1.Anticoagulation effect of LMWH combination with warfarin in treatment of intracranial venous sinus thrombosis at early stage

Objective To explore the anticoagulation effect of low molecular weight heparin (LMWH) combination with warfarin at the early stage at the treatment of intracranial venous sinus thrombosis.Methods 80 cases of intracranial venous sinus thrombosis patients in people's hospital of Xinjiang Uygur autononous region from January 2010 to December 2014 were chosen to be analyzed retrospectively.37 cases in observation group were treated with low molecular heparin (LMWH) in combination with warfarin, and 43 cases in control group were treated with warfarin.The clinical curative effect between two groups was compared post-treatment.Results The effective rate in observation group was 91.89%, which was significantly higher than 67.44% in control group (P<0.05) .The recanalization rate of involved intracerebral venous in observation group was 89.19%, which was significantly higher than 67.29% in control group(P<0.05).After treatment, the prothrombin time (PT), thrombin time (TT) and activated partial thromboplastin time (APTT) were higher and fibrinogen (FBG) in both groups was lower than those pre-treatment(P<0.05).The PT and APTT was higher and FBG was lower in observation group than those in control group(P<0.05).Conclusion Low molecular weight heparin in combination with warfarin worked well than single warfarin in the treatment of intracranial venous sinus thrombosis.
2.Role of histamine on the expression of tight junctions gene in airway epithelia

OBJECTIVE To investigate and elucidate the role of histamine on the regulation of tight junctions(TJs) gene expression of the airway epithelial cells. METHODS We cultured primary human bronchial epithelial cells from healthy and asthmatic individuals in the presence of histamine for 96 hours in air-liquid interface (ALI) culture system. The TJs expression on the level of mRNA and protein was determined by RT-qPCR and immunofluorescence (confocal microscopy), respectively. RESULTS A RT-qPCR analysis revealed that the mRNA expression of several TJs were up- or down-regulated by histamine. However, examination by confocal microscope revealed no detectable alterations in the morphology of the tight junctions after stimulating with histamine. CONCLUSION Our findings that histamine could regulate the mRNA expression of TJs of bronchial epithelial cells. That might contribute to the pathogenesis of chronic airway inflammatory diseases.
3.The value of CT and MRI in the diagnosis of hepatocellular adenoma

Objective To study the value of CT and MRI in the diagnosis of hepatocellular adenoma(HCA).Methods The comprehensive imaging features of HCA in 11 patients confirmed by pathology were analyzed retrospectively in our hospital.Results 11 patients presented with a total of 18 lesions, and plain CT showed low or slightly low density in all lesions, with irregular high density area in two lesions.The CT values of the lesions were significantly higher than in adjacent normal liver in the arterial phases(P<0.05).On T1WI, two lesions showed low or slightly low signal intensity, and 16 lesions showed slight hyperintensity.On T2WI, 14 lesions showed hyperintensity, and 4 lesions showed hypointensity.All lesions showed significant enhancement in arterial phase of dynamic enhanced scanning.Pseudocapsule enhancement was revealed in 17 lesions.For 18 lesions, ADC values were measured on DWI, and the ADC values of the lesions were higher than the value of surrounding normal liver parenchyma.Conclusion Dynamic contrast-enhanced CT combined with MRI and ADC value measurement can provide valuable information in diagnosis of HCA.
4.Effects of Jinzhuyalong Ershiwuwei coral capsule on PLA2 in degenarated cervical intervertebral discs

Objective To study the anti-inflammation mechanism of Jinzhuyalong Ershiwuwei coral capsule on cervical spondylosis by investigating Jinzhuyalong Ershiwuwei coral capsule on PLA2 in degenerated cervical intervertebral discs.Methods 60SD rats (SPF) were divided into a control group (15),a medicine model control group (15),a medicine modle 1 group (15),and a medicine model 2 group (15) by random lottery; Medicine model 1,2 groups were given Jinzhuyalong Ershiwuwei coral capsule powder 0.33 × 5 g/kg 0.66×5 g/kg,and added 1 ml distilled water dilution lavage,l/d,90 d,The unbalance of power cervical spondylosis animal models were chosen and C4~5 neck intervertebral discs were taken after 90 days.Millipore colorimetry methods were used to measure PLA2 activity in C4~5 cervical intervertebral discs.Results The PLA2 activity of the cervical intervertebral disc (12.37 ± 4.21) IU/mg · min-1 in the model group was significantly higher than the control group(2.56± 1.19)IU/mg· min-1 (P<0.05).The cervical intervertebral disc PLA2 activity of medicine model group 1 (8.56± 2.13)IU/mg · min-1 and medicine model group 2 (5.58 ±2.39)IU/mg· min-1 was lower than the model control group(12.37±4.21)IU/mg· min-1,with significant differences (both P<0.01).Conclusion Jinzhuyalong Ershiwuwei coral capsule could down regulate PLA2 activity in degenerated cervical intervertebral discs,and also decrease the production of some inflammaroty mediators.
5.The safety and feasibility of dual axis rotational angiography for the diagnosis of coronary artery disease in aged Chinese patients

