1.Different expressing levels of osteoprotegerin and receptor activator of nuclear factor-?B ligand from osteoblasts of young, adult and aged mice

Objective To investigate different expressing of OPG and RANKL expression from osteoblasts stimulated with estrogen of young, adult and aged mice changing with age. Method We employed RT-PCR and ELISA methods to measure the OPG/RANKL expression from osteoblasts before and after co-cultured with 17?-estradiol. Results OPG expressing from osteoblasts of the yound and adult mice was significantly increased after stimulated with estrogen, however, RANKL expressing wasn′t decreased significantly, compared to from osteoblasts of the aged. There were no significantly difference among osteoblasts co-cultured with methyl-piperidino-pyrazile (MPP). Conclusion Our results suggest that estrogen may exert its anti-resorptive effects on bone, at least in part, by increasing OPG and decreasing RANKL expression in osteoblasts. Decreased OPG and increased RANKL may be one of important reeasons of postmenopausal osteoporosis.
2.Treatment of Cervical Spondylopathy in Nerve Root Pattern by Electroacupuncture plus Tuina Therapy

In the treatment of 126 cases of cervical spondylopathy by electroacupuncture on Jiaji (Ex-B 2),Jianyu (LI 15), Quchi (LI 11) and Hegu (LI 4), in combination with Tuina therapy, the results showed cure in 74 cases, improvement in 49 cases, failure in 3 cases and the total effective rate in 97.6%, after four courses of the treatment.
3.Study on the epiemiological patterns of HIV infection in HongKong by a mathematic model

Objective To study the epidemiological patterns of HIV infection prevalent in HongKong.Method The epidemiological patterns of HIV infection in HongKong were analyzed with a mathematic model SI by changing its initial condition,which was used to test the related data.Results The changeable patterns of susceptible accumulative numbers of HIV infection in HongKong and the epidemiological curves and the curve of infection velocity were obtained.Conclusion The epidemic pattern of HIV infection in Hongkong and local infection velocity from 1987 to 2004 were well described using the epidemic model SI.An accumulation of 2 733 cases of HIV infection will be predicted in the year of 2005.The epidemic peak occurred in 2002 to 2004 and the overall HIV epidemic will last for 50 years approximately.
4.EFFECTS OF SELENIUM-ENRICHED PROBIOTICS ON IMMUNITY AND ANTIOXIDATION FUNCTION IN MICE

Objective: To investigate the effects of selenium-enriched probiotics on immunity and antioxidation function in mice. Method: One hundred healthy KM mice, female and male in half, were randomly divided into control, sodium selenite, selenium-enriched yeast (Se yeast), selenium-enriched probiotics (Se probiotics) and probiotics groups. One milliliter of water, sodium selenite (2 ?g Se/ml), Se yeast (2 ?g Se/ml), Se probiotics (2 ?g Se/ml) or probiotics were respectively supplemented to five groups in oral (ig) every day. Whole experiment lasted for 28 d. During the experiment, immunity and antioxidation functions were measured respectively. Results: The activation of peritoneal macrophage and thymus index in Se probiotics group were significantly higher than those of other four groups. The spleen index of Se probiotics group was higher than that of control, sodium selenite and probiotics group. The spleen lymphocyte transformation rate in Se probiotics group was significantly or very significantly higher than that in control or probiotics group. In addition, blood GSH-Px and plasma SOD activity in Se probiotics group were significantly higher than those in control and sodium selenite group. Plasma MDA concentration in Se probiotics group was very significantly lower than that in control and sodium selenite group. Conclusion: Selenium-enriched probiotics supplementation could significantly enhance immunity and antioxidation function in mice.
5.EFFECTS OF SELENIUM ENRICHED PROBIOTICS ON ANTIOXIDATIVE ACTIVITIES AND IMMUNE FUNCTIONS IN WEANLING PIGLETS

