1.Clinical comparison of different blood collection methods in routine blood test results

Objective To compare clinical value of peripheral blood collection and venous blood collection methods in blood tests.Methods 467 patients who received blood test were selected.The venous blood and peripher-al blood were collected,the indicators of blood test were detected,and the test results were analyzed.Results The white blood cell count (WBC),red blood cell count (RBC),platelet count (PLT),red blood cell hematocrit (HCT),hemoglobin (HB)of venous blood were (5.7 ±1.2)×109 /L,(3.72 ±0.48)×109 /L,(218.0 ±57.5)× 109 /L,(41.8 ±19.8)g/L,(115.7 ±13.8)g/L respectively,those of peripheral blood were (6.7 ±3.4)×109 /L, (3.62 ±0.72)×109 /L,(204.0 ±87.7)×109 /L,(32.7 ±20.7)g/L,(103.6 ±19.6),the differences were statis-tically significant (t =5.994,2.497,2.885,6.865,10.908,all P <0.05).The mean red cell hemoglobin concentra-tion (MCHC),mean red cell volume (MCV),mean erythrocyte hemoglobin (MCH)and other indexes had no signifi-cant differences(all P >0.05).Conclusion The test results in patients with venous and peripheral blood was quite different,venous blood test results obtained with greater value in clinical use,it is recommended that routine blood test used venous blood.
2.Statistical analysis and learning advice of Nursing Research self-study examination papers during the past five years

Objective To analyze the subject frequency and score proportion of the national unified examination papers of Nursing Research according to the topic content,in order to offer proposals for the reformation and optimization of the examination,and provide self-test-takers with references of improving learning methods and results.Methods The papers of the subject of Nursing Research during October 2008 to January 2013 had been statistically analyzed following the syllabus.Results Comprehensive coverage of knowledge point existed in the papers of the recent five years.The theme of measurement of research tools (5.8%) and basic procedures of nursing research (7.0%) took a smaller proportion compared with nursing research design (16.0%) and nursing papers writing(16.3%).Conclusions The contents of the 10 papers of the past 5 years concentrate high attentions and emphasis on the comprehension and application of the basic theory and methods,which accord with the goals and requirements of this subject in the training of the research ability.
3.Analysis of the status quo of voluntary blood donation and confidential blood donation after voluntary blood donation in Jiaxing

Objective This paper analyzes the situation of confidential blood discarding in voluntary blood donation in Jiaxing in 2011-2016 years,and provides scientific evidence for ensuring the blood safety.Methods During 2011-2016 years,15 voluntary blood donors who required confidential blood donation in the blood bank were investigated for their reasons and characteristics.Results The blood discarding rate was 0.011%.15 cases were abandoned in the confidential blood discarding of voluntary blood donation in Jiaxing,in which 14 blood donors' 4 infectious disease markers were negative,1 blood donor was HIV positive,18-30 years old unmarried males are the main category of secrecy abandoned blood donors,the number of secondary donors who have high-risk behaviors or diseases are relatively small.The causes of confidentiality were mainly high risk behaviors (27%) and feculent sexual history (20%).Conclusion Blood donors' conscience to report the truth is quite important to improve the safety of blood transfusion,it is a must to prepare the training of staffs,to broaden the back way,strengthen the propaganda and education,strengthen the consultation work before the blood donation and provide a private security environment of abandoned blood for blood donors.
4.Experimental study on apoptosis of ovarian cancer cell line SKOV-3 induced by triptolide

Objective To investigate the effect of triptolide (TP) on the apoptosis of ovarian cancer cell line SKOV-3 and its molecular mechanism.Methods Ovarian cancer cell line SKOV-3 was cultured and treated with different doses of TP (1 × 10-6 mg/ml,1 × 10-5 mg/ml,1 × 10-4 mg/ml,1 × 10-3 mg/ml),Iscoves modification of DMEM (IMEM) medium without TP was negative control.At the 12 h,24 h and 48 h after treatment,apoptosis rate was determined by terminal dexynucleotidyl transferase (TdT) mediated dUTP nick end labeling (TUNEL) kit.At the 48 h after treatment,mRNA expression of apoptosis molecules and invasion molecules were determined by fluorescence quantitative polymerase chain reaction (PCR) kit.Results (1) At the 12 h,24 h,and 48 h after treatment with different doses of TP,at the same treatment time,the higher the TP dose was,the higher the apoptosis rate of SKOV-3 cells was;at the same treatment dose,the longer the treatment time was,the higher the apoptosis rate of SKOV-3 cells was;and TP increased the apoptosis rate on dose dependent and time dependent manner;(2) After 48 h treatment with different doses of TP,the higher the TP dose was,the higher the mRNA expression of cytochrome C (CytC),Bax,and Caspase-3 was,the lower the mRNA expression of Bcl-2,matrix metalloproteinases (MMP)2,MMP7,MMP9,N-cadherin,and vimentin was;TP increased mRNA expression of CytC,Bax,and Caspase-3 and decreased mRNA expression of Bcl-2,MMP2,MMP7,MMP9,N-cadherin,and vimentin on dose dependent manner.Conclusions TP can induce the apoptosis of ovarian cancer cells,activation of mitochondrial apoptosis pathway and inhibition of cell invasion are molecular mechanism of TP promoting apoptosis.
5.Effect of Emodin on the Apoptisis and Intracellular Ca~(2+) Concentration of Hepatic Stellate Cell-T_6

