1.Determination of Turbidity of Water by Hach 2100-N Scattering Turbidimeter

Turbidities of water samples were determined by Hach 2100-N scattering turbidimeter in this assay.This method showed a good linearship at range of 0~100 NTU with r=0.9999 for water samples,and also revealed good precision compared with the transmissometry.The data obtained by this method showed no significant differences in the standard series at the range of 0~6.5 NTU and the water samples at the range of 0~5 NTU.
2.Tanshinone IIA prevents high glucose-induced human umbilical vein en-dothelial cell apoptosis

AIM:To investigate the effect of tanshinone IIA on the apoptosis of human umbilical vein endothe -lial cells (HUVECs) after high glucose treatment.METHODS:The cell viability was determined by MTT assay.The cell apoptotic rate was examined by flow cytometry with Annexin V-FITC/PI double staining .The expression of Bcl-2 and Bax, and the release of mitochondrial cytochrome C ( Cyt C) were analyzed by Western blotting .RESULTS: Tanshinone IIA significantly inhibited high glucose-induced decrease in cell viability and increased the cell apoptosis .Additionally, after tanshinone IIA treatment , Bax expression and the release of mitochondrial Cyt C were significantly inhibited , while Bcl-2 expression was increased .CONCLUSION: Tanshinone IIA prevents high glucose-induced endothelial cell apoptosis via mitochondria-dependent pathway .
3.Imageology study of sellar turica and its potential role on growth of pituitary adenoma

Objective To study imageology anatomy concerned of sellar turica and explore its potential role on growth of pituitary adenomas. Methods According to classification of sinus sphenoidalis,103 normal imageology data of sellar turica region were analyzed, including 83 cases with entire saddle type sinus sphenoidalis and 20 cases anti-saddle type sinus sphenoidalis. Furthermore, the clinical data of 45 cases of pituitary adenomas with extension into sinus sphenoidalis were retrospectively studied. Results The relationship between sellar floor's shapes and sinus sphenoidals'types was that the larger the sinus sphenoidalis was, the deeper the floor was(98.8%). The relationship between sellar floor's shapes and the superior surface of hypophysis was that most of depressed sellar floors were companied with depressed superior surfaces of hypophysises(93.8%). The anteroposterior shapes of sellar turica in midsagittal can be divided into 4 types such as bag-type, kettle-type, tube-type, and sifter-type. 45 patients suffering from pituitary adenoms with extension into sinus sphenoidalis all had entire saddle type sinus sphenoidalis. Conclusions The imageology anatomy concerned of sellar turica could explain the phenomena of pituitary adenomas with extension toward sellar floor and sinus sphenoidalis. Referring to the literatures concerned, it indicated that the imageology anatomy was the key factor of affecting the growth of pituitary adenomas.
4.Determination of the dissolution of Breviscapinun Dispersive Tablet by HPLC

AIM: To establish a method for determing the dissolution of Breviscapin Dispersive Tablet. METHODS: Breviscapin release was investigated in phosphate BS(pH7.4) by the third method for the release determination (Chinese Pharmacopoeia 2005 edition).Breviscapin content was assayed by HPLC and the release percentage was then calculated. RESULTS: The average recovery of scutellarin was 101.93%,RSD=(1.09)%.Good linear relationship was shown in the concentration range of 0.007-0.18 mg/mL,r=0.999 9.The dissolution was almost unaffected by rotation rate,but greatly affected by the type of solvent. CONCLUSION: The method is sensitive,accurate and quick for the quality control of Breviscapin Dispersive Tablet.
5.THE METABOLIC BALANCE OF ZINC AND COPPER IN DIABETES MELLITUS

Zinc(Zn) and copper(Cu) balance tests were conducted in patients with diabetes mellitus. The results revealed that the negative balance of Zn and excessive positive balance of Cu were much improved after the management of diabetes mellitus. In addition, a close relationship was found between serum glucose concentration and the disorder of Zn and Cu balance, i.e. serum Zn level, apparent intestinal absorption rate and balance status of Zn were reversely related to serum glucose concentration, whereas urinary Zn, serum Cu level and apparent intestinal absorption of Cu were positively correlated to serum glucose concentration. Poly-factor analysis suggested that hyperzincuria and malabsorption of Zn were the main factors concerning negative Zn balance.
6.The mechanism of resistance of Pseudomonas aeruginosa to beta-lactam antibiotics and clinical significance.

