1.Preventive Effect of Ephedrine on the Hypotension Spinal Anesthesia

Objective: To evaluate the preventive effect of ephedrine on hypotension under spinal anesthesia in patients with different age. Methods: Sixty patients scheduled for spinal anesthesia were randomly allocated to four groups with 15 each. Group Ⅰ and Ⅱ were adult patients aged from 25 to 55 years old and group Ⅲ and Ⅳ were geratic aged patients of 6575 years old. Every patient received a fluid loading of 7ml/kg before spinal anesthesia. The ephedrine was administrated to group Ⅱ and IV patients in a dose of 10mg i v and 30 mg i m and same amount of normal saline was administrated to group Ⅰ and Ⅲ as soon as spinal anesthesia was performed. Results:Thirty patients in the placebo groups developed a maximal decrease in MAP exceeding 20%. Ten of these (2 in group Ⅰ and 8 in group Ⅲ ) developed a decrease in MAP exceeding 35% and required treatment. In aged groups, all patients in group Ⅲ had a decrease in MAP exceeding 20 %, 53 % of whom, the decrease exceeded 35 %. Conclusion:Prophylactic use of ephedrine is effective in the prevention of hypotension during spinal anesthesia, especially in aged patients.
2.Effect of Eucommiae ulmoides on the Microstructural and Ultrastructural Structure of Femur and Lumbar Vertebrae in Ovariectomized Rats

OBJECTIVE: To evaluate the effect of Eucommiae ulmoides on the microstructural and ultrastructural change of femur and lumber vertebrae in ovariectomized rats.METHODS: 80 4-month-old female SD rats were randomized to 4 groups: sham operation group,model group,alfacalcidol group,E.ulmoides group.In the sham operation group,the abdominal cavities of the rats were cut open to expose ovaries before being sutured.However,the rats in the other 3 groups were ovariectomized bilaterally.At 10 days after operation the rats were administered intragastrically with corresponding drug.After 3 months,femur and lumber vertebrae were dissected and fixed in 4% glutaral for 3 days,then decalcified in 4% EDTA for 4 weeks,and then the structure of femur and lumber vertebrae was observed under light microscope and transmission electron microscope(TEM).RESULTS: Observed under light microscope,the femur and lumber vertebrae were full of fat in the ovariectomized rats and in which the density of bone trabecula decreased significantly;however,in alfacalcidol-treated rats and E.ulmoides-treated rats,the amount of fat decreased markedly while the density and thickness of bone trabecula increased.Observed under TEM,in the ovariectomized rats,rough endoplasmic reticulum(RER),cytolysosome and mitochondrial were plentiful inside osteoclasts in femur and lumber vertebrae,partial osteoblasts atrophied,the amount of RER and other cellular organ decreased significantly and apoptosis appeared for partial cells;however,in alfacalcidol-treated group and E.ulmoicdes-treated group,the above changes improved significantly.CONCLUSION:E.ulmoides has remarkable beneficial effect on osteoporosis in ovariectomized rats.The mechanism might be attributed to its inhibition on the synthesis of fat and the activity of osteoclasts,promotion on the activity of osteoblasts and inhibition on the apoptosis.

Persistent hyperinsulinemic hypoglycemia in infants is one of the most common causes of persistent hypoglycemia in infants. The knowledge of molecular defects leading to persistent hyperinsulinemic hypoglycemia in infants has been rapidly growing in recent years. According to the responsible genes, this ailment can be divided into five types. However, no molecular defect can be yet found in as many as 50% of the patients.
4.A strategy for rapidly constructing the single and dual-site targeting plasmid-based RNAi vector

Objective To develop a rapid method for construction of RNAi vector.Methods After inversely inserting the U6 and H1 polymerase Ⅲ promoters into the pBluescriptⅡ backbone vector,only with two short reverse complementary oligo nucleotides,the RNAi vector with one interference cassette could be constructed.Using two isoaudamers MunⅠ and EcoRⅠ with sticky ends,several cassettes could be fused together to form the ultimate multiple-site targeting RNAi vector.Using this method,we constructed RNAi vectors targeting green fluorescent protein(GFP) and firefly luciferase(LUC) gene separately or both for the test of knock down property of these vectors.Results Constructed single and two site targeting RNAi vectors got desirable knockdown effects,in which single site targeting vector could get about 90 percent knock down efficiency whereas two site targeting vector reached about 87.5 percent knock down efficiency either.Conclusion Our RNAi vector construction strategy is efficient and time-saving so that it can be used for knocking down several genes simultaneously in the same cell.
5.The inhibitory effect of chemotherapeutic drugs on the proliferation of Bel-740 2 cells

Objective To evaluate the in vitro effect of che motherapeutant on hepatic carcinoma. Methods The inhibitory effect of 5-Fu, DDP and 4-PA on Bel-7402 cel ls proliferation was studied by MTT colorimetry. Resu lts (1) Bel-7402 was on an active proliferation by the treat ment of 5-Fu and DDP for half an hour, and the proliferation inhibited after 12 hours; but the proliferation was inhibited by 4-PA after half an hour of tr eatment. (2) The inhibitory rates increase with time and were over 50% after 48 ~72 hours of treatment. (3) There was temporarily active proliferation of Bel- 7402 cells in the early period of chemotherapy by 5-Fu and DDP. Conclusion Continuous chemotherapy is effectiv e for the inhibition of Bel-7402 cells proliferation.
6.Long-term follow-up results of Perthes disease in older children treated with surgical containment methods

