1.Expression of human platelet factor 4 mRNA and its relation with angiogenesis and lymph node metastasis in papillary thyroid carcinoma

Objective To study the expression of human platelet factor 4 (hPF4)mRNA and its relationship with angiogenesis and lymph node metastasis in papillary throid carcinoma(PTC). Methods RT-PCR was used to detect the expression of hPF4 mRNA in 40 cases of PTC and 10 cases of normal tissue surrounding carcinoma. Immunohistochemical stain was performed to detect microvessel density(MVD) whose endothelia were marked by antibody CD105. Results The positive-expression rate of hPF4 mRNA was 80 % respectively in normal tissue surrounding carcinoma. The positive-expression rate of hPF4 was 47.5 % in PTC cases. Objective To investigate the significance of the expression of interleukin-10(IL-10),interleukin-13(IL-13)and interleukin-15(IL-15)in serums of patients with hepatitis B.Methods The expression of IL-10,IL-13 and IL-15 in serums of 109 patients was measured by ELISA.Results The serum levels of IL-10,IL-13 in patients with moderate degree chronic hepatitis B(CHB)group were significantly higher than that in patients with chronic severe hepatitis group,severe degree CHB group,acute hepatitis group and normal group.The serum level of IL-15 was increased in patients with acute hepatitis、moderate degree CHB group,severe degree CHB group and chronic severe hepatitis compared with normotensives.The proportion of IL-15/IL-10 and IL-15/IL-13 in patients with chronic severe hepatitis group,severe degree CHB group and acute hepatitis group were higher than that in patients with moderate degree CHB group and normotensives.The serum level of IL-15 and the proportion of IL-15/IL-10 and IL-15/IL-13 in dead group were significantly higher than that in improving group with chronic severe hepatitis.Conclusion There is an abnormal cell-mediated immune response in patients with hepatitis B.Combining detection on the levels of serum IL-10,IL-13 and IL-15,and the proportion of IL-15/IL-10 and IL-15/IL-13 would nicely show the cellular active status and be helpful to the prediction of prognosis and direction of therapy in patients with hepatitis B of different kinds of clinical type.
3.Screening of probiotic properties of twelve strains of lactobacilli by in vitro techniques

Objective: To study the probiotic properties of 12 selected strains of lactobacilli.Methods: The probiotic properties of 12 selected strains of lactobacilli were investigated for their resistance to pH 2.5 and 0.3% oxgall,their adhesion to Caco-2 cells and antimicrobial activity against enteric pathogenic bacteria.Results: The 12 strains all showed relatively high resistance to acid and bile salt and varied antimicrobial activities against pathogenic bacteria of intestinal infection.Lactobacillus johnsonii JCM1022 exhibited a broad inhibitory effect on enteric pathogenic bacteria,especially on enteropathgenic E.coli and Salmonella enteritidis,while Lactobacillus reuteri JCM1081 possessed a high activity of adhesion to Caco-2 cells(495.07?80.03).Conclusion: Among the 12 strains,Lactobacillus johnsonii JCM1022 and reuteri JCM1081 are qualified candidates for clinical application.
4.Clinical application of pneumatic lithotripsy under ureteroscope

Objective To explore the value of pneumatic lithotripsy under ureteroscope in the management of ureteral calculus . Methods The clinical data of 360 cases of ureteral stones treated ureteroscopically by pneumatic lithotripsy were retrospectively analyzed. Results Stones were successfully fragmented on one session in 85% of the cases (306/360). Complications were observed in 2 5% of the cases (9/360), including 6 cases of ureteral perforation and 3 cases of urinary infection. Conclusions Pneumatic lithotripsy under ureteroscope is safe, effective and minimally invasive.

Objective To study the expression of thrombospondin(TSP) mRNA and its relationship with angiogenesis and lymph node metastasis in papillary thyroid carcinoma(PTC).Methods RT-PCT was utilized to detect the expression of TSP1 and TSP2 mRNA in 40 cases of PTC and 10 cases of normal tissue surrounding carcinoma.Immunohistochemical stain was performed to detect microvessel density(MVD) whose endothelia were marked by anti-CD105 McAb.Results The positive-expression rate of TSP1 mRNA,TSP2 mRNA was 80.0%,90.0% respectively in normal tissue surrounding carcinoma.The positive-expression rate of TSPI mRNA was 45.0%.The positive-expression rate of TSP2 mRNA was 47.5%.The MVD value of PTC cases with TSP1 and TSP2 mRNA positive expression was significantly lower than that of cases without TSP1 and TSP2 mRNA expression(P

Objective To analyze the clinical effects of internal radiation of iodine-125 seed implantation in the treatment of advanced pancreatic carcinoma.Methods 22 patients with unresectable and advanced pancreatic carcinoma were treated by the internal and interstitial radiation using iodine-125 seed implantation.Results Survival of 20 months was observed in 5 patients,10 ~ 20 months in 9 patients,2 ~10 months in 8,patients with an average of (13.47 ± 8.12) months.Among the 22 patients,complete response was obtained in 5 patients,partial response in 8 patients,no response in 7 paticnts,and the other showing PD.The response rate( CR + PR) was 59%.Conclusions Intra - operative internal radiation by iodine - 125 seed implantation does show some therapeutic effects for advanced pancreatic carcinoma.
7.Detection of Two Types of Aminoglycoside-modifying Enzymes Genes in Chryseobacterium spp

