1.Exploration of Optimal Scheme and Theoretic Mechanism for Treatment of Chronic Ulcerative Colitis

Objective To explore optimal proposal and theorical mechanism for treatment of chronic ulcerative colitis(UC) with the therapeutic instrument inducting combined energies into anus.Methods 76 patients with mid-range chronic UC were sampled and divided into two groups randomly.Patients in both groups were all withdrawn from drugs and treated with the therapeutic instrument inducting the combined energies into the anus by choosing different combined energies.There were 41cases in the treated group and they were treated with intracavity electrod synchronizing with electrotherapy,heat therapy,magnetic therapy and massor.35 cases in the control group were treated with intracavity electrod synchronizing with heat therapy and magnet therapy.The two groups were compared after twelve months.Results The general improvement rate and cure rate of the two groups showed great significant difference(P
2.Expression and the related factors of E-selectin in insulin-resistant rat endothelium

Objective To discuss the expression and the related factors of E-selectin in insulin-resistant(IR)rat endothelium. Methods Wistar rats were randomly divided into 3 groups: high fat feed,high fructose feed and the control feed(HF group,HG group and NC group). IR rat model was established by high fat feeding or high fructose feeding for 4 weeks and was evaluated with euglycemic clamp technique. E-selectin protein expression was detected by Western-blot analysis. Results The expression of E-selectin protein in vessel endothelium was increased in HF group and HG group compared with that in NC group (P<0.01), and higher in HF group than in HG group (P<0.05). E-selectin protein expression was correlated significantly with TG, FFA, FIns and HOMA-IR in HF group and HG group (P<0.05). Conclusions IR may cause the increased expression of E-selectin in endothelium of rats fed with high fat or fructose.
3.Infrared Spectral Analysis of Yinhuang Medicinal Broth and Its Membrane Fraction

Objective To discuss difference of Yinhuang medicinal broth and its membrane fraction from macroscopic quality,provide reasonable experimental basis for membrane system.Methods Method of infrared spectral analysis was used to compare absorption peak position,peak intensity,correlation coefficient of spectrogram of Yinhuang medicinal broth and membrane fraction,judge separation efficiency by overall analysis the spectrogram.Results There was a lots of same peak position at Yinhuang medicinal broth and final membrane fraction.Their coefficient correlation was 0.832 7.Conclusion After impurity removal and concentration by two kind of membrane,Yinhuang medicinal broth lose a little ingredient, so the selection of membrane system is reasonable.
4.Determination of cucurbitacin B in Pedicellus Melo by capillary zone electrophoresis

AIM:To develop a new method to determine cucurbitacin B in Pedicellus Melo by capillary zone(electrophoresis(CZE).) METHODS:All the samples were analysed on a fused silica capillary(75 cm?75 ?m I.D.) by using 50 mmol/L sodium borate solution(containing 5% acetonitrile) as the background electrolyte with the running voltage at 12.0 kV and detection wavelength of 265 nm.And clorprenaline hydrochloride was applied as the internal standard. RESULTS:The results showed a good linear correlation between relative peak area of cucurbitacin B(y) and its concentration(x) over the range of 0.15~1.2 mg/mL.Regression equation was y=(0.003 4x)+0.058,r=0.999 7,and the average recovery for baicalin was 100.2% with the RSD at(2.11%). CONCLUSION:The method is rapid,simple,reproducible and produces low pollution.It serves as a novel way to determine cucurbitacin B.
6.Influence of Severe Chyle Blood Serum to the Three Types of Common Biochemical Detection Method

Objective Aim to explore how severe chyle blood serum effects on the End-point method of colorimetry,velocity method,immunological transmission turbidimetry(TIA)three types of common biochemical tests and their different influ-ences.Methods Collected 20 normal appearance serums,divided each pooled serum into A,B,C,D,E,F and G 7 experimen-tal groups and one control group.Each group contented for 1ml serum.Added intralipid 10,20,40,80,160μl and 320μl to the experimental groups from A to G in turn to prepare into different concentrations of simulate chylous samples.Tested each group by the End-point method of colorimetry,velocity method and TIA which represented by Glucose (Glu),uric acid (UA),Total bilirubin (TBil),aspartate aminotransferase (AST),gamma-glutamyl transpeptidase (GGT),lactate dehydro-genase (LDH),hypersensitive C reactive protein (hs-CRP),Apolipoprotein A1 (apoA1 )andβ2-microglobulin (β2-MG). Used pair T test for both experimental group and their control group.Results In terms of TIA,there were statistical differ-ences between all the experimental groups and their control groups in the three projects (t=-2.842~29.465,P<0.05).In terms of the End-point method of colorimetry,there were statistical differences between all the experimental groups and their control groups (t=-34.942~13.884,P<0.05)except for A group in Glu experiment (t=1.875,P=0.068>0.05).In terms of velocity method,there were no statistical differences between all the experimental groups and their control groups (t=-1.532~1.619,P>0.05)except for the B and C groups in GGT experiment (t=2.234&5.006,P<0.05).Conclusion Severe chyle blood serum had significant influence on the End-point method of colorimetry and TIA,as well as less influ-ence on velocity method.
7.Effect of contact system in monocyte adhesion induced by urokinase-type plasminogen activator