To study the resistant mechanism and clinical significance of pseudomonas aeruginosa to beta-lactam antibiotics, the outer membrane permeability rate of 30 P. aeruginosa strains to 5 beta-lactam antibiotics was measured and their production of beta-lactamase and the beta-lactamase genes they carried detected. Furthermore, the relationship between the permeability, beta-lactamase and the clinical effects of beta-lactam antibiotics was observed. By using 14C-penicillin and liquid-scintillant isotope assay, the affinity of penicillin binding proteins (PBPS) was measured and their roles in the resistant mechanism studied. It was revealed that the permeability rate was higher in sensitive strains than in resistant ones (P < 0.05). All strains harbored 1-4 beta-lactamase genes and produced beta-lactamase. Higher permeability rate and higher degree of stability to beta-lactamase indicated better clinical therapeutic effects. The affinity of PBPs changed little without regard to the permeability and beta-lactamase. These results suggested that the permeability of outer membrane and beta-lactamase, but not PBPs, played important roles in the resistant mechanism of P. aeruginosa to beta-lactam antibiotics and affected the clinical therapeutic effectiveness of some patients.

7.Modified isolation and culture methods of human ovarian granulosa cells

BACKGROUND:To build up an effective method of isolating and culturing granule cels is a pivotal step to enhance fertilization-embryo transfer rate. Current studies mainly focus on the isolation methods of human ovarian granulosa cels rather than cel counting, purity and subsequent growth. OBJECTIVE: To establish the effective methods of isolating, purifying and culturing human ovarian granulosa cels in vitro. METHODS: Folicular fluid was harvested from women undergoing fertilization-embryo transfer procedures. Human ovarian granulosa cels were obtained from the folicular fluid by lysis treatment, precipitation method or density gradient centrifugation. Granulosa cel mucus masses were digested with type I colagen enzyme or hyaluronidase and then cultured in the culture medium with or without autologous folicular fluid. RESULTS AND CONCLUSION: Lysis treatment yielded the largest amount of granulosa cels compared to the precipitation method and density gradient centrifugation (P > 0.05,P < 0.05, respectively). Cels prepared by the three methods showed the same cel viability. After 24 hours of culture, the precipitation method obtained the largest amount of adherent granulosa cels (P < 0.05); and the density gradient centrifugation obtained the least amount of cels (P < 0.05). Compared with type I colagen enzyme, hyaluronidase took less time to digest the cels thoroughly. Autologous folicular fluid could promote the growth and survival of granulosa cels. These findings indicate that the precipitation method, though time-consuming, can obtain the highest cel viability and harvested the largest amount of granulosa cels after culture; hyaluronidase is more suitable for digesting granulosa cel mucus mass than type I colagen enzyme; autologous folicular fluid added into the culture medium is more conducive to granulosa cel growth.

9.Expression of tumor necrosis factor alpha and its relationship with infiitrating lymphocytes in lichen planus lesions

Objective To investigate the expression of tumor necrosis factor alpha(TNF-α)and its relationship with infiltrating lymphocytes in lichen planus(LP).Methods Tissue specimens were obtained from the lesions of 60 patients with LP and normal skin of 20 human controls.Immunohistochemical SP method was used to detect the expression of TNF-α,and infiltrating lymphocytes were counted in TNF-α-positive tissue sections.Results TNF-α was expressed in 72% of the LP specimens but in none of the control specimens(P < 0.01).Positive staining for TNF-α was mainly located in the membrane of prickle cells,cytoplasm or membrane of dermal infiltrating lymphocytes.The expression of TNF-α in LP was uncorrelated with age,sex or disease course(all P > 0.05),but was positively correlated with infiltrating lymphocyte number (rs =0.47,P < 0.01).Conclusion TNF-α seems to play a certain role in the pathogenesis of LP.
10.Study on histopathological changes of hepatic portal area in elderly men with type 2 diabetes mellitus

Objective To study the histopathological features of hepatic portal area in autopsy specimens of elderly men with type 2 diabetes mellitus (T2DM).Methods One hundred and three autopsy cases with T2DM (diabetic group) and contemporary 48 non-diabetic cases matched by gender and age (control group) were selected in the study.The histopathological changes of hepatic portal area were observed by histochemical and immunohistochemical methods.Results (1) In diabetic group, endothelial denudation, eosinophilie deposits, eccentric intimal thickening and luminal stenosis were found in hepatic interlobular artery (arteriole, the lumen diameter < 100 μm).The medial smooth muscle cells proliferated and migrated into intima, and fibroplasia occurred in adventitia.62.1% (64/103) of the patients with T2DM had microangiopathy and there was statistically significant difference between the two groups (χ2= 27.14, P< 0.01 ).(2) Hyaline collagen fiber tissues deposited around interlobular artery, interlobular vein and interlobular bile ducts, resulting in enlargement of the portal area and the secondary atrophy and disappearance of portal triad.65.0% (67/103) of the patients with T2DM had this pathological changes in the portal area and there was statistically significant difference between the two groups(χ2= 23.27,P<0.01).(3) Terminal portal area was notable.Conclusions Mieroangiopathy can be observed in the hepatic portal areas of elderly men with T2DM, which may induce the fibrotic tissue hyperplasia, the atrophy of the portal triad and the sclerosing portal area.