Objective: To investigate the effects of selenium enriched probiotics on antioxidative activities and immune functions in weanling piglets. Methods: Twenty-four healthy weanling piglets (Duroc?Landrace?Yorkshire ) were divided into 3 groups. One served as control group (C) and two were supplemented different selenium (Se) sources namely selenium enriched probiotics (T1) and Na selenite (T2) as test groups respectively . The experiment lasted for 60 days. The blood GSH-Px activity, blood Se concentration, serum superoxide dismutase (SOD) activity, serum malonaldehyde (MDA) level, serum antibody level of swine fever, and the tissue content of selenium were determined. Results: Blood GSH-Px , Se concentration and serum SOD activity of T1, T2 groups were higher than those of control group after 60 d Se supplementation, and serum MDA content was markedly lower than control group. At the same time, the serum antibody level of swine fever, the tissue content of selenium in T1, T2 group were much higher than control group, and T1 group was higher than T2 group. Conclusion: Organic Se supplementation could significantly enhance immune function and antioxidative activities of weanling piglets.
6.Value of heart-type fatty acid-binding protein in predicting prognosis of patients with sepsis

Objective To study the predictive value of heart-type fatty acid-binding protein (H-FABP) in the improvement of survival rate in patients with severe sepsis.Methods There was a prospective case-control study in 50 patients with severe sepsis from October 2014 to October 2015.According to the international guidelines for severe sepsis and septic shock set in 2012,all patients were divided into sepsis group (n =16),severe sepsis group (n =14) and septic shock group (n =20),and these patients were further divided into to survival group (n =22) and non-survival group (n =28) as per patients surviving within 28 days.Gender,age,ethnic,and Acute Physiology and Chronic Health Evaluation score (APACHE Ⅱ) were recorded.H-FABP,B-type natriuretic peptide (BNP),creatine kinase (CK),creatine kinase isoenzyme (CK-MB),and troponin-T (cTn-T) were determined within 6 hours after admission.The statistical software SPSS21.0 was used to do t test,test,and the receiver operating characteristic curve for comparison between groups.Results H-FABP in septic shock group was higher than severe sepsis group and sepsis group (P < 0.01).The 28-day mortality of septic group (80%) and severe sepsis group (71.4%) were higher than sepsis group (12.5%) (P < 0.01).Non-survival' s H-FABP,BNP,cTn-T,CK,CKMB was higher than survivals (P < 0.05).Through the ROC of H-FABP (AUC=0.748,P=0.003,95% C I:0.605-0.890) and BNP (AUC =0.714,P=0.010,95% CI:0.573-0.856),it shows that H-FABP is better than BNP.when H-FABP take 9.902 ng/mL,the sensitivity of 82.1%,specificity of 63.6%.H-FABP,BNP has a certain value in 28-day mortality prediction.Conclusions Sepsis shock has significantly higher 28-day mortality than sepsis and severe sepsis.Compared with BNP,H-FABP has greater predictive value in sepsis patients,it will increase along with the aggravation of the disease;it can also predict 28-day mortality.
7.Effects of benvitimod on levels of interleukin-4 and -10, interferon-γ and transforming growth factor-βin patients with atopic dermatitis

Objective To detect levels of interleukin?4(IL?4), IL?10, interferon?γ(INF?γ)and transforming growth factor?β(TGF?β)in the culture supernatant of peripheral blood mononuclear cells (PBMCs)from patients with atopic dermatitis(AD), and to evaluate regulatory effects of benvitimod on these cytokines. Methods PBMCs were isolated from 20 AD patients and 20 healthy controls. Then, PBMCs from AD patients were equally divided into 4 groups to be cultured with phosphate?buffered saline (PBS group), 400 nmol/L benvitimod solution (benvitimod group), 250 nmol/L dexamethasone solution (dexamethasone group) and 10 nmol/L tacrolimus solution (tacrolimus group), respectively. Enzyme?linked immunosorbent assay(ELISA)was performed to detect levels of IL?4, IL?10, INF?γand TGF?βin the culture supernatant of PBMCs. Results Compared with healthy controls, patients with AD showed significantly higher level of IL?4(83.4 ± 12.2 vs. 44.3 ± 5.7 pg/ml, P<0.05), but lower levels of INF?γ(12.5 ± 2.3 vs. 25.4 ± 3.4 pg/ml, P<0.05)and IL?10(144.4 ± 4.1 vs. 189.9 ± 6.5 pg/ml, P<0.05)in peripheral blood. However, no significant difference in the level of TGF?βwas observed between the patient group and control group(178.9 ± 17.40 vs. 158.7 ± 18.30 pg/ml, P>0.05). Compared with PBS, 400 nmol/L benvitimod could decrease the expression of IL?4(50.2 ± 10.1 vs. 83.3 ± 12.2 pg/ml, P < 0.05), IL?10 (124.7 ± 17.5 vs. 144.4 ± 4.1 pg/ml, P>0.05)and INF?γ(9.56 ± 5.1 vs. 12.5 ± 2.3 pg/ml, P>0.05), but increase the expression of TGF?β(203.6 ± 15.3 vs. 178.9 ± 17.4 pg/ml, P>0.05)by PBMCs. In addition, no significant differences in the expression of IL?4, IL?10, INF?γ or TGF?β were observed between the benvitimod group and dexamethasone group or tacrolimus group(all P>0.05). Conclusion Benvitimod can regulate the expression of IL?4, IL?10, INF?γand TGF?βby PBMCs in patients with AD.
8.Construction of a recombinant Escherichia coli for high trehalose production.

