1.Mitochondria, Reactive Oxygen Species and Apoptosis

Mitochondria are involved not only in energy metabolism but also in free radical metabolism. Superoxide anion can be generated through a way of electron leak of respiratory chain and the reactive oxygen species (ROS ) can be formed in the further reactions of O2* in mitochondria. The role of mitochondria in anti-oxidant functions and cell apoptosis is discu ssed in terms of electron leak of respiratory chain, uncoupling of oxidative pho sphorelation, mitochondrial pore, Box- or/and PTP-mediated release of cytochro me c from mitochondria and so on. The signaling act of ROS is emphasized in the regulation of cell apoptosis.
2.Minimally invasive fixation of complex proximal tibial fractures by uniaxial and polyaxial locking plates

Objective To investigate the clinical application of minimally invagive fixation of complex proximal tibia fractures by uniaxial and polyaxial locking plates. Methods The clinical data of 38 patients with minimally invagive fixation of complex proximal tibia fractures by uniaxial(n=21)and polyaxial(n=17)locking plates from January 2008 to June 2009 were retrospectively analysed,and the union rates and function recovery were compared between groups.Results All patients were followed up for 3 to 20 months,with an average of 12 months.All patients had bone union.The time of fracture union for fixation by uniaxial locking plates was 10 to 20 weeks,with an average of 14 weeks;and that for fixation by polyaxial locking plates was 11 to 18 weeks,with au average of 13 weeks.Evaluated by Johner-Wruhs method, there were 14 "excellent" cases,5 "better" eases,2 "good" cases and 0 "poor" case for fixation by uniaxial locking plates (rate of "excellent and better",90.4%),and there were 11 "excellent" cases,5 "better" cases,1 "good" cage and 0 "poor" case for fixation by polyaxial locking plates (rate of "excellent and better",94.1%). Conclusion Minimally invagive fixation of complex proximal tibia fractures by uniaxial and polyaxial locking plates is stable,has less effects on bone blood supply,high bone union rate and favourable function recovery,and is an effective way in the treatment of complex proximal tibia fractures.Fixation by uniaxial locking plate has a better mechanical intensity,while fixation by polyaxial locking plate can adjust the screw angle according to fracture situation,which can be clinically applied accordingly.
3.Single allograft lung transplantation in 4 cases of COPD

Objective To report the management of single allograft lung transplantation in 4 cases of COPD. Methods Four COPD cases receiving lung transplantation were analyzed respectively. There was diffuse emphysema in double lung in one case and in the remaining 3 cases severe diffuse emphysema that mixed together partially to be bulla but not to the 1/3 of the thoracic cavity. All the cases received the corpse donor. One case with diffuse emphysema received the single-right lung transplantation, one case received simultaneous single-lung transplantation and lung volume reduction and the remaining 2 cases received LVRS 47 days and 28 days after single-left lung transplantation respectively. The inductive treatment with the daclizumab or antithymocyte globulin (ATG) was done and the rejection was prevented with the Tacrolimus, mycophenolate and prednisone. Results The operations were successful in the 4 cases. One case had the acute rejection 5 days after operation and reco- vered after intensive treatment with Methylprednisolone. Two cases discharged successfully and lived to today and the first case lived more than 2 years. Two cases died 74 days and 77 days after lung transplantation respectively. Conclusion In late stage COPD, single allograft lung transplantation is an effective treatment to the bilateral emphysema patients and the lung volume reduction should be done simultaneously to the patients with the bullae that may be ruptured.
4.Effect of Retention-Enema of Bowels-Unblocking and Heat-Purging Mixture Combined with Electro-acupuncture for Peritonitis After Operation: An Observation of 32 Cases

To investigate the effect of retention-enema of Bowels-Unblocking and Heat-Purging Mixture (BHM) combined with electro-acupuncture (EA) for peritonitis after operative.In admission order, 32 cases of acute diffuse peritonitis were allocated to group A and were treated with retention-enema of BHM and EA based on routine western medicine 6 hours after operation.Other 28 cases in group B were treated with routine western medicine only. Recovery time of bowel sound,passing flatus and defecation was observed after operation as well as post-operative complications, multiple organs dysfunction syndrome (MODS) and multiple organ failure (MOF).After operation, recovery time of bowel sounding,passing flatus and defecation was 20.32?4.34, 29.43?9.66 and 40.69?13.69 hours in group A respectively, which differed from 30.66?6.06, 40.66?17.33 and 66.48?17.58 hours in group B (P
5.An Experiment of Using Nitinol Alloy Momemorial Stent to Treat the Stenosis of the Rabbi

