1.Serum NO and NOS in Different Kinds of Cerebral Vascular Stenosis

Objective To search for the mechanism of NO and TNOS/iNOS in cerebral vascular stenosis,the changes of these serum factors in different kinds of cerebral vascular stenosis were studied. Methods Biochemistry assays were adopted,to analyze serum NO and TNOS/iNOS in different vascular stenosis group. Results Serum NO contents of ' Mix' (P=0.000) or ' MCA' (P=0.001) significantly increased. INOS activity of ' In' (P=0.004) and ' MCA' (P=0.002) markedly increased. And TNOS activity of ' In' group significantly weakened (P=0.032),but the TNOS activity of ' Mix' (P=0.011) and ' MCA' (P=0.018) group markedly intensified. Conclusion NO and TNOS/ iNOS partly participate in the occurrence mechanism of cerebral vascular stenosis. And the study provided theory basis for studying multiple factors of cerebral vascular stenosis.
2.Observation on sterile effect of sterile adhesive tape on opened infusion bottle mouth

Objective To discuss the sterile effect of sterile adhesive tape on opened infusion bottle mouth. Methods From February to May, 2009, 400 infusion bottles were chosen and divided into the observation group and the control group with 200 bottles in each. The bottle mouth in the observation group was covered with sterile adhesive tape, while the control group used iodophor. The incidence of infusion reaction, disposable materials and time consuming of nurses were compared. Results No infusion reaction occurred in both groups. But disposable materials and time consuming was less in the observation group compared with the control group. Conclusions Application of sterile adhesive tape in sterilization of opened infusion bottle mouth is safe, reliable, simple and economic, it can save time and increase work efficiency of nurses.
3.The effect of hyperoxaluria and ureteral obstruction on renal tubular epithelial cell apoptosis rats

Objective To investigate the role of hyperoxaluria and unilateral complete ureteral obstruction in the development of renal tubular epithelial cell apoptosis. Methods Forty male Wistar rats were divided into four groups randomly,with ten rats in each.The groups were named A(control),B(hyperoxaluric),C(obstructive) and D(hyperoxaluric+obstructive).The rats were sacrificed 2 weeks after operation;left nephrectomy was then performed.The tissue sections obtained from kidneys were examined under light microscopy for the presence and the degree of crystal deposition in the tubular lumen.Apoptotic changes in renal tubular cells were examined using the terminal deoxynucleotidyl transferase(TdT)-mediated dUTP in situ nick and labeling (TUNEL) method. Results Oxalate crystal deposition was evident in the renal tubular lumen in groups B and D,while there was no evidence in groups A and C.With respect to apoptotic changes,the mean values of the apoptotic cell count were 1.1?1.1 in A and 31.3?8.0 in B.The values for C and D were 145.0?33.4 and 275.8?51,respectively.There was statistically significant difference between all goups ( P

5.Analysis of early complications of free gracilis muscle transfer in reconstruction of brachial plexus injury

Objective:To explore the causes and preventive measures of early complications after free gracilis muscle transfer in reconstruction of brachial plexus injury, and to improve the postoperative function of the transferred gracilis muscle.Methods:Patients were recruited from August, 2005 to December, 2016. All 111 patients of brachial plexus injury underwent reconstructive surgery using 122 free gracilis flaps. Early postoperative complications, including recipient site, donor site and systemic complications, were closely observed and recorded. Outcome measurements included incidence and timing of flap compromise, type of flap compromise, causes of vascular occlusion and salvage rate. The postoperative strength of gracilis was evaluated according to the BMRC score. The data were statistically analyzed. The difference was statistically significant if P<0.05. Results:The survival rate of 122 free gracilis transfers was 98.4% (120/122). Early complications occurred in 32 cases (including 2 complications in 6 patients) : 27 cases with recipient site complications (84.4%), 4 with donor site complications and 7 with systemic complications. Among the 32 cases of complications, 17 flap compromises caused by vascular obstruction and 15 of them were salvaged completely after exploration. Flap crisis was the main issue that affected the postoperative function of gracilis muscle, and 58.8% (10/17) of patients with vascular crisis showed muscle strength above M 3 after surgery. The main causes of vascular crisis were venous tortuosity and venous thrombosis, which had nothing to do with operation time and intraoperative blood loss. Conclusion:Flap crisis is the main factor affecting the postoperative function of gracilis. The rate of flap salvage can be tremendously increased by early detection, re-exploration and effective management of the flap crisis.
6.CD44s expression study in colorectal cancer tissues

