1.EFFECT OF DL-HOMOCYSTEINE ON EXPRESSION OF VCAM-1 AND ADHESION BETWEEN ENDOTHELIAL CELLS AND MONOCYTES IN VITRO

Objective: To investigate the effect of DL-homocysteine (Hcy) on expression of VCAM-1 and the adhesion between endothelial cells and monocytes in cultured rat aortic endothelial cells. Method: Rat aortic endothelial cells were cultured in vitro and treated by 0-5.0 mmol/L Hcy for 24 h. The activity of SOD and GSH-Px, the contents of MDA in the medium and the rates of adhesion between endothelial cells and monocytes were determined. The expressions of NF-?b,VCAM-1 protein and the VCAM-1 mRNA were detected by immunocytochemical and reverse transcriptase PCR respectively. Results: Compared with control group and 0.01 mmol/L Hcy , 0.1-5.0 mmol/L Hcy significantly inhibited the activity of SOD and GSH-Px, increased the contents of MDA and the expression of NF-?B and VCAM-1mRNA, enhanced the rates of adhesion between endothelial cells and monocytes. Conclusion: Homocysteine may play a crucial role in atherosclerosis by increasing the expression of NF-?B,VCAM-1mRNA and enhancing the rates of adhesion between endothelial cells and monocytes.
2.THE PROTECTIVE EFFECTS OF FOLIC ACID,VB_6,VB_(12) TO RAT AORTIC ENDOTHELIAL CELLS INJURIED BY DL-HOMOCYSTEINE

Objective:To investigate the cross protective effects of folic acid,VB6,VB12 on rat aortic endothelial cells(RAEC) from injury induced by DL-homocysteine(Hcy).Method:RAEC were cultured in vitro,and the cross effects of folic acid,VB6,VB12 were studied by MTT.According to the results of orthogonal design,the activities of SOD,GSH-PX,NOS and the contents of MDA,NO in the media were compared.Results:The results of orthogonal design indicated that the inhibition of Hcy to RAEC could be relieved by folic acid,VB12 excluding VB6.The interaction existed between folic acid and VB12,as well as VB6 and VB12.The inhibition of Hcy to the enzymes(SOD、GSH-PX、NOS) could be relieved by three vitamin projects.Folic acid,VB12,VB6 could inhibit the reduction of NO and decrease the production of MDA.Conclusion:The simultaneous addition of folic acid,VB6 and VB12 may be an excellent strategy for protection of RAEC injury induced by DL-homocysteine.
3.Relationship between expression of RANTES and microvascular density in tissues of primary hepatocarcinoma

Objective To investigate the relationship between the expression of regulated-upon activation normal T expressed and secreted(RANTES)and microvascular density(MVD)in hepatocarcinoma(PHC)tissues,and explore its clinical significance. Methods The samples of PHC tissues and adjacent tissues in 47 patients were collected.The expression of RANTES in tissues was detected by immunohistochemistry,and MVD was calculated.The relationship among the expression of RANTES,MVD and clinicopathological features was analysed. Results The positive expression rate and expression score of RANTES in PHC tissues were significantly higher than those in adjacent tissues(55.32% vs 19.15%,P<0.01;1.89±1.77 vs 0.77±1.29,P<0.01).MVD in PHC tissues was significantly higher than that in adjacent tissues(67.30±13.68 vs 37.20±10.58,P<0.01).MVD of PHC tissues in patients with metastasis was significantly higher than that in patients without metastasis[(73.50±13.77)/HP vs (64.10±12.68)/HP,P<0.05],while there were no correlations among MVD,expression of RANTES and the other clinicopathological features of PHC.MVD was positively correlated with the expression score of RANTES in PHC tissues(r=0.386,P<0.05). Conclusion RANTES might be closely related to the angiogenesis in PHC.
4.Study of the method used to induce the model of type 2 diabetic mellitus rat and the characteristic of the nephropathy

