1.Ex vivo expansion of human cord blood CD_(34)~+ cells with hematopoietic growth factors

Objective: To investigate the role of recombinant human hematopoietic growth factors(HGFs) in expansion of purified CD34+ cells derived from human cord blood.Methods: A sterilized separation system for purification of CD34+ hematopoietic stem / progentior cells(HSC/HPC) was established by means of StemsepTM according to the strategy of negative selection on immunomagnetic separation.CD34+ cells were expanded with various combination of cytokines in a 21-day liquid culture systems.Results: A combination of seven HGFs including FL,Tpo,SCF,IL-3,GM-CSF,G-CSF and Epo was identified as the most potent combination of those tested,and it increased total nucleated cell by 1627.57?52.93 fold,total progenitor cells(CFCs) by 56.78?8.13 fold and CD34+ cell by 41.09?4.11 fold,respectively.Among the HGFs examined,FL and Tpo were the most potent ones for expanding cord blood CD34+ cells.Maximal expansions of CFCs and CD34+ cells were observed during the first two weeks of culture.Conclusion: Under conditions of appropriate HGFs combination and expansion duration,it is possible to expand cord blood CD34+ cells ex vivo for transplantation and therapeutic gene transfer.
2.STUDY ON THE SIGNAL TRANSDUCTION AND REGULATION IN ERYTHROPOIESIS BY TOTAL SAPONINS OF PANAX GINSENG

Objective To investgate the signal transduction and regulation in erythropoiesis by total saponins of Panax ginseng(TSPG) to clarify the mechanism for Panax Ginseng promoting hematopoiesis.Methods The colony-forming ability of mononuclear cells(MNCs) from human umbilical cord blood was assayed in methylcellulose assay systems,various dilutions of TSPG(0,10-100mg/L) were then added to each methylcellulose culture mixture;MNC proliferation in response to erythropoietin(Epo) was measured by MTT;To investigate whether the preferential differentiation of MNCs into erythroid lineage cells was caused by an increase in the expression of erythropoietin receptor(EpoR) on the cells,immunoblotting analyses were performed;Immunoprecipitation of JAK_2/STAT_5 was used for the effect of TSPG on Epo/EpoR-induced tyrosine phosphorylation of JAK_2/STAT_5. Results 1.TSPG(10-75mg/L) can promote the colony formation of BFU-E,CFU-E,and the preferential differentiation into erythroid lineage cells was induced from 25mg/L of TSPG;2.Using the colorimetric MTT assay,MNCs exhibited proliferous responses to Epo(2?10~3-5?10~4IU/L) reaching maximum at 5?10~3IU/L Epo;3.The addition of TSPG did not increase the expression of EpoR;4.After MNCs were incubated in the presence of with or without TSPG for 24 h,the cells were stimulated with Epo for 0,2,5,30min,and tyrosine phosphorylation JAK_2/STAT_5 was measured.The pretreatment with TSPG enhanced Epo-induced tyrosine phosphorylation of JAK_2 and STAT_5(STAT_(5a) and STAT_(5b)).Conclusion In the present study,we have shown that the Epo/EpoR-mediated signals are enhanced,and JAK_2/STAT_5 signals plays an essential role in the effect of TSPG on erythropoiesis,whereas the expression level of EpoR remains unchanged.

5.Effects of the methods of treating Shaoyang by dispersing and dispelling wind and relieving pain on thrombosis in vitro and function of platelet aggregation in rats

