3.Dectin-1 mRNA expression in lung of different immune status mice infected with Aspergillus fumigatus

Objective To study dectin-1 mRNA expression in lung of different immune status mice infected with Aspergillusfumigatus, and to explore the influence of immunosuppressive agent on the expression of dectin-1 and its relationship with the progression of disease. Methods Mice were divided into four groups which were normal control, immuncompromised, immuncompromised with A.fumigatus inoculation and immuncompetent with A.fumigatus inoculation groups. We explored the kinetic mRNA expression of dectin-1 in lung of different groups by real-time quantitative PCR. Pulmonary fungal burden assessment was performed to reflect the progressing of disease during the experimental time course. Results On day 3 after inoculation, pulmonary fungal burden of the immuncompromised mice was higher than that of the immuncompetent group. On day 1 and 3, dectin-I mRNA expression in lung of the immuncompromised group was much lower than that of the normal control. On day 3, dectin-1 mRNA expression in lung of the immuncompetent mice infected with A.fumigatus was much higher than that of the immuncompromised with A.fumigatus inoculation and the normal control groups ( P < 0.05 ). Conclusion During the infection, expression of dectin-1 in lung of the immuncompetent group was strikingly increased, which may play an important role on the defence to A.fumigatus invasion. Cyclophosphamide inhibited the expression of dectin-1 in lung of mice which may be one of the mechanisms of the invasive pulmonary aspergillosis development induced by eyclophosphamide.
4.Manifestations of cerebral developmental venous anomalies and its associated lesions in MRI

Objective To evaluate the application value of different MR sequences in cerebral developmental venous anomalies (DAVs),and to explore the relationship between DVAs and its associated lesions.Methods MRI findings in 38 patients of DAVs were analyzed retrospectively.Imaging performance and characteristics of DAVs and its associated lesions in different MR sequences were analyzed.Results In all of the 38 cases,3 cases were multiple DVAs without associated lesions,1 case was combined right lateral ventricle hemorhage,1 case was combined with multiple cavernous hemangioma,8 cases were single DVAs with solitary cavernous hemangioma,5 cases were combined with astrocytoma,1 case was combined with intracranial hematoma,and 19 cases were single DVAs with no comorbidity.DVAs in 16 cases showed strip or small patchy hypo-intensity lesions and 15 cases were not visible on T1WI;16 cases showed strip or small patchy hypo-intensity,5 cases showed strip or flocculent hypo-intensity and 10 cases could not seen on T2WI;19 cases showed patchy or strip hypo-intensity and 8 cases showed iso intensive signal on DWI;8 of the 10 cases who performed susceptibility weighted imaging (SWI) examination showed dendritic low signal,showing a typical caput medusae sign,2 cases showed no lesions on SWI;30 cases underwent 3D-T1WI enhanced scan showed clearly all lesions of DAVs,19 cases of them showed typical caput medusae sign and large draining veins.Conclusion Routine MR sequence can demonstrate part of the DVAs and associated peripheral lesions,DWI showed lesions of DAVs is superior to routine MR sequence,SWI and enhanced 3D-T1WI can accurately diagnose DVAs and show the venous drainage.
5.Analysis of millimeter-wave dosimetry in cell culture dishes with finite-difference time-domain technique.

Analysis is carried out on millimeter-wave (MMW) dosimetry in culture dishes used in experiments on MMW biological effects at the cellular level. Finite-difference time-domain (FDTD) technique is employed to compute the 6 mm MMW power density (PD) irradiated into cells in a typical culture dish and the MMW power absorption density (PAD) of cells, followed by the qualitative and quantitative analyses on the outcomes. Indicated in the results, distributions of MMW PD irradiated into cells and the PAD of cells are complicated with evident inhomogeneity so that MMW dosimetry varies a lot in different position whose influence on the experimental outcomes is not neglectable. Consequently, the precise determination of dosimetry is of great importance to be conducted in related experiments.

6.Power density analysis on millimeter waves irradiated into cell monolayers in culture dishes.

Biological effects of millimeter waves (MMWs) at the cellular level are explored in experiments using culture dishes containing cell monolayers with MMW irradiated from the underneath. Analysis is carried out for culture dishes with diameters much larger than the MMW wavelength to discover the relation between the portion of the incident MMW power density (PD) irradiated into the cell monolayer and the interfering factors such as the bottom thickness of the culture dish, the MMW wavelength, and the electromagnetic parameters of the dish and culture solution. With the help of the finite-difference time-domain (FDTD) technique, another analysis is performed to display the effect of the dish geometries upon the MMW PD irradiated into the cell monolayer in a specific culture dish with a diameter similar to the MMW wavelength. The results indicate that rigorous analysis, precise measurement and accurate calculation of MMW PD are essential accompaniments for these experiments, and large-diameter culture dishes are more preferred in experiments to small-caliber ones.