Objective To compare the effects of nitinol alloy memorial stent with silastic tube in treating the stenosis of the rabbit.Methods 16 rabbits with external ear canal(EEC) stenosis were randomly divided into two groups.One group was implanted with skin on the EEC wound while the other not.By self-comparison method nitinol alloy memorial stem was implanted in a rabbit's one ear and the silastic tube in the other.After days 5,15,30,and 60 later,the diameters of the external ear canal (with two materials planted) were measured respectively in the skin-planting group.In the naked group scar tissues were harvested and tested separately according to the planting material.HE coloration were used to study fibroblast hyperplasty while RT-PCR were applied to detecting the TGFβ1mRNA expression.The two brackets were compared according to their effect to scar hyperplasty.Results The EEC diameters using nitinol alloy memorial stem were found more spacious than using silastic tube.HE coloration showed the fibroblast hyperplasia was more mitigatory by using the nitinol alloy memorial stent.RT-PCR also found the TGFβ1mRNA expression was low by using same material.Conclusion The nitinol alloy memorial stent shows obvious superiority over silastic tube in external ear canal stenosis therapy.
6.To promote colonic adaptation of short bowel rats by enteral nutrition plus fiber treatment

Objectives:To study the colonic adaptation of short bowel rats by enteral nutrition plus fiber treatment. Methods:The short bowel rats were randomized into two groups:feed by Pepti-2000(EN) and by Pepti-2000 plus fiber(EF);and another normal rats as control(CONT) group.The nutritional states and morphologic changes of the colon were investigated. Results:From postoperative days(PODs) 15,the weight of EF group was increasing and greater than EN group after PODs 18.The net nitrogen balance in EF group was almost the same as CONT,but greater than EN group.All the mucosal thickness,villous height and surface area of the colon in EF group were greater than CONT and EN group(P<0.001).DNA index and percentage of S phase cells of the colonic mucous in study groups were higher than CONT group(P<0.05).Also the levels of colonic mucous IGF-1 mRNA in study groups were higher than in CONT group(P<0.05). Conclusions:Enteral nutrition plus fiber can not only promote nutritional states;but enhance colonic adaptation to the short bowel rats.
7.Human umbilical cord blood stem cells differentiate into nasal ciliated epithelial cells

BACKGROUND:Damage to nasal ciliated epithelial cels can lead to a severe injury in nasal biological function. Compared with other adult stem cels, human umbilical cord blood stem cels have better differentiation potential.
OBJECTIVE: To explore the feasibility of human umbilical cord blood stem cels differentiating into nasal ciliated epithelial cels through in vitro culture and induction techniques.
METHODS:Normal and healthy umbilical cord blood samples were colected to isolate human umbilical cord blood stem cels, folowed by identification and subculturein vitro. Umbilical cord blood stem cels at passage 3 were infected with recombinant adeno-associated virus carrying enhanced green fluorescent protein and cultured using air liquid interface culture method. Thereafter, PCR assay was employed for detecting MUCS expression in cultured stem cels at 1 and 2 weeks after induction, and immunofluorescent staining for FOXJ1 was performed at 3 weeks.
RESULTS AND CONCLUSION:After subculture, passage 3 umbilical cord blood stem cels that could express stem cel surface markers were visible in a uniform shape and had good refraction. After 3 hours of gene transfection, green fluorescence issued from the passage 3 cels were visible, and the cel positive rate was up to 96.2% until 48 hours, indicating good transfection efficiency. RT-PCR findings showed that MUC8 mRNA had no expression in the umbilical cord blood stem cels, but expressed strongly in the nasal ciliated epithelial cels, whose expression was weak at 1 week of culture and increased at 2 weeks. Additionaly, the positive expression of FOXJ1 red fluorescence was observed under the transfection of green fluorescent protein. These results suggest that human umbilical cord blood stem cels could differentiate into nasal epithelial cels under suitable conditions.
8.Expression and clinical significance of MTDH, HIF-1α and TKTL1 in laryngeal carcinoma.

