1.Comparative Study on Tryptic Peptide Mapping of Globin Beta Chains and the Genotype from Nineteen Inbred Strains of Mice

This study represents the first description of structural variations in the globin beta chains of 19 strains of inbred mice. The trypic pep tide mapping of beta chains of globin from BALB/c, BALB/cA, BALB/cJ, BALBcJ-nu, BALB/nu/ + ,BALB/cyJax,B10,B10A/sui,C57BL/6,C57BL/10snJ,SMMC/B,SMMC/C, C3H/He, CFW, DBA/2,SWI,Swars/nu/+,SSB and 129/sv strains of mice was established. The results showed that the position of peptide spots upon fingerprint mapping of beta chains was obviously different in comparison with each other. The different patterns of variability and polymorphism found in the various mouse species may then have come into being as a consequence of the action of selective forces on the various new beta chains which were created by some exchange processes.
2.Effect of achyranthes bidentata polysaccharides on induction and differentiation of CD4~+T cells

AIM: To study the immunity modulation mechanism of achyranthes bidentata polysaccharides (ABPS). METHODS: (1) PBMC from patient with asthma or lung cancer were cultured with 800 mg/L ABPS for 18 h. IFN-? and IL-4 mRNA was detected by RT -PCR. (2) Th cells were cultured with serially diluted ABPS for 48 h and with 800 mg/L ABPS for various times. Cultured supernatant was harvested at diffevent time points and RNA was extracted, respectively. IFN-? or IL-4 transcriptional and translational levels were assay by RT -PCR and ELISA. 3.Application of Analytic Hierarchy Process in Evaluation Index System of Additional Essential Medicine

OBJECTIVE:To provide reference for the selection of new edition of Essential Medicine List by quantitative com-prehensive evaluation for additional essential medicine. METHODS:The evaluation factors were established according to Delphi method,and Analytic Hierarchy Process(AHP)was conducted to compare the factors in pairs,construct the judgment matrix,cal-culate the weight coefficients of every factor and do the consistency test. RESULTS:The evaluation model was composed of 6 first-class indexes(including effectiveness,safety,stability,availability,affordability and adaptability)and 24 second-class index-es,and the weight of each index to the target layer was calculated. Among them,the weight of validity and security is the top two. CONCLUSIONS:The weight coefficient can be determined scientifically and reasonably by AHP,and the reliability and operability of the evaluation index system can be improved. It is a useful attempt to explore the selection system of additional essential medi-cine.
4.Evidence-based Review on Yanhuning Injection in the Treatment of Epidemic Mumps

OBJECTIVE:To systematically evaluate therapeutic efficacy of Yanhuning injection in the treatment of epidemic mumps,explore valid way of evidence-based review,provide reference for the selection of new essential medicine list. METH-ODS:Retrieved from PubMed,CJFD,Wanfang database and VIP,randomized controlled trials(RCTs)about Yanhuning injection alone or combined with other positive drugs(trial group)vs. other positive drugs(control group)in the treatment of epidemic mumps. Meta-analysis was conducted by using Rev Man 5.3 statistical software after quality evaluation by modified Jadad scale. RE-SULTS:Totally 26 RCTs were included,involving 2613 patients. Results of Meta-analysis showed,overall response rate [OR=5.28,95%CI(3.92,7.11),P<0.001] of trial group was significantly higher than that of control group;defervescence time [MD=-1.97,95%CI(-2.83,-1.12),P<0.001],swelling disappearance time [MD=-2.05,95%CI(-2.53,-1.58),P<0.001] and pain disappearance time [MD=-1.39,95%CI(-2.02,-0.75),P<0.001] of trial group were significantly shorter than those of control group,there was statistical significance. CONCLUSIONS:Yanhuning injection has good curative effect for epidemic mumps,and can significantly shorten defervescence time,swelling disappearance time and pain disappearance time.
5.Value of ultrasound in diagnosis of lung atelectasis/consolidation in multiple trauma patients with mechanical ventilation

