Objective To investigate the role of cytochrome C in the apoptosis of hepatoma cell induced by flurouracil,and to analyze the distribution of cytochrome C in cytosol and the relationship between the cytochrome C distribution and Caspase-9 activation.Methods The human hepatoma HepG2 cells were treated with flurouracil at 1?10-2mol/L and 2,4,8,16,24h respectively.The chang of cytochrome C in HepG2 apoptosis was detected by using Fluorescent Assay Kit;and proteolytic cleavage of caspase-9 and distribution of cytochrome C in cytosol or in mitochondria was analyzed by Western blot.Results Four hours after cells exposure to flurouracil,the caspase-9 activity in HepG2 cells increased gradually and reached the peak at 16h,compared with the control groups,the difference was significant(P

Objective To determine whether the important apoptosis-related genes of p53 and bcl-2 participate in the regulation of apoptosis of human liver cancer cells by establishing the model of apoptosis of human liver cancer cells induced by mitomycin C (MMC). Methods The selected human liver cancer cell line HepG2 was treated with 8mg/ml MMC. The morphological changes of the cells were observed using fluorescent and electron transmission microscopy and the DNA fragments of the cells analyzed by agarose gel electrophoresis to testify cell apoptosis. The expression of p53 and bcl-2 genes was detected by flow cytometry. Results The morphological observation and biochemical examination showed that MMC induced the HepG2 cells to undergo apoptosis. After administration of MMC, the expression of p53 gene changed obviously while that of bcl-2 gene was not apparent. Conclusion p53 plays an important role in the regulation of apoptosis induced by MMC in HepG2 cells. This regulation may be an early event in apoptotic signal pathway. Meanwhile, the apoptosis of the cells is p53-dependent. However, bcl-2 gene may not participate in the control of this apoptosis.
3.Treatment of Hepatic Cavernous Hemangioma:A Report of 47 Cases

Objective To summrize the effects of various treatment methods,including operation,transcatheter arterial embolization(TAE),and percutaneous ethanol injection(PEI) on hepatic cavernous hemangioma(HCH),in order to select an optimal therapy strategy for HCH.MethodsClinic data of 47 patients with 58 hepatic hemangiomas treated in our hospital in recent 10 years were analyzed retrospectively.Results There were 26 males and 21 females with mean age 42?2 5 years(10~72 years old)in this series.The diameters of these lesions were 3~21 5cm(mean diameter 8 5?1 8cm).Of them,obvious symptoms appeared in 39 patients,malignancy could not excluded in 6,and the size of HCH increased rapidly in 2.Operations were performed in 40 patients,including 2 patients failed to PEI;TAE was conducted in 7cases.There was no mortality or major morbidity occurred in the operative patients,except right pleual effusion in one case and subdiaphragmatic fluid collections in another case(2/40).Neither is mortality nor morbidity seen in the TAE treated case.Conclusions Operation is an effective procedure for HCH.But for more diffuse lesions or lesions located in special region,TAE is a more favorable alterative selection.Because PEI has risk of hemorrhage and hardener injection under ultrasonography guide has uncertain outcome,so it should be chosen cautiously.
4.Analysis of the Application of Cephalosporin in Our Hospital during 2010-2014

OBJECTIVE:To provide reference for rational application of cephalosporins in the clinic. METHODS:The applica-tion of cephalosporins antibiotics in our hospital during 2010-2014 was analyzed dynamically in respects of consumption sum, DDD,DDC,DDDs,etc. RESULTS:During 2010-2014,the proportion of total consumption sum of antibiotics in total consump-tion sum kept below 30%,and decreased year by year,decreasing from 27.35% in 2010 to 14.70% in 2014. Among antibiotics, the consumption sum and DDDs of cephalosporins were the highest;consumption sum decreased year by year,decreasing from 44.50% in 2010 to 30.49% in 2014;DDDs increased year by year,increasing from 11.00×104 in 2012 to 14.22×104 in 2014. The consumption sum and DDDs of second generation cephalosporins were the highest. The consumption sum of cefotiam ranked the first place;DDDs of cefprozi ranked the first place during 2010-2011,while cefotiam ranked the first place during 2012-2014. DDC of cefpirome was the highest during 2010-2011,and that of cefamandole was the highest during 2012-2014,indicating it was expensive. The serial number ratio of cefprozi and cefuroxim was always >1 during those years,which indicated it was cheap and used frequently;that of most drugs were close to 1. CONCLUSIONS:The application of cephalosporins in our hospital tend to be rational,but the propaganda and supervision of rational use of antibiotics should be further strengthened.
5.Study on Contamination Level of Polychlorinated Biphenyls in Retail Livestock Food in a City