AIM:To study the effect of contact system in monocyte adhesion induced by urokinase-type plasminogen activator (U-PA). METHODS:U937 cells were labeled with [3H] thymidine in the culture medium, test reagents were added into the cell suspension in 24-well plates. The counts?min -1 were counted in a liquid scintillation counter. RESULTS:U-PA-induced monocyte adhesion was inhibited by high-molecular-weight kininogen and kallikrein, and promoted by factor Ⅻ and plasminogen. CONCLUSION:these contact system proteins may be important modulators of U-PA-induced monocyte adhesion, a process which is involved in many pathophysiological events.
8.Mutagenicity of Enphorbia lunulata decoction in vitro

Aim To assay the mutagenicity of Enphorbia lunulata(EL) decoction and to modify the Ames test for evaluation the mutagenicity of herbal medicine samples.Method The mutagenicity of EL decoction was assayed by standard Ames test; the teratogenicity was done by mammalian bone marrow chromosomal aberration test. In modified Ames test system,the influence of histidine EL decoction was excluded by additional negative control, in which the test media was supplied with histidine (histidine amount equaled to the histidine in different concentration of EL decoction).Result The mutagenicity of EL decoction was positive in the standard Ames test. The teratogenicity of EL decoction was negative in mammalian bone marrow chromosomal aberration test. By the modified Ames tests,the mutagenicity of EL decoction was negative.Conclusion The standard Ames test is not suitable for evaluating the mutagenicity of EL decoction, but the modified Ames test is. The mutagenicity in vitro and the teratogenicity in vivo of EL decoction are all negative.
9.The role of apparent diffusion coefficient in the differentiation between benign and malignant bone tumors

Objective To explore the role of apparent diffusion coefficient (ADC) value of diffusion-weighted imaging (DWI) in the differentiation between benign and malignant bone tumors. Methods Echo planar imaging DWI was performed in 18 patients with benign tumor or tumorous lesion and 26 patients with malignant tumor of bone. Three b-values (0, 500 and 1000 s/mm2) were applied. The lowest, highest, and whole ADC values were measured for each lesion, respectively. Results The lowest ADC values of benign bone tumor[ mean( 1.28±0. 49) ×10-3mm2/s ] were significantly higher than that of malignant tumor [ mean ( 0. 92±0. 35 )×10-3mm2/s, t = 2. 839, P < 0. 01 ]. The whole ADC values of benign bone tumor [ mean (1.62±0. 51 ) ×10-3mm2/s] were significantly higher than that of malignant tumor [ mean ( 1.21±0. 36) ×10-3mm2/s, t = 3. 092, P < 0. 01 ]. However, there were much overlapping between benign and malignant bone tumor in the lowest and whole ADC values. There was no significant difference for the highest ADC values between benign [. Mean ( 2. 02±0. 55 )×10-3mm2/s] and malignant bone tumor[ mean( 1.71±0. 65 ) ×10-3mm2/s, t = 1. 669, P > 0. 05 ]. Excluding cases of bone cyst and aneurismal bone cyst, the lowest, highest, and whole ADC values of benign bone tumor was (1.11± 0. 31 )×10-3mm2/s, ( 1.88±0. 49)×10-3mm2/s, and( 1.45±0. 35 )×10-3mm2/s, respectively. There was no significant difference for the lowest, highest, or whole ADC values between benign and malignant bone tumor (t = 1. 728, 0. 964, and 2. 012, respectively, P > 0. 05). Conclusion ADC value is useless for the differentiation between benign and malignant bone tumors.
10.The diagnostic value of gluocose-6-phosphate isomerase in patient with rheumatoid arthritis

Objective To investigate the diagnostic value of gluocose-6-phosphate isomerase (G6PI) detected by an enzy-me linked immunosorbent assay (ELISA) in patients with rheumatoid arthritis (RA). Methods The G6PI was detected by ELISA in serum samples from 106 patients with RA, 53 non-RA controls with various rheumatic diseases, and healthy individuals. The level of rheumatoid factor (RF), anti-CCP antibodies and AKA were also assessed in RA patients. The correlation analysis beween G6PI and anti-CCP, IgM-RF. G6PI, anti-CCP, IgM-RF and AKA were carried out between patients with erosion and with non-erosion diseases . Results ① G6PI serum level of patients with RA was (1.61 ±1.20) μg/ml, and was (0.11 ±0.17) in patients with other rheumatic diseases, and (0.06±0.07) μg/ml in healthy individuals. There was statistical significant difference between RA patients and patients with other rheumatic diseases (P<0.05). Receiver operator curve analysis (ROC) showed an opitium cut off level for C6PI at 0.225 μl/ml. The sensitivity of G6PI was 0.868, the specificity was 0.853 in RA. C6PI was associated with RF, but was not associated with anti -CCP. C6PI ws not associated with disease activity index by Spearman' s correlation analysis. The association between above parameters with bone erosion was not detected, however. Conclusion C6PI is abnormally increased in some RA so it may be a new diagnostic marker for RA. G6PI has a reasonable sensitivity (86.8%) and with high specificity(85.3%) to RA and it is valuable for RA diagnosis. C6PI is associated with RF, but not completely overlaps. C6PI is not associated with diseases activity. No association is found between G6PI and bone erosions.
