1.Study on the clinical effect and immunologic function of Moxifloxacin combined with small dose of hormone in the treatment of Ventilator pneumonia in elder

Objective To investigate the clinical effect and immunologic function of Moxifloxacin combined with small dose of hormone in the treatment of Ventilator pneumonia in elder.Methods 64 cases of Ventilator pneumonia in our hospital were collected and randomly divided into experiment group and control group, 32 cases each.Two groups were given conventional treatment, the control group received Methylprednisolone Sodium Succinate 1 mg/kg qd, the experiment group was given Methylprednisolone Sodium Succinate 1 mg/kg qd, and Moxifloxacin Hydrochloride and Sodium Chloride Injection 400 mg qd.Two groups of patients were continuous treated for 10 days.After treatment,T lymphocyte subsets, NK cells, white blood cell count, C reactive protein, clinical symptoms disappeared time, mechanical ventilation time, ICU length of stay and mortality rate were compared. Results After treatment, the total effective rate in the experiment group 75% was higher than the control group 50%( P <0.05 ).The levels CD3 +, CD4 +, CD4 +/CD8 +and NK cell in two groups increased(P<0.05), levels of CD8 +decreased(P<0.05),levels of WBC, CPR and PCT decreased in the two groups(P<0.05), and compared with the control group, the levels CD3 +, CD4 +, CD4 +/CD8 +and NK cell in the experiment group were higher(P<0.05), levels of CD8 + were lower(P<0.05),levels of WBC,CPR and PCT were lower(P <0.05), the rales disappeared time, cough disappeared time, fever disappeared time were significantly shorter than the control group(P <0.05), the duration of mechanical ventilation and the length of hospital stay were significantly shorter(P<0.05).Conclusion Moxifloxacin combined with small dose of hormone in the treatment of Ventilator pneumonia in elder was significantly effective, and it can relieve inflammation, prevention of infection control, enhance immune function.
2.miR-363 downregulates Mcl-1 expression and induces apoptosis in HepG2 cells

AIM:To determine the effect of microRNA-363(miR-363) on HepG2 cells.METHODS:Bioin-formatic analysis was conducted to identify if the Mcl-1 was regulated by miR-363.The expression of Mcl-1 and miR-363 was detected by real-time PCR in normal liver cell line LO2 and hepatocellular carcinoma cell line HepG2, Huh7 and PLC. miR-363 was transfected into the HepG2 cells, and then the level of Mcl-1 was measured.The relative viability was evalua-ted by MTT assay after the HepG2 cells were transfected with miR-363, and the apoptosis was analyzed by flow cytometry with Annexin V/PI staining.RESULTS:Bioinformatic analysis identified that there was a putative target site in the Mcl-1 mRNA for miR-363.Transfection of miR-363 mimics suppressed Mcl-1 expression in the HepG2 cells.Transfection of miR-363 mimics inhibited the cell viability as well as inducing cell apoptosis in HepG2 cells.CONCLUSION: Over-ex-pression of miR-363 significantly inhibits the cell viability and induces apoptosis in HepG2 cells, and the mechanism may be related to the downregulation of Mcl-1 caused by miR-363 transfection.
3.Comparative Proteomics Analysis on Splenic Lymphocytes in Migraine Rat Model with Hyperactivity of Liver-yang