Objective To evaluate the clinical safety and feasibility of dual-axis rotational coronary angiography (XperSwing) in diagnosing coronary artery disease in aged Chinese patients through comparing it with standard coronary angiography. Methods During the period from April 2011 to December 2012, a total of consecutive 151 patients with an age ≥ 65 years underwent diagnostic coronary angiography at authors’ hospital. The patients were randomly divided into the standard angiography group (n = 75) and XperSwing group (n = 76). The used dose of contrast, radiation exposure dose and operation time of each patient were recorded. Besides, the occurrence of procedure-related symptoms and arrhythmia during the procedure was recorded, and the blood pressure (BP), heart rate (HR) and creatinine level immediately before-and-after the procedure were estimated. The number of additional acquisition needed to be performed was used to evaluate the efficacy of the two methods. Results Coronary angiography was successfully accomplished in all patients of the two groups, and no significant difference in the number of additional acquisition existed between the two groups (P > 0.05). The used dose of contrast in XperSwing group was reduced by 44% when compared with that in the standard angiography group, i.e. (29.28 ± 5.06) ml vs(22 755.97 ± 11 239.22) mGycm2, (P<0.001). No statistically significant differences in the occurrence of procedure-related symptoms and arrhythmia during the procedure, in the operation time as well as in the postoperative changes of BP and HR existed between the two groups (P > 0.05). No contrast- induced nephropathy occurred in both groups, although the difference in postoperative changes of creatinine level between the two groups was statistically significant (P < 0.05). Conclusion This study indicates that in diagnosing coronary artery disease in aged Chinese patients, dual-axis rotational coronary angiography is more safe and effective than conventional coronary angiography, as XperSwing can significantly reduce the contrast dose and radiation exposure for the patients.
6.Bionic biphasic ceramic bioactive bone for segmental bone defects

BACKGROUND:The bioceramics has the ideal pore size, high porosity and the through-hole rate, can provide the ideal physiological activity space for the bone cel repair, and can obviously improve bone conduction. OBJECTIVE:To explore the bone conduction and bone induction in the repair of bone defects in the stage of bone defect of bionic biphasic ceramic bioactive bone. METHODS:A total of 20 New Zealand white rabbits were randomly divided into bioactive glass and biomimetic biphasic ceramic bioactive bone groups, and were used to construct the animal bone damage model. They were given the repair with bioactive glass and biomimetic biphasic ceramic bioactive bone. RESULTS AND CONCLUSION:At 4 weeks after model establishment, scanning electron microscopy demonstrated that dense periosteal tissue was observed in the biomimetic biphasic ceramic bioactive bone group. At 8 weeks, dense combination was found, and no obvious fissure existed. At 12 weeks, complete bone demarcation blurred, showing a natural transition. Moreover, the binding site was very dense. There were a large number of new bone tissues, bone trabecula was regular and connected to a piece. The bone material has been largely degraded. Bone defects were repaired completely. The bone density was close to normal bone. At 8 weeks, in the bioactive glass group, the binding site presented obvious fissure. At 12 weeks, the fissure had been connected, but the binding was not tight as compared with the bionic biphasic ceramic biologic active bone group. The bone defect got preliminary repair. A smal number of new bone formed trabecular bone, but could not connect or traverse. There was no recanalization of the marrow cavity. A few continuous bone cal us traversed the broken end. These data demonstrate that bionic biphasic ceramic bioactive bone has good bone conduction, bone induction and biocompatibility in the repair of segmental bone defects.
7.The protective role of peroxisome proliferator activated receptors-α and pathomechanism in D-galactosamine/lipopolysaccharide-induced acute liver failure in rats

