1.SOMATOSTATIN IN SYNAPTIC STRUCTURES OF SUBSTANTIA GELATINOSA OF THE SPINAL CORD

The distribution of somatostatin (SRIF) in substantia gelatinosa of the rat spinal cord was studied by means of immuno-electron microscopy. The ultrastructural features showed that nerve terminals containing SRIF take part in forming presynap- tie elements of axe-somatic, axe-dendritic and axo-axonic synapses. The immune- reactive products locatl at the external membrane of mitochondria, around the small clear synaptie vesicles and in the large granular vesicles. Most of synaptic vesicles are round or ovoid in shape. Only a few of them are flattened. Based on the ultrastructural characteristics mentioned above and related experimental results the authors believe that SRIF in substantia gelatinosa of the rat spinal cord is probably involved as a neurotransmitter instead of neuromodulator.
2.THE PEPTIDERGIC INNERVATION OF CEREBRAL VESSELS——LIGHT MICROSCOPIC AND IMMUNOELECTRON MICROSCOPIC STUDY

Using the immunohistochemical and immunoelectron microscopic methods, the localization and pattern of vasoactive intestinal polypeptidergic nerves in the wall of cerebral vessels were investigated. The results revealed that the VIP-like immunoreactive (VIPLI) fibres were located in the adventitia or at the adventitia-media border. No synaptic contacts were identified among the nerve terminals or between nerve terminal and smooth muscle cell. VIPLI terminals directly apposed to the smooth muscle cells with a distance of 100 nm.The observations mentioned above indicate that there is peptidergic innervation in the wall of the cerebral vessels besides traditional adrenergic and cholinergic innervations. The regulatory function of VIP-containing nerves to the cerebral vessels may be performed by affecting the smooth muscle directly through the nonsynaptic release. In addition, present study identified and discussed the distribution and function of substance P(SP) in the cerebral vessels.
3.Effects of Yifei Kangliu Oral Liquid on cell cycle and protein-nucleic acid synthesis of experimental lung cancer

OBJECTIVE: To explore the effect of the traditional Chinese medicine Yifei Kangliu (YFKL) Oral Liquid on the proliferation, cell cycle and protein-nucleic acid synthesis of murine Lewis lung cancer cell and human lung adenocarcinoma cell line SPC-A-1. METHODS: The inhibiting rates of tumor growth were calculated by weighing the weight of tumor inoculated in vivo, combined by counting cancer cells in vitro. The ratio of the cell cycle and exponents of DNA, RNA, and protein were measured by flow cytometry (FCM). RESULTS: The inhibiting rate of tumor growth in the treated group with YFKL Oral Liquid was 30.38% (P<0.05). The proportion of cells in S phase of the treated groups with YFKL Oral Liquid was lower than that of the control group. In the group with most significant result, 72% of the cells were stagnated in G0/G1 phase. The inhibiting rates of DNA, RNA and protein in murine Lewis lung cancer were 7.4%, 23.73% and 23.31% respectively. In SPC-A-1 cell line, the inhibiting rates were 9.3%, 10.1% and 14.7% respectively, demonstrating amplified effects on lower levels. CONCLUSION: YFKL Oral Liquid significantly inhibited the proliferation of murine Lewis lung cancer cell and human lung adenocarcinoma cell line SPC-A-1 by blocking the cancer cells entering the proliferative phase resulted from its inhibition of DNA.
4.Changes in serum levels of Th22 cell-related cytokines and complements in patients with drug eruption before and after treatment

