Chinese Journal of Tissue Engineering Research 2013;(37):6664-6670
BACKGROUND:The glial scar is a major obstacle to the regeneration of the central nervous system, which plays a physical and chemical barrier role to central nervous system regeneration. How to effectively inhibit glial scar formation and promote axonal regeneration has great significance.
OBJECTIVE:To explore the effect of the immune response in the formation of the glial scar after spinal cord injury.
METHODS:A computer-based online search was performed in the PubMed database and the CNKI database for the articles on the mechanisms of spinal cord injury, immune cel s and spinal cord injury as wel as the glial scar formation after spinal cord injury from January 1990 to January 2013. The key words were “spinal cord injury, immunity, glia scar”in Chinese and English. The languages of the articles were limited in Chinese and English. Final y, 72 articles were included for the further analysis according the inclusion criteria.
RESULTS AND CONCLUSION: After spinal cord injury, the effect of autoimmune response in which stil been much debated. Studies showed that spinal cord injury can cause suppression of cel ular immunity and non-specific immune function, causing immune function decreasing. Monitoring immune indicators and improving immune function have great significance in promoting the ful rehabilitation of patients with spinal cord injury. Glial scar is the most important factor to prevent the repairing after spinal cord injury. Immune and inflammatory response is the main reason to secondary spinal cord injury. Degree of repair after spinal cord injury mainly depends on the protection of the residual neurons in order to keep them off secondary damage. However, autoimmune T cel s can protect the residual neurons, the specific effect of T lymphocytes in glial scar formation after spinal cord injury is stil unclear, and further exploration is needed.

BACKGROUND: The oriented migration of bone marrow mesenchymal stem cells may depend on the interaction between local chemotactic factors and cellsurface receptors. However, which chemotactic factors may mediate the oriented migration of bone marrow mesenchymal stem cells towards the fracture site remains unclear. OBJECTIVE: To tag autologous bone marrow mesenchymal stem cells, evaluate its role in bone healing, and detect the highly expressed factors associated with migration of bone marrow mesenchymal stem cells in the microenvironment. METHODS: The fluorescence/chimeric C57BL/6 mouse models were established, then left shankbone fracture models were also produced. The percentages of green fluorescent protein positive cells to al cells at the fracture site and the percentage of osteoblasts differentiated from bone marrow mesenchymal stem cells to al the osteoblasts were detected at different time points. The role of bone marrow mesenchymal stem cells in the fracture repairing was evaluated. The levels of chemotactic factors protein expression at the fracture site in different time points were detected with immunohistochemistry technology. RESULTS AND CONCLUSION: The percentage of green fluorescent protein positive cells to al cells at the fracture site was (3.011±0.911)%, (9.031±0.145)%, (12.064±0.145)% at 1, 5, 14 days postoperatively; and osteoblasts differentiated from bone marrow mesenchymal stem cells accounted for 50% of al the osteoblasts. After fracture, the stromal cel derived factor-1, colony stimulating factor, hepatocyte growth factor, monocyte chemoattractant protein-1, and matrix metal oproteinases-9 were expressed to varying degrees in the microenvironment, while the expression of granulocyte colony-stimulating factor was negative. The expression of stromal cel derived factor-1 in the fracture microenvironment was the highest, mainly due to the migration of bone marrow mesenchymal stem cells. Experimental findings indicate that, autologous bone marrow mesenchymal stem cells participate in and play an important role in bone healing. The stromal cel derived factor-1 plays an important role in promoting bone marrow mesenchymal stem cells migration and promoting bone healing.

Objective To study the mechanism of the protective effect of lycium barbarum polysaccharide on light aging resistance rats by using the metabolic profile and metabolic target analysis technique. Methods Ultraviolet irradiation induced Wistar rats were induced to produce skin photo aging model and 24 rats were randomly divided into three groups, including control goup, model group, Lycium barbarum polysaccharide group (LBP). After modeling for 24 hours, LBP group was conducted with Lycium barbarum polysaccharide solution of 10 mg/kg. Control group and model group were given same volume of stroke-physiological saline solution for 14 days. The biochemical indexes such as rat serum antioxidant activity of related substances and MDA were measured in model group and drug group; the urine metabolomics study was also investigated for the mechanism of lycium barbarum polysaccharide against light aging band. Results Compared with the model group, the LBP group rats total superoxide dismutase activity (301.51 ± 42.56 U/mgvs.93.41 ± 56.31 U/mg), hydroxyproline (8.91 ± 5.78μg/mgvs.4.74 ± 1.54μg/mg) content significantly increased (P<0.05), but the malondialdehyde (8.54 ± 6.41 nmol/mgvs.21.31 ± 6.58 nmol/mg) decreased without any statistics difference (P<0.05). The urine metabonomics results showed that LBP could regulate the skin photoaging of multiple metabolic pathways and key metabolic enzymes in the process, such as peanut four acid, tyrosine, taurine, citric acid and hippuric acid, L-cysteine, inositol, threonine etc. Conclusions In the process of skin photoaging in rats, multiple metabolic pathways in vivo were disordered, and Lycium barbarum polysaccharide could play a protective role by regulating the key metabolic enzymes in the network.

Objective To describe the diagnostic approach and clinical outcomes of fetal intraabdominal extralobar pulmonary sequestration (IEPS).Methods Three cases of IEPS were diagnosed prenatally by ultrasound in our center. The data of these 3 cases and 43 cases in prior studies from 1986 to 2016 were retrospectively reviewed. The prenatal sonographic features, treatment, and outcomes were collected. Results The gestational age of diagnosis IEPS was (24.5±5.9)weeks,the male-to-female ratio was 3-4∶1,78.3% (36/46) of IEPS masses located on the left side,47.8% (22/46) of lesions were homogenous and hyperechoic,8.7% (4/46) of cases associated anomalies,78.3% (36/46) of patients were preformed resection and postoperative recovery and subsequent course were uncomplicated.Feeding arteries were detected in 15.2% (7/46) cases and confirmed by surgery or autopsy. Conclusions Prenatal ultrasound has the diagnostic accuracy of IEPS.Feeding arteries noted on color Doppler are strongly indicators of IEPS.Postsurgical outcomes are favorable.
