1.Effects of Hrs on Apoptosis and Cell Proliferation in Colon Cancer Cell Lines

Objective To study the effects of Hrs on apoptosis and cell proliferation in colon cancer cell lines HCT?116 and SW?480. Methods The expression of Hrs in HCT?116 and SW?480 was knocking down by shRNA. Cell apoptosis was detected by FACS and cell proliferation was de?termined by MTT,incucyte and soft agar. Results The apoptosis was significantly increased after Hrs knocking down in both HCT?116 and SW?480,and the cell proliferation was inhibited. Conclusion Hrs is involved in the apoptosis and proliferation of colon cancer cells.
2.Determination of Trace Amounts of Sulfide by Flow-injection Chemiluminescence Method

A new flow-injection chemiluminescence method for the determinstion of trace amounts of sulfide was developed. It is based on the catalysis of sulfide to chemilumincence reacion between luminol and hydrogen peroxid. The linear range extends from 2.0 × 10-9 to 8.0 × 10-7 g/mL and the detection limit is 5.6 × 10-10 g/mL.The relative standard deviation (RSD) is less than 5% for 1.0 × 10-7 g/mL sulfide solution in eleven repeated measurements. The method has the advantages of simplicity, rapidity, sensitivity and has also been applied to the determination of trace amounts of sulfide in water samples.
3.Improvement on micro-manipulation of fertilized eggs of mice

Objective: To improve the micro-manipulation of mouse fertilized eggs,promoting its survival rate and productivity in pseudo-pregnant mice. Methods: DNA fragments of definite concentration and size were used for micro-manipulation with conventional and improved methods, then their survival rate of manupulated eggs and productivity of pseudo-pregnant mice were analyzed and compared. Results: With improved methods, the survival rate of eggs increased from 60.3% to 79.5%, the productivity of pseudo-pregnant mice and offspring increased from 35.7% to 56.8% and 10.1% to 14.1% respectively. Conclusion: The improved methods of micro-manipulation can be used in transgenic mouse production.
4.Development of pMTR1 plasmid-based transgenic mice model for studying gene mutation in vivo

Objective: To establish C57BL/6 transgenic mice lineages containing pMTR1 plasmids as an efficient animal model for studying gene mutation in vivo . Methods: The linearized pMTR1 DNAs, constructed by molecular cloning, were injected into 572 fertilized eggs of C57BL/6 mice by microinjection. The manipulated embryos were transferred into the oviducts of 45 pseudopregnant mice respectively, from which 35 offsprings were obtained. The genomic DNAs of these offsprings were analyzed with PCR and genomic Southern blotting. Four mice whose genomes integrated with copies of intact pMTR1 vectors were chosen as founders to establish transgenic mice lineages. The recovery of pMTR1 plasmid were conducted on the genomic DNAs of F1 or F2 transgenic offsprings. Results: It was showed that intact and functional pMTR1 plasmid could be efficiently recovered from the genomic DNAs of these mice. Conclusion: (1) The transgenic mouse lineages containing copies of a stably integrated pMTR1 plasmid is established. (2) The transgenic mice are suitable models for studying gene mutation in vivo .
5.Role of parecoxib in protection of learning and memory ability in rats with transient global cerebral ischemia/reperfusion inj ury

Objective To explore the role of parecoxib in protection of learning and memory ablility in rats with global cerebral ischemia/reperfusion injury. Methods Male Wistar rats were randomly divided into a sham group (S group), a ischemia/reperfusion group (I/R group), and a ischemia/reperfusion with parecoxib group (I/R+PA group). Global cerebral ischemia was induced by Pulsinelli 4-vessel occlusion. 72 h after reperfusion, Morris water maze was used to assess spatial learning and memory ability for consecutive five days. The histological changes in the hippocampus were detected by Nissl staining. Results As compared with the I/R group, rats in S guoup and I/R+PA group had a significantly shortened escape latency (P<0.05), and had significantly longer dwelling time in the former platform quadrant and more frequent cross-platform movement in the Morris water maze test. Conclusions Parecoxib plays a role in protection of learning and memory ability in rats with global cerebral ischemia/reperfusion injury.
6.Clinical Observation of Edaravone Combined with Butyl Phthalide in the Treatment of Acute Ischemic Stroke

OBJECTIVE:To observe clinical efficacy and safety of edaravone combined with butyl phthalide in the treatment of acute ischemic stroke (AIS). METHODS:258 AIS patients were randomly divided into observation group and control group, with 129 cases in each group. Both groups were given routine treatment as antiplatelet,improving microcirculation,controlling blood pressure,lowering blood glucose,regulating blood lipid,keeping plaque stable,nourishing brain cells. Control group was additionally given Butyl phthalide capsules orally,200 mg,tid. Observation group was additionally given Edaravone injection 30 mg added into Sodium chloride injection 100 ml,ivgtt,bid,on the basis of control group. Both groups continuously received 14 days of treatment. The serum inflammatory factor,miR-222 and neurologic impairment score of 2 groups were observed before treatment,7,14 d after treatment. Clinical efficacies and the occurrence of ADR were compared between 2 groups. RESULTS:There was no statistical significance in the serum inflammatory factor,miR-222 and neurologic impairment score between 2 groups before treatment(P>0.05). The serum inflammatory factor and neurologic impairment score of 2 groups were decreased significant-ly 7,14 d after treatment,while serum levels of miR-222 were increased significantly;the observation group was significantly bet-ter than the control group,with statistical significance(P<0.05). Total effective rate of observation group was 92.2%,which was significantly higher than that of control group (69.8%),with statistical significance (P<0.05). No obvious ADR was found in 2 groups. CONCLUSIONS:Edaravone combined with butyl phthalide is effective in the treatment of AIS,and can significantly de-crease serum inflammatory factor level,promote the expression of miR-222 and improve neurologic function with good safety.
7."Influences of ""Antiphlogistic No.1"" on the degranultion of Activated RBL-2H3 cells"