OBJECTIVE To study 6 kinds of aminoglycoside-modifying enzymes (AMEs) genes in Chryseobacterium spp isolates. METHODS The isolates were identified by API20NE Gram-negative identification cards,and the susceptibility of antimicrobial agents was detected by MIC kits ( bioM?rieux ) 6 AMEs genes of 2 strains of Chryseobacterium spp were measured by PCR,and verified by DNA sequencing and sequence analysis. RESULTS In the 2 strains,2 kinds of resistant genes [aac(6′)-Ⅱ and ant(2″)-Ⅰ] were positive,and 4 kinds genes of AMEs [aac (6′)-Ⅰb,aac(3)-Ⅱ,ant(3″)-Ⅰ and aac(3)-Ⅰ] were negative.The amplicons were purified,sequenced and analyzed with BLAST 2.0 and found to be identical to aac(6′)-Ⅱ and ant(2″)-Ⅰ. CONCLUSIONS There are AMEs in Chryseobacterium spp isolates. This is the first report on AMEs genes [coexistance of aac(6′)-Ⅱ and ant(2″)-Ⅰ] in Chryseobacterium spp.
8.Forty Types of Resistant-related Genes in a Pan-resistant Pseudomonas aeruginosa

OBJECTIVE To study 40 kinds of resistant-related genes in a pan-resistant Pseudomonas aeruginosa.METHODS To detect the susceptibility of antimicrobial agents by MIC,40 resistant-related genes including 29 ?-lactamases genes,porin oprD2 genes,6 aminoglycoside-modifying enzymes(AMEs)genes,chlorhexidine/sulfadiazine resistant gene(qacE△1-sul1)and intergron(intⅠ1,2,3),etc,form 1 strain of P.aeruginosa were measured by PCR,and verified by DNA sequencing.RESULTS In the strain,there were positive of 6 kinds of resistant-related genes(blaTEM,blaOXA10,aac(6′)-Ⅱ,aac(3)-Ⅱ,qacE△1-sul1 and intⅠ1),but without oprD2 genes.Twenty-seven kinds of ?-lactamases genes,4 kinds of AMEs(aac(6′)-Ⅰb,aac(3)-Ⅰ,ant(3″)-Ⅰ and ant(2″)-Ⅰ),and 2 kinds of intⅠ(intⅠ2 and intⅠ3) were negative.CONCLUSIONS The multi-resistant mechanisms of pan-resistant P.aeruginosa are mainly related to 7 kinds of resistant-related genes(blaTEM,blaOXA10,oprD2,aac(6′)-Ⅱ,aac(3)-Ⅱ,qacE△1-sul1 and intⅠ1).
9.Correlation between CD24 expression and its relation with Ki67,Bcl-2 in breast carcinoma

Objective To investigate the expression of CD24 and Ki67 in the invasive breast ductal car-cinoma tissues, so as to assess the role of CD24 protein in the carcinogenesis and progression of breast canc-er. Methods The expression of CD24 and Ki67, Bcl-2 was examined by immunohistochemically in 86 breast cancer specimens and 30 non-cancer specimens ( ElivisionTM Plus). The relationship between CD24 expression and Ki67, Bcl-2, patient age, tumor size, histopathologic grade, axillary lymphnode metastasis, and clinical stage of breast cancer was analyzed. Results In 86 eases of invasive breast ductal carcinoma, the positive incidences of CD24 was 87.2% (75/86), which distinguished apparently from those of the con-trol group( P <0.01 ). In breast carcinoma tissues, the expression of CD24 was positively related to axillary lymphnode metastasis(P <0.01 ), but was not related to patients age,tumor size, histopathologic grade and clinical stage(P>0.05). CD24 expression was positively related to Ki67 (P<0.01 ) and negatively relat-ed to Bcl-2(P>0.05). Conclusion CD24 expression was related to the proliferation, invasion and me-tastasis of breast cancer cells and it can serve as an important marker for predicting biological behavior and prognosis of tumors.
10.Expression of CD24 in esophageal squamous cell carcinoma and its relationship with prognosis

Objective To study the significance of CD24 protein expression in esophageal squamous cell carcinoma and its relationship with clinical pathological parameters and clinical prognosis.Methods The expression of CD24 protein was studied in 90 esophageal squamous cell carcinoma samples by EnVision immunohistochemical analysis and to study relationship of CD24 with clinical pathological parameters and prognosis.Results The expression of CD24 in esophageal squamous cell carcinoma was significantly higher than that in adjacent normal tissue (x 2 =0.832,P < 0.05).The expression of CD24 was not related with the age of patien,sexuality and the histologic grade (x2 =0.741,0.361,0.930,8.858,all P > 0.05),but with clinical stage of the cance (x2 =10.804,P =0.005),lymph node metastasis (x2 =12.349,P =0.000) and depth of tumor invasion (x2 =13.94,P =0.007).Significant difference in survival between the different CD24 expression group,the OR (95 % CI) values were 2.105 (61.022-69.273),2.313 (55.108-64.176),1.286 (32.388-37.429),the survival of patients with strong expression of CD24 was inferior to that of weak expression of CD24,depth of tumor invasion and lymph node metastasis were independent prognostic factors of esophageal squamous cell carcinoma.Conclusion CD24 may be related to the malignant process of esophageal squamous cell carcinoma.Overexpression of CD24 indicates poor prognosis in esophageal squamous cell carcinoma patients.CD24 may become a new independent prognostic factor for esophageal squamous cell carcinoma.
