1.Inhibitory effect of extracts from asparagus filicinus rhizome on proliferation of human osteosarcoma cells

Objective To investigate the inhibitory effect of extracts from asparagus filicinus rhizome on prolifieration of human osteosarcoma Saos-2 cells and its molecular mechanism .Methods MTT assay was used to detect the cytotoxic activity and growth inhibition of three different extracts from asparagus filicinus rhizome against Saos-2 cells ;plate colony formation assay was per-formed to detect active fraction of asparagus filicinus rhizome on the anchorage dependent growth of Saos-2 cells ;the cell cycle alter-ation was determined by propidium iodide staining and flow cytometry analysis ;the alteration of protein expression level of COX-2 was determined by using Western blotting .Results Ethyl acetate fraction of asparagus filicinus rhizome (AF-A) exerted the potent cytotoxicity on Saos-2 cells(IC50 =26 .7 μg/mL);AF-A induced the inhibitory effect on the anchorage dependent growth of Saos-2 cells in a dose dependent manner(P<0 .05);Saos-2 cells treated by AF-A at the concentration of 30 .0 and 100 .0 μg/mL for 48 h induced the increase of percentages of S phage from (31 .8 ± 4 .8)% in the control group to (43 .7 ± 2 .5)% and(51 .9 ± 1 .9)% ,the difference showing statistical significance (P< 0 .05) .Western blotting showed that AF-A at different concentrations decreased COX-2 protein expression .Conclusion AF-A posseses the inhibitory effect on the proliferation and growth of human osteosarcoma cells in vitro ,and its mechanism might be associated with the induction of S phage arrest and the inhibition of COX-2 protein ex-pression level .
2.Identification of nm23-H1 as a metastatic suppressor and prognostic factor in nasopharyngeal carcinoma by proteomic analysis

Objective:To identify proteins associated with nasopharyngeal carcinoma (NPC) metastasis,and provide scientific basis for the prevention and cure of NPC.Methods:A two-dimensional gel electrophoresis and mass spectrometry were performed to screen for differential proteins between highly metastatic 5-8F and non-metastatic 6-10B NPC cell lines.Western blot was used to confirm the differential proteins.We used siRNA to inhibit the expression of differential protein nm23-H1 to determine the association of nm23-H1 with NPC in vitro invasive ability.Immunohistochemistry and statistics were used to evaluate the correlation of nm23-H1 expression with clinicopathological features and clinical outcomes in paraffin-embedded archival tissues including 93 cases of primary NPC and 20 cases of cervical lymphonode metastatic NPC (LMNPC).Results:A total of 15 differential proteins in the 2 cell lines were identified by a proteomic approach,and 3 differential proteins were selectively confirmed.Downregulation of nm23-H1 by siRNA significantly increased the in vitro invasive ability of 6-10B.Significant nm23-H1 downregulation was observed in LMNPC compared with primary NPC.nm23-H1 downregulation in primary NPC was positively correlated with lymphonode and distant metastasis,advanced clinical stage and recurrence.Survival curves showed that patients with nm23-H1 downregulation in primary NPC had a poor prognosis.Multivariate analysis confirmed that nm23-H1 expression level in primary NPC was an independent prognostic indicator.Conclusion:nm23-H1 behaves as a metastasis suppressor in NPC,and nm23-H1 downregulation is a biomarker for poor NPC prognosis.
3.The application of cardiopulmonary resuscitation skill training in pre-job training for clinical interns and its value of quality control

Objective To explore the application of cardiopulmonary resuscitation skill training in pre-job training for clinical interns and its value of quality control. Methods A total of 136 clinical interns in Chongqing Qianjiang Central Hospital who came from Chongqing Medical University, Jishou University, and Hubei University for nationalities were included in this study from June 2015 to June 2017. The test score, pass rates and make-up pass rates after 3 months of pre-job training of cardiopul-monary resuscitation were compared between training and evaluation group (2016 session) and control group (2015 session). We analyzed the correlation of retraining and evaluation of cardiopulmonary resuscitation and test score of pre-job training and its value of predicting the effectiveness of pre-job training. Results There were 65 clinical interns in 2016 session and 71 clinical interns in 2015 session. After 3 months training, the test score, pass rates and make-up pass rates in retraining and evaluation group were significantly higher than that in the control group, respectively (t=15.594,P=0.000; χ2=22.859, P=0.000; χ2=6.179, P=0.018). Conclusions Car-diopulmonary resuscitation skill training could enhance the effectiveness of pre-job training for clini-cal interns in non-affiliated teaching hospital and increase interns' ability to carry out cardiopulmonary resusci-tation and improve the quality of pre-job training for clinical interns. It could be used as an effective quality control strategy of clinical teaching
