Acrylamide(AA)is a chemical used in many industries around the world and more recently was found to form naturally in foods cooked at high temperatures.Acrylamide was shown to be a neurotoxicant,reproductive toxicant,and carcinogen in animal species.Only the neurotoxic effects were observed in humans and only at high levels of exposure in occupational settings. The present review is a synopsis of research on the genotoxicity and reproductive toxicant of acrylamide.

5.Effects of Simvastatin on Oxidative Stress and Cell Apoptosis in Aged Mice with Myocardial Ischemia-re-perfusion

OBJECTIVE:To study the effects of simvastatin on oxidative stress and cell apoptosis in aged mice with myocardi-al ischemia-reperfusion (IR). METHODS:Aged mice were randomly divided into sham operation group (phosphate buffer solu-tion),model group(phosphate buffer solution)and simvastatin low-dose,medium-dose and high-dose groups(2.5,5 and 20 mg/kg) with 14 mice in each group. Those groups were given relevant medicine intraperitoneally before modeling for 7 d,once a day. IR model was induced in those groups except for sham operation group. The area ratio of myocardial infarction,myocardial cell apop-tosis rate,activity of myocardial tissue apoptosis gene Caspase-3,the protein expression of Bax and Bcl-2,Akt phosphorylation, serum concent of MDA and activity of SOD were all detected. RESULTS:Compared with sham operation group,the area ratio of myocardial infarction,myocardial cell apoptosis rate,Caspase-3 activity,the protein expression of Bax and MDA content were all increased in model group,while the protein expression of Bcl-2,Akt phosphorylation and SOD activity were decreased(P<0.01). Compared with model group,the area ratio of myocardial infarction,myocardial apoptosis rate,Caspase-3 activity,the protein ex-pression of Bax and MDA content were all decreased in simvastatin high-dose group,while the protein expression of Bcl-2,Akt phosphorylation and SOD activity were increased (P<0.01). There was no statistical significance in above indexes in simvastatin low-dose and medium-dose groups (P>0.05). CONCLUSIONS:Simvastatin can relieve myocardial IR injury in aged mice,and the mechanism of which may be associated with inhibiting myocardial cell apoptosis and the generation of oxidative stress.
6.Establishment and preliminary application of measuring method for three-dimensional kinematics of the hindfoot joint in vivo

Objective To establish a new method for measuring the 3-D kinematics of hindfoot joints in vivo by using reverse engineering software together with the theory of rigid body kinematics.Methods CT images were gathered from 9 feet of 5 healthy volunteers in both the initial position (neutral position) and the terminal position (extremely inversion-adduction-dorsiflexion position).The 3-D digital modules of hindfoot joints in the initial position and terminal position were established with MIMICS 10.01 software.The data of reconstructed digital modules was inputted into the GEOMAGIC 10.0 software in STL format for twice registration,and then their relatively displacement and changes of angle in 3-D space between the two positions were calculated Results The rotation range of the tibiotalar joint was 3.89° ±2.77° in eversion,5.29°±4.47° in abduction,10.77°+5.70° in dorsiflexion,and the relative displacement was 0.78±0.59 mm towards lateral ankle,0.18±0.75 mm towards the hindfoot,(0.65±0.71) mm towards the proximal limbs;the range of subtalar joint was 16.46°±2.94° in inversion,12.77°±1.81° in adduction,6.33°±4.32° in plantarflexion,and the relative displacement was 5.50±1.45 mm towards medial ankle,1.96±1.77 mm towards forefoot,0.43±1.18 mm towards distal limbs; the range of talonavicular joint was 38.82°±5.98° in inversion,19.71°±6.33° in adduction,5.09°±6.89° in plantarflexion,and the relative displacement was (9.77±1.73) mm towards medial ankle,3.13±1.29 mm towards hindfoot,4.64±1.42 mm towards proximal limbs.Conclusion This method measuring 3-D kinematics of hindfoot joints in vivo is non-invasive and easy to operate.