Objective To evaluate the long-term results of Chiari osteotomy in the treatment of Legg-Calv?-Perthes disease for the patients of 8 years old or above and discuss the factors that influence the prognosis of this disease. Methods There were 116 hips of 103 patients of Perthes disease treated with Chiari osteotomy between June 1986 and December 2002. All patients were 8 years old or above. 72 hips of these cases acquired follow-up, the meaning follow-up duration was 97 months(range, 38-196 months). According to lateral pillar classification, 30 hips were classified as group B, 22 hips as B/C group, 20 hips as C group. There were 50 hips in male, 22 hips in female, 47 patients between 8 and 10 years old, 22 cases between 10 and 14 years old. X-ray showed 25 hips with subluxation of femoral head preoperatively. Results According to the revised Stulberg classification system: (1)31 hips had Stulberg classⅠ,Ⅱ outcome, in which B group accounted for 19 of 30 hips, the hips in group C had the worst results: of 20 hips, only 3 had a Stulberg class Ⅰ or Ⅱ outcome, the results for the hips in lateral B/C border group were intermediate between those groups B and C. 33 hips had Stulberg class Ⅲ, in which B group had 10 hips, B/C group had 11 hips, C group had 12 hips. 8 hips had Stulberg class Ⅳ or Ⅴ, B group had 1 hip, B/C group had 2 hips, C group had 5 hips. (2)All cases had a satisfied containment, no one had subluxation after Chiari osteotomy. (3)48% of male patients had a Stulberg class Ⅰor Ⅱ outcome, in contrast, only 23% of female had the comparable outcome. (4)55% of the group of 8 to 10 years old patients had a Stulberg class Ⅰ or Ⅱ outcome, in comparison with 17% from the group of 10 to 14 years old cases. Conclusion The patients older than 8 years at the onset of disease and in the lateral pillar B or C group should be treated with surgical containment, Chiari osteotomy can fulfill such task. The gender, the age of onset, especially the lateral pillar classification strongly correlate with the outcome of Perthes disease, and the latter can help us to evaluate the severity degree of the disease and the prognosis.
7.The change of MTH2 expression with aging in hippocampi of senescence-accelerated mouse P8

Objective To observe the expression of oxidized purine nueleoside triphosphatase MTH2 in the hippocampi of senescence-accelerated mouse P8(SAMP8). Methods Randomly selected SAMP8 and its control strain SAMRl aged 1-,4-,8-and 12-month and performed immunohistochemisry and Western blot analysis to explore the expression changes of MTH2 in the 2 groups of mice. Results The MTH2 protein was detected in cytoplasm and nuclei of neurons throughout the whole hippocampus of SAMRl at all ages.There were no obvious changes of SAMR1 in both CA1 and CA3 subregions,but an age-related decline of MTH2 protein in both regions of SAMP8 was found.The optieal density(OD)value of Cal hippoeampus(0.0473),CA3 hippoeampus (0.0510)were significantly less in 8-month old mice than in the 4-month old[CA1:0.0619,CA3:0.06803(all P<0.05).By immunoblot analysis,the grey values of MTH2 expression were 0.529,0.313,0.032 in hippoeampus of 4-12 months-old SAMP8 mice,and significantly Iower than in the age-matched SAMR1 mice(all P<0.05). Conclusions MTH2 deficiency may be a new factor involved in the accelerated aging and in deficits of learning and memory ability in SAMP8.
8.The Study of WT1 Gene Expression in the Urine from Patients with Renal Diseases by Semi-nested PCR

Objective To investigate the clinical significance of WT1 gene expression in the urine. Methods The expression of WT1 gene in the urine of patients with chronic glomerular nephritis(CGN), rheumatic diseases,diabetes mellitus(DM) and healthy subjects was detected by semi-nested PCR. Results The positive rates of WT1 gene expression in CGN with proteinuria,DM with proteinuria, DM without proteinuria and rheumatic diseases were 46.7% (14/30),44.4% (16/36),5.95% (5/84) and 16%(4/25), respectively. WT1 expression in the urine was not related to the degree of proteinuria, hematuria, renal function or clinical course of diseases. WT1 expression in the urine of the healthy subjects or cystitis patients was negative. Conclusion The measurement of WT1 mRNA in the urine could play an important role in early diagnosis of progressive renal damage.
9.Therapeutic Effect of Jianpi Tiaozhong Xiaopi Recipe for Functional Dyspepsia with Spleen Deficiency and Qi Stagnation

Objective To observe the effect of Jianpi Tiaozhong Xiaopi Recipe ( JTXP) for the treatment of functional dyspepsia ( FD) with spleen deficiency and Qi stagnation. Methods A total of qualified 110 cases were evenly randomized into Chinese medicine group and western medicine group. Chinese medicine group was given modified JTXP orally, and western medicine group was given Mosapride and Lansoprazole orally. After treatment for one continuous month, therapeutic effect was evaluated and the improvement of clinical symptoms before and after treatment was observed. After follow-up for half a year, the recurrence of FD was observed. Results ( 1) After treatment for one month, the scores of abdominal distension and fullness, gastric vague pain, poor ap petite, belching and hiccup were obviously decreased in both groups compared with those before treatment ( P<0.05) , and the relief of the symptoms in Chinese medicine group was superior to that in western medicine group (P<0.05). (2) The total effective rate was 94.5% in Chinese medicine group and was 81.8% in western medicine group, the difference being significant ( P<0.05). ( 3) The results of follow up showed that recurrence of FD was shown in 3 cases (5.8%) of Chinese medicine group and in 8 cases (17.8%) of western medicine group, the difference being significant ( P<0.05). ( 4) During the treatment, no obvious adverse reaction was shown in both groups. Conclusion JTXP exerts certain therapeutic effect for the treatment of FD with spleen deficiency and Qi stagnation.