Objective To explore the effect of emodin on the apoptisis and intracellular Ca2+ concentration of Hepatic Stellate Cell-T6.Methods The immortalized rat hepatic stellate cell line HSC-T6 was commonly allocated into normal control group and treated groups with different concentrations emodin(the extreme concentration are 10?mol/ml, 20?mol/ml, 40?mol/ml respectively). The cell apoptosis rate were analyzed by flow cytometry, and intracellular Ca2+ concentration of HSC-T6 were detected by Furo-3/Am fluorescence assay and confocal laser microscopy.Results After intervention of different concentration emodin,the apoptosis rate of HSC-T6 is 4.0?0.5%, 4.3?0.6%, 6.9?1.1% respectively,and there are statistical significance compared with the control group(P
6.Management of limb length discrepancy during total hip replacement

Objective To investigate the management of limb length discrepancy during total hip replacement. Methods Forty-four hips of 44 patients undergone unilateral primary total hip replacement between July 1997 and July 2000 were included in this study. There were 30 males and 14 females. The ages of the patients at the surgery ranged from 42 to 80 years(mean, 65.8 years). Evaluation of the limb length discrepancy through clinical measurements and radiographic templates was done preoperatively to anticipate the optimal implantation position of the components and determine the level of expected femoral neck cut to maintain the equality of the limbs. During the surgery, the distance between two reference points made at pelvis and femur were measured before femoral neck osteotomy was performed based on preoperatively estimated parameters. The distance between the two points was measured again after insertion of the trail components in order to adjust the offsets of the femoral neck and head. Results Of the 30 patients with shortened limb ranging 1.5 to 5 cm preoperatively, only 4 had limb length discrepancy between 1.5 and 2 cm after surgery. Of the 14 patients with equal limb length preoperatively, only one limb had been lengthened more than 1.0 cm postoperatively compared with its contralateral limb. Conclusion The preoperative measurements, templating and intraoperative corrections are helpful in minimizing limb length discrepancy during total hip replacements and correcting preoperative limb length discrepancy as well.
7.Ipsilateral femoral neck and shaft f ractures

Objective To elucidate the clinical characteristics and diagnostic outline of ipsilateral femoral neck and shaft fractures.Methods A retrospective study was carried ou t on 7patients with ipsi-lateral femoral neck and shaft fractures associated injuries of the knee;the duration of delay in diagnosis o f the femoral neck fractures,the managem ent of treatment and final clinical o utcomes were reviewed.Results All of the femoral shaft fractures occur red in the middle third and the femora l neck fractures in the base of the neck.Two patients were associated w ith knee injuries.The duration of delay in diagnosis of the femoral neck fractures ranged from 1to 75days.Si x femoral shaft fractures and 5neck f ractures were treated operatively.The mean follow-up period was 15.6mo nths,and all of the shaft and neck fra ctures were eventually healed without the evidence of avascular necrosis of the femoral head in spite of2mal-unions of the femoral neck.Conclusion In case of femoral shaft fracture resulting from high-energy trauma,an A-P pelvic film should be routinely taken.The patients with negative findings on the hip radiographs continue to complain of hip pain following open reduction and intern al fixation of the femoral shaft fractures should undergo CT or MR imaging examination.Once the femoral neck f racture is recognized,operative re duction and fixation should be em-ployed immediately.The outcome of o perative treatment is much superior to that of non-operative.
8.Quantitative estimation of regional brain gray nuclei iron content in idiopathic restless legs syndrome patients by phase values