Objective To investigate the direct effect ofAntiphlogistic No.1 on the degranultion of RBL-2H3 mast cells stimulated by anti-DNP IgE and DNP-BSA complex.Methods The effect of Antiphlogistic No.1 on stabilization of RBL-2H3 cell membrane was assessed by degranultion inhibition rate.β-hexosaminidase and IL-4 released from RBL-2H3 cells were detected by PNAG colorimetric assay and ELISA.Results β-hexosaminidase and cytokine IL-4 production releases from RBL-2H3 cells was reduced after they had been treated with Antiphlogistic No.1.The inhibition rate of β-hexosaminidase release was 42.47% at the concentration of 8 μg/ml (P<0.01) and dose dependently increased to 52.40%,68.26% and 72.15% respectively when drug concentration up to 80 μg/ml,800 μg/ml,8000 μg/ml,while inhibition rate of IL-4 generation were 13.87%,23.27%,31.95%,39.99% (P<0.01).Antiphlogistic No.1 maximum inhibition rate of β-hexosaminidase release is 84.48% of dexamethason maximum inhibition rate while Antiphlogistic No.l maximum inhibition rate of IL-4 is close to dexamethason maximum inhibition rate.Conclusion Antiphlogistic No.1 could stabilize the cellular membrane of RBL-2H3 and provide protection against Ⅰ type allergic response.
8.Applicability of a natural swelling matrix as the propellant of osmotic pump tablets.

The purpose of this study is to investigate the applicability of a natural swelling matrix derived from boat-fruited sterculia seed (SMS) as the propellant of osmotic pump tablets. The sugar components, static swelling, water uptake and viscosity of SMS were determined and compared with that of polythylene oxide (WSR-N10 and WSR-303). Both ribavirin and glipizide were used as water-soluble and water-insoluble model drugs. Then, the monolayer osmotic pump tablets of ribavirin and the bilayer osmotic pump tablets of glipizide were prepared using SMS as the osmotically active substance and propellant. SMS was mainly composed of rhamnose, arabinose, xylose and galactose and exhibited relatively high swelling ability. The area of the disintegrated matrix tablet was 20.1 times as that at initial after swelling for 600 s. SMS swelled rapidly and was fully swelled (0.5%) in aqueous solution with relative low viscosity (3.66 +/- 0.03) mPa x s at 25 degrees C. The monolayer osmotic pump tablets of ribavirin and the bilayer osmotic pump tablets of glipizide using SMS as propellant exhibited typical drug release features of osmotic pumps. In conclusion, the swelling matrix derived from boat-fruited sterculia seed, with low viscosity and high swelling, is a potential propellant in the application of osmotic pump tablets.
9.Identification of Salvia shandongensis new species based on sequences of the plastid psbA-trnH intergenic region.

To identify Salvia shandongensis and its relatives at molecular level, the psbA-trnH intergenic region of three species including Salvia shandongensis, Salvia miltiorrhiza and S. miltiorrhiza f. alba were amplified and sequenced. Sequences were assembled with CodonCode Aligner. The K2P genetic distances between Salvia shandongensis and its relatives were calculated and UPGMA tree was performed by MEGA5.0. The results indicated that the lengths of psbA-trnH regions of Salvia shandongensis were about 391 bp, while the lengths of psbA-trnH regions of Salvia miltiorrhiza and S. miltiorrhiza f. alba were about 386 bp. The psbA-trnH sequences showed considerable variations between species and thus were revealed as a promising candidate for barcoding of Salvia shandongensis and its relatives. The intra-specific genetic distances of Salvia shandongensis were 0, while the intra-specific genetic distances of Salvia miltiorrhiza and S. miltiorrhiza f. alba were 0.002 and 0.001 respectively. Additionally, the genetic distance of Salvia shandongensis and Salvia miltiorrhiza ranged from 0.034 to 0.04, and the genetic distance of Salvia shandongensis and S. miltiorrhiza f. alba ranged from 0.005 to 0.008, the intra-specific genetic distances of Salvia shandongensis were much smaller than that of Salvia miltiorrhiza and S. miltiorrhiza f. alba; clustering results showed that there were obvious differences between Salvia shandongensis, Salvia miltiorrhiza and S. miltiorrhiza f. alba, which was consistent with morphological characteristics. This study not only firstly provides the scientific basis for establishing the taxonomy position in molecular level and revealing their genetic relationships of S. shandongensis, S. miltiorrhiza and S. miltiorrhiza f. alba; but also provides DNA molecular identification scientific basis for the development of new medicinal plant resources of Salvia shandongensis. Our results suggest that the psbA-trnH intergenic spacer region can be used as a barcoding to identify Salvia shandongensis, Salvia miltiorrhiza and S. miltiorrhiza f. alba.
10.Reflection on teaching of extracellular matrix in current medical cell biology

Extracellular matrix (ECM)is not only one of current hot-points in medical cell biology,but also one tricky part for undergraduates to learn. This article compared chapters of ECM in medical cell biology courses and spotlighted that ECM chapter was often neglected in some domes-tic universities. Then it analyzed the possible causes,such as variable arrangement of ECM section in current textbooks. Lastly,the article recommended several suggestions,including giving a timely re-vision of the old ECM knowledge,designing an appropriate strategy for teaching,enumerating certain representative diseases to improve the ECM education. It appealed our teachers to pay more attention to how to make the module of ECM master well in near future.
