1.Clinical application of Danhong injection combined with low-tnoiecular-weight heparins in treatment of unstable angina pectoris

Objective To observe the efficacy of Danhong injection combined with low-molecularweight heparins in treating unstable angina pectoris.Methods One hundred and three cases of unstable ansina pectoris were enrolled and divided into group A and B,Group A were given Danhong injection and lowmolecular-weight hepafins,Group B were given routine treatment.Results The overall response rate of group A was 90.7% and it was 74.0% for group B.The difference was significant (x2 =4.90,P<0.05).Conclusion Treatment with Danhong combined with low-molecular-weight heparins produced a good outcome in the patients with unstable angina pectoris.
2.Interleukin-4in Experimental Murine Systemic Candidiasis

Objective To investigate the change of interle ukin -4(IL -4)in experimental murine systemic candidiasis.Methods In the dexamethasone -induced immuno -suppressed murine systemic can-didiasis models and control healthy mice,two -site ELISA and RT -PCR were applied to determine the levels of IL -4protein or mRNA expression in th e spleens respectively.Colony form ing units(CFUs )of infected kidneys were determined with the plating dilution method,and the mean survival t ime (MST)of mice was also recorded.Results The IL -4concentration in the spleen of healthy mice was 80.1?19.1pg /g.I n contrast,in those infected with lethal dose of yeasts t he concentrations of IL -4were 124.8?24.1pg /g and 262.8?21.8pg /g on 3days or 7days after infection respectively.Meanwhile,the fungal lo ads in kidneys were(21.25?6.31)?102CFUs and(57.52?10.41)?102 CFUs on 3days or 7days after infectio n(P

4.Expression of CD80?CD86 and CTLA-4 in Peripheral Blood Mononuclear Cells of the Patients with Systemic Lupus Erythematosus

Objective To investigate the role of CD80/CD86 and CTLA-4 in the pathogenesis of systemic lupus erythematosus(SLE) and its clinical significance. Methods Applying RT-PCR technique to semiquantitatively analyze CD80/CD86 and CTLA-4 mRNA expression on peripheral blood mononuclear cells (PBMC) from 32 active SLE patients. Results Compared with normal controls, the percentage of positive CD86 expression in active SLE, accounted for 90.63% (29/32), was significantly increased (P 0.05, both). Conclusion The abnormal expression of CD86 and CTLA-4 might play an important role in the pathogenesis of SLE.
5.Expression of p63 and cyclooxygenase-2 and their correlation in skin tumors.

To study the expression of p63 and cyclooxygenase-2 (cox-2) in skin tumors and evaluate the correlation between p63 and cox-2, the expressions of cox-2 and p63 were measured by streptavidin-peroxidase complex immunohistochemical technique in 17 cases of skin squamous cell carcinoma (SCC), 19 cases of Bowen's disease(Bowen), 11 cases of actinic keratosis(AK), 12 cases of seborreic keratosis(SK) and 13 specimens of normal skin. Our results showed that the expression of p63 in skin squamous cell carcinoma, Bowen's disease and actinic keratosis were significantly higher than that in seborreic keratosis, while the expression of p63 in seborreic keratosis was significantly higher than that in normal skin. The expression of cox-2 in skin squamous cell carcinoma, Bowen's disease and actinic keratosis were significantly higher than that in seborreic keratosis, while no statistical difference was noted in the expression of cox-2 between seborreic keratosis and normal skin. Cox-2 expression was positively correlated with the high p63 expression in malignant skin tumors. The increased expression of cox-2 and p63 may play an important role in the development of skin tumors and work synergetically in malignant skin tumors.
6.Expression of Th17 cytokines in skin lesions of patients with psoriasis.

In order to investigate the role of Th17 cytokines in the pathogenesis of psoriasis, reverse transcriptase-polymerase chain reaction (RT-PCR) was used to detect the expression of IL-17, IL-23 (p19/p40), and IL-6 in skin lesions and non-lesions of the patients with psoriasis and skin tissues of normal subjects. The results showed that the mRNA expression levels of IL-17, IL-23p19, IL-23p40 and IL-6 in psoriasis lesion were significantly higher than those of non-lesions (1.231 +/- 0.843 vs 1.003 +/- 0.044, 1.166 +/- 0.142 vs 0.765 +/- 0.133, 1.125 +/- 0.104 vs 0.730 +/- 0.103, 1.186 +/- 0.222 vs 0.976 +/- 0.122, respectively, all P < 0.05). Meanwhile, The expression levels of IL-17 mRNA, IL-23p19 mRNA, IL-23p40 mRNA and IL-6 mRNA were higher in non-lesions than those in normal skin tissues (1.003 +/- 0.044 vs 0.620 +/- 0.104, 0.765 +/- 0.133 vs 0.584 +/- 0.078, 0.730 +/- 0.103 vs 0.000 +/- 0.000, 0.976 +/- 0.122 vs 0.656 +/- 0.121, respectively, all P < 0.05). The overexpression of Th17 cytokines in the skin lesions of patients with psoriasis may indicate that Th17 cytokines play a very important role in the immunopathogenesis of psoriasis.
7.Expression of TLR9 and its mRNA in the lesions of lichen planus.

