1.The application of vitamin D in essential hypertension treatment

Objective To investigate the standard supplement of a certain dose of vitamin D for blood pressure control in pa-tients with essential hypertension.Methods 100 cases of patients with essential hypertension were randomly divided into 2 groups voluntarily,50 cases in each group.One group were set as control group,in which patients underwent a standard treatment.The other group was experimental group,in which 25 μg/d vitamin D was administrated in addition to standard treatment.Serum 25-hydroxyvitamin D[25 (OH)D]concentrations were measured by enzyme-linked immunosorbent assay(ELISA)and patients were followed up for one year to observe the situation of blood pressure controll.Mean and variable coefficient(CV)of systolic and dias-tolic blood pressure were calculated and compared between groups.Results In experiment group,the serum 25 (OH)D concentra-tion was (110.3±24.5)μg/L,systolic pressure was (144±16)mm Hg,variable coefficient of systolic pressure(CV1)was 11.1%, diastolic pressure was (87±9)mm Hg,variable coefficient of diastolic pressure(CV2)was 10.3%.While in control group,the 25 (OH)D concentration was (30.1±14.9)μg/L,systolic pressure was (145 ±26)mm Hg,CV1 was 17.9%,diastolic pressure was (87±10)mm Hg,CV2 was 12.4%.Serum 25(OH)D concentration and CV1 were statistically different between the two groups(P<0.01),CV2 was not statistically different(P >0.05).Conclusion Administrate quantitative vitamin D in addition to standard treat-ment is helpful for systolic pressure control in patients with essential hypertension but is meaningless for diastolic pressure control.
2.Diagnostic value ofβ-HCG and fβ-HCG of adverse pregnancy

Objective To explore the diagnostic value of β‐human chorionic gonadotropin (β‐HCG) and its free β‐subunit (fβ‐HCG) of adverse pregnancy .Methods Chemiluminescence technique was used for the detection of serum levels ofβ‐HCG ,fβ‐HCG and progesterone in women with early adverse pregnancy (threatened abortion group ,missed abortion group and ectopic pregnancy group) and normal pregnant women (control group) ,respectively .The 48 h doubling rate ofβ‐HCG of each group was calculated . And the results were analyzed .Results Comparing with threatened abortion group ,there were significant differences in the levels of β‐HCG ,fβ‐HCG ,progesterone and doubling rate both of missed abortion group and ectopic pregnancy group (P<0 .05) .How‐ever ,there was no difference in these indicators between missed abortion group and ectopic pregnancy group ( P> 0 .05 ) . Conclusion T he changes of β‐HCG and fβ‐HCG can reflect adverse pregnancy .
3.Arthroscopic-assisted anterior cruciate ligament reconstruction

In oder to make the bone tunnel in optimal position, effective and firm fixation, early functional recov- ery and minimally operative injury we reported a new technique in reconstructive anterior cruciate ligament(ACL)using patellar tendon autograft fixed with interference screw. With the assistance of arthroscopy 74 patients with ACL rupture were treated between October 1997 and April 1999. The clinical results showed that the operative injury was minimal, fixed bone grafts were accurate and the reconstructed ACL was isometricity. The reconstructed ligament could be fixed biologically and firmly because of used patellor tendon autograft with bone grafts at two end. Clinical experience of ar- throscopic - assisted ACL reconstruction was summarized.
4.Establishment of Anterior Instability of Gienohumeral Joint on Rabbits

