1.Improved mesh pelvic reconstruction in treatment of Female Pelvic organ prolapse

Objective To explore the modified mesh in female pelvic floor reconstruction for pelvic organ prolapse and clinical effects of the application.Methods 78 patients with pelvic organ prolapse,line modified mesh pelvic floor reconstruction surgery(study group)and vaginal hysterectomy and vaginal repair of anterior and posterior wall(control group)treatment,clinical efficacy was compared.Results The mean operating time,blood loss,average body temperature,catheter indwelling time,residual urine volume,and postoperative hospital stay showed no significant difference(P>0.05).No postoperative infection,vaginal hematoma,mesh erosion occurred.Two groups of patients before discharge degree of POP-Q points are 0°or Ⅰ degree,to cure the standard.Vaginal length of the control group was significantly shorter than after treatment(P<0.05),while no significant change in the observation group(P> 0.05).Control group,the recurrence rate was significantly higher than the observation group(P<0.05),while the observation group of sexual satisfaction was better than the control group(P<0.05).Conclusion The modified mesh treatment of female pelvic floor reconstruction is an effective method of pelvic organ prolapse,the surgical curative effect is better,the recurrence rate is lower,worthy of clinical application.
2.Prediction of thermal environment via revision of PMV index with body temperature

PMV (Predicted Mean Vote) is a widely used index for evaluating the thermal environment. However, few studies have been conducted to take physiological values directly as evaluating indices. This paper assumes a linear relation between body temperature and both sweating rate and heat produced by shivering, and introduces the linear relation into the human heat balance equation to revise the classic PMV. And the assumption of linear relation is subsequently proved. The revised PMV possesses the same characteristic of dependent heat load as that of the classic one, and moreover it is convenient to be calculated.
3.The primordial study of application of primer dimer in the gene clone

Objective According to principle of forming primer dimer,a new cloning gene method was established. Methods Two and more oligonucleotide with complementary partnership in the 3′ end were synthesized by artifical method,oligonucleotide,Taq DNA polymerase and dNTP were mixed in the proportion of reasonable,then expanded and extended by PCR,result was identifed by agarose gel electrophoresis.Results A clear special band was appeared in the agarose gel. Conclusion Short fragment target gene less than 500 bp may be synthesized by this method.
4.Investigation of the correlation between posterior neck pain and lumbar epidural pressure during percuta-neous endoscopic lumbar discectomy

Objective To investigate the correlation between posterior neck pain and lumbar epidural pressure (LEP)during percutaneous endoscopic lumbar discectomy (PELD).Methods A prospective study was performed on 86 patients undergoing PELD,46 males,40 females,aged 1 9-71 years,with ASA physical status of Ⅰ or Ⅱ.Each patient received lumbar epidural anesthesia.Lum-bar epidural pressure (LEP)was monitored continuously through a lumbar epidural catheter which was connected to a pressure transducer.LEP before the operation (LEPbase ),LEP at the time of pos-terior neck pain (LEPpain )and maximal LEP (LEPmax )were recorded.Results Thirty patients (34.9%)complained of posterior neck pain during the procedure.The lowest LEPmax was 31.0 mm Hg,and the highest LEPmax was 77.0 mm Hg.The LEPmax in patients with neck pain [(60.6± 8.8)mm Hg]was significantly higher than LEPmax in patients without neck pain [(50.7 ± 9.5 ) mm Hg](P <0.01 ).Patients with higher LEPmax had higher probabilities of having posterior neck pain (P <0.01).Conclusion Patients with higher LEPmax had higher probabilities of having posterior neck pain.
5.Clinical relevance of distolingual roots and periodontal status in mandibular first molars:a cross-sectional study employing CBCT analysis

Objectives:Distolingual root of the permanent mandibular first molar(PMFM-DLR)has been frequently reported,which may complicate the treatment of periodontitis.This study aimed to assess the morphological features of PMFM-DLR and investigate the correlation between the morphological features of PMFM-DLR and periodontal status in patients with Eastern Chinese ethnic background.Materials and methods:A total of 836 cone beam computed tomography(CBCT)images with 1497 mandibular first molars were analyzed to observe the prevalence of PMFM-DLR at the patients and tooth levels in Eastern China.Among them,complete periodontal charts were available for 69 Chinese patients with 103 teeth.Correlation and regression analyses were used to evaluate the correlation between the morphological features of DLR,bone loss,and periodontal clinical parameters,including clinical attachment loss(CAL),probing pocket depth(PPD),gingival recession(GR),and furcation involvement(FI).Results:The patient-level prevalence and tooth-level prevalence of DLR in mandibular first molars were 29.4%and 26.3%,respectively.Multiple linear regression analysis suggested that bone loss at the lingual site and CAL were negatively affected by the angle of separation between distolingual and mesial roots in the transverse section,while they were significantly influenced by age and the angle of separation between distobuccal and mesial roots in the coronal section.Conclusions:The prevalence of PMFM-DLR in Eastern China was relatively high in our cohort.The morphological features of DLR were correlated with the periodontal status of mandibular first molars.This study provides critical information on the morphological features of DLR for improved diagnosis and treatment options of mandibular molars with DLR.
6.Tetrahedral DNA nanostructures synergize with MnO2 to enhance antitumor immunity via promoting STING activation and M1 polarization.

Stimulator of interferon genes (STING) is a cytosolic DNA sensor which is regarded as a potential target for antitumor immunotherapy. However, clinical trials of STING agonists display limited anti-tumor effects and dose-dependent side-effects like inflammatory damage and cell toxicity. Here, we showed that tetrahedral DNA nanostructures (TDNs) actively enter macrophages to promote STING activation and M1 polarization in a size-dependent manner, and synergized with Mn2+ to enhance the expressions of IFN-β and iNOS, as well as the co-stimulatory molecules for antigen presentation. Moreover, to reduce the cytotoxicity of Mn2+, we constructed a TDN-MnO2 complex and found that it displayed a much higher efficacy than TDN plus Mn2+ to initiate macrophage activation and anti-tumor response both in vitro and in vivo. Together, our studies explored a novel immune activation effect of TDN in cancer therapy and its synergistic therapeutic outcomes with MnO2. These findings provide new therapeutic opportunities for cancer therapy.