BACKGROUND: Migraine is usually located in the course of Shaoyang channel, and the curative effect of treatment with methods of treating Shaoyang disease by dispersing and dispelling wind (DW) as well as relieving pain (RP) on migraine is prominent, but how about the effects of these two ways on thrombogenesis in vitro and the function of platelet aggregation (PA)?OBJECTIVE: To probe into the effect of treatment of Shaoyang disease and methods of DW and RP on thrombogenesis in vitro and function of PA throughthe experiments of thrombogenesis in vitro and function of PA in rats, and investigate the compatible significance of DS and DW as well as RP through formula compatible experiment.DESIGN: Completely randomized grouping design and controlled trial.SETTING: Department of Prescription, College of Basic Medical Sciences,Heilongjiang University of Chinese Medicine.MATERIALS: The experiment was conducted in the Laboratory of Department of Prescription, Heilongjiang University of Traditional Chinese Medicine from March to August 2000. Sixty healthy adult Wistar rats were randomly divided into 6 groups: Normal control group, Shaoyang treatment group (group Ⅰ ), DW and RP group (group Ⅱ ), high-dose and whole-prescription group (group Ⅲ), low-dose and whole-prescription group (group Ⅳ), and positive control group with 10 rats in each group.METHODS:① Positive control group:Gastric perfusion of 0.034 g/mL compound divaside slice suspension (made by Harbin Second Biochemical Pharmacy co., Ltd) was given to rats (0.39/kg); Group Ⅲ and Ⅳ (ingredients: Chaihu 20 g, huangqin 10 g, banxia 15 g, gancao 10 g, chuanqiong 20 g, tianma 15 g, xixin 5 g, quanxie 5 g, wugong 5 g): Gastric perfusion of 1.5, 0.75 g/mL of complete prescription was given to rats (17.40, 8.70 g/kg);Group Ⅰ (ingredients: Chaihu 20 g, huangqin 10 g, banxia 15 g, gancao 10 g) and group Ⅱ (ingredients: Chuanqiong 20 g, tianma 15 g, xixin 5 g, quanxie 5 g, wugong 5 g): Gastric perfusion of 0.75 g/mL agents of treatment with DS and DW wind antidyne was given to rats as 8.70 g/kg; Normal control group: Gastric perfusion of normal saline of the same volume wasgiven to rats.Intervention on rats of all groups lasted 12 days.② Experiment of thrombogenesis in vitro: The cephalic artery of one side was seperated 2 hours after the last time of administration. The distal part was deligated, the blood current in proximal part was blocked with bulldog clamp and the arterial cannula was inserted into the cephalic artery. Loosened the bulldog clamp, blood of 1.8 mL was collected and put into the rotary ring of thrombogensis meter to rotate for 15 minutes at 17 r/min,and then poured the thrombus, measured the length and humid weight of thromb. After that,dried the humid thromb and measured the dry weight.③ Determination of PA:The packing fraction (PF) in the 1st and 5th minutes as well as the maximal packing fraction (MPF) and assembling inhibition ratio of platelet-rich plasma within 5 minutes were recorded with test.MAIN OUTCOME MEASURES: Effect of treating Shaoyang disease nephelometry.④ Differences of measurement data were compared with tand methods of DW as well as RP on thrombogenesis in vitro and the function of PA.RESULTS: A total of 60 rats were involved in the analysis of results. ①The length, humid weight and dry weight of thrombogenesis in vitro in group Ⅲ and Ⅳ, positive control group and group Ⅱ were obviously lower than those in group Ⅰ (P < 0.05). ② The PF in 1st, 5th minute and the maximal aggregathe normal control group (P < 0.05-0.01); The humid weight and dry weight of thromb in group Ⅳ were significantly lower than those in tion (MA) of blood platelet in rats of each group were significantly lower than those in the normal control group (P < 0.05-0.01), and the curative effect on rats in the positive control group, group Ⅲ and Ⅳ as well as group Ⅱ were better than that in group Ⅰ.CONCLUSION: There are no significant effects of dispersing thrombogenesis in vitro and inhibiting PA by only treating Shaoyang disease. The effect of combined prescription of DW and RP as well as treatment of Shaoyang disease are obviously enhanced.

The concept of golden mean (Zhong Yong), i.e. moderation, is the highest principle of moral cultivation in Confucianism. It entails an exclusive methodology in addressing politics, philosophy and everything in the universe. This concept has permeated into all aspects of traditional Chinese culture. As an ancient therapy, tuina is an important part of Chinese medicine and has its characteristic theory and methods. Just like Chinese medicine, tuina has long been greatly influenced by the concept of golden mean. To better promote the development of tuina, it’s necessary to investigate the historical evolution of tuina and the concept of golden mean as well as the influence of ‘golden mean’ idea on tuina manipulation, mechanism and treatment.

Objective:This article collected, organized, and summarized the medical literatures of cupping therapy from three major Chinese databases including China National Knowledge Infrastructure (CNKI), Chongqing VIP (VIP), and WanFang, with cupping, moving cupping, flash cupping, pricking cupping, or medicinal cupping as the retrieve title or key words, and the retrieve range was from 1953 to July 15, 2015. This article was to give an overall review of cupping therapy from the clinical application and mechanism research based on its application form, indications, and mechanism research progress. The results showed that there are various types of cupping methods; its indications cover multiple subjects such as internal medicine, external medicine, gynecology and pediatrics; and its major action mechanisms include negative pressure suction, increasing pain threshold, promoting dermal blood flow and improving body immunity.