OBJECTIVE:
To investigate the functional mechanism of metadherin (MTDH), hypoxia-inducible factor-1 alpha (HIF-1α) and transketolase-like gene 1 (TKTL1) and their interaction with each other in laryngeal carcinoma development.
METHOD:
Thirty laryngeal carcinoma samples and 20 samples of para-carcinoma tissue were taken from the patients received operation treatment. The expression levels of MTDH, HIF-1α and TKTL1 were detected in all the samples by SP immunohistochemical methods. The data were analyzed by the SPSS13.0 statistical software.
RESULT:
The positive expression rate of MTDH, HIF-1α and TKTL1 in the 30 cases of laryngeal carcinoma was 56.67%, 60.00% and 63.33%, respectively, which was 20.00%,10.00% and 15.00% respectively in the para-carcinoma tissue, the difference of the positive expression rate of the tested objects between these two groups was statistically significant (P < 0.05 or P < 0.01). Of the 30 cases of laryngeal carcinoma, the positive expression rate of MTDH, HIF-1α and TKTL1 in the cases with lymphnode metastasis was 84.62%, 84.62% and 79.62%, respectively, compared with the rate in those without lymph nodes metastasis, which was 35.29%, 41.18% and 35.29%. The difference of the positive expression rate of the tested objects between these two groups was statistically significant (P < 0.05). In the tissue of poorly differentiated squamons cell carcinoma verified by pathology, the positive expression rates of MTDH and HIF-1α was 73.68% and 84.21%, respectively, while that in the tissue of well differentiated squamous carcinoma was 27.27% and 18.18%. The difference of the positive expression rate of the tested objects between these two groups was statistically significant (P < 0.05 or (P < 0.01). The positive expression rate of TKTL1 in laryngeal carcinoma at stage I-II was 25.00% and that at stage III-IV was 72.22%, the difference between which was statistically significant (P < 0.05). The positive expression rate of HIF-1α in laryngeal carcinoma at stage I-II was 33.33% and that at stage III-IV was 77. 78%, the difference between which was statistically significant (P < 0.05). The expression of MTDH, HIF-1α and TKTL1 showed no relationship with age,smoking amount or the tumor location (P > 0.05). The Spearman's rank correlation analysis showed that the positive expression rates of MTDH and HIF-1α in laryngeal carcinoma had positive correlation (r = 0.384, P < 0.05); the positive expression rates of TKTL1 and HIF-1α in laryngeal carcinoma had positive correlation (r = 0.508, P < 0.01); But there was no significant correlation between the positive expression rates of MTDH and TKTL1 in laryngeal carcinoma (r = -0.107, P > 0.05).
CONCLUSION
It suggested that these three proteins may have close relationship with the occurrence, invasion and metastasis of the laryngeal carcinoma. MTDH and TKTL1 may be expected to be new clinical targets for laryngeal carcinoma treatment and it could offer theoretical basis for the prognosis of the laryngeal carcinoma.

9.Production of nitric oxide and change of nitric Oxide synthase activity in brains mitochondria of the rats with focal cerebral ischemia/reperfusion

AIM and METHOD:To determine the production of nitric oxide(NO) and change of NO synthase(NOS) activity in mitochondria isolated from the rat brains of the ischemia/reperfusion rat model produced by transient occlusion of middle cerebral artery on the following the points: 2 h after occlusion of artery and 30 min, 2h, 4h after reperfusion. RESULTS: After the occlusion of middle cerebral artery, the respiratory control rate(RCR) of mitochondria significantly decreased and slightly increased at 4h after reperfusion. Meantime, the production of NO in mitochondria increased significantly. But with the increase of perfusion, production of NO gradually decreased and reached normal level as in the control group. It also shows that cerebral ischemia increased NOS's activity significantly in the mitochondria and still kept a higher level than the control group although it decreased gradually after reperfusion. But the iNOS's activity did not show obvious change. The change of total NOS's activity depends on the change of cNOS's activity. CONCLUSION: The activation of NO/NOS system in the mitochondria might play an important role in the reperfusion injury during reperfusion of ischemic brain.
10.Therapeutic Effect of Tonglieshu capsules for Nonbacterial Prostatitis

[ Objective ] To investigate the therapeutic effect of Tonglieshu capsules (mainly composed of Radix Linderae, Semen Litchi, Semen Vaccariae, Rhizoma Corydalis, Rhizoma Sparganii, Rhizoma Curcumae, Herba Taraxaci, etc. ) for nonbacterial prostatitis. [Methods] Forty SD rats were randomized to 4 groups: normal control group (group A) , model group (group B), Qianlietong group (group C) and Tonglieshu, Capsules (group D). Except those in group A, rats in other groups were injected with Xiaozhiling Injection directly into the prostatic glands to establish nonbacterial prostatitis. After treatment with Tonglieshu capsules for 4 weeks, white blood cells counting, density of lecithin corpuscle and inflammatory cells infiltrating into interstitium and glandular lumen of prostatic tissues were observed. [Results] WBC counting was decreased and density of lecithin corpuscle increased in group D (P 0.05) . [Conclusion] Tonglieshu capsules has an effect in treating chronic prostatitis by decreasing the WBC counting, reducing infiltration of inflammatory cells and increasing the density of lecithin corpuscle in prostatic tissues.
