1.Lumbar spinal implants for degenerative lumbar spondylolisthesis: biocompatibility evaluation
Chinese Journal of Tissue Engineering Research 2015;19(16):2608-2612
BACKGROUND:Spinal internal fixation is widely used in the treatment of lumbar spondylolisthesis, with the purpose of lumbar fusion, reconstruction of lumbar sagittal sequences, relieving spinal cord or nerve root compression. OBJECTIVE:To review the biocompatibility of lumbar spinal implants applied in the treatment of degenerative lumbar spondylolisthesis. METHODS:A computer-based retrieval of PubMed and CNKI was performed for relevant articles published from January 2000 to January 2015. The keywords were biocompatibility, degenerative lumbar spondylolisthesis, implant in English and Chinese, respectively, which appeared in the title and abstract. RESULTS AND CONCLUSION:Lumbar spinal implants commonly used include pedicle screws and interbody fusion cages. The fusion cages are mainly classified as biological fusion cage, metal fusion cage, composite fusion cage and absorbable fusion cage. Internal fixation implants made of different materials have different biocompatibilities, but also develop different stabilities. With the development of bionics, 3D printing technology, biomechanics and materials science, lumbar spinal implants wil have better biocompatibility, bone substitute capability and biomechanical properties.

3.Detection of Mycobacterial16S rRNA Gene and M.tuberculosis DNA in Skin Lesions of Sarcoidosis with Polymerase Chain Reaction
Chinese Journal of Dermatology 1995;0(04):-
Objective To investigate the possible role of mycobacterial infection in the pathogenesis of sarcoidosis. Methods DNA fragments of mycobacterial16S rRNA gene and the complex-specific insertion sequence IS6110of M.tuberculosis were detected by polymerase chain reaction(PCR)in formalin-fixed,paraffin-embeded skin biopsy specimens from12patients with sarcoidosis. Results Mycobacterial common sequence16S rRNA gene was found in6patients,among which5were further confirmed as M.tuberculosis, the other one was atypical mycobacteria. The IS6110DNA fragment amplified by PCR was confirmed by DNA sequencing. Conclusion The above findings suggest an aetiological role of mycobacterial infection, especially M.tuberculosis,in some patients with sarcoidosis.
4.Cloning of Human Ro60 cDNA and Construction of Baculovirus Expression Vector
Chinese Journal of Dermatology 1994;0(02):-
Objective To construct a baculovirus expression vector for expression of human Ro60 cDNA. Methods Ro60 cDNA was cloned from Hela cell RNA with RT-PCR and was inserted into plasmid pFastBac HTc. Plasmid pFastBac HTc-Ro60 was then transformed into E. coli DH10Bac competent cells for transposition. The constructed recombinant was determined by both blue-white colony screening and PCR analysis. Results A 1.56kb Ro60 cDNA was obtained from Hela cell RNA and was successfully inserted to plasmid pFastBac HTc. DNA sequencing and restriction enzyme analysis revealed that both open reading frame and sequence of the cDNA were identical to Ro60 sequence previously published. Specific transposition and virus recombination were obtained in the vector Bacmid-Ro60. Conclusion Ro60 cDNA has been successfully cloned and constructed to baculovirus expression vector, which is useful for protein expression and further study of this autoantigen in the pathogenesis of lupus erythematosus.
5.Detection and identification of Mycobacteria with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP)from patients with Mycobacterial skin infections
Journal of Peking University(Health Sciences) 2003;0(05):-
Objective: To establish a rapid approach to the detection and identification of Mycobacteria from lesions of patients with suspected Mycobacterial infections. Methods: Specimens were obtained from five patients suspected to have Mycobacterial infections. DNA extracted from clinical samples was amplified by nested PCR. The PCR products were digested with Hha Ⅰ, MboⅠ, and BstUⅠ restriction enzymes and applied to PAGE. The species of Mycobacteria were determined by restriction fragment length polymorphism (RFLP) analysis. Identification of Mycobacteria culture was also performed in 3 patients. Results: M. marinum was found in two patients diagnosed as swimming pool granuloma. M. tuberculosis was found in one patient diagnosed as infectious skin granuloma. All these 3 Mycobacteria were confirmed by Mycobacteria culture. A strain of M. tuberculosis and a strain of M. fortuitum were detected in remain two patients. Conclusion: The results above indicate that PCR-RFLP analysis is rapid and reliable in detection and identification of different Mycobacteria species from skin tissues. Application of this method will be helpful for early diagnosis and treatment of Mycobacteria skin infections.
