1.Recent advances in the study of Nrf2 and inflammatory respiratory diseases.

Nuclear factor-erythroid 2 related factor 2 (Nrf2) is an ubiquitous and important transcription factor. It regulates antioxidant response elements (AREs)-mediated expression of antioxidant enzyme and cytoprotective proteins. A large body of research showed that Nrf2-Keap1 (Kelch-like ECH-associated protein 1, Keap 1)-ARE signaling pathway is involved in the endogenous antioxidant defense mechanisms. Nrf2 increases the expression of a number of cytoprotective genes, protects cells and tissues from the injury of a variety of toxicants and carcinogens. As a result, Nrf2 enhances the expression of glutathione and antioxidants such as superoxide dismutase and glutathione S-transferase, and subsequently scavenging free radicals. Air pollution especially from PM2.5 particles, is associated with an increasing morbidity of inflammatory pulmonary diseases and their deterioration. More and more studies demonstrated that Nrf2 was a novel signaling molecule in the modulation of inflammatory responses in these inflammatory respiratory diseases, such as asthma, acute lung injury (ALI) and COPD. Therefore, Nrf2 targeting might be a therapeutic target, which will provide clinical benefit by reducing both oxidative stress and inflammation in asthma, acute lung injury (ALI) and COPD. This review focused on the relationship between Nrf2 and inflammatory respiratory diseases and oxidative stress.
2.Application of damage control resuscitation in treatment of severe multiple injuries combined with traumatic hemorrhagic shock

Objective To investigate the clinical value of damage control resuscitation ( DCR) in the treatment of severe multiple injuries combined with traumatic hemorrhagic shock. Methods A retrospective analysis was done on 27 patients with severe multiple injuries combined with traumatic hemorrhagic shock treated by DCR. Another 32 patients treated with traditional aggressive/normotensive fluid resuscitation were used as control. Lactic acid clearance time, coagulation, diffuse intravascular coagulation (DIC) morbidity, and mortality were observed and compared between two groups. Results Compared with traditional aggressive/normotensive fluid resuscitation, DCR had better curative effect, shorter clearance time of lactic acid, more rapid recovery of blood coagulation function and lower incidence of DIC morbidity and lower mortality for patients with severe multiple injuries combined with traumatic hemorrhagic shock. Conclusions In the treatment of severe multiple injuries combined with traumatic hemorrhagic shock, the use of DCR can remarkably improve the survival rate of patients and also provide a new way for resuscitation and rescue of other types of shock patients.
3.Exploration of the relationship between serf-efficacy sense and psychological health of nursing staff

Objective To explore the relationship between general self-efficacy sense and psycho-logical health of nursing staff. Methods 230 healthy nursing staff were investigated with self-rating de-pression scale, serf-rating anxiety scale and general serf-efficacy scale. Correlation analysis and t test were adopted to analyze the data. Results There was a negative correlation between the scores of general self-efficacy scale and the scores of self-rating anxiety and depression scale. Moreover, the scores of anxi-ety and depression of nursing staff with higher self-efficacy were higher than those with lower self-effica-cy. Conclusions Self-efficacy sense has positive effects on the level of psychological health of nursing staff. The higher self-efficacy sense will help maintain higher psychological health state.
4.Related factors of suicide ideation in military personnel

Objective:To explore the prevalence rate of suicidal ideation and its risk and protective factors in military personnel.Methods:A total of 2071 military personnel were investigated by stratified random cluster sampling in May to August of 2015.The Question 7 of Beck Depression Scale (BDI) was used to test suicide ideation (item 7 score ≥ 1 was dovetailed in suicide ideation group).Family,social demographic and occupational factors were assessed with the self-made general characteristics questionnaire.The data were analyzed with multiple stepwise logistic regression.Results:There were 382 (18.4%) military personnel whose scores of item 7 were ≥1.Family history of suicide (OR =42.97,95% CI =29.14-63.35),surrounding suicide (OR =2.46,95% CI =1.75-3.46),drinking (OR =1.64,95% CI =1.31-2.07) and level of education (OR =1.38,95% CI =1.01-1.90) were risk factors of suicidal ideation.Protective factors were (OR =0.60,95% CI =0.50-0.72) and station (OR =0.15,95% CI =0.03-0.64).Conclusion:It suggests that in suicidal prevention for military,more attention is deserved paying to those military personnel who have family history of suicide,surrounding suicide,drinking habit,and are stationed in remote areas and air force.
5.Studies on the radiation resistance of esophageal cancer stem cells and the expressions of relative proteins