Objective To determine the role and mechanism of peroxisome proliferator activated receptors (PPAR) α in a mouse model of D-galactosamine/lipopolysaccharide (D-GalN/LPS)-induced acute liver failure(ALF).Methods Firstly,C57BL/6 mice were randomly divided into control group(n =8),ALF 2h group(n =8),ALF 4h group (n =8),ALF 6h group (n =8).Secondly C57BL/6 mice were randomly divided into control group(n =8),ALF group(n =8),WY14643 group(n =8).To induce ALF,the mice were injected intraperitoneally with D-GalN (700 mg/kg) and LPS (10 μg/kg).WY14643 (6 mg/kg),the selective agonist of PPAR α,was administered via tail vein two hours prior to D-GalN/LPS exposure.Two,four,and six hours after D-GalN/LPS treatment in the first study,mice were anesthetized and blood was collected,6h after D-GalN/LPS treatment in the second study,blood was collected.The liver tissue was harvested for histology and mRNA extraction.Serum levels of ALT and AST were measured to evaluate the hepatic damage.Inflammatory cytokines (TNFα,IL-1β,IL-6) and chemokines (CXCL-1,CXCL-10) were detected by real-time quantitative PCR.Differential protein expression of p-NF-κBp65,p-JNK,p-ERK,p-p38 in inflammatory pathways was detected by Western blotting.Significance of inter-group differences was assessed by one-way ANOVA,and pairwise comparison was performed by the least significant difference test.Results The gene and protein expression of PPAR α were gradually reduced during the development of ALF.Compared with the model group,the liver architecture was better preserved almost with normal morphology in WY14643-treated mice.Serum ALT and AST levels in WY14643-treated group were significantly lower [ALT:(555 ±62)U/L vs (2 898 ±822) U/L,P <0.05; AST:(791 ±58) U/L vs (3 013 ±997)U/L,P < 0.05].The expression of proinflammatory cytokines and chemokines was significantly suppressed during the activation of PPAR α.In the second study,the levels of gene expression of proinflammatory cytokines and chemokines were detected in control group,ALF group and WY14643 group respectively as followings:TNFα (0.161 ± 0.085,7.996 ± 1.068,3.346 ± 0.94,P < 0.05),IL-1β(0.041 ±0.002,3.657 ±0.904,0.176±0.089,P<0.01),IL-6 (0.018 ±0.008,1.762 ±0.589,0.163±0.0487,P <0.05),CXCL-1 (0.063 ±0.008,7.881 ±0.966,2.737 ±0.864,P <0.01),CXCL-10 (0.054 ±0.005,5.671 ±0.948,2.578 ±0.804,P <0.05).Conclusion Our findings first demonstrate that PPARα protects liver from injury in an ALF mouse model by suppressing inflammatory response,indicating PPARα as a potential new therapeutic target for ALF.
8.Expressions of EZH2 and Ki-67 in ameloblastoma and their clinical significance

Objective To study the relationship between EZH2 and Ki-67 expression and both in ameloblastoma and normal oral mucosa.Methods EZH2 and Ki-67 were detected by immunohistochemical SP method in 50 cases of ameloblastoma (30 cases primary,20 cases recurrence) and expression in 20 cases of normal oral mucosa.Results The positive expression of EZH2 protein in recurrent ameloblastoma was (36.25±7.24) %,significantly higher than that of primary ameloblastomas (25.26±4.28) % (P < 0.001).Ki-67 in recurrent positive expression in ameloblastoma was (34.96±5.28) %,significantly higher than that of primary ameloblastomas (29.68±3.27) % (P < 0.05).There was positively corTelated on EZH2 and Ki-67 (P < 0.05),but not on the size of tumor and gender (P > 0.05).Conclusions There are high expression of EZH2 and Ki-67 in ameloblastoma,and their play key role in the occurrence and development of ameloblastoma.They can be used as a reference index for the prognosis.The combined detection of EZH2 and Ki-67 protein expression with clinical evidence and its prognosis may contribute to the process of ameloblastoma and recurrence.

OBJECTIVE:To establish a method for the contents determination of gallic acid,naringin,hesperidin,neohesperi-din and baicalin in Qinghou liyan granule. METHODS:UPLC was performed on the column of ACQUITY UPLC BEH C18 with mobile phase of acetonitrile- 0.1% phosphoric acid(gradient elution)at a flow rate of 0.40 ml/min,detection wavelength was 280 nm,column temperature was 30 ℃,and injection volume was 1 μl. RESULTS:The linear range was 0.048-0.480 μg for gallic ac-id(r=0.999 3),0.012-0.120 μg for naringin(r=0.999 9),0.016-0.160 μg for hesperidin(r=0.999 8),0.022-0.220 μg for neo-hesperidin(r=0.999 9)and 0.072-0.720 μg for baicalin(r=0.999 2);limit of quantitation was 2.4,1.6,1.8,1.8,1.6 ng,limit of detection was 7.5,5.0,5.6,5.6,5.0 ng;RSDs of precision,stability and reproducibility tests were no lower than 3.0%;recover-ies were 96.64%-102.02%(RSD=2.00%,n=6),95.86%-102.56%(RSD=2.86%,n=6),97.24%-102.54%(RSD=2.10%,n=6),97.44%-102.60%(RSD=2.40%,n=6)and 96.91%-103.13%(RSD=2.62%,n=6). CONCLUSIONS:The method is rapid and accurate,and can be used for the simultaneous determination of gallic acid,naringin,hesperidin,neohesperidin and baicalin in Qinghou liyan granule.