To investigate the role of toll-like receptor 9 (TLR9) in the pathogenesis of lichen planus, the expressions of TLR9 and its mRNA in the lesional skin of lichen planus were detected by immunohistochemical technique (SP) and RT-PCR. As control, normal skin of healthy volunteers was also tested. The immunohistochemical study showed that the expression of TLR9 in the lesional skin of lichen planus was significantly higher than that in the normal controls. The results of RT-PCR showed that both skin lesions and normal controls had TLR9 expression. In skin lesions, the expression level of TLR9 mRNA was 1.6075+/-0.0930, which was significantly higher than that in normal controls (P<0.001). These findings indicated that up-regulated expression of TLR9 and its mRNA might be involved in the pathogenesis of lichen planus.

BACKGROUND: Cell apoptosis and expression of related apoptotic gene are found in rats with acute cerebral ischemia-reperfusion injury.OBJECTIVE: To study the effects of ganglioside on cell apoptosis in rats with acute cerebral ischemia-reperfusion injury.DESIGN: Randomized control animal experiment.SETTING: Department of Neurology, China-Japan Friendship Hospital of Jilin University.MATERIALS: The experiment was conducted in the Animal Laboratory of China-Japan Friendship Hospital of Jilin University at April 2002.Forty-eight healthy male Wistar rats aged 3-4 months with the body mass of (220±50) g were selected and randomly divided into ischemia-reperfusion group and ischemia-reperfusion + administration group (intraperitoneal injection of ganglioside GM-1 at 30 minutes before ischemia) with 24 rats in each group, and each group was subdivided into three groups according to the reperfusion time: 3-hour, 6-hour and 24-hour with 8 rats in each time-point.METHODS: ①Rat models of cerebral ischemia-reperfusion were established. ②Diphenylamine method was adopted to detect changes of DNA splitting rate in brain tissues at 3 hours,6 hours and 24 hours after cerebral ischemia.100 mg of cerebral cortex was made into 10% homogenate by adding into 0.9 mL of splitting fluid,which was then put in the centrifuge tube for repeated freezing and melting. The supernatant and deposit were collected.DNA splitting rate = supernatant absorption/(supernatant absorption + deposit absorption). ③Immunohistochemical method was used to study the expression of protein kinase Cδ (PKCδ) as well as changes of ganglioside after administration. MAIN OUTCOME MEASURES: Changes of DNA splitting rate in cerebral cortex of rats as well as intensity of PKCδ expression.RESULTS: One rat in the normal saline control group died for exceeding anesthetization at the 6th hour of reperfusion, and 2 rats died at the 24th hour of reperfusion, which were supplemented respectively. With the time of reperfusion increasing, changes of DNA splitting rate significantly increased, which peaked at the 24th hour. The expression of PKCδ peaked at the 6th hour of reperfusion and gradually decreased. The DNA splitting rate and PKCδ expression were remarkably decreased at corresponding time-points in the ganglioside group.CONCLUSION: Ganglioside can inhibit cell apoptosis and reduce PKCδ expression after cerebral ischemia-reperfusion to protect cerebral ischemia-reperfusion injury.
10.The implicit memory of individual depression in percept-driven process under the influence of sad mood induction

Objective To investigate the effect of sad mood to implicit memory of depressed individuals under the condition of percept-driven process. Methods Using the percept-driven implicit paradigm improved by Paller,26 depression subjects and 25 normal subjects completed study-test task and reported the gender of different emotional faces. Results ①There were main effects of time(F = 4.61, P<0.05)and mood state(F= 21.61, P <0.05) ,significant interaction of time and emotion(F = 4. 13, P<0. 05) ,no significant difference of group on visual analogue scale(VAS) mood ratings. ②Among the accuracy rate of subjects' gender judging to different emotional faces,there were significant main effects on time(F = 4.12, P<0.05)and experiment type (F = 20.55, P < 0.05) , and there was significant interaction of time and experiment type (F=31.72,P<0.05). ③Further simple effects analysis showed that the positive((80 ± 13)%vs(92 ± 10)% , F=65.06, P<0.05) ,negative((58 ± 12)%vs(91 ±10)%,F=10.00,P<0.05),neutral((84±16)%vs(88±9)%, F= 12.49, P < 0. 05) faces when the presentation time was 3600ms in experiment type had significant simple effects; the positive ((76 ± 12)%vs(85±10)%,F=54.72, P<0.05) ,neutral((82 ± 10)% vs(80 ± 10)% , F = 54.57, P<0.05)faces when the presentation time was 300 ms in experiment types had significant simple effects, while the accuracy of neutral faces had significant simple effect between the two groups. Conclusion Sad mood enhance the implicit memory of both depressed and normal subjects to positive and negative faces,and the accuracy rate in implicit processing to neutral faces of depressed group were lower than that of normal group.