Objective To study the radiation sensitive enhancement ratio (SER) of NS398 on esophageal cancer stem cells and adherent tumor cells and analyze the radioresistance related protein expressions.Methods ECA109 esophageal cancer stem cells were cultured in serum-free medium.Expression levels of cell surface maker CD44 + and CD271 + were analyzed by flow cytometry.MTT assay was used to detect cell proliferation after the treatments with NS398 and irradiation(0,4 and 8 Gy).The sensitization effects of NS398 on the parental cells and its spheroid were evaluated by clone formation assay.Western blot assay was performed to determine protein expressions.Results Serum-free medium was successfully applied to isolate the cancer stem cells with spherical properties.CD271 + in the spheroid cells was notable higher than that in the parent cells (t =3.81,P < 0.05).After irradiation,the proliferation rate of parental cells was higher than that in spheroid cells.After the combination treatment of NS398 and irradiation,SF2 value of parental cells was lower than spheroid cells(t =2.91,P < 0.05)and the SER of NS398 on parental cells was greater than spheroid cells.The expressions of Bmi-1,c-Myc,β-catenin and Cyclin D1 in spheroid cells were higher than those in parental cells (t =8.09,7.90,7.50,7.15,P<0.05).Cyclin D1 expression levels under both cell situations increased after 4 Gy irradiation (t =9.74,6.67,P <0.05).Compared to the 4 Gy irradiation alone group,the β-catenin and Cyclin D1 expression levels in both parental cells (t =10.15,12.12,P < 0.05) and spheroid cells (t =3.23,7.45,P < 0.05) decreased in the combination group.Conclusions Esophageal cancer stem cells with high level of CD271 can be isolated with serum-free medium and it is radioresistant where β-catenin and its downstream proteins may be involved.
6.PI3K/Akt/NF-κB regulate ABCB1/P-glycoprotein-mediated multidrug resistance in colon carcinoma cells

Background and purpose: Multidrug resistance (MDR) is the dominating obstacle to the chemotherapy. There is strong evidence that the phosphoinositide 3-kinases (PI3Ks) signaling pathway is involved in MDR phenotype, however, the mechanism of MDR occurrence is still unknown. This study tended to investigate the regulating effect of PI3K/Akt signaling pathway and its downstream target genes in P-glycoprotein (P-gp) (ABCB1 gene encoding)-mediated MDR in human colon carcinoma HCT-116/L-OHP cells. Methods:Pretreatment with PI3K selective inhibitor LY294002 (20μmol/L) for 2 h, the sensitivity of L-OHP was evaluated by the CCK-8 (cell counting kit-8) assay in HCT-116/L-OHP cells, and the expressions of P-gp, LRP, MRP-2, Akt, p-Akt, IκB and p-IκB were evaluated by Western blot. The activity of ABCB1 promoter was evaluated by chromatin immunoprecipitation analysis (CHIP). Results: After inhibiting the activity of PI3K/Akt signaling pathway, the IC50 value of L-OHP decreased from(157.48±16.73)μg/mL to (53.68±3.18)μg/mL in HCT-116/L-OHP cells, and the reversal index was 2.93 (P<0.01). The expressions of P-gp, p-Akt and p-IκB were down-regulation compared with the concrol group (P<0.01), but the expressions of LRP, MRP-2, Akt and IκB didn't change signiifcantly. CHIP result has conifrmed that NF-κB protein could bind to the region of ABCB1 gene promoter in HCT116/L-OHP cells. Conclusion:Blocking of PI3K/Akt/NF-kB signal pathway could increase the drug sensitivity to MDR cells, inhibit the phosphorylation of p-Akt and p-IκB, and reversing ABCB1/P-glycoprotein-mediated multidrug resistance in colon carcinoma cells.
7.Effect of WS070117M1 on chronic obstructive pulmonary disease in mice and the underling mechanisms of anti-inflammation.

The aim of this study is to investigate the anti-inflammatory effect of the adenosine derivative N6-(3-hydroxylaniline) adenosine (WS070117M1) on cigarette smoke plus LPS (lipopolysaccharide)-induced chronic obstructive pulmonary disease (COPD) in mice and its mechanism. COPD model was established by exposing male BALB/c mice to cigarette smoke and challenged with LPS inhalation. Supernatants of bronchoalveolar lavage fluid (BALF) were harvested and IL-1β, IL-6, IL-8 and TGF-β1 levels were measured by ELISA (enzyme-linked immunesorbent assay). The number of total white blood cells and neutrophils in bronchoalveolar lavage fluid was counted separately. Lung tissue was stained with Mayer 's hematoxylin and eosin for histopathologic examination. pAMPKa protein expression and distribution of lung tissue were analyzed by immunohistochemistry method. In vitro, levels of AMPKα phosphorylation in phorbol-12- myristate-13-acetate (PMA) differentiated THP-1 cells was detected by immunohistochemistry, IL-8 level in supernatants of cigarette smoke condensate stimulating PMA differentiated THP-1 cells was measured by ELISA. The results showed that WS070117M1 treatment significantly activated AMPKa in the lung tissue. It also resulted in down regulation of IL-1β, IL-6, IL-8 and TGF-β1 levels in bronchoalveolar lavage fluid and IL-8 level in cigarette smoke condensate stimulating PMA differentiated THP-1 cells. In addition, WS070117M1 could inhibit the recruitment of total white blood cells and neutrophils. These results suggest that WS070117M1 may alleviate the airway inflammation by activating AMPK in the lung tissue.
8.Correlationship between visuosaptial ability and air traffic management ability