Objective To investigate changes in serum levels of Th22 cell ? related cytokines and complements in patients with drug eruption before and after treatment, and to explore their possible roles in the occurrence and development of drug eruption. Methods This study included 35 patients with drug eruption, and 35 sex?and age?matched healthy controls. Five milliliters of peripheral blood samples were collected from the controls and patients before and after treatment. Enzyme?linked immunosorbent assay(ELISA)was performed to measure serum levels of interleukin 22(IL?22)and IL?13, and the cytometric bead array(CBA)system was used to determine serum levels of tumor necrosis factor?α(TNF?α) and complement components C3a, C4a and C5a. Results Before treatment, the patients with drug eruption showed significantly higher serum levels of IL?22(40.85 ± 14.56 vs. 29.09 ± 8.66 ng/L, t=5.549, P<0.05), IL?13(869.94 ± 463.39 vs. 372.92 ± 151.75 ng/L, t=6.071, P<0.05), TNF?α(1.03 ± 0.64 vs. 0.44 ± 0.31 ng/L, t=4.321, P<0.05), complement C3a(55.21 ± 32.98 vs. 42.44 ± 14.26 ng/L, t=2.832, P<0.05), C4a(285.11 ± 123.91 vs. 237.00 ± 63.57 ng/L, t=2.257, P<0.05), and C5a(279.68 ± 127.72 vs. 215.98 ± 65.38 ng/L, t=2.495, P<0.05)compared with the controls. After treatment, the serum levels of IL?22, IL?13, TNF?α, complement C3a, C4a and C5a in patients decreased to(32.72 ± 11.77)ng/L,(456.21 ± 123.22)ng/L,(0.64 ± 0.39)ng/L,(45.47 ± 21.11)ng/L,(241.86 ± 84.12)ng/L and(239.61 ± 103.51)ng/L respectively, with a significant difference between the pretreatment and posttreatment values of these proteins(t = 4.443, 5.197, 3.572, 3.213, 2.728 and 4.772, respectively, all P ≤ 0.01). Additionally, the serum levels of IL?22 and IL?13 were still significantly higher in the patients than in the controls(both P < 0.05), while there were no significant differences in the serum levels of TNF?α, complement C3a, C4a or C5a between the patients and controls after treatment(all P > 0.05). Correlation analysis showed positive correlations between complement C3a and C4a serum levels(r = 0.660, P < 0.05), between C3a and C5a serum levels(r = 0.404, P < 0.05), between C4a and C5a serum levels(r = 0.501, P < 0.05), and between IL ? 22 and TNF ? α serum levels(r = 0.573, P = 0.005), but negative correlations between IL ? 22 and complement C3a serum levels(r = -0.490, P = 0.005), in patients before treatment. Conclusion The activation of Th22 cell?related cytokines and complements may play important roles in the occurrence and development of drug eruption, and IL?22 may participate in the regulation of complements.
5.Establishment of Na+-glucose cotransporter 2 heterologous expression system in HEK293 cells

Objective To establish heterologous expression system of Na+-glucose cotransporter 2 (SGLT2) gene.Methods Human SGLT2 cDNA from normal kidney, generated by RT-PCR,was subclonedintoPEXL-GFP vector andtransfectedinto HEK293cells. After 24hours of incubation, the expression of SGLT2-GFP fusion protein was detected by Western blotting and laser confocal microscopy.Transport activity of SGLT2-GFP fusion proteins in cultured human HEK293 cells was evaluated with the uptake test of glucose analogue.ResultsSGLT2-GFP fusion protein was expressed in cultured human HEK293 cells.Furthermore, confocal microscopy using green fluorescent protein(GFP) revealed a punctate membrane pattern of SGLT2.Glucose analogue uptake increased in HEK293 cells transfected with SGLT2-GFP at least by 3.5 folds compared with HEK293 cells transfected with GFP vector only(P<0.01).Conclusion Heterologous expression of SGLT2 gene in HEK293 cells is successfully established, which provides valuable approach for the functional and pathological study of SGLT2 gene.
6.Effects of perforated bovine amnion combined with recombinant bovine basic fibroblast growth factor on degree Ⅱ burn wounds A comparison with imperforated bovine amnion and vaseline gauze dressing

OBJECTIVE: To observe the clinical effect of perforated bovine amnion combined with recombinant bovine basic fibroblast growth factor (rb-bFGF) on degree Ⅱ burn wounds.METHODS: A total of 43 patients with small and medium-size thermal burn were collected, and the area of testing wound was 1% -2%. The wounds with the same nature were divided into three groups: perforated bovine amnion (treatment group), bovine amnion (control 1 group), and vaseline gauze dressing (control 2 group). All the three groups combined with rb-bFGF. RESULTS: Compared with control 1 group (P < 0.01) and control 2 group (P < 0.05), the treatment group could obviously decrease the healing time of deep degree Ⅱ burn wounds. For superficial degree Ⅱ burn wounds, compared with the control 2 group, the treatment group could also decrease the healing time; however, there was no significant difference between treatment group and control 1 group (P > 0.05). Dressing was not changed frequently, and the pain was relieved. Rash or other adverse effects were not detected in the three groups.CONCLUSION: The combination of perforated bovine amnion and rb-bFGF can obviously promote the healing of burn wounds.
7.Morphological changes of pancreatic cancer cells transfected by dendrimer modified carbon nanotubes

