Sphingosine-1-phosphate (S1 P) is a kind of bioactive messenger which is catalyzed by sphingosine kinases (SphK) and mediates various biological behaviors through binding to sphingosine-1-phosphate receptors(S1PR).Researches demonstrate that the aberration of SphK,S1P and S1PR is closely related to the occurrence and development of cancers,such as leukemia,breast cancer,colorectal cancer and lymphoma.The interaction between S1PR1 and signal transducer and activator of transcription 3 (STAT3) is shown to take part in the pathogenesis of malignant tumors as well.Therefore,they are considered to have the potential tumor-treatment value.

ObjectiveTo investigate the effect of montelukast on the secretion of mucus protein in lipopolysaccharide (LPS) induced human bronchial epithelial cells.MethodsPrimary human bronchial epithelial cells were isolated and identiifed in vitro. LPS (1μg/mL) was used to induce cell inlfammatory response. Montelukast (50 μmol/L, 20μmol/L, 10μmol/L) was used as intervention. The concertration of mucin-5 subtype AC (MUC5AC) in cell supernatants was measured by ELISA. The expression levels of MUC5AC mRNA and protein were determined by RT-PCR and Western-blot. DCFH-DA lfuorescent probe was used to detect reactive oxygen species (ROS). To further elucidate the mechanism, NF-κB (p65)、IκBα、ERK1/2 phosphorylation be-fore and after montelukast intervention were determined by Western-blot.ResultsMontelukast decreases the expression levels of MUC5AC mRNA and protein in a dose-dependent manner in LPS induced human bronchial epithelial cells. Meanwhile, mon-teluskast suppresses ROS generation and NF-κB (p65)、IκBα、ERK1/2 phosphorylation.Conclusions In response to LPS in-duced inlfammation, montelukast decreases the expression level of MUC5AC in vitro, which may be related to NF-κB and ERK activation.

[Summary] Extracellular calcium is essential for the regulation of a variety of biological processes. Calcium-sensing receptor (CaSR) plays a central role in maintaining Ca2+ homeostasis, while G-protein α-11 (Gα11 ) subunit and adaptor-related protein complex 2 sigma (AP2σ) are also involved in CaSR signaling transduction. Loss- or gain-of-function mutations of these encoding genes cause different types of familial hypocalciuric hypercalcaemia (FHH) and autosomal dominant hypocalcaemia (ADH). Calcimimetic and calcilytic drugs are useful in treating these FHH and ADH disorders. The incidence and mortality of colorectal cancer (CRC) increased rapidly in recent decades and become enormous challenges in China.Lack of effective early warning of molecular markers and dynamic monitoring technology in term of early diagnosis,treatment evaluation,dynamic recurrence and metastasis monitoring are the clinical diagnosis and treatment bottlenecks of CRC.Traditional diagnosis and treatment of CRC rely on a single level of patient information with low accuracy.Based on the system of biology medical model,to carry out a joint diagnostic model,will overcome the traditional problems through a number of multi-level information integration of the joint diagnosis model,will significantly improve the sensitivity and specificity of diagnosis of CRC.The major challenge in patients with advanced and metastatic CRC is the instability of the tumor genome and the treatment-induced resistance during chemotherapy and targeted therapy.It is necessary to carry out continuous dynamic biopsy in order to accurately guide the development of treatment decisions.Compared with the pathological examination of traditional surgical specimen,liquid biopsy,such as circulating tumor cells,circulating tumor DNA detection technology,with noninvasive,real-time dynamic monitoring,could evaluate the efficacy of treatment,and guide the precise individual diagnosis and treatment.Today,the new strategy and new technology need to undergo clinical trials urgency,through technology optimization,reduction of costs and improvement of detection accuracy,would quickly extended to clinical applications in future.

Objective:The study is to investigate whether macrophage migration inhibitory factor(MIF)-173 single nucleotide polymorphism(SNP) correlates with inflammatory bowel disease(IBD) in Chinese Han population in Zhejiang Province.Methods:MIF-173 SNP alleles were typed using tetra- primer amplification refractory mutation system(ARMS) and restriction fragment length polymorphisms(RFLP)-PCR in 142 healthy subjects and 98 patients with inflammatory bowel disease(IBD),respectively.At the same time,the PCR products were cloned and sequenced.Results:We detected three kinds of genotypes at the MIF-173 locus,no deviation from Hardy-Weinberg equilibrium were observed.The result was the same from tetra-primer ARMS and RFLP-PCR.Statistical analysis showed,the frequency of MIF-173 CC genotype was significantly higher in patients with ulcerative colitis(UC) 15.5% than in healthy individuals 5.6%(P