Trehalose, a compatible solute, is widely used in food, cosmetics, pharmaceutical products and organ transplantation. Nowadays, trehalose is mostly produced by enzymatic synthesis with many secondary products and lowpurity. In this study, high amount of trehalose was produced by recombinant E. ccli fermentation. First, a bifunctional trehalose gene TPSP was amplified from genome of C. hutchinscoii. Second, an expression vector pTac-HisA containing TPSP was constructed and transformed into the host E. coli. Expression of this bifunctional enzyme-TPSP converted glucose to trehalose. The result suggested that TPSP from C. hutchinsonji has been successfully expressed in E. ccoi. High amount of extracellular trehalose generated from glucose by whole-cell catalysis and After optimization, the production of trehalose in shake flasks was improved to 1.2 g/L and the relative conversion rate reached 21%. The production in bioreactor reached 13.3 g/L and the relative conversion rate reached 48.6%. It is the first time to realize the functional expression of the bifunctional enzyme-TPSP of C. hutchinsonii in E. coli and achieved the conversion form glucose to trehalose. This study laid a foundation for industrial large-scale production of trehalose.

9.Coefficients between TEOAEs and DPOAEs in infants

bjective:To investigate the coefficients of TEOAEs and DPOAEs based on various pass/fail criteria. Method:Thirty-six Australia infants were tested (age range: 1~6 months) using both TEOAE and DPOAE. Cohen′s Kappa (K) were used to analyze different criteria.Result:There is a significant low agreement in the screening outcomes between TEOAEs and DPOAEs, and different criteria for TEOAEs.Conclusion:The findings from the study of coefficients among commonly used screening pass/fail criteria suggested that there is no simple relationship among them. The possible reasons for weak agreement were discussed.
10.Necessity of replanning for the change of anatomy and dosimetry during intensity modulated radiotherapy for nasopharyngeal carcinoma

Objective To study the dynamic change of anatomy and dosimetry distribution and its influence during intensity modulated radiation therapy(IMRT) for nasopharyngeal carcinoma(NPC). Methods From June 2006 to August 2007,12 patients with stage Ⅲ-Ⅳ NPC receiving initial IMRT concurrently combined with chemotherapy were included in the study. The target volumes and,involving organs were contoured on the first set of spiral CT images. When IMRT planning was finished,a second CT scans was acquired to rectify the treatment center. Weekly spiral CT images acquired during the treatment period according to the same treatment center were fused with the first CT images. In order to determine the best time of replanning, modified plan target volume( PTV1 ) and normal organs( parotids and outline) were contoured on the fusion interface by a single physician. Changes of each structure throughout treatment period were measured by a system software. Then the dose distributions were computed and evaluated for replanning CT using the same beams arrangement in the original plan. Cumulative dose was estimated compared with the original plan. Results The volume of outlines and parotids decreased gradually, and the change came to peak in the fifth week. So the fifth and first week CT scans were selected as research objectives. No significant changes in maximal and mean dose was observed in the brainstem, spinal cord or mandible. Despite volume changes,D99 and D95 of the PTV1 did not change siguificantly(P>0.05). D95 of the bilateral parotids increased significantly ( PL = 0.03,PR = 0.01 ). Conclusions During IMRT for NPC, the volume of PTV1, the outlines and parotids decrease,and the change come to peak in the fifth week. Comparing to the first treatment plan, the dose parameters of the parotids increase significantly in the second plan based on the fifth week CT,but those of the brainstem,cord,mandible and PTV1 change slightly.