ObjectiveTo discuss the feasibility of the method that can be used to induce the model of type 2 diabetic mellitus rat and explore the characteristic of the nephropathy.The rats were fed with the high sucrose,high fat and high energy feed for a long time and then it was injected with the low dose STZ.Methods30 SD rats were selected and then it was randomly divided them into 2 groups,the control group (10 rats) and the model group (20 rats).The model group was fed with the high calorie feed forl0 weeks to induce insulin resistance and then the rats were induced to type 2 diabetes mellitus by injection of streptozocin (30 mg/kg).The rats of the model group were continually fed with the high calorie feed for 2 months.Before the end of this study,the 24-hours microalbuminuria,serum creatinine and blood urea nitrogen were detected and the periodic acid Schiff staining on the kidney were also measured.ResultsAfter the rats of the model group were established,the levels of the bodyweight,the cholesterol,the triglyceride and the insulin were [ (468.7 ± 8.8 ) g,( 1.92 ± 0.27 ) mmol/L,( 1.32 ± 0.34) mmol/L,(38.81 ± 5.39 ) mU/L ] respectively,all of them were higher than the levels in the normal group,which were [ (436.9 ± 7.4) g,(1.16 ±0.17)mmol/L,(0.8 ±0.18)mmo1/L,(21.43 ±4.19)mU/L],respectively( t =9.755,8.077,4.437,8.902,P < 0.01 ).After injection of STZ for 2 weeks,the levels of the blood glucose,the insulin and the insulin resistance of the diabetes mellitus rats were [ ( 19.31 ± 1.55 ) mmol/L,( 31.31 ± 8.60) mU/L,(26.55 ± 6.33) ] respectively,it was higher than levels inf the normal group,which was[ (5.45 ±0.69) mmol/L,( 19.97 ± 3.26) mU/L,(4.82 ± 0.84) ] ( t =26.383,3.951,10.719,P < 0.01 ).Before the end of this study,the levels of the blood glucose,insulin resistance,24-hours microalbuminuria,serum creatinine,blood urea nitrogen of the diabetes mellitus rats were [ ( 19.27 ± 1.97 ) mmol/L,( 16.70 ±7.51 ),(72.49 ± 8.53 ) mg/24 h,( 74.76 ± 8.38 ) μmol/L,( 19.09 ± 4.21 ) mmol/L],it was higher than the levels in the normal group,which were [ (5.62 ±0.65) mmol/L,(5.45 ± 1、33),( 15.26 ±2.20) mg/24 h,(40.81 ± 1.97) μmol/L,(9.87 ±2.13) mmol/L,t =20.961,4.657,20.623,12.495,6.352,P <0.01 ].And the pathological changes of the diabetes mellitus rats kidney tissues were the most serious through themethod of periodic acid-Schiff's staining (PAS).ConclusionsThe model of type 2 diabetic mellitus rat was constructed through the way of feeding the SD rats with high sucrose,high fat and high energy feed for a long time and low dose STZ.The diabetic mellitus rats had the symptom of drinking more,eating more and diuresis,and the character of this model had high levels of albuminuria,serum creatinine,blood urea nitrogen.The incrassated glomerular mesangium,the crescent-shaped focus and the glomerulosclerosis were also observed through the PAS.
6.MRI findings of neurologic complications in the enterovirus 71-infected hand-foot-mouth disease

Objective To explore the imaging characteristics of neurologic complications associated with the enterovirus 71 ( EV71 ) epidemic by analyzing 25 cases and reviewing the literature. Methods Twenty-five cases of hand-foot-mouth disease with neurologic complications during the recent EV71 outbreaks of Hainan province were studied for the clinical features and imaging findings, and literature were reviewed.Results In 5 cases, acute flaccid paralysis associated with EV71-infected hand-foot-mouth disease was related to the linear high signal in the spinal cord on sagittal images. Two cases showed symmetrical, welldefined hyperintense lesions in the spinal cord on T2WI transverse. Strong enhancement of the ventral horns and root was seen on the contrast-enhanced axial T1WI. In brainstem encephalitis, all lesions presented with significant hyperintensity on T2WI and hypointense on T1WI in the posterior portions of the medulla oblongata, midbrain, and pons. The manifestations of aseptic meningitis (AM) on MRI have no characteristics, but subdural effusion, meningeal enhancement and hydrocephalus can be the indirect signs of AM. Conclusions MRI is an effective method to investigate neurologic complications associated with the EV71 epidemic. Posterior portions of the medulla oblongata and pons, bilateral ventral horns of spinal involvement are characteristic findings of enteroviral encephalomyelitis.