Objective To explore the relationship between visuospatial abilities of the students ma-jor in air traffic management and their performances,and then provide theories of their vocational classified. Methods 104 healthy male college students were tested by the cognitive laterality battery (CLB),which included visuospatial tests and verbal-sequential tests.At the same time,their performances assessed by air traffic management stimulators.Results Subjects with higher scores achieved by operating on air traffic management stimulators have higher visuospatial abilities,which was confirmed by the visual spatial scores (A) ((0.183±0.094) vs (0.060±0.338), t=41.57, P=0.000) and cognitive laterality quotient (CLQ) ((0.087±0.108) vs (-0.096±0.101), t=43.14, P=0.000)).However,compared with others in lower scores group,their scores on the phonological tests (P) reduced ((0.096±0.081) vs (0.157±0.092), t=6.81, P=0.012) ) .Moreover,there were the significant positive correlation between their performance on air traffic management stimulators and A ( r=0.749, P<0.01) ,CLQ ( r=0.663, P<0.01) ,cognitive performance quotient (CPQ) ( r=0.394, P<0.01).Conclusion For air traffic management occupation,the higher visual spatial abilities were the base of having the higher performance.Thus the visuospatial abilities may be as a good index of selecting and vocational classified for air traffic management students.
9.Effect of sleep deprivation on learning and memory network in healthy adults by fMRI

Objective To observe the neural mechanism of learning and memory network disturbance induced by 36 hours sleep deprivation(SD).Methods 18 young male healthy volunteers were enrolled in this study.Resting state functional magnetic imaging(rs-fMRI) data was collected by blood oxygen level dependence functional magnetic imaging(BOLD-fMRI) during rest wakefulness(RW) and 36 hours SD.Bilateral hippocampus were selected as seed points by REST software automated anatomical labeling template.And then compared the differences of functional connectivity(FC) between bilateral hippocampus and whole brain regions by using paired-t test within statistical parametric mapping 8 (SPM8) software.Finally,correlation analysis had been done between significant FC of brain regions and WMS MQ values (SD-RW).Results Compared with RW,the bilateral hippocampus showed positive connected with the right dorsal posterior cingulate cortex,and showed negative functional connectivity with bilateral postcentral gyrus.The WMS MQ was significantly decreased after 36 hours SD.The connection between bilateral hippocampus and the right posterior cingulate cortex showed negative correlation with the deductionofWMS MQ (left:r=-0.463,P=0.015;right:r=-0.546,P=0.003).Conclusion SD has a significant effect on bilateral hippocampal FC.The FC between bilateral hippocampus and right dorsal posterior cingulate cortexis significant increased,while between bilateral postcentral gyrus is decreased after 36 hours SD.The former may be an important neurological mechanism in learning and memory ability regulation after 36 hours sleep deprivation.
10.Purification and characterization of deoxyribonuclease from earthworm Eisenia foetida

Objective:To purify a kind of deoxyribonuclease from earthworm Eisenia foetida (named earthworm DNase, EDNase) and study its characteristics. Methods: Acetone precipitation, ion-exchange chromatography, high performance liquid chromatography, SDS-PAGE, CapiUary electrophoresis isoelectric focusing and MALDI-TOP MS were used for the study. Results: This purified protocol improved 137-fold purification and 45.6 % recovery of enzyme activity. The molecular mass of EDNase was estimated to be 63 000. Mg2+ , Mn2+ and Ca2+ were strong inhibitors of EDNase, while Na+ slightly increasd the enzyme activity. The enzyme was completely stable in the pH range from 4. 4 to 5.2 and had a pH optimum of 4.8. The optimum temperature was 37℃ and the enzyme was stable up to 40 ℃. The pI of the enzyme was 6. 20. Km and Vmax for the enzyme were 1.52 g/L and 4. 89 mg/(mL ·min), respectively, with calf thymus DNA as substrate. The enzyme was able to degrade chromosemal DNA, linear λbacteriophage DNA as well as supereoiled plasmid DNA, but didn' t display any RNase activity. Conclusion: This kind of deoxyribonuclease possesses unique characteristics, which is different from the deoxyribonucleases which we have known before.
