1.The Inhibitory Effect of OGT Gene Expression on The Level of tau Phosphorylation

To investigate the effect of OGT expression inhibited by RNAi on the alteration of tau phosphorylation and glycosylation level in HEK293T cells.The siRNAs targeting OGTgene(OGT-siRNA1~3) were designed and chemically synthesized.The OGT-siRNA1~3 were transfected into HEK293T cells via lipofectamine2000.The efficacy of RNA interference was detected by RT-PCR.The fluorescence of GFP/OGT was counted by fluorescence microscopy after pEGFP/OGT and OGT-siRNA1~3 cotransfected to HEK293T cells for 24 h.The level of tau phosphorylation and glycosylation were detected by Western blot after Plasmid pCI/tau441 and the siRNA cotransfected HEK293T cells for 48 h.OGT-siRNA3(100 nmol/L) could effectively downregulate the expression of OGT gene compared with Mock group(80.0% at mRNA level and 51.3% at protein level).The level of phosphorylation at various sites of the tau was significantly upregulated and the glycosylation was downregulated following the inhibition of OGT gene expression.There is an apparent negative correlation between the modification of phosphorylation and glycosylation of tau protein, the deficient in glucose uptaken/metabolism may be an important pathogenesis of AD.
2.Correlation between motor skills and focused with shifting attention in preschool children

Objective:
To investigate correlations between motor skills with focused and shifting attention among preschool children, and to provide basis for the overall development of preschool children aged 4-6 years.
Methods:
During March to June 2020, a total of 165 preschool children aged 4-6 years were selected and investigated with subscales of the Bruininks Oseretsky Test of Motor Proficiency 2nd Edition (BOT-2) for agility and limb bilateral coordination assessment, and subscales of the Movement Assessment Battery for Children test 2nd Edition (MABC-2) for throwing and catching skill assessment, as well as balance subscale for the assessment of limb movement coordination, hand eye coordination, agility, and balance. The Kansas Reflection Impulsivity Scale for Preschoolers for Pre schoolers (KRISP) and Dimensional Change Card Sort (DCCS) were used to assess focused and shifting attention levels. Correlation and linear regression analyses were performed for statistical analysis.
Results:
Except for one handed throwing skills and shifting attention, there were statistically significant gender differences between boys and girls in total score of body movement coordination, total score of hand to eye coordination, hand to hand connection, total score of agility of movement, total score of movement balance, static support, walking on tiptoe, foot to foot jump and focused attention( t =-6.86, -2.00 ,-3.15,-3.75,-3.00,-2.95,-2.18,-2.11,-3.21, P <0.05), and girls were better than boys; children s total score of body movement coordination, total score of hand to eye coordination, hand to hand connection, agility of movement, walking on tiptoe and focused attention improves with age( r =0.47,0.41,0.47,0.51,0.16,0.31, P <0.05); After excluding the interference of gender and age, total score of body movement coordination was significantly correlated with preschool children s focused attention( β=0.31,95%CI =0.08-0.39, P <0.01).
Conclusion
Limb movement coordination and focused attention are significantly and positively correlated among preschool children aged 4-6 years. Therefore, preschool education and family activities should focus on designing and developing limb movement coordination related games and courses to enhance the focused attention of and form favorable attention quality in preschool children aged 4-6 years.
3.Effect of systematic intervention on improving of nurses’ capability of identifying events in electrocardiographic monitoring

Objective To study the effect of systemic intervention on strengthen nurses’ capability of identifying events in ECG monitoring, so as to make sure the safety of patients.Methods Set up ECG monitoring quality management group, formulated alarming standard values of alarm, set sequence, unit’s default value etc.And all the nurses received the training of different level.Meanwhile, the capability of early-warning in nurses of different seniorities were observed and assessed, and data was statistically analyzed.Results The capability of early-warning in nurses of different seniorities were all improved, alarming settings time of nurses with 1-4 nursing ages was shortened from 105 s to 42 s, that of nurses with 5-9 nursing ages was shortened from 87 s to 31 s, that of nurses with more than 10 nursing ages was shortened from 68 s to 25 s.Alarm response time of nurses with 1-4 nursing ages was shortened from 92 s to 57 s, that of nurses with 5-9 nursing ages was shortened from 54 s to 42 s, and that of nurses with more than 10 nursing ages was shortened from 30 s to 25 s. Conclusions The systematic intervention can effectively improve alarm setting correctly and nurse’ s capability of early-warning, and ensure quality of critical patients’ care.
4.Analysis of the impact of bone quality on the accuracy of computer-aided design of implant guide plate

Objective To explore the influence of different bone quality around the implant on the implantation accuracy,the com-puter-aided design of implant guide plate was used for patients with poor bone quality to evaluate its accuracy.Methods Retrospective analysis of relevant clinical data from 29 single-tooth implant patients with relatively poor bone quality meeting inclusion criteria was conducted.Using 3Shape Implant Studio software,implant plans were designed and fully guided digital templates were fabricated before surgery.Implantation was guided by the templates throughout,with torque and ISQ values recorded to evaluate the initial stability of the implant.Postoperative implant positions were reconstructed using CBCT images,and the three-dimensional deviation of implant place-ment was evaluated in conjunction with the preoperative design.Bone quality around the implants was also recorded and analyzed before and after surgery.Results In this study,digital guide plates were used in patients with poor bone quality,and the initial stability of implants was≥20 N·cm except for three cases.In cases of poor bone quality,the accuracy of the fully guided digital templates was as follows:cervix deviation(0.94±0.59)mm,apex deviation(1.40±0.81)mm and angle deviation 4.10°±2.99°.Bone quality had a greater impact on angle deviation(P<0.05).The higher the proportion of D3 bone was around the implant body 1/3,the smaller the deviation of neck,apex and angle was,while the higher the proportion of D5 bone,the greater the deviation of implantation.Conclu-sion For patients with poor bone quality,under the premise of restoration-oriented,the proportion of D3 bone around the implant can be increased by computer-aided design,and the initial stability of the implant can be improved by guiding the implantation with digital guide plate.Attention should be paid to the influence of bone on the angle deviation of implant during the application of guide plate.
5.Characteristics of high cardiovascular risk in 360 000 adults in Northwest China

【Objective】 To assess the prevalence and treatment of high cardiovascular disease (CVD) risk, and identify individual characteristics related to high CVD risk. 【Methods】 Based on the data of the China Patient-Centered Evaluative Assessment of Cardiac Events (PEACE) Million Persons Project (MPP) from 2016 to 2019, this study enrolled local residents aged 35 to 75 years from 39 counties or districts in Northwest China. Rates of high CVD risk and individual characteristics were assessed in the overall study population. Statin and aspirin use was also evaluated among those at high risk for CVD. Multivariable mixed models were fitted to evaluate the relationship between individual characteristics and high CVD risk. 【Results】 Among 364 537 participants, the average age was (54.6±9.7)years, and 5.8% was at a high risk for CVD. Multivariate mixed models showed that individuals who were currently using alcohol, overweight or obese tended to have a high risk for CVD, while married persons, those with a higher education level or a higher household income were correlated with a lower risk for CVD (all P<0.05). Among high-risk persons, hypertension was the most prevalent risk factor (98.1%), and only 1.3% and 3.5% reported their use of statins and aspirin, respectively. 【Conclusion】 Of the 364 537 participants, about 1 in 17 had a high risk for CVD. Among those at a high CVD risk, only less than 4% reported taking statins or aspirin. These findings indicate that there is still much room for risk mitigation in this population in China.