Objective To investigate the relationship between standard CD44(CD44s)expression and tumor stage and prognosis in colorectal cancer.Methods CD44s expression was detected by immunohistochemistry in tumor tissues and normal mucosa specimens from 74 cases of primary colorectal cancer.Results Of 74 cases,the expression of CD44s in colorectal cancer tissue and normal mucosa specimens was documented in 42% and 16%,respectively.The expression of CD44s in primary tumors significantly increased in stage Ⅲ and Ⅳ than in stage Ⅰ and Ⅱ(39% vs.6%,x~2=8.46,P<0.01).A statistically significant correlation was observed between the overall survival and CD44s expression(x~2=17.82,P<0.01).Furthermore,a poor prognosis of CD44s-positive tumors was observed in patients with stage Ⅲ and Ⅳ colorectal cancer(x~2=16.23,P<0.01),but not in patients with stage Ⅰ and Ⅱ colorectal cancer(x~2=1.34,P>0.05).Multivariate analysis indicated that TNM stage and CD44s expression were independent predictors of overall survival in colorectal cancer.Conclusion CD44s overexpression is involved in the progress of colorectal cancer and predicting the prognosis.
7.Injection of human umbilical cord derived mesenchymal stem ceils into the vitreous of rabbits

Objective To observe the survival of human umbilical cord derived mesenchymal stem cells (hUC-MSCs) after injection into the vitreous of rabbits,and the animal safety under those procedures.Methods Twenty-seven pigmented rabbits were randomly divided into 3 groups (intravitreal injection 1 week group,2 weeks group and 4 weeks group),each with 9 rabbits.For each animal the right eye was the experimental eye receiving hUC-MSCs injection,while the left eye was the control eye receiving cuhure medium.The rabbit eyes were examined by slit-lamp microscope,indirect ophthalmoscopy,fundus photography,fundus fluorescence angiography(FFA)and Tono-pen tonometer before and after injection.hUC-MSCs were labeled by CM-Dil in vitro,and their survival status was measured by eonfocal fluorescence microscopy,light microscope and transmission electron microscope at 4 weeks after injection.Results Four weeks after injection,a large number of the hUC-MSCs were still alive in the vitreous cavity.The overall condition of those rabbits was good.The anterior segment and retina of experimental eyes were normal,without hyperfluorescence,hypofluorescenee and leakage in the retina at 1,2 and 4 weeks after injection.There was no significant difference on lOP before and after injection at different time points (P>0.05),and no obvious changes at cornea,anterior chamber angle,lens,retinal structure by.light microscope and transmission electron microscope examination.Conclusion hUC-MSCs can survive in the rabbit vitreous for four weeks;intravitreal injection of hUC-MSCs was safe and feasible.
8.Effectiveness of comprehensive intervention on rural community patients with diabetes

Objective To evaluate the impacts of comprehensive diabetes intervention to provide evidence for effective diabetic control in rural communities.Methods Diabetic patients from four communities at Wujin District of Changzhou City were selected.One hundred and seventy-three patients from two communities received comprehensive intervention,including medication therapy,diabetes education,and diet.exercise,and behavior instruction(intervention group);142 patients from the other two communities got medication therapy only (control group).All the participants were interviewed with structured questionnaire and phyrsical examination.The second epidemic investigation was conducted at 4 years to evaluate the effects of the intervention.Results At the end of the intervention,the awareness rates of normal fasting plasma glucose,major diabetes risk factors,healthy diet,and therapeutic strategies in the intervention group were 90.8%.74.6%,89.6%,and 59.5%,respectively,which were significantly higher than those in the control group (all P<0.05).For the intervention group,the rates of diet behavior or physical exercise change were 68.8% and 58.4%.respectively.which were much higher than those of the control group(all P<0.05).The average levels of systolic pressure,diastolic pressure,fasting plasma glucose,and total cholesterol were (130.6±10.4)mm Hg(1 mm Hg=0.133 kPa),(78.4±7.5)mm Hg,(6.2±3.1)mmol/L,and (4.5±0.9)mmol/L,respectively,in the intervention group,which were lower than those in the control group(all P<0.05).Conclusion Comprehensive intervention shows significant effects on rural community patients with diabetes.
9.Analysis on the prevalence and influencing factor of metabolic syndrome in rural elderly population in Changzhou of Jiangsu province

