Objective To observe the tissus implanted into the damaged articular cartilage of rabbits with induced autogeneic mesenchymal cells. Methods The autologous mesenchymal cells derived from bone marrow of New Zealand rabbits were harvested. With basic fibroblast growth factor(bFGF, 25 ng/ml) and transforming growth factor beta 1(TGF-? 1, 2 ng/ml), the cells were induced and expanded in cell culture. The induced cells with absorbable gelatin sponge as a carrier were then implanted into the damaged articular cartilage in rabbits as experimental group. The absorbable gelatin sponge without cells were served as control. Specimens were harvested at the end of 4, 12 and 24 week after implantation, and were stained with toluidine blue. Results By RT- PCR, it was confirmed that there was expression of typeⅡ procollagen mRNA in the induced mesenchymal cell. After implantation, it was difficult to macroscopically distinguish the repaired tissues from the normal cartilaginous tissue in the experimental group in 24 weeks. While the defect of articular cartilage was filled with white and swampy tissue in the control group at the same time. Histologically, the defective area of the articular cartilage was replaced by the formation of neo- cartilage which showed positive staining of toluidine blue in 4 weeks in the experimental group. The neo- cartilage was modeled to normal cartilage tissues in 12 weeks and was similar to the surrounding cartilage in 24 weeks. But in the control group, the defect of articular cartilage was not repaired by cartilage tissue at every stage and were replaced by fibrocartilage which was shown weakly positive staining of toluidine blue in 24 weeks. Conclusion The transplant of the induced autogeneic mesenchymal cells derived from bone marrow might promote repair of articular cartilage, and restore its structure and function.

OBJECTIVE:To evaluate the role of PDCA cycle management on standardizing clinical use of atomization inhala-tion drug. METHODS:Using a retrospective method,300 medical records of atomization inhalation drug use were collected from our hospital during Jan.-Dec. in 2013(non-intervention group),200 medical records collected during Apr.-Jun. in 2014(one cycle intervention group),and 180 medical records collected during Oct.-Dec. in 2014(two rounds of cycle intervention group). The use of atomization inhalation drugs was compared before and after the application of PDCA cycle management. Before and after two cy-cle intervention,4 types of medical staff were investigated on cognition of the related knowledge of atomization inhalation drugs, as physicians, pharmacists, nurses and mechanic. RESULTS:After two rounds of PDCA cycle management and intervention, the proportion of unsuitable route of administration of antibiotics, long acting glucocorticoid,non-atomization inhalation dosage form of resolving phlegm drugs, protease decreased from 54.0%,62.0%,59.7%,44.7% before intervention to 0.6%,1.1%,15.0%, 1.1% after intervention;those phenomena had not been found, such as unsuitable route of administration of TCM injection and non-atomization dosage form of theophyllines, unsuitable atomization frequence, unsuitable indication, drug mixture for preventing and controlling symptom, etc. The proportion of medical staffs being familiar with medical device, drug classification, drug selec-tion for prerention and treatment and the proportion of hospital drug stock to 93.4%,86.7%,92.4%,96.2% after intervention from 38.0%,79.0%,49.0%,39.5%before intervention. CONCLUSIONS:The application of PDCA cycle management can effec-tively regulate the use of atomization inhalation drugs in our hospital;the method can be promoted and applied.

Objective:To study the different feeding practices of Chinese infants within four months of age in 2002. Method:The method of multi-steps cluster sampling was adopted. In order to ensure the sample size sufficient additional subjects were included in this survey. Results:More than two thirds (71.6%) of infants within four months of age had been almost exclusively breastfed, 65.5% and 74.4% in urban and rural areas respectively. The average percentage of mixed feeding was 23.0% (27.0% in urban and 21.2% in rural areas); and that of artificial feeding was 5.4% (7.5% in urban and 4.4% in rural areas). The rate of breastfeeding decreased with the increase of family income. Conclusion:Although the rate of almost exclusive breastfeeding is increasing, there is still far away from the aim of 85% in 2001-2010 National Outline for Children.

OBJECTIVE:To prepare m-nisoldipine solid dispersion from poorly-soluble m-nisoldipine so as to improve its solubility and dissolution rate in vitro. METHODS: Solid dispersions of m-nisoldipine were prepared by coprecipitation method with poloxamer as carrier; Differential scanning calorimetry (DSC) was used to determine the status of nimodipine in carrier. The solubility and the dissolution rate of the solid dispersion in vitro were studied. RESULTS: DSC analysis indicated that eutectic mixture was formed from m-nisoldipine and poloxamer. The solubility of m-nisoldipine and the its solid dispersions prepared from m-nisoldipine and poloxamer at different ratio (1∶3, 1∶5, 1∶7) were 0.89, 4.50, 15.35, and 23.03 mg?L-1, respectively, and their 120 min dissolution rates were 26.80%, 38.57%, 41.38%, and 45.92%, respectively. In the same ratio, the dissolution rates of the solid dispersions were higher than those of their physics mixtures. CONCLUSIONS: The solid dispersion of m-nisoldipine prepared with poloxamer as carrier can increase the solubility and dissolution rate in vitro.

Journal of Pharmaceutical Analysis 2010;22(3):175-178
Objective To analyze proliferation and differentiation of glial fibrillary acid protein (GFAP)- and nestin-positive (GFAP+/nestin+) cells isolated from the subventricular zone following fluid percussion brain injury to determine whether GFAP+/nestin+ cells exhibit characteristics of neural stem cells. Methods Male Sprague-Dawley rats, aged 12 weeks and weighing 200-250 g, were randomly and evenly assigned to normal control group and model group. In the model group, a rat model of fluid percussion brain injury was established. Five days later, subventricular zone tissue was resected from each group and made into single cell suspension. After serum-free neural stem cell medium culture and subsequent serum-induced differentiation, cell type, proliferation and differentiation capacities were determined by immunofluorescence staining and flow cytometry. Results At 3-7 days after fluid percussion brain injury, nestin+/GFAP+ cells in the single cell suspension from the model group significantly outnumbered those from the normal control group (P<0.01). In the model group, an increased number of small neurospheres with smooth cell edge and bulged center formed after primary culture, and were clearly visible with the increase of culture time and medium replacement. After several passages, many clonal spheres were obtained, suggesting strong self-proliferatiing capacity. Neurospheres from the model group differentiated into astrocytes, neurons and oligodendrocytes. Conclusion GFAP+/nestin+ cells isolated from the adult rat subventricular zone after fluid percussion brain injury are thought to be neural stem cells because of their self-renewal and multi-differentiation capacities.