Objective To explore the molecule mechanisms of migraine with hyperactivity of liver-yang related proteins. Methods Animal model of migraine with hyperactivity of liver-yang was established through electrical trigeminal ganglion stimulation,and by administering orally with Fuzi decoction. The total proteins of splenic lymphocytes in the rats were separated by immobilized pH gradient-based two-dimensional gel electrophoresis. With coomassie blue staining,the 2-DE images were analyzed by PDQuest 7.0 software. Matrix-Assisted Laser Desorption/Ionization-Time of Flight Mass Spectrometry (MALDI-TOF-MS) and database were used to identify the differential proteins. Result Ten differential spots protein were identified,and they were chloride intracellular channel 1,thioredoxin peroxidase Ⅰ,thioredoxin peroxidase Ⅱ,sequence 7 from patent WO9709348,Rho GDP dissociation inhibitor (GDI) alpha,actin related protein 2/3 complex neurofilament light polypeptide,proteasome subunit beta type and heat shock protein-27,annexin A1. Conclusion These protein may be involved in the development of migraine with hyperactivity of liver-yang. These data provide useful clues for elucidating the mechanisms of migraine with hyperactivity of liver-yang.
4.Clinical significance of detection of GDF3,PSA combined with MUC1 for diagnosis of breast cancer

Objective To investigate the clinical significance of detection of growth differentiation factor 3(GDF3), prostate specific antigen(PSA) combined with mucin 1(MUC1)for diagnosis of breast cancer.Methods50 cases of breast cancer were selected as the breast cancer group, 50 cases of benign breast disease were selected as the benign breast disease group, and 50 healthy people were selected as the normal group.GDF3, PSA, MUC1 and tumor markets carbohydrate antigen(CA)15-3,CA125,carcinoembryonic antigen(CEA) were detected, and sensitivity, specificity and accuracy were analyzed.ResultsCompared with benign breast disease group and normal group, GDF3, PSA and MUC1 in breast cancer group were higher(P<0.05), CA15-3, CA125 and CEA were higher(P<0.05).The sensitivity, specificity and accuracy of PSA+MUC1 were higher than other group(P<0.05),the sensitivity, specificity and accuracy of combined detection of GDF3, PSA and MUC1 were higher(P<0.05).ConclusionDetection of GDF3 combined with MUC1 and PSA for early diagnosis of breast cancer has more diagnostic significance.
5.Advances in research on relation of ubiquitin-proteasome pathway to malignancy

Ubiquitin-proteasome pathway is an important mechanism regulating many processes of cellular biology,and also a potential target for abnormal regulation associated with malignancy. This pathway may up-regulate or down-regulate the expression of some tumor-inhibitory genes, transcriptional factors and cyclins,and alter the generation of MHC-I-restricting antigen peptides through the activity of specific proteasome, and consequently,participates in the genesis and progression of malignancy.
6.Comparison of immediate PTCA and elective PTCA on infarct related artery after spontaneous recanalization

Objective To compare the clinical outcomes of immediate PTCA and elective PTCA on infarct related artery after spontaneous recanalization. Methods One hundred and eighteen patients with SR between January 1996 and March 2001 were enrolled. According to TIMI flow, the patients were divided into 2 study populations. The two study populations were divided into 2 groups according to immediate PTCA or elective PTCA, respectively. Results Mortality and reinfarction were similar between the 2 groups two weeks after heart attack. However, recurrent ischemic events were more frequent in elective PTCA group than in immediate PTCA group according to TIMI flow grade 2 (1/24 vs 4/12,P=0.034). 7.Construction of recombinant plasmid of Pseudomonas aeruginosa exotoxin A and its secreting expression in E. coli

Objective To clone and to express the full-length Pseudomonas aeruginosa exotoxin A gene and to purify the expressed protein using affinity chromatography. Methods Exotoxin A gene was amplified from the recombinant plasmid pSK/PEA-T vector and subcloned into the pMAL-P2X vector. Then the recombinant plasmid pMAL- PEA was transformed to E.coli BL21. After induction with IPTG, the expressed protein was analyzed by SDS-PAGE and purified with affinity chromatography. Results The recombinants expressed the fusion protein at a level of about 20% of total cell proteins, and 80% of the fusion protein was secreted into the supernatant. Conclusion Successful expression and purification of PEA are of significance for in-depth study of the pathogenic mechanism of Pseudomonas aeruginosa and preparing immunotoxin.
8.Comparison of uterine NK cell phenotypes and T lymphocytes between uterine basal decidua and peripheral blood in human term pregnancy