8.Effect of different transfection reagents and injection methods in mice testicular injection on the expression of exogenous gene.

The purpose of this study was to study the effect of three different transfection reagents (Lipofectamine™ LTX & PLUS™, Lipofectamine 2000 and Nano-PAMAM-D) and three different testicular injection methods (rete testicular injection, seminiferous tubules injection and testicular interstitial injection) on the efficiency of production transgenic mice. After the mixtures of plasmid DNA (pEFP-C1) and transfection reagent were injected with different testicular injection methods, the sperm density, vitality, positive sperm rates and PCR positive transgenic mice rate were examined 30 days after injection. The results showed that the damage degree from slight to serious of three transfection reagents was Lipofectamine™ LTX & PLUS™, Lipofectamine 2000, and PAMAM-D. The sperm positive rates with green fluorescence of these three groups were 35.65%±0.69%, 12.86%±0.35% and 10.04%±0.20%, respectively. The PCR positive rates of transgenic newborn mice were 29.17%, 13.70% and 5.88%, respectively. Among the groups of different testicular injection methods, the damage degree from slight to serious was rete testicular injection, seminiferous tubules injection, and testicular interstitial injection, whereas the sperm positive rates with green fluorescence were 35.13%, 15.13%, and 0%, respectively. The PCR positive rates of transgenic newborn mice among different testicular injection groups were 33.3%, 12.5%, and 0.0%. The combination of rete testicular injection and Lipofectamine™ LTX & PLUS™ had the lowest toxicity and highest transgenic efficiency in the production of transgenic mice.

9.Anticancer activity of Luteolin and its synergism effect with BCG on human bladder cancer cell line BIU-87

Objective: To investigate the anticancer activity of Luteolin (Lu) and its synergism effect with bacillus calmette-guerin (BCG) on human bladder cancer cell line BIU-87. Methods: Cultured BIU-87 cells were treated with different concentrations of Lu alone or the combination of Lu with BCG. MTT assay was used to measure the cell proliferation inhibition, and IC50 was calculated. Cell cycle and apoptosis were analyzed by lfow cytometry with propidiumiodide (PI) staining and Annexin-V FITC/PI dual parameter markers to clarify the mechanism of inhibiting cell proliferation and inducing apoptosis. Caspase-3 and phosphorylated c-Jun N-terminal kinases (P-JNK) expression were measured to detect the apoptosis signal pathways of Lu in cancer cells. Results: Both Lu and BCG apparently inhibited the cell proliferation and induced the apoptosis dose-dependently, and microscope observation showed morphological changes in the apoptosis. Flow cytometry indicated that Lu arrested the cell cycle at G2 phase (P<0.05). It sensitized BCG-induced cytotoxicity and cell apoptosis, and upregulated expression of caspase-3 and activation of JNK (P<0.05). Conclusion: As an effective anticancer agent, Lu can sensitize the effect of BCG by inducing the cell cycle arrest and apoptosis. hTis synergism effect is achieved by activation of caspase-3 and JNK. Combination of Lu with BCG may be one of the potential treatment for bladder cancer.
10.Free flap transplantation in the reconstruction of large area defects in head and neck

Objective To review clinical experience of free flap transferring in the reconstruction of head and neck defects. Methods Fifteen free anterolateral thigh flaps, 5 free deep inferior epigastric flaps and 3 free lateral arm flaps were used to reconstruct large area head and neck defects. Results All 23 free flaps were alive with good appearance. 1 case survived by performing embolectomy after suffering arterial crisis and delayed recovery of wound took place in 2 cases. Conclusion Free flap transplantation is an ideal choice for reconstructing large area cranio-facio-cervical defects.