The paper proposes and realizes a programmable wireless neural stimulation system which can be used as a solution of functional electrical stimulation to treat neural diseases. The system is composed of two parts: controller and neural stimulator. The controller can transmit pulse parameters to the stimulator wirelessly, and the stimulator can generate bidirectional pulses with charge balance. The simulator takes use of ADCs to sample on the bidirectional pulse output, which compared with preset amplitude to the DAC output voltage to realize the voltage calibration. Through the test, the whole system works stably and the output of the biphasic charge balanced circuit is definite. The stimulator output ranges from 0 to 5 V ajustably, and the frequency ranges from 1 Hz to 200 Hz ajustably, while the pulse width ranges from 500 μs to 1500 μs ajustably. The duration of the stimulation can be set from 10 s to 10 min.

Objective To investigate clinical effect of recombinant human growth hormone (rhGH)for inter-venting central precocious puberty in children,to provide reference for clinical treatment.Methods 80 patients with growth deceleration slows CCP were selected,when height growth rate below 4cm/year,40 patients on the basis of joint GnRHa used rhGH treatment were selected as the observation group,and the remaining 40 patients refused to use rhGH,continued to use of GnRHa treatment were selected as the control group.After treatment predicted height,height,bone age,growth rate and other indicators were compared.Results In observation group,the actual height Ht, predicted height PAH,the growth rate Gv,serum insulin -like growth factor -1 (IGF -1 )after treatment were (1 44.48 ±6.59)cm,(1 54.94 ±4.52)cm,(4.43 ±0.64)cm/6months,(1 32.25 ±8.84)ng/mL,compared to before treatment,the differences were statistically significant (t =6.548,P <0.01 ;t =5.734,P <0.01 ;t =28.869,P <0.01 ;t =20.65,P <0.01 ),compared with the control group,the differences were significant (t =3.943,P <0.01 ;t =4.759,P <0.01 ;t =28.247,P <0.01 ;t =20.882,P <0.01 ),there were no differences in other indices;hor-mones FSH,LH and other indicators of the two groups before and after treatment,and blood sugar,thyroid indicators showed no abnormality.Conclusion Recombinant human growth hormone in the treatment of central precocious puberty deceleration in children does not increase bone age,which will help improve the growth rate and predicted height,which worthy of clinical application.

Objective To evaluate tension-free repair in the management of recurrent inguinal hernia. Methods From 1/1993 to 12/2002, 163 patients with recurrent inguinal hernia underwent reoperation, clinical data were reviewed. Results The male: female ratio was 138: 25, age from 34 to 76 years. The primary surgery had been traditional hemiorrhaphy without prothesis in 142 cases (87. 1 % , including 65 cases of Bassini ,35 cases of McWay and 42 cases of Shouldice) , a tension-free procedure in 12 cases(7. 4% , including 3 cases of laparoscopic mesh repair) and unknown techniques in 9 cases(5. 5% ). The average interval from the initial surgery to recurrence was 36?14 months (range from 3 months to 10 years). Recurrent hernias were treated with a Shouldice repair in 71 cases(43. 6% ) , a tension-free repair in 92 cases(56. 4% , including a laparoscopic mesh repair in 3 cases). All these 163 cases(100% ) were followed up for 56 ?1 months (range from 30 months to 12 years) after the second surgery. It was found that after reoperation the recurrence rate of Shouldice procedure and tension-free repair was 16. 1% (11/71) and 2. 2% (2/92) respectively (x2 = 8. 327 ,P

Objective To evaluate the distribution of pathogens causing bone and joint purulent infections and the bacterial resistance to antibiotics,and to provide reference of clinical antibiotic therapy.Methods A total of 514 patients who had bone and joint purulent infections in Department of Orthopedics of Shangrao People’s Hospital from Jan 2009 to Jun 2013 were retrospectively analyzed. Results 296 strains of Gram-negative bacteria were isolated,the infection rate was 47.2%,among which 296 strains of Escherichia coli were the most common infection pathogen,the infection rate was 15.6%,followed by Acinetobacter baumannii,Pseudomonas aeruginosa,and the infection rates were 11.8%,11.3%.331 strains of Gram-positive bacteria were isolated,the infection rate was 52.8%.Staphylococcus epidermidis,Staphylococcus aureus,were the common infection pathogens,following the infection rates were 21.5%,13.7%.Gram-negative bacteria had the highest sensitivity to imipenem,while Gram-positive bacteria had sensitivity to vancomycin.Conclusion Gram-positive bacteria is the main pathogens with bone and joint purulent infections.Selection of antibiotics according to the drug sensitive test has important clinical significance.