Objective To establish the animal model(rabbit) of shoulder instability for providing the experimental basis of related studies.Methods shoulder joints of 6 skeletally matured New Zealand white rabbits were measured with the anatomical characteristics and joint motion range in order to determine the operative approach,the similarities of skeletal structure,surrounding muscles,joint capsule and ligaments between rabbit and human were compared.Eighteen rabbits,weighing between 2.5 and 3kg were randomized into 2 groups(experimental group 12 and control 6).In experimental group,instability model was established by means of the operation procedure as follows:the glenohumeral joint of right shoulder was exposed by cutting the subscapularis tendon,then the anteroinferior labrum and surrounding capsular ligament were excised.For control group,sham operation was performed by only curing the skin and superficial tissue,the subscapularis tendon and glenoid structures was kept intact.The specimens of 6 rabbits were harvested 6 weeks after operation.The remaining rabbits of both experimental and control groups were killed 12 weeks post the operation for specimens processing.The motion range and instability were measured in all specimens.Results The gross anatomy of shoulder ioint in rabbit is similar to human.In experimental group,the stability is showed less than control,grade 2 anterior instability was found.The motion range was restricted in all directions,such as external and internal rotation,extension and abduction compared to control group.Conclusion Rabbit is suitable to establish animal model for shoulder instability and related iniuries,the animal model which was designed by this study could provide valuable information for the research of anterior shoulder instability.
5.Clinical studies of vitamin D intervention in patients with hypertension

Objective To investigate the effect of controlling blood pressure in treating patients with hypertension by taking quantitative vitamin D to provide support for the clinical treatment of patients with hypertension.Methods 100 cases with hypertension were randomly divided into the two groups voluntarily,50 cases in each group and one group with a simple standard treatment was considered as the control group,and the other one was the study group,25μg/d vitamin D was taken besides standard treatment.Each group content of 25-hydroxyvitamin D [25 (OH)D] level were measured by Enzyme-linked immunosorbent assay (ELISA) and lasted for one year to detect the situation of controlling blood.Mean and variable coefficient of systolic and diastolic blood pressure were calculated and compared each other.Results After treatment,the content level of 25 (OH)D was (110.3 ± 24.5)nmol/L,systolic pressure was (144 ± 16) mmHg,variable coefficient was 11.1%,diastolic pressure was (87 ± 9) mmHg,variable coefficient was 10.3% in experiment group;while the content of level of 25 (OH)D was (30.1 ± 14.9)mol/L,systolic pressure was (145 ± 26)mmHg,variable coefficient was 17.9%,diastolic pressure was (87 ± 10)mmHg,variable coefficient was 12.4% ;the content of level of 25 (OH)D,variable coefficient of systolic pressure had statistical difference (t =12.47,x2 =6.72,P < 0.01),variable coefficient of diastolic pressure had no statistical difference (P > 0.05).Conclusion Taking quantitative vitamin D besides standard treatment is benefit for controlling the level of systolic pressure in patients with hypertension but is meaningless to the level of diastolic pressure.
6.Arthroscopic subacromial decompression for subacromial impingement syndrome

Objective To explore the clinical results of arthroscopic subacromial decompression (ASD) for treatment of subacromial impingement syndrome. Methods Twelve cases with subacromial impingement syndrome from October 1998 to November 2003 were retrospectively studied. There was one case at degree Ⅰ, five at degree Ⅱ and six at degree Ⅲ according to Neer classification. Preoperative X-ray of obverse shoulder joint and outlet supraspinatus muscle was made. Seven cases was examined with ultrasonography and 10 with MRI or MRA. All cases were treated with arthroscopic subacromial decompression and arthroscopic acromioplasty and six cases underwent simultaneously arthroscopic suture of the rotator cuff. Results The follow up for 1-6 years (average 26 months) showed significant statistical difference upon preoperative UCLA score and postoperative UCLA score that were 16.9?4.0 and 32.5?1.4, respectively (t=-14.027,P﹤0.01). The results were excellent in two cases and good in ten. All cases were satisfied with the operation. Conclusions ASD is shown to be an effective procedure for subacromial impingement syndrome and has the advantages of minimal trauma and rapid postoperative recovery.
7.Development and evaluation of HEK293 cells stably expressing CRFR1

