Objective To clone survivin gene, prepare its monoclonal antibody(McAb) and check its expression in liver carcinoma cells. Methods Survivin gene cDNA was amplified by RT-PCR from human breast carcinoma cell line MCF-7 and constructed into prokaryocytic expression vector.Fusion-protein of human Survivin was expressed and used for immunizing BALB/C mice. The spleen cells from immunized mice were fused with SP2/0 cells and selectively cultured with HAT medium.ELISA and Western blot were used to screen and identify the McAbs.Immunocytochemical staining was applied for survivin expression in liver carcinoma cells. Results The full survivin gene was cloned. The hybridoma cell that secret specific monoclonal antibody against human surviving was identified. The immunoglobulin subclasses of the McAbs were IgG1.Western blot showed that the McAbs against survivin can specifically react with MS-Survivin fusion protein. The positive reaction was found in hepatocarcinoma cell line HepG2 and hepatocarcinoma tissue by immunocytochemical staining. Conclusion The McAbs against the human Survivin were successfully prepared by a MS2-Survivin fusion protein expressed by E.coli. and the McAb had positive reaction with HepG2 and hepatocarcinoma tissue. It may be a useful tool to study the functions of survivin and check clinical cancer samples.

Objective To study the action and mechanism of staphylococcal exfoliative toxin A(E-TA)on pemphigus foliaceus antigen(PFA)and pemphigus vulgaris antigen(PVA)expressed on cultured human keratinocytes. Methods Stratified human keratinocytes were incubated with ETA and then stained with sera from patients with pemphigus foliaceus or pemphigus vulgaris as the first antibodies and FITC-la-beled sheep anti-human IgG as the second antibody.Total protein was harvested from the cells pretreated with ETA and run on SDS-PAGE for Western blot with the same antibodies.Simultaneously, supernatants of the keratinocytes before and after ETA treatment were collected for detection of the levels of IL-1?,IL-6with ELISA kits. The caseinolytic activities of the supernatants were tested by spectrometry in which casein was used as a non-specific substrate. Results Down-expression of PFA was shown after ETA treatment while no change of PVA expression was found. The high intensity and continuous linear appearance of fluo-rescent staining before ETA treatment became weak and discontinuous after ETA treatment, which were re-covered gradually in24hours.The degradation of proteins recognized by PF sera after ETV treatment was revealed by Western blot. The decreasing tendency of IL-1?concentration was found in the supernatants of cell culture after ETA treatment, but IL-6level was too low to be detected.Increased caseinolytic activities were found in the supernatants, and declined36hours after ETA treatment. Conclusions ETA acts on PFA expressed on keratinocytes in vitro, which is reversible along with withdrawal of ETA.The mechanism of E-TA act on PFA may be related to proteolytic action instead of promoting cytokine secretion.

Objective:
To explore health self-management, adaptation, and perceived stress among nursing interns in medical colleges and universities under the normal prevention and control of COVID-19, and to provide a scientific basis for the health management of practice nursing students.
Methods:
Medical colleges in the Inner Mongolia region were selected to study 263 nursing student interns by cluster random sampling. The survey was conducted using the Health Self-Management Ability Assessment Scale, the Chinese College Students Adjustment Scale and the Perceived Stress Scale.
Results:
The total score of health self-management ability of nursing students during internship was (138.31±20.13). The total score of health self-management ability of nursing students with different educational backgrounds and different residence places was statistically significant ( F/t =3.51, 2.46, P < 0.05 ); the total adaptive score was (3.17±0.72); during the epidemic, the total score of perceived stress was (11.52±2.79), and the difference in the total score of perceived stress among nursing students with different educational backgrounds was statistically significant ( t=4.05, P <0.05). Pearson analysis showed that there was a significant negative correlation between health self-management ability, adaptation, and perceived stress among nursing students during the epidemic period ( r =-0.31, -0.35, P <0.01).
Conclusion
There was a close relationship between perceived stress of nursing students and health self-management and adaptation; the educators in medical colleges should actively explore strategies to improve the health self-management ability and adaptation level of nursing students to enhance the psychological adjustment ability of nursing students to maintain good physical and mental health.

OBJECTIVETo explore the clinical significance of eukaryotic initiation factor 4 E (eIF4E) and mammalian target of rapamycin (mTOR) expressions in esophageal squamous carcinoma tissues.
METHODSClinicopathological data and paraffin samples of resected tumor tissue from 148 patients with esophageal squamous carcinoma undergoing resection in our department between January 2010 and December 2012 were collected retrospectively. Expressions of eIF4E and mTOR were detected in above carcinoma tissues, counterpart para-carcinoma tissues (1 cm distance to carcinoma) and normal tissues (5 cm distance to carcinoma) with Western blot and immunohistochemistry. Their relevance with clinicopathological features was analyzed.
RESULTSExpression of mTOR located mainly in cytoplasm and elF4E mainly in cellular membrane, presenting as yellow grains. These two markers showed strong expression in carcinoma tissues and weak or none in para-carcinoma tissues. In esophageal squamous carcinoma tissues, counterpart para-carcinoma tissues and normal tissues, mTOR protein expression was 85.8% (127/148), 35.1% (52/148) and 3.4% (5/148), eIF4E protein expression was 93.9% (139/148), 35.1% (52/148) and 12.8% (19/148), with a downtrend respectively (all P<0.05). Expressions of mTOR and eIF4E were associated with tumor invasion depth and lymphatic metastasis (all P<0.05), while mTOR expression was associated with differentiation degree (P=0.003), but eIF4E expression was not. Both expressions were not associated with gender, age, and tumor size (all P>0.05).
CONCLUSIONSExpressions of eIF4E and mTOR are up-regulated in esophageal squamous carcinoma tissues, which may be associated with tumor malignance and lymphatic metastasis of esophageal squamous carcinoma. Combined detection of two markers may be helpful to predict the tumor malignance and the prognosis of patients.

Journal of Biomedical Engineering 2003;20(1):101-103
This is an in vitro study designed to assess the effect of temperature variation on the heart function of the five-strip eyebrow squirrel--amammal of the hibernating kind. A heart model of exsomatizing action and for determining the physiological parameters with four physiological recorders was used to measure the LVSP, LVEDP and (+/- dP/dtmax), respectively. It was found that the curves of the squirrel's heart mechanics property in relation to temperature variation are similar to the curves of the non-hibernating animals'. But, the squirrels' heart contraction function changes with in a much wider range, suggesting that their hearts are more tolerant of temperature variation.