10.A clinical analysis of patients with hepatitis B virus related acute-on-chronic liver failure treated by combination of traditional Chinese and western medicine

Objective To observe the clinical curative effect of combination of traditional Chinese and western medicine for treatment of patients with hepatitis B virus (HBV) related acute-on-chronic (subacute) liver failure (ACLF). Methods A prospective randomized controlled trial was conducted; 66 cases of HBV-ACLF patients were randomly divided into two groups: a test group (44 cases) and a control group (22 cases). Conventional western medicine treatment was given to both groups; the patients in test group additionally received the traditional Chinese medicine (TCM) in accord to the principles of differentiation of syndromes in TCM, in cases with damp-heat and blood stasis syndrome with yellow appearance, Liangxue Jiedu Huayu decoction (Paeoniae Radix Rubra 60 - 150 g, Artemisiae Scopariae Herba 30 - 90 g, Gardeniae Fructus 9 - 12 g, Hedyotis diffusa Willd 20 - 30 g, Salviae Miltiorrhizae Radix et Rhizoma 30 g, Atractylodis Macrocephalae Rhizoma 30 g, Rubiae Radix et Rhizoma 30 - 45 g, Siegesbeckiae Herba 30 - 45 g, Bletillae Rhizoma 15 g ) was given, in cases with Qi deficiency and blood stasis with yellow appearance, Yiqi Jiedu Huayu decoction (Astragali Radix Preparata Cum Melle 30 g, Pseudostellariae Radix 15 g, Artemisiae Scopariae Herba 30 - 60 g, Polygoni Cuspidati Rhizoma et Radix 15 - 30 g, Salviae Miltiorrhizae Radix et Rhizoma 30 g, Aconiti Lateralis Radix Preparata 10 - 15 g, Atractylodis Macrocephalae Rhizoma 30 g, Rubiae Radix et Rhizoma 30 - 45 g, Siegesbeckiae Herba 30 - 45 g, Gigeriae Galli Endothelium Corneum 20 g) was given, the dosage in both groups being 1 dose daily, one dose was prepared to a water decoction 250 - 300 mL which was divided into two parts, one part taken twice a day; the control group received only western medicine treatment. After 2 weeks of treatment, the clinical comprehensive curative effect, the syndrome score efficacy, and the changes of main indexes of liver function,cholinesterase (ChE), albumin (Alb), prothrombin activity (PTA) were observed in the two groups.Results The clinical total efficacy in the test group was significantly higher than that in the control group [75.0% (33/44) vs. 45.5% (10/22),P < 0.05]. The syndrome score total efficacy in the test group was also higher than that in the control group [75.0% (33/44) vs. 63.6% (14/22)], but the difference was not statistically significant (P > 0.05). After treatment, the alanine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin (TBil) and direct bilirubin (DBil) were all lower than those before treatment in both groups, while the ChE, Alb and PTA were higher than those before treatment, and the degree of changes was more significant in the test group [test group: ALT (U/L): 63.28±99.28 vs. 574.58±571.08, AST (U/L): 86.84±92.88 vs. 438.20±482.74, TBil (μmol/L): 161.90±178.34 vs. 269.46±95.10, DBil (μmol/L): 115.32±126.51 vs. 209.12±79.78, ChE (U/L): 4 239.14±1 505.00 vs. 3 341.49±1 609.40, Alb (g/L): 32.65±4.77 vs. 29.73±3.31, PTA: (69.69±44.92)% vs. (32.84±7.47)%; control group: ALT (U/L): 93.28±93.86 vs. 365.24±376.98, AST (U/L): 126.26±121.35 vs. 287.17±301.04, TBil (μmol/L): 226.80±187.38 vs. 281.02±103.73, DBil (μmol/L): 172.50±147.32 vs. 227.96±87.20, ChE (U/L): 4 484.66±1 886.53 vs. 3 918.77±1 417.77, Alb (g/L): 33.17±4.76 vs. 30.47±3.03, PTA: (63.80±36.80)% vs. (33.96±6.32)%,P < 0.05 orP < 0.01].Conclusion The combination of TCM and western medicine for treatment of HBV-ACLF can improve liver function, and the prognosis is superior to using western medicine treatment alone.
