali, Rhizoma loptidis, Flos Lonicerae, etc.) on rat's colitis induced by trinitrobenzene sulfonic acid (TNBS) and investigate its mechanism. METHODS: The rat's colitis was induced by TNBS and ethanol, one week later the rats were given the medicine for 14 days. The 21st day the mucosal damage index (CMDI) and histopathology (HP) of the colon were observed. Meanwhile the level of myeloperoxidase (MPO), nitric oxide (NO), superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), maiondiadehyde (MDA), catalase (CAT) were detected. RESULTS: CMDI and HP, biochemistry in model rats showed that the models were successful. Chang'ankang groups significantly alleviated CMDI, HP, and improved the high level of MPO, NO and MDA, promoted the content of SOD, GSH-PX, CAT. CONCLUSION: Treatment with Chang'ankang shows beneficial effects on rat's ulcerative colitis by alleviating damage of free radical and NO and leukocyte.