Objective To investigate the prevalence and influencing factor of metabolic syndrome(MS)in rural elderly population. Methods With randomized cluster sampling,investigations including questionnaires,physical examination,fasting plasma glucose,blood lipids were performed in rural population aged over 60 years in Changzhou city in 2004-2005. Results A total of 4976 elderly people were enrolled in the study.The prevalence of MS was 24.1%,and 10.3%in men,34.3%in women by international diabetes federation(IDF)2005 definition.86.2%individuals had one or more of the metabolic disorders.Logistic regression analysis showed that sex,smoking,sweetmeat,family history of essential hypertension(EH)and tea drinking were influencing factors of MS. Conclusions The prevalence of MS is high in rural elderly population,especially in females.
10.The role and mechanism of resveratrol on trabecular meshwork cells induced by H2O2 and TGF-β2

Objective To investigate hydrogen peroxide (H2O2) and transforming growth factor-β2 (TGF-β2) induced fibronectin (FN), collagen 1 (COL1), nuclear factor (NF)-κB P65 proteins and interlukin (IL)-1βgene expression in human trabecular meshwork cells (HTMCs), and the interventional mechanism of resveratrol (RSV). Methods (1) HTMCs with 70 to 80%confluency were divided into 5 groups. The experimental groups were treated with serum-free medium and with H2O2 at concentrations of 150, 300, 450 and 800μmol/L. The control group was treated with 0μmol/L H2O2. The protein levels of FN, COL1, NF-κB P65 and NF-κB P65 phosphorylation (P-NF-κB P65) were measured by Western blot assay. The expression of IL-1βgene was measured by qPCR. (2) HTMCs were divided into 3 groups. The control group was treated withserum-free medium and without H2O2 and RSV. The H2O2 group was treated with 300μmol/L H2O2. The H2O2+RSV group was treated with 300μmol/L H2O2 and 25μmol/L resveratrol (RSV). The expressions of proteins and genes mentioned above were detected in three groups. NF-κB P65 nuclear translocation was assessed by immunofluorescence technique. (3) HTMCs were divided into 3 groups. The control group was treated with serum-free medium and without TGF-β2 and RSV. The TGF-β2 group was treated with 5μg/L TGF-β2. The TGF-β2+RSV group was treated with 5μg/L TGF-β2 and 25μmol/L RSV. The expressions of proteins and genes mentioned above were detected in three groups. Results (1) Compared with control group, the protein levels of FN and P-NF-κB P65 were significantly increased in 150, 300, 450 and 800μmol/L groups,the expression levels of COL1 protein and IL-1β gene were significantly increased in 300, 450 and 800 μmol/L groups (P <0.05). There were no statistical significances between other indicators. (2) The expression levels of FN, COL1, P-NF-κB P65 proteins and IL-1βgene were significantly higher in H2O2 group than those in control group, and which were significantly lower in H2O2+RSV group than those in H2O2 group. Compared with control group, only the expression of IL-1βgene was decreased in H2O2+RSV group (P < 0.05). NF-κB P65 was only expressed in cytoplasm in control group, while it was expressed in both cytoplasm and nucleus in H2O2 group. Compared with H2O2 group, NF-κB P65 was mainly expressed in cytoplasm. (3) Compared with control group, the expressions of FN, COL1, P-NF-κB P65 proteins and IL-1β gene were significantly increased in TGF-β2 group (P < 0.05). Compared with TGF-β2 group, the indicators mentioned above were significantly decreased in TGF-β2+RSV group (P<0.05). Conclusion H2O2 and TGF-β2 can upregulate the expression of FN, COL1, P-NF-κB P65 proteins and IL-1βgene in HTMCs, which may be involved in the development and progression of glaucoma. RSV can inhibit the influence of H2O2 and TGF-β2 in HTMCs and exert a protective effect on glaucoma.