Objective To evaluate the value and significance of bedside ultrasound in diagnosis of lung atelectasis/consolidation in multiple trauma patients with mechanical ventilation.Methods Bedside thoracic ultrasound and chest computed tomography(CT)were performed in 81 multiple trauma patients with mechanical ventilation admitted to the emergency intensive care unit(EICU).CT result was regarded as the "golden standard" to evaluate the value of ultrasound in diagnose lung atelectasis/consolidation.At the same time,the ultrasound was used to dynamically monitor the lung recruitment effect of the therapeutic measures.Results CT detected 154 regions of lung atelectasis/consolidation of 324 lung regions in 81 patients,while ultrasound detected 126 regions that were divided into 87 complete regions and 39 incomplete regions according to different sonogram in the breathing cycle.The sensitivity,specificity,positive predictive value,negative predictive value and accuracy of ultrasound were 81.8%,100%,100%,85.9% and 91.4% respectively.A concordance test showed a very high concordance between ultrasound and CT,with a Kappa coefficient of 0.825(P=0.031).Ultrasound found that 39 regions with incomplete lung atelectasis/consolidation were completely recruited and 62 out of 87 regions with complete lung atelectnsis/eonsolidatian gained visible recruitment within three days after different therapeutic measures,with total effective rate of 80.2%.Conclusion Bedside ultrasound can continuously monitor and guide the therapeutic measure to recruit the lung and is a convenient,safe,direct-viewing and accurate method for diagnosis of lung atelectasis/cansolidation in multiple trauma patients with mechanical ventilation.
6.Effect of Cold Stress on Connexin43 Protein Expression With Drug Intervention in Neonatal Rats’ Myocardial Cells

Objective: To study the effects of acute cold stress on connexin43 (Cx43) protein expression with drug intervention, and cell to cell conduction with its mechanism in neonatal rats’ myocardial cells.
Methods: The primary neonatal rats’ myocardial cell culture was conducted in 4 groups. Group① , the cells were normally cultured, Group②, the cells were cultured at 4℃, Group③, the cells were cultured at 0℃and Group④, the anti-arrhythmia peptide (AAP 10) was added in Group②and Group③. The apoptosis rate of myocardial cells was evaluated by lfow cytometry assay, mRNA and protein expressions of CX43 were examined by RT-PCR and Western blot analysis, and CX43 phosphorylation product (P-CX43) was detected.
Results: Compared with normally cultured cells, the myocardial cell apoptosis rate was obviously increased by acute cold stress at 4℃and 0℃with time extension. The mRNA expression of Cx43 was decreased at varying degrees at 4℃and 0℃stimulation, the protein expression of Cx43 was decreased at varying degrees at 4℃and 0℃stimulation with time extension, and P-Cx43 level was decreased. While the APP 10 intervention may obviously elevate the protein levels of Cx43 and P-Cx43.
Conclusion: Acute cold stress could reduce the protein expression of CX43 and P-CX43, while APP 10 intervention may elevate such expression and improve the cell to cell conduction in neonatal rats’ myocardial cells.
7.Clinical value of CT 3D-imaging in guiding interventional occlusion therapy for PDA

Objective To discuss the clinical application value of 256-slice helical CT 3D-imaging in guiding interventional occlusion therapy for patent ductus arteriosus (PDA).Methods A total of 40 patients with sonography-proved PDA were randomly divided into group A (angiography group) and group B (CT-guided group) with 20 patients in each group.For the patients of group A,occlusion of PDA was performed based on the intraoperative angiography findings;and for the patients of group B,occlusion of PDA was carried out according to CT examination results.Intraoperative cardiac ultrasound monitoring was adopted and the curative effect was evaluated.Results The morphology of PDA demonstrated on CT 3Dimaging in group B was highly consistent with the configuration of PDA displayed on intraoperative angiography in group A.The most narrow diameters of PDA in group B and group A were (3.88±1.59) mm and (3.63±1.41) mm respectively,and the lengths of PDA in group B and group A were (6.1±1.06) mm and (6.82±0.74) mm respectively;the differences between the two groups were not statistically significant (P>0.05).The time spent for surgery in group B and group A was (34.3±9.11) min and (17.33±5.81) min respectively,and the intraoperative X-ray radiation doses in group B and group A were (33.93±11.0) mGy and (66.48±9.77) mGy respectively;the differences between the two groups were statistically significant (P<0.001).In group B,the preoperative X-ray radiation dose from CT examination was (119.79±29.45) mGy,when it was added to the intraoperative X-ray radiation dose the total cumulative radiation dose of group B was strikingly higher than that of group A.Conclusion Contrast-enhanced 256-slice helical CT scan and 3D-imaging technique can replace intraoperative angiography to get accurate anatomical imaging information of PDA,which are very helpful for the performance of interventional occlusion of PDA,meanwhile,it can effectively reduce the damage to the punctured artery and shorten the operation time.However,the radiation dose is a factor that should be taken into consideration.(J Intervent Radiol,2017,26:206-209)
8.Effect of cell loss in the CA3 area on hippocampal neurogenesis in chronically epileptic mice