Objective To observe the process of polyamidoamine dendrimer modified single walled carbon nanotubes complexes (CNT-PAMAM-D) entering into pancreatic cancer BxPC3 cells, and to evaluate its safety as a vector. Methods The CNT-PAMAM-D were prepared by ultrasound, mixing and rinsing, and the morphology was characterized by atom force microscopy and transmission electron microscope. Then the prepared CNT-PAMAM-D was incubated with human pancreatic cancer cell line BxPC3 cells for 12, 48, 72hours, and then the cells were collected. Distribution of CNT-PAMAM-D in cells and cell ultrastructure was observed by transmission electron microscopy. Results After the combination of PAMAM-D and CNT, the surface of CNT was surrounded by dendrimer and nanocomposite particles with a size of 20 nm was formed. It was showed that CNT-PAMAM-D could be transfected into human pancreatic cancer cells by cell pinocytotic way, and it entered cytoplasm at 12 h. With the extension of the transfecting time, CNT-PAMAM-D could enter lysosomal and nucleus, but the morphology and ultrastructure ocf BxPC 3 cells was not significantly changed. Conclusions The CNT-PAMAM-D is a highly effective and safe nano vector.
8.The neuropsychological and cortical metabolic changes in two patients with Benson's syndrome

Objective Standard neuropsychological assessment plus structural and functional imaging were used in accurate diagnosis of Benson's syndrome (posterior cortical atrophy).Method Serial neuropsychological screening and integrative assessments of visual spatial function, 3D structural MRIimaging and functional FDG-PET imaging were used in two cases of Benson' s syndrome.Results The clinical signs were agnosia, optic ataxia, apraxia, alexia, agraphia and prosopagnosia.MRI imaging revealed bilateral parietal and occipital lobe atrophy.FDG-PET imaging revealed low metabolism in the posterior cortex.The agraphia was constructive: the words were correct but written in the wrong location.Conclusion Standard neuropsychological assessments can recognize the disease nature.When combined with the structural and functional imaging, a correct diagnosis of Benson's syndrome can be made.
9.Cortex function and neuropsychological changing of non fluent aphasia type-frontal temple dementia

Objective To analyse the neuropsychological changing of nonfluent aphasia frontotemporal dementia (nf-FTD) with the FDG-PET, MRI and fMRI. Methods The language and other cognitive function were dynamically estimated by standard neuropsychological assessment which modified by Chinese. Compared with FDG-PET and fMRI, the relationship between the neuropsychologieal changing and the cortex function was studied. Results All the 5 patients met the criteria of nf-FTD by Association for Frontotemporal Dementias, with core syndrome and other supptted syndrome. And still they had the good word reading, calculation by pen, and the un-speechmemory. And they also had the related well living ability. They all had the related cortext metabolism support. Conclusions nf-FTD patients preserve the word reading, visuapacial ability and calculation by pen. These can help us to give the effective advise about the patients training and caring and to give them the chance to show their real social skill.
10.Clinical analysis of 217 patients with metastatic carcinoma of bone

Objective To analyze the clinical characteristics of patients with metastatic carcinoma of bone. Methods The clinical data of 217 patients with metastatic carcinoma of bone was retrospectively analyzed. The clinical features were summarized,including primary tumor,age distribution,first symptom,time of onset and bone metastatic site. Visual analogue scale(VAS)was used to evaluate the difference of pain in patients with bone metastases before and after zoledronic acid treatment. Results The bone metastasis occurred more frequently in men(male:female = 1.28:1). The majority of primary tumors were lung cancer(51.61%)and breast cancer (15.67%),respectively. The peak ages of the patients with metastatic carcinoma of bone were between 50 and 70 years old. Most of the patients with metastatic carcinoma of bone initially showed varying degrees of pain ,some of the patients with dysfunction,pathological fracture,or local lumps as the initial symptoms,only a tiny minority of patients were inadvertently found. Onset time was often within 3 months ,which accountings for 54.84%. Bone metastases were mostly in the spine,pelvis,femur and ribs,and most of the bone metastases were multiple. The VAS score of 108 patients who with bone metastases pain treated by zoledronic acid was 0~4(1.31 ± 1.07),with statistically significant differences compared with pretherapy(P<0.01). Conclusion In order to reduce the pain of patients,and to improve the quality of life and prolong the survival time,bone metastases should be diagnosed early and given comprehensive treatment.