OBJECTIVETo analyze naringin, naringenin and its metabolites in rat plasma after intragastric administration of exocarpium Citri grandis alcohol extract.
METHODRat blood samples were collected 1.0 hour after oral administration of 50 g x kg(-1) exocarpium Citri grandis alcohol extract and analyzed by UPLC-Q-TOF with MS(E) function. The post-acquisition data were processed using Metabolynx.
RESULTNaringin (M1), naringenin (M2), naringin-5-O-glucuronide (M3), naringin-4-O-glucuronide (M4), glucuronide conjugate of naringenin (M5), naringin-4-O-sulfate (M6), methylated conjugate of hydroxylated naringenin (M7), glucuronide and sulfate conjugate of naringenin (M8), glucuronide conjugate of hydroxylated naringenin (M9) in rat plasma were detected. M3, M4, M6 were first reported as the metabolites of naringin. M7, M9 were first reported as the metabolites of naringenin.
CONCLUSIONThe results indicated that naringin, naringenin can be metabolited as the forms of glucuronidation, sulfation and naringenin can also be metabolited as the forms of methylation with hydroxylation and glucuronidation with hydroxylation in vivo after administration.

8.Genetic quality analysis of 24 domestic inbred mouse strains by microsatellite DNA

Objective To analyze the genetic quality of 24 domestic inbred strains mice using microsatellite loci panel.Methods Previously selected 30 microsatellite loci of mouse with high polymorphism and more allele numbers were used to synthesize corresponding fluorescently-labeled primers.Then the genomic DNA samples of each mouse were amplified by PCR and the products were analyzed by STR scanning to genotype the inbred strains of mice.Results Out of the 24 inbred strains, 15 inbred strains showed the same genotype within one strain at 30 loci.Among different strains, microsatellite loci indicated polymorphism which could be used to distinguish different strains.However, the rest 9 strains demonstrated polymorphism within strains.Conclusions Our stuoly provides a useful microsatellite panel to detect genetic quality of inbred mice and distinguish different strains with the optimized microsatellite loci.
9.Construction of evaluation index system for full-time scientific researchers in Level-three general hospital

Objective:To establish a set of comprehensive appraisal indicator system for the full-time scientific research personnel in Level-three general hospitals, improve the management of full-time scientific research personnel at hospital.Methods:Firstly, an initial indicator system was established by literature review and in-depth interviews with experts. Secondly, Delphi method was used to screen the indicators to confirm the indicator system. Finally, Analytic Hierarchy Process (AHP) was used to determine the weights of indicators at all levels, consistency tests were also conducted.Results:A set of comprehensive appraisal index system for full-time scientific research personnel in Level-three general hospitals was established, including four first-level indicators, which covered the comprehensive quality, scientific research capacity, scientific research performance and academic impact, as well as other 17 secondary indicators and 53 third-level indicators. Among these indicators, the scientific research performance has the largest weight value (0.5224), and according to the consistency test results, CR was less than 0.1. Besides, through the consistency test, the weight assignment is reasonable.Conclusions:The appraisal index system of full-time scientific research personnel in Level-three general hospitals is reliable. It can be used as a tool for evaluation of full-time scientific research personnel, which also provide reference for other hospitals to improve the management of full-time scientific research personnel.
10.Correlation between talonavicular joint movement and changes of the medial longitudinal arch.

OBJECTIVETo explore the characteristics of the talonavicular joint movement in vivo and its effects on changes of the medial longitudinal arch.
METHODSFoot CT images in the initial position (neutral position) and terminal position (maximum varus-adduction-dorsiflexion position) were acquired from 9 cases (5 healthy volunteers, including 4 males and 1 female) during foot varus-adduction-dorsiflexion motion. Based on the principle of rigid body kinematics, the CT data were reconstructed and analyzed with Mimics and Geomagic reverse engineering software. The changes of the talonavicular joint in three-dimensional in 6 degrees of freedom were calculated to determine its correlation to the medial longitudinal arch angle.
RESULTSDuring foot varus-adduction-dorsiflexion motion, the talonavicular joint underwent varus-adduction-plantarflexion motion, with the motion range of 38.82∓5.98° in varus, 19.71∓6.33° in adduction, and -5.09∓6.89° in plantarflexion. During talonavicular joint motion, the medial shift of the navicular was significantly correlated to the changes of foot medial longitudinal arch (P<0.05).
CONCLUSIONDigital technology can solve the problem of measurement of talonavicular joint three-dimensional motion in vivo. Though as a ball-and-socket joint, the talonavicular joint mainly rotates around the sagittal axis, and its movement is a major factor to cause changes of foot medial longitudinal arch.