Aim ToconstructHEK293cellsstablyex-pressing corticotropin releasing factor receptor 1 ( CRFR1 ) , and evaluate its function by the cAMP as-say.Methods CulturedHEK293cellsweretransfect-ed with CRFR1-expressing vector by Lipofectamine 2000 and were selected by using G418 . CRFR1 ex-pression was detected by Western blot, RT-PCR and immunofluorescence.Results Westernblot,RT-PCR and immunofluorescence data revealed that the HEK293 cells expressed CRFR1 protein stably. The dose-responsive relationship experiment revealed that CRF induced a CRFR1-mediated cAMP production in HEK293 cells with EC50 =(5. 64 ± 0. 05) × 10 -10 mol ·L-1.Conclusion HEK293celllinesstablyex-pressing CRFR1 were constructed successfully, which would provide a cellular model to facilitate the research on the biological function of CRFR1 and CRFR1-targe-ted drug screening.
8.Application of intestinal fatty acid binding protein and blood procalcitonin for diagnosis of traumatic intestinal rupture in early stage

Objective To investigate the role of intestinal fatty acid binding protein (IFABP) and blood procalcitonin (PCT) in diagnosis of traumatic Intestinal rupture in early stage.Methods The clinical data of 58 patients with abdominal injuries admitted from May 2012 to April 2016 were retrospectively analyzed.All 58 patients were divided into intestinal rupture group (n =21) and nonintestinal rupture group (n =37).The concentrations of IFABP and PCT were detected,analyzed and compared between two groups at different intervals.Results The IFABP and PCT in intestinal rupture group were significantly higher than those in non-intestinal rupture group.The IFABP and PCT in intestinal rupture group significantly decreased after operations.There were significantly differences in IFABP and PCT between two groups at admission,4 hours after admission,preoperative period,and 24 hours after operation.However,these differences disappeared at 72 hours after operation.At the same time,the accuracy rate 92.4%,sensitivity 96.3%,specificity 72.8% found in combination of these two biomarkers were significantly higher than those of IFABP and PCT measured separately.Conclusions The combination of IFABP and PCT detection can be used as an indicator for the diagnosis of traumatic intestinal rupture in the early stage.
9.Damage control surgery in severe abdominal trauma

Objective To investigate the utilization of damage control surgery (DCS) in treatment of the severe abdominal traumas.Methods The clinical data of 56 patients with severe abdominal traumas treated by DCS (Emergency operation,fluid resuscitation and definitive operation were used gradually.)from January 2011 to January 2013 in Department of General Surgery,Taizhou People's Hospital were retrospectively analyzed.Results Of the 56 patients,52 were cured and discharged from hospital,the clinical curative rate was 92.9%; 4 patients dead.Conclusions Abdominal traumas under going operations by applying the damage control surgery significantly increased the clinical curative rate.
10.Generation of fully human anti-vascular endothelial growth factor 165 monoclonal antibody with antitumor efficacy from transgenic five-feature mice

Aim: To prepare a fully human anti-VEGF_(165)(vascular endothelial growth factor 165) monoclonal antibody with antitumor activity from five-feature mice which express human immunoglobin loci. Methods: A routine method for the generation of monoclonal antibodies( mAbs) against the human VEGF_(165) was developed. The immunizing effect between five-feature mice and BALB/c was observed and the mAb was purified through MBP IgM affinity chromatography. The effect of mAbs on antitumor was tested ire vitro by T24 cell line. Results: Four hybri-domata secreting mAbs steadily were isolated successfully, and the serum titer of mAb in BALB/c mice was almost 10 times higher than that in five-feature mice. The indirect ELISA method for mAb titer determination was also established. The anti-VEGF_(165) mAb was purified to homogeneity by precipitation with ammonium sulfate followed by the affinity chromatography on MBP IgM purification column. Moreover, both purified human IgM V_2, V_(75) and mouse ascites were characterized by SDS-PAGE and Western blotting. Proliferation of T24 cell line was considerably inhibited by V_2 and V_(75). Conclusion: Five-feature mice could be used to produce fully human monoclonal antibody. The fully human anti-VEGF mAb is potential in the cancer treatment.