Objective To investigate the applicative value of enhanced 3D multi-echo GRE T2*-weighted angiography(ESWAN) sequence phase values in evaluating brain gray nuclei iron content in idiopathic restless legs syndrome(RLS) patients, providing imaging basis in diagnosis and treatment of idiopathic RLS. Methods In our institute from June 2012 to September 2016,forty-five RLS patients were selected as the RLS group, and 45 healthy volunteers as the control group. ESWAN sequence was performed and serum ferritin values were obtained in all patients and volunteers. The raw data of ESWAN was postprocessed , where the phase maps were obtained. Phase analysis was performed on localized brain gray nuclei regions of interest (substantia nigra, red nucleus, dentate nucleus, thalamus, pallidum, putamen and caudate nucleus ) selected on phase maps. Differences between the 2 subject groups were evaluated using ANCOVA including age as a covariate. Results The phase values of the substantia nigra, thalamus, pallidum and putamen in the RLS group were (-0.087 ± 0.021), (-0.053 ± 0.012), (-0.161 ± 0.008), (-0.125 ± 0.019) radians , respectively. The phase values of the substantia nigra, thalamus, pallidum and putamen in the control group were (-0.127 ± 0.007), (-0.066 ± 0.007), (-0.166 ± 0.007), (-0.150 ± 0.010) radians, respectively. There were significant differences between the two groups (F=142.492, 37.988, 10.558, 60.725;P<0.05). Indicating reduced iron content in several regions of brain gray nuclei of the patients with RLS. Serum ferritin concentration between the RLS patients (157.02±95.78)μg/L and healthy controls (175.49 ± 38.65)μg/L was not significant (F=1.353,P>0.05). Conclusions Phase values can make a quantitative assessment of brain gray nuclei iron content in RLS patients, our results supported the hypothesis of reduced brain iron content in RLS patents , which may have an important role in the pathogenesis of the disorder. However, iron content change in some brain regions was not correlated with serum ferritin concentration changes.
9.Biotransformation of dihydroartemisinic acid by suspension culture cell of Artemisia annua

Objective To investigate the biotransformation of dihydroartemisinc acid(Ⅰ) by cultured cells of Artemisia annua.Methods Dihydroartemisinic acid was added to the suspension cells of A.annua which had been pre-cultured for 14 d and co-cultured for another 2 d.The biotransfromed products were detected with TLC and HPLC,and isolated by various chromatographic methods.Results The chemical structures of biotransformed products were elucidated on the basis of spectroscopic data.3-?-Hydroxy-dihydroartemisinic acid(Ⅱa) and 3-?-hydroxy-dihydroartemisinic acid(Ⅱb) were obtained after 2 d administration of dihydroartemisinic acid to the suspension cells of A.annua.Ⅱa and Ⅱb yields could reach a maxium mole ratio of 2.6% and 15.7% after 1 d incubation,respectivly.Conclusion It is the first time for the biotransformation of dihydroartemisinic acid to epimeric 3-hydroxyartemisinic acid by using suspension culture cell of A.annua.The results indicate that cells of A.annua have the ability to hydroxylate dihydroartemisinic acid region selectively.
10.Comparative study on effection of risperidone to the metabolism in schizophrenia patients

Objective To explore the affection and related factors of risperidone on the metabolic indexes in schizophrenia patients.Methods 56 cases hospitalized schizophrenia patients were treated with risperidone for 16 weeks.Waistline,blood glucose and blood lipid,blood pressure and other indicators were observed before and after treatment,risk factors that caused metabolic syndrome (MS) and abnormal indexes were analyzed,and 36 healthy persons as control group.Results The rate of MS in patient group was obviously higher than that in control group ( 12.50% vs 0%,P =0.026).Waistline,triglyceride ( TG),high density lipoprotein ( HDL-C ),and 2 hours postprandial blood glucose (2hPG) in patient group ( (4.15 ± 3.14)cm in man and (4.41 ± 2.86)cm in woman,(0.26 ±0.51 ) mmol/L,(-0.09 ±0.19) mmol/L,(0.49 ±0.84) mmol/L) increased compared with that in control group ( ( 0.76 ± 1.09 ) cm in man and ( 0.64 ± 0.81 ) cm in woman,( 0.04 ± 0.15 ) mmol/L,(-0.01 ± 0.12)mmol/L,(0.04 ± 0.35 ) mmol/L) at the 16th weekend,and difference was significantly (P < 0.05 ).Logistic regression analysis showed that the risk factors were family history of diabetes Odds Ratio (OR) 20.34 (95% CI 1.97 ~210) and age OR 1.19(95%CI 1.03 ~ 1.38),the basis waistline OR 1.26 (95% CI 1.04 ~ 1.52) and family history of obesity OR 4.27(95% CI 1.21 ~ 15.1 ),the basis TG OR 73.88 (95% CI 3.41 ~ 16.01 ) and the basis systolic pressure OR 1.16 (95% CI 1.00 ~ 1.33 ),the basis waistline OR 1.41 (95 % CI 1.08 ~ 1.84 ) and age OR 1.53 (95% CI 1.02 ~ 1.31 ) in developed MS,abnormal rate of waistline,TG,and 2hPG.Conclusion Risperidone can cause abdominal obesity,metabolic disorders of glucose and lipid even MS.So it shoud be used carefully for the patient who is old and has family history of diabetes and obesity posture.