Objective To investitgate the level of polychlorinated biphenyls in retail livestock meat.Methods Totally 28 samples of three kinds of meat including of beef,pork and mutton were collected during Aug-Dec.,2008.Twelve dioxin-like polychlorinated biphenyls(DL-PCBs) and six marker PCBs were extracted by Soxhlet extractor,cleaned up by FMS and qualitative and quantitative analyzedsis by isotope dilution high-resolution gas chromatography/high resolution mass spectrometry(HRGC/ HRMS).Results Levels of total 18 compounds including of 12 DL-PCBs and indicator PCBs in beef,pork and mutton were 1 607.60 pg/g fat,435.82 pg/ g fat,and 505.17 pg/g fat respectively,DL-PCBs concentration were 456.19 pg/g fat,46.37 pg/g fat and 77.50 pg/g fat respectively.Maker-PCBs concentration were 1 151.41 pg/g fat,389.45pg/g fat and 427.67 pg/g fat respectively.The levels of PCB toxicity equivalent(TEQ)(the median value)were:0.84 pg/g fat in beef,0.15 pg/g fat in pork,0.28 pg/g fat in mutton respectively.The TEQ concentration of PCB(median value) was lower than the EU action level limit value.In all samples,PCB118 was the greatest contributor to the total contaminated concentration,and PCB126 was the dominant contributor to toxicity equivalent(TEQ).Six of 28 samples exceeded the limit value of the EU action level.Conclusion The results show that PCBs contamination exists in a city from the six samples exceeding the EU standard limit.It should take measures to explore the cause of contamination of PCBs compounds and control the pollution in order to guarantee food safety effectively.

Objective To determine the effect of caspase 9 in the apoptosis of hepatoma cells induced by flurouracil , and to investigate the activity alteration and proteolytic cleavage of caspase 9. Methods The human hepatoma HepG2 cells were treated with flurouracil for 2,4,8,16,24h respectively. The caspase 9 activity was detected using caspase 9 Fluorescent Assay Kit.Proteolytic cleavage of caspase 9 was analyzed by Western blot.The apoptotic rates of HepG2 cells induced by flurouracil with or without caspase 9 inhibitor treatment were measured by flow cytometry. Objective To establish a method to measure Angiotensin Converting Enzyme activity in serum by MECC. Method Mixture of serum and substrate Hip-His-Leu had been incubated for 120 min at 37℃, substrate Hip-His-Leu was digested into two parts, Hip and His-Leu. The reaction was ended by 0. 1 mmol/L HC1 , the production was analyzed by MECC directly to detecte content of Hip in production, and calculated ACE activity. Running buffer was 20 mmol/L pH 9. 0 boric acid-borate buffer (including SDS 50 mmol/L) , capillary column was 75?m i. d.?37 cm, injection was 3s by press, voltage was 16 kV, running time was 7. 5 min, detected by UV detector at 200 nm, tempreture was 20℃. Result The within-run and between-run CV was 2. 7% and 5. 2%. The detection limits of ACE activity was 0. 2 IU/L ( singal/noise = 3). The ACE activity and absorption was linearly related from 2. 4 to 72 IU/L. The mean value of ACE was 5. 2-21. 9 IU/L (x?1. 96 s) in 50 normals. Conclusion It was a one of rapid, precise methods for determination of ACE activity in serum.

Objective: To review the clinical effect of early enteral nutrition in patients with carcinoma of esophagus after operation.Methods: Nutrient canal was indwelt through Fresubin(TP) was given through the nasal-intestinal tube in 120 cases with carcinoma of esophagus after operation.Results: The complications such as anastomotic leakage,lung infection and incisional infection in the patients receiving early enteral nutrition were much less than those in control group.No death occurred.The time for bowel function recovery was much shorter.Conclusion: Early enteral nutrition can promote the recovery of gastrointestinal function,improve the nutrition,decrease the occurrence of complications and raise the successful rate of operation in operative patients with esophagus carcinoma.
10.Prophylactic cranial irradiation for non-small cell lung cancer: a systematic review

ObjectiveTo determine whether prophylactic cranial irradiation (PCI) has a role in the management of patients with non-small cell lung carcinoma (NSCLC)treated with radical intent.MethodsWe searched The Cochrane Library,MEDLINE,EMbase,CBM,CNKI and VIP.The quality of the includedstudieswascriticallyevaluated.Dataanalyseswereperformed usingtheCochrane Collaboration's RevMan 5.1 software.ResultsFour randomized controlled trials involving 905 patients met the inclusion criteria.The results meta-analyses showed the incidence of brain metastases was lower in PCI group compared with the observation group ( x2 =1.98,P =0.000 ) ; but there is no evidence of 1-year overall survival (OS) benefit ( x2 =1.12,P =0.880).Only RTOG 2009 provides prospective data:There were no significant differences in global cognitive function (P =0.600) or ADL ( P =0.880) after PCI,but there was a significant decline in immediate recall (P=0.030) and delayed recall (P =0.008 ) at 1 year,At 1 year,there was no significant differences in QOL after PCI ( P =0.050).Conclusions This systematic review show significantly decreases the risk of BM without improving 1-year OS in NSCLC patient receiving prophylactic cranial irradiation.There is insufficient evidence to support the use of PCI in clinical practice.Where possible,patients should be offered entry into a clinical trial.