Objective To study the influence of cell loss in the CA3 area on hippocampal neurogenesis in mice with temporal lobe epilepsy .Methods Newly born cells in the subgranular zone (SGZ ) of dentate gyrus were labeled by the proliferation marker bromodeoxyuridine (BrdU ) at 2 months after pilocarpine‐induced status epilepticus .Double labeling of BrdU+ NeuN was carried out to compare neuronal differentiation between type Ⅰand Ⅱ mice ,which represented partial cell loss and almost complete cell loss in the CA 3 area ,respectively .DCX and BrdU single staining were made to analyze the proliferation of progenitor cells in SGZ and the survival of the newly born cells .Results When compared with that of the type Ⅱ mice ,the number of double labeled cells of BrdU+NeuN in the subgranular zone‐granule cell layer (SGZ‐GCL) in the type Ⅰ mice increased significantly ( P<0 .05) .However ,the percentages of double labeled cells in all the BrdU + cells were found to be similar between the two groups (P>0 .05) .No significant difference in the number of DCX + cells in SGZ was found between the typeⅠ and Ⅱ mice (P>0 .05) .At 6 weeks after BrdU labeling ,more remaining BrdU+ cells were found in the SGZ‐GCL in the type Ⅰ mice when compared with their counterparts in the type Ⅱ mice (P<0 .05) .Conclusion The cell loss in the CA3 area of pilocarpine‐induced epileptic mice contributes to the declined hippocampal neurogenesis by exerting negative effects on the survival of newly born cells .
9.Preliminary analysis of hidden hemorrhage after primary unilateral total hip arthroplasty

Objective To analyze the hidden blood loss following primary total hip arthroplasty (THA) and put forward corresponding treatment measures.Methods A retrospective study was done on the 138 patients with hip disease managed with THA for the preliminary unilateral replacement from January 2009 to January 2011.There were 48 males and 90 females,at age range of 45-71 years (mean,62 years).The pre-and post-operative blood routine and intra-and post-operative blood loss and transfusion were analyzed and hidden blood loss during peri-operation period was evaluated.Results All the patients were trnsfused with autologous blood intra-operatively.Intra-operative blood loss was (495 ± 105) ml and postoperative drainage volume was ( 121 ±29) ml.In addition,116 patients were given red cell suspension (RCS) and blood plasma to raise the level of hemoglobin and the RCS transfusion volume was (600 ± 225) ml.The total blood loss was (1 521 ±156) ml including hidden blood loss of (822 ±96) ml (57.2%).No obvious differences were observed in the indicators between males and females.Conclusions Hidden blood loss is an important factor that affects the prognosis of the patients treated by THA.In order to secure their safety in perioperative period and benefit their recovery,assessment of the amount of bleeding in THA should fully take the hidden blood loss into consideration and prompt treatment measures should be taken to fully and effectively recover their effective circulation blood volume.
10.Influence mechanism of glial cell line-derived neurotrophic factor on the proliferation of spermatogonial stem cells

Objective To investigate the molecular mechanisms of glial cell derived neurotrophic factor in promoting proliferation of spermatogonial stem cell.Methods RNAi expression vectors,targeted at GDNF,were constructed and transfected into SSCs from 5 to 7 days old mice.The SSCs with highest effectiveness of GDNF interfere was set as study group.And the SSCs without GDNF interfere was considered as control group.The ELISA method was used to compare the proliferative rate between study group and control group.Flow cytometry,RT-PCR were used to detect the expression of GDNF,RTKs,Fyn and FAK's mRNA,and the apoptosis of SSCs.Results From 1 to 4 days after transinfection,the absorbable A value in study group was 0.45 ± 0.02,0.68 ± 0.03,1.12 ± 0.03,2.24 ± 0.04,respectively.Meanwhile,the same item in control group was 0.46 ± 0.03、0.73 ± 0.02、1.32 ± 0.05、1.15 ± 0.06,respectively (P < 0.05).There were significant different between experiment groups (25.43 ± 1.91) % and control group (5.61 ± 0.16)% in the apoptosis rates of SSCs (P < 0.05).Significant differences were noted between experimental group and control group(P < 0.05).The mRNA expression rates of GDNF was (12.32 ± 1.22) % in study group and (54.25 ± 1.34)% in control group (P <0.01).The mRNA expression rates of RTKs and Fyn and FAK in study group and control group were (16.24 ± 1.35)% vs (45.35 ± 1.37)%,(18.32 ±1.34)% vs (38.37 ± 1.55)%,(20.04 ± 1.65)% vs (43.27 ± 1.28)%,respectively (P <0.05).Conclusions The glial cell line derived neurotrophic factor was important in course of SSCs' proliferation,which may up-regulating the expression of RTKs,Fyn and FAK.
