Aim To investigate the effects of heme oxygenase-1(HO-1) on brain damage due to cerebral ischemia/reperfusion.Methods 120 male mice were divided into four groups randomly and the cerebral ischemia/reperfusion(I/R) model was established through drawing out and reperfusing 40% of the whole blood volume in combination with clamping the carotid arteries for 20 min after anesthesia with 40 g?L-1chloral hydrate(400 mg?kg-1 ip).The mice were intracerebraventricularly(icv) injected with 3(l of150 ?mol?L-1 hemin,150 ?mol?L-1 ZnPPIX,or artificial cerebrospinal fluid(ACSF),respectively 16 h before the model established.Sham operation group only received anesthesia and operation without I/R and icv injection. The hippocampal HO activity and cerebral cortex xanthine oxidase(XO) activity, the levels of malonaldehyde(MDA) and reactive oxygen species(ROS),hippocampal neuron apoptosis, and hippocampal HO-1 expression were determined by spectrophotometer,TUNEL method, and Western blot, respectively. Results The increased hippocampal HO-1 expression and HO activity, decreased XO activity,MDA and ROS levels, and diminished hippocampal apoptosis were observed in the hemin group, compared with ACSF group(P0.05),but inhibited the activity of HO with the elevation of XO activity,MDA and ROS levels,as well as cellular apoptosis(vs ACSF,P

AIM:To study the reversal effects of multidrug resistance by transfecting tumor necrosis factor ?(TNF-?) cDNA and multidrug resistant 1(MDR1) gene antisense RNA into multidrug resistant breast cancer cell line MCF-7/ADR.METHODS:The recombinant vector of enhanced green fluorescent protein(EGFP) with MDR1 antisense RNA and recombinant vector of red fluorescent protein(DsRed2) with TNF-? cDNA were constructed by RT-PCR and DNA recombinant techniques. The recombinant vectors were transfected into multidrug resistant breast cancer cell line MCF-7/ADR.The cell growth curves, cell apoptosis rates,MDR1 gene expression at mRNA and P-gp levels, and the sensitivity to ADR were determined before and after the transfection.RESULTS:After the transfection, cells showed lower growth rate, higher apoptosis rate, lower MDR1 expression at mRNA and P-gp levels, and the sensitivity to ADR increased significantly.CONCLUSION:Transfection of TNF-? cDNA and MDR1 antisense RNA into multidrug resistant breast cancer cells may have good effects on reversal of multidrug resistance.

Objective To observe the efficacy and toxicities of RF transparent heating (RTH) combined with transcatheter hepatic arterial chemoembolization (TACE) in the treatment of advanced primary hepatic carcinoma. Methods In a randomized manner, 69 patients with advanced primary hepatic carcinoma were divided into two groups: study group (TACE+RTH) 34 cases and control group (TACE alone) 35 cases, the control group were treated with DDP 80mg, FU 1000mg and E-ADM 60mg, E-ADM was used with iodized oil embolism 10ml. Results The total effective rate in the near future were 70.59% and 45.71%, the overall survival rates of 0.5, 1.0, 1.5, 2.0 years were 82.35%, 73.53%, 58.82%, 38.24% in study group and 74.29%, 75.14%, 45.71%, 22.86% in control group, respectively. Toxicities were similar comparing with the two groups. Conclusions RTH combined with TACE in the treatment of advanced primary hepatic carcinoma is better than TACE alone, at the same time TACE +RTH method is no increasing toxicity and is an effective safe combined one.