7.Influence of Huiyang Shengji Ointment and Its Modified Formulae on Serum Interleukin-1α and Thromboxane B2 in Diabetic Rats with Chronic Skin Ulcers

Objective To observe the influence of Huiyang Shengji Ointment and its modified formulae on interleukin-1 (IL-1α) and thromboxane B2 (TXB2) in diabetic rats with chronic skin ulcers, and explore the mechanism for promoting the healing of ulcer.Methods Six out of 160 rats were randomly selected as a blank group, without any further processing. The remaining rats were made diabetic model and randomly divided into five groups after 2 weeks:1 d, 3 d, 5 d, 7 d and 14 d groups. Then, these groups were further divided into normal group (Vaseline ointments), model group (Vaseline ointments), Huiyang Shengji Ointment group (whole formula Ointment), Wenyang Yiqi group (Yiqi group, modified Wenyang Yiqi formula ointments) and Huoxue Shengji group (Huoxue group, modified Huoxue Shengji formula ointments). Normal group and model group were given Vaseline ointments;whole formula group, Yiqi group and Huoxue group were given corresponding ointment. Normal group used the method of skin excision, while other groups used STZ injection-hydrocortisone interference-skin excision-foreign body embedded preparation of composite factors for chronic skin ulcer model. After the appropriate treatment period, the rats were executed and tested for the contents of IL-1α and TXB2 in serum by enzyme-linked immunosorbent assay of five time points.Results In treatment 3 d, the contents of IL-1α in Yiqi group were significantly higher than the blank group, model group, whole formula group and Huoxue group (P<0.05). In treatment 5 d, the contents of IL-1α in whole formula group were significantly higher than the blank group and model group (P<0.05). In treatment 7 d, the contents of IL-1α in each treatment group were significantly higher than blank group and model group (P<0.05), and the whole formula group was higher than the Yiqi group and Huoxue group. In treatment 14 d, the contents of IL-1α in model group and Huoxue group were lower than the blank group (P<0.05). In treatment 3 d, the contents of TXB2 in normal group and the whole formula group were higher than the blank group (P<0.05). In treatment 5 d, the contents of TXB2 in whole formula group were higher than the blank group and the normal group (P<0.05). In treatment 7 d, the contents of TXB2 in Yiqi group were higher than the blank, the model, the whole formula and Huoxue groups (P<0.05). In treatment 14 d, the contents of TXB2 in Huoxue group were higher than the blank and model group (P<0.05), and the contents of TXB2 in the blank group and normal group was lower than those treatment groups (P<0.05).Conclusion Huiyang Shengji Ointment and its modified formulae could promote inflammation, stimulate secretion of inflammatory cytokines, while Yiqi Wenyang ointments played a more active role in promoting inflammation of the early phase of wound surface.
8.Association of the TGF-β1 gene promoter polymorphisms with hepatocellular carcinoma

ObjectivesTo study the relationship between the allele frequencies and genotype distribution of transforming growth factor-beta 1(TGF-β1) gene promoter polymorphisms in Chinese patients with heptocellular carcinoma(HCC),and to analyze the association of the serum levels and genotype of TGF-β1 with HCC.MethodsThe polymorphisms of TGF-β gene,including polymorphisms of TGF-[β1 gene -800G/A、-509C/T,were analyzed by the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP)methods in 102 patients with HCC and 110 healthy controls,and the serum level of TGF-β1 was determined by enzyme-linked immunosorbent assay (ELISA). ResultsThe HCC group showed significantly higher serum levels of TGF-β1 than control group [(51.06 ± 9.74)μg/L,(22.12 ± 8.67 )μg/L,t=22.884, P<0.01], The distributions of TGF-β gene -800G/A polymorphisms were not different significantly between HCC group and control group, but TGF-β1 -509C/T gene polymorphism was significantly different. The relative risk suffered from HCC of C allele was 1.822 times of the T allele (OR=1.822,95 %CI:1.238-2.682,t=22.884,P<0.01), the serum level of TGF-β1 T allele carriers was significantly higher than that of no carriers [(53.52:±:10.07)μg/L,(43.57±9.89)μ.g/L,t=3.898, P<0.01]. ConclusionTGF-β1 gene -509C/T polymorphism is associated with HCC, and T allele may be a risk factor for HCC, in which the TGF-β1 T allele carriers may have increased risk by enhancing the TGF-β1 expression in the pathogenesis of HCC.

10.Protective effect of hawthorn leaf procyanidins on cardiomyocytes of neonatal rats subjected to simulated ischemia-reperfusion injury.

OBJECTIVETo study the direct effect of hawthorn leaf procyanidins on cardiomyocytes subjected to ischemia-reperfusion injury and elucidate their therapeutic mechanism on ischemic heart diseases.
METHODCultured cardiomyocytes of neonatal rats were subjected to anoxia-reoxia injury which simulated the ischemia-reperfusion injury in vivo, and hawthorn leaf procyanidins were applied. The therapeutic effect was valued by LDH leakage and MTT test. For a further mechanism study, contents of MDA and activities of SOD in cardiomyocytes were measured.
RESULTHawthorn leaf procyanidins in 24-60 mg x L(-1) significantly and dose-dependently inhibited LDH leakage (compared with the model group, all P<0.01 ) and cell viability decrease (compared with model group, 24-48 mg x L(-1) groups all P<0.05; 60 mg x L(-1) group, P<0.01) in cardiomyocytes induced by anoxia-reoxia injury. Furthermore, hawthorn leaf procyanidins in 24-60 mg x L(-1) significantly inhibited the increase of MDA content (compared with the model group, all P<0.01) and the decreased of SOD activity (compared with the model group, 24 mg x L(-1) group, P<0.05; other groups all P<0.01) in cardiomyocytes undergoing anoxia-reoxia injury.
CONCLUSIONHawthorn leaf procyanidins have a significant therapeutic effect on the simulated ischemia-reperfusion injury of cultured neonatal rat cardiomyocytes, which may relate to their anti-oxidation effects. And the direct protective effect of hawthorn leaf procyanidins on cardiomyocytes subjected to ischemia-reperfusion injury may be one of the key mechanisms of its therapeutic effect on ischemic heart diseases.
